Rough vaccines in animal brucellosis: structural and genetic basis and present status.

Brucellosis control and eradication requires serological tests and vaccines. Effective classical vaccines (S19 in cattle and Rev 1 in small ruminants), however, induce antibodies to the O-polysaccharide of the lipopolysaccharide which may be difficult to distinguish from those resulting from infection and may thus complicate diagnosis. Rough attenuated mutants lack the O-polysaccharide and would solve this problem if eliciting protective immunity; the empirically obtained rough mutants 45/20 and RB51 have been used as vaccines. Strain 45/20 is reportedly unstable and it is not presently used.

Retinal structure and function in an animal model that replicates the biochemical hallmarks of desmosterolosis.

Desmosterolosis is a rare, autosomal recessive, human disease characterized by multiple congenital anomalies in conjunction with grossly elevated levels of desmosterol and markedly reduced levels of cholesterol in all bodily tissues. Herein, we evaluated retinal sterol composition, histology, and electrophysiological function in an animal model that exhibited the biochemical features of desmosterolosis, produced by treating pregnant rats and their progeny with U18666A, an inhibitor of desmosterol reductase. Treated rats had cataracts, were substantially smaller, and had markedly high levels of

Identifying behavioral structure from deep variational embeddings of animal motion.

Quantification and detection of the hierarchical organization of behavior is a major challenge in neuroscience. Recent advances in markerless pose estimation enable the visualization of high-dimensional spatiotemporal behavioral dynamics of animal motion. However, robust and reliable technical approaches are needed to uncover underlying structure in these data and to segment behavior into discrete hierarchically organized motifs. Here, we present an unsupervised probabilistic deep learning framework that identifies behavioral structure from deep variational embeddings of animal motion (VAME).

Structural basis for the modulation of voltage-gated sodium channels by animal toxins.

Animal toxins that modulate the activity of voltage-gated sodium (Nav) channels are broadly divided into two categories-pore blockers and gating modifiers. The pore blockers tetrodotoxin (TTX) and saxitoxin (STX) are responsible for puffer fish and shellfish poisoning in humans, respectively. Here, we present structures of the insect Nav channel NavPaS bound to a gating modifier toxin Dc1a at 2.8 angstrom-resolution and in the presence of TTX or STX at 2.6-Å and 3.2-Å resolution, respectively. Dc1a inserts into the cleft between VSDII and the pore of NavPaS, making key contacts with both domai

Recent advances in the structure and function of the parathyroid gland in animals and the effects of xenobiotics.

Parathyroid glands in animals consist of a single basic type of secretory cell concerned with the elaboration of one hormone. Parathyroid hormone is the principal hormone involved in the minute-to-minute fine regulation of blood calcium in mammals. A larger biosynthetic precursor of parathyroid hormone is first synthesized on ribosomes of the endoplasmic reticulum in chief cells. Pre-proparathyroid hormone is rapidly converted to proparathyroid hormone in the Golgi apparatus. Active parathyroid hormone is packaged into membrane-limited secretory granules that are stored in the cytoplasm until

Panoramic view of retinal and choroidal circulation and structures in vivo based on ultra-widefield OCT/OCTA: from animal to human.

This study aimed to compare retinal and choroidal circulation and structural parameters between humans and five commonly used experimental animals (rats, mice, guinea pigs, rabbits, and monkeys) using ultra-widefield swept-source optical coherence tomography (SS-OCT) and SS-OCT angiography (SS-OCTA). The study found that the six species exhibited unique patterns in retinal and choroidal vessel flow distribution. Rats and mice demonstrated significantly higher retinal and choroidal vessel flow density (VFD) compared to humans, while retinal vessels in rabbits are confined to a broad horizontal

Selective GPR17 antagonism enhances structural and functional recovery in animal models of demyelination.

Myelination, driven by differentiation of oligodendrocyte precursor cells, is critical for metabolic and structural support and efficient axonal signal transmission in neurons. Loss of myelin is a hallmark of multiple sclerosis and other devastating demyelinating disorders. As demyelination persists, neurons become increasingly vulnerable, leading to neurodegeneration and chronic disability. Restoring myelin through endogenous repair mechanisms offers a promising therapeutic approach to mitigate progressive neuronal loss. One key regulator of myelination is the G protein-coupled receptor 17, G

Research Note: The structure and diversity of antibiotic resistance genes in animal house environment.

In recent years, a series of public health issues caused by the spread of antibiotic resistance have been widely concerned. The indoor air of livestock and poultry houses is considered to be one of the main sources of environmental contamination of ARGs. This study characterized the micro-organisms and ARGs in the air particulate matter of chicken houses using metagenomics. The study successfully detected 761 different subtypes of resistance genes including aminoglycosides, tetracyclines, MLSB etc., 4 types of mobile genetic elements, and various pathogenic microorganisms from the aerosols in

Transient Receptor Potential (TRP) and Thermoregulation in Animals: Structural Biology and Neurophysiological Aspects.

This review presents and analyzes recent scientific findings on the structure, physiology, and neurotransmission mechanisms of transient receptor potential (TRP) and their function in the thermoregulation of mammals. The aim is to better understand the functionality of these receptors and their role in maintaining the temperature of animals, or those susceptible to thermal stress. The majority of peripheral receptors are TRP cation channels formed from transmembrane proteins that function as transductors through changes in the membrane potential. TRP are classified into seven families and two

MORPHOMETRIC ASSESSMENT OF STRUCTURAL CHANGES IN THE VASCULAR BED OF DUODENUM IN ANIMALS WITH OBSTRUCTIVE CHOLESTASIS.

Aim of the research - comprehensive morphometric assessment of the structural and spatial organization of the circulatory system of the duodenum in obstructive cholestasis in intact animals. Three days after the simulated obstructive cholestasis in 3 experimental animals, on examination of radiographs there is a tendency to reorient the arterial and venous bed of the duodenum. Analysis of morphometric parameters also showed a tendency to constriction of arteries and varicose veins. Some narrowing of the arteries was evidenced by a decrease in the diameters of the main trunks of the vascular or

The population structure of Acinetobacter baumannii isolated from animals: an emerging zoonotic threat.

To investigate the population structure of Acinetobacter baumannii in animals and the transmission pattern between humans and animals, we conducted a molecular epidemiological study by supplementing the data from poultry combined with data from 509 animal isolates and 2,218 human isolates in the NCBI GenBank database. Antibiotic susceptibility testing and genomic analysis showed that A. baumannii isolated from the brains of dead poultry embryos was universally sensitive to 19 antibiotics and genetically diverse. The pathogen has been isolated from 33 animal species from 17 countries worldwide.

Abnormal vestibular brainstem structure and function in an animal model of autism spectrum disorder.

Autism spectrum disorder (ASD) is a neurodevelopmental disorder that includes several key neuropathological changes and behavioral impairments. In utero exposure to the anti-epileptic valproic acid (VPA) increases risk of an ASD diagnosis in human subjects and timed in utero exposure to VPA is a clinically relevant animal model of ASD. Many human subjects with ASD have cerebellar hypoplasia, fewer Purkinje cells, difficulties with balance, ophthalmic dysfunction and abnormal responses to vestibular stimulation and such vestibular difficulties are likely under reported in ASD. We have recently

Relationship between scapular position and structural strength of rib cage in quadruped animals.

Determining scapular position is a major issue in reconstructing the skeletal systems of extinct quadruped archosaurs and mammals, because the proximal portion of the scapulae has no direct skeletal joint with the vertebrae or ribs. When quadrupeds stand or walk, their trunk is suspended between the forelimbs by the serratus muscles, which arises from the lateral sides of the "thoracic" ribs and inserts into the proximal portion of the costal surface of the scapula. Therefore, the "thoracic" ribs are subjected to a static or dynamic vertical compression between the lifting force from the muscl

Ferritin mRNA translation, structure, and gene transcription during development of animals and plants.

Changes in transcription/accumulation of ferritin mRNA accompany cell differentiation and development as cells establish the pattern of iron storage and metabolism that matches their specialized features. Signals which induce changes in ferritin gene expression include hormones (thyrotropin in thyroid cells), monokines (tumor necrosis factor alpha in myoblasts, and preadipocytes), phagocytosis and/or inducing agents (erythrophagocytosis in macrophages, phorbol esters in promyelocytes, and dimethyslulfoxide in proerythroblasts), heat shock (in red cells) and light (in leaf mesophyll). Tertiary structure of animal tRNATrp in solution and interaction of tRNATrp with tryptophanyl-tRNA synthetase.

Alkylation in beef tRNATrp of phosphodiester bonds by ethylnitrosourea and of N-7 in guanosines and N-3 in cytidines by dimethyl sulfate and carbethoxylation of N-7 in adenosines by diethyl pyrocarbonate were investigated under various conditions. This enabled us to probe the accessibility of tRNA functional groups and to investigate the structure of tRNATrp in solution as well as its interactions with tryptophanyl-tRNA synthetase. The phosphate reactivity towards ethylnitrosourea of unfolded tRNA was compared to that of native tRNA. The pattern of phosphate alkylation of tRNATrp is very simil

Discovery of Diphenyl Ether Derivatives as Novel BKCa Channel Activators: Structure-Activity Relationship, Cryo-EM Complex Structures, and In Vivo Animal Studies.

The BKCa channel, a large-conductance calcium-activated potassium channel, plays a crucial role in maintaining the homeostasis of the micturition cycle and airway-related functions. In this study, we optimized a novel BKCa channel activator, 4d, with a diphenyl ether structure identified from library screening. This led to the discovery of potent activators, 10b (EC50 = 0.12 μM, cell-based assay) and 51b, an orally bioavailable derivative. Compound 10b demonstrated potent in vivo efficacy in a spontaneous hypertensive rat (SHR) of urinary incontinence model, while compound 51b showed dose-depe
