Synthesis, characterization, antimicrobial, cytotoxic and carbonic anhydrase inhibition activities of multifunctional pyrazolo-1,2-benzothiazine acetamides.

The advent of antibiotic resistance in microorganisms requires the discovery and synthesis of novel antibiotics. At the same time, human pathogens are contributing to chronic and persistent inflammation. Motivated by these two concerning issues, new antibiotic drug candidates are synthesized by incorporation of benzothiazine, pyrazole, and amide moieties in a new scaffold to create multifunctional derivatives of pyrazolo-1,2-benzothiazine. The presented compounds have been synthesized and analyzed using spectroscopic and spectrometric techniques including FTIR, HRMS, 1H and 13C NMR spectroscop

Aryl amino acetamides prevent Plasmodium falciparum ring development via targeting the lipid-transfer protein PfSTART1.

With resistance to most antimalarials increasing, it is imperative that new drugs are developed. We previously identified an aryl acetamide compound, MMV006833 (M-833), that inhibited the ring-stage development of newly invaded merozoites. Here, we select parasites resistant to M-833 and identify mutations in the START lipid transfer protein (PF3D7_0104200, PfSTART1). Introducing PfSTART1 mutations into wildtype parasites reproduces resistance to M-833 as well as to more potent analogues. PfSTART1 binding to the analogues is validated using organic solvent-based Proteome Integral Solubility Al

Pyrimidine-based sulfonamides and acetamides as potent antimicrobial Agents: Synthesis, Computational Studies, and biological assessment.

A series of novel sulfonamide and acetamide derivatives of pyrimidine were synthesized and their antimicrobial activities were assessed. Based on the Microbroth dilution method, the minimum inhibitory concentration (MIC) of the synthesized compounds demonstrated moderate to good levels of antifungal and antibacterial activity. Structure-activity relationship analysis suggested that the presence of electron-withdrawing groups, such as halogens, nitrile, and nitro groups, on the pyrimidine ring contributed to the enhanced antimicrobial potency, while electron-donating substituents led to a decre

Design and synthesis of α-phenoxy-N-sulfonylphenyl acetamides as Trypanosoma brucei Leucyl-tRNA synthetase inhibitors.

Human African trypanosomiasis (HAT), caused by the parasitic protozoa Trypanosoma brucei, is one of the fatal diseases in tropical areas and current medicines are insufficient. Thus, development of new drugs for HAT is urgently needed. Leucyl-tRNA synthetase (LeuRS), a recently clinically validated antimicrobial target, is an attractive target for development of antitrypanosomal drugs. In this work, we report a series of α-phenoxy-N-sulfonylphenyl acetamides as T. brucei LeuRS inhibitors. The most potent compound 28g showed an IC50 of 0.70 μM which was 250-fold more potent than the starting hi

2-Chloro-N-(4-hy-droxy-phen-yl)acetamide.

The title compound, C8H8ClNO2, is significantly distorted from planarity, with a twist angle between the planes through the hy-droxy-benzene and acetamide groups being 23.5 (2)°. This conformation is supported by intra-molecular C-H⋯O and N-H⋯Cl contacts. In the crystal, N-H⋯O hydrogen-bonding contacts between acetamide groups and O-H⋯O contacts between hydroxyl groups form tapes propagating parallel to [103].

Identification of the Acetamide-Chalcone Derivative That Selectively Inhibits ABCG2 Transport Activity.

Multidrug resistance (MDR) remains a major obstacle to effective cancer chemotherapy, often driven by the overexpression of ABC transporters such as ABCG2. In this study, we identified the synthetic chalcone derivative, B5, as a selective ABCG2 inhibitor. Among the 20 chalcone analogs evaluated, B5, bearing an acetamide group at R1 and an ethoxy substituent at R2, was the only compound that produced more than 50% inhibition of ABCG2-mediated Hoechst 33342 transport at 10 μM, while showing no activity against ABCB1. B5 exhibited an IC50 of 2.7 μM for ABCG2 inhibition, consistent with the low-mi

Coupling nitrate electrochemical reduction and nitrite oxidation of ethanol for acetamide synthesis.

Electrochemical acetamide synthesis under ambient conditions offers a sustainable route for converting waste nitrate into valuable chemicals. Conventional methods, limited to standalone reduction or oxidation processes, typically achieve low Faradaic efficiencies (<40%) and yields (<0.2 mmol h⁻¹ cm⁻²). Here, we present a tandem reaction system coupling cathodic reduction and anodic oxidation in a full-cell electrolyzer to enhance acetamide production. At the cathode, nitrate is first reduced to nitrite, which subsequently oxidizes ethanol to acetaldehyde. This acetaldehyde reacts in situ with

Metal-Free Aminocarbonylation of p-Quinone Methides with Isocyanides: Synthesis of Sterically Hindered α-Arylated Acetamides.

The synthesis of sterically hindered α-arylated acetamides generally requires a multistep reaction sequence and is also difficult to access due to steric constraints. This protocol allows the synthesis of sterically hindered α-arylated acetamides in moderate to high yields via 1,6-addition of isocyanides to p-quinone methides in the presence of BF3 .OEt2 . The present transformation features transition metal-free conditions, avoiding the use of toxic carbon monoxide, broad substrate scope, mild reaction conditions, and operational simplicity.

Activity refinement of aryl amino acetamides that target the P. falciparum STAR-related lipid transfer 1 protein.

Malaria is a devastating disease that causes significant morbidity worldwide. The development of new antimalarial chemotypes is urgently needed because of the emergence of resistance to frontline therapies. Independent phenotypic screening campaigns against the Plasmodium asexual parasite, including our own, identified the aryl amino acetamide hit scaffold. In a prior study, we identified the STAR-related lipid transfer protein (PfSTART1) as the molecular target of this antimalarial chemotype. In this study, we combined structural elements from the different aryl acetamide hit subtypes and exp

2-((1H-indol-3-yl)thio)-N-phenyl-acetamides: SARS-CoV-2 RNA-dependent RNA polymerase inhibitors.

Severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) is the causative agent of Coronavirus Disease 2019 (COVID-19) pandemic. Despite intensive and global efforts to discover and develop novel antiviral therapies, only Remdesivir has been approved as a treatment for COVID-19. Therefore, effective antiviral therapeutics are still urgently needed to combat and halt the pandemic. Viral RNA-dependent RNA polymerase (RdRp) of SARS-CoV-2 demonstrates high potential as a reliable target for the development of antivirals. We previously developed a cell-based assay to assess the efficiency of co

Complementary hydrogen bonding of a carboxylato-barbiturate with urea and acetamide: experimental and theoretical approach.

A barbiturate derivative, 4-(2,4,6-trioxo-tetrahydro-pyrimidine-5-ylidenemethyl)-benzoic acid (L1) possessing functional complementarity to amides has been synthesized and characterized. Its binding separately with urea and acetamide was monitored using UV-vis, fluorescence and (1)H-NMR spectroscopic titrations. Experiments suggested stronger binding of L1 with urea as compared to acetamide. The solid adducts of L1 prepared separately with urea and acetamide were also characterized using IR, (1)H-NMR spectral and PXRD techniques. Theoretical studies on hydrogen bonded complexes of L1-urea and

N-(3,5-Di-chloro-4-meth-oxy-phen-yl)acetamide.

The title compound, C9H9Cl2NO2, crystallizes in the triclinic space group P1 with six mol-ecules in the asymmetric unit. Structure determination at 100 K from a two-component twin crystal shows that the six crystallographically independent mol-ecules adopt closely similar conformations. The di-chloro-phenyl and acetamide planes form a dihedral angle of ca. 10.0° (mean of six mol-ecules), while the meth-oxy substituent is oriented nearly orthogonal to the aromatic ring (mean dihedral angle = 88.5°). In the extended structure, the mol-ecules are linked by N-H⋯O hydrogen bonds into infinite chain

Acetamide: a low-cost alternative to alkyl imidazolium chlorides for aluminium-ion batteries.

We propose a new electrolyte for rechargeable aluminium-ion batteries based on a room-temperature eutectic mixture of acetamide and aluminium chloride. When diluted with dichloromethane, the electrolyte shows similar cycling performance to the more traditional 1-ethyl-3-methylimidazolium chloride-based electrolytes for a fraction of the cost.

Monitoring 2-phenylethanamine and 2-(3-hydroxyphenyl)acetamide sulfate in doping controls.

2-Phenylethanamine (phenethylamine, PEA) represents the core structure of numerous drugs with stimulant-like properties and is explicitly featured as so-called specified substance on the World Anti-Doping Agency (WADA) Prohibited List. Due to its natural occurrence in humans as well as its presence in dietary products, studies concerning the ability of test methods to differentiate between an illicit intake and the renal elimination of endogenously produced PEA were indicated. Following the addition of PEA to the Prohibited List in January 2015, retrospective evaluation of routine doping contr

Copper(I)-catalyzed asymmetric allylic alkylation of α-sulfanyl acetamides and esters with Morita-Baylis-Hillman carbonates.

Chiral α-sulfanyl carbonyl compounds are attractive synthetic targets owing to their potential bioactivities. Herein, we report a copper(I)-catalyzed enantioselective allylic alkylation of α-sulfanyl acetamides and esters with Morita-Baylis-Hillman (MBH) carbonates, giving a diverse array of chiral α,α-disubstituted amides and esters in high yields, enantioselectivities and E/Z ratios. This reaction shows broad substrate scope, excellent scalability, and versatile downstream derivatization of the products.

Assessing p-tolyloxy-1,3,4-oxadiazole acetamides as lipoxygenase inhibitors assisted by in vitro and in silico studies.

The underlying correlation between the inflammation, innate immunity and cancer is extensively familiar and linked through a process mediated by three enzymes; cyclooxygenase (COX), lipoxygenase (LOX) and cytochrome P450 (CYP450). The ever increase in the reported side effects of the antiinflammatory drugs against the targeted enzymes and the resistance developed afterwards compels the researchers to synthesize new effective molecules with safer profile. On the basis of these facts, our ongoing research on 1,3,4-oxadiazole derivatives deals with the synthesis of a new series of N-alkyl/aralky/
