Plumbagin protects against cataract formation in STZ-induced diabetic rats: Establishing insights into the molecular mechanism.

BACKGROUND: Our previous study demonstrated that plumbagin has the potential to reduce the lens aldose reductase and mitigate the cataract formation in the glucose-induced ex vivo model. Therefore, we explored the molecular insights into the anticataract action of plumbagin using the STZ-induced in vivo diabetic rat model. METHODS: Albino rats (Wistar rats, 150-180 g body weight, 15-18 weeks old, of either sex) were used in the study for assessment of the anticataract action of plumbagin against the STZ-induced diabetic cataract rat model. STZ was intraperitoneally injected at a dose of 60 mg

Unveiling the anti-glycation properties of acetylsalicylic acid: targeted inhibition of albumin glycation.

Glycation due to increased blood sugar levels aggravates diabetic complications. Glycation hampers the structural and functional integrity of human serum albumin (HSA). Acetylsalicylic acid (ASA)/aspirin exhibits anti-glycation properties. Although the precise molecular mechanism of ASA in glycation has not yet been conclusively demonstrated, acetylation has been considered the central mechanism underlying its biological action. The present study aims to unveil the specific mechanism of action of ASA on glycated HSA through meticulously designed approaches. Fluorescence and UV-visible spectros

Pleiotropic Properties of Valsartan: Do They Result from the Antiglycooxidant Activity? Literature Review and In Vitro Study.

Valsartan belongs to angiotensin II type 1 (AT1) receptor blockers (ARB) used in cardiovascular diseases like heart failure and hypertension. Except for its AT1-antagonism, another mechanism of drug action has been suggested in recent research. One of the supposed actions refers to the positive impact on redox balance and reducing protein glycation. Our study is aimed at assessing the antiglycooxidant properties of valsartan in an in vitro model of oxidized bovine serum albumin (BSA). Glucose, fructose, ribose, glyoxal (GO), methylglyoxal (MGO), and chloramine T were used as glycation or oxida

Design and evaluation of multi-target AGEs/ALEs inhibitors for potential application in skin anti-aging.

The main causes of skin aging include both intrinsic and extrinsic factors such as oxidative stress triggered by reactive oxygen species (ROS), modification of collagen and elastin by reactive carbonyl species (RCS), UV exposure, lifestyle habits and environmental pollutants. Skin aging is the most visible manifestation of senescence and is unavoidable. In particular, delaying age-related changes in facial appearance is a key objective for older adults. To address this issue, naturally occurring compounds including vitamin C, vitamin E, coenzyme Q10, resveratrol, niacinamide, carnosine, retino

Investigation of in vitro antibacterial, antiglycation, and antioxidant activities of selamectin/chlorogenic acid system and their interaction with immunoglobulin G.

The interaction mechanism between chlorogenic acid (CGA) and/or selamectin (SEI) and immunoglobulin G (IgG) was studied by multispectral techniques and molecular docking, and the effect of the complex formation on the protein's functional properties was then evaluated. The results showed that CGA or SEI binds to the different site on IgG chain through different binding forces, which induces different influence on the protein's secondary structure. The interaction of IgG with CGA is higher than that of IgG with SEI due to the multiple hydroxyl groups, the flexible and compact structure of CGA.

A multimodal caffeic acid-derived alkyl-amide antidiabetic agent: targeting α-glucosidase, α-amylase, oxidative stress, and protein glycation.

Historically, natural products have been a primary source of new drugs and lead compounds. However, their direct application as therapeutic agents remains limited. Given the multifactorial nature of diabetes, therapeutic agents with multi-targeting properties offer a promising solution for effective management. Herein, we developed a library of seven lipophilic derivatives of caffeic acid (CA) and evaluated their multimodal antidiabetic activities. Among these, the most potent compound, CA14, showed an IC50 of 1.94 μM, representing a 113-fold improvement over acarbose (IC50 219.70 μM). Further

2-Deoxy-D-ribose induces cellular damage by increasing oxidative stress and protein glycation in a pancreatic beta-cell line.

2-Deoxy-D-ribose (dRib) is a sugar with a high reducing capacity. We previously reported that dRib induced damage in pancreatic beta-cells. The aim of this study was to investigate the mechanism of dRib-induced beta-cell damage. 2-Deoxy-D-ribose provoked cytotoxicity and apoptosis within 24 hours in HIT-T15 cells. Three antiglycating agents-diethylenetriaminepentaacetic acid, aminoguanidine, and pyridoxamine-dose dependently inhibited dRib-triggered cytotoxicity and significantly suppressed apoptosis induced by dRib. 2-Deoxy-d-ribose increased intracellular reactive oxygen species and protein

Water-Soluble Products of Photooxidative Destruction of the Bisretinoid A2E Cause Proteins Modification in the Dark.

Aging of the retina is accompanied by a sharp increase in the content of lipofuscin granules and bisretinoid A2E in the cells of the retinal pigment epithelium (RPE) of the human eye. It is known that A2E can have a toxic effect on RPE cells. However, the specific mechanisms of the toxic effect of A2E are poorly understood. We investigated the effect of the products of photooxidative destruction of A2E on the modification of bovine serum albumin (BSA) and hemoglobin from bovine erythrocytes. A2E was irradiated with a blue light-emitting diode (LED) source (450 nm) or full visible light (400-70

The Glycine-Proline-Hydroxyproline Tripeptide Reduces Cell Death Caused by Glycative Stress.

INTRODUCTION: Long-term intake of Collagen Tripeptide (CTP), which is rich in Glycine- X-Y sequences, improves blood vessel elasticity and reduces the serum levels of cytotoxic glyceraldehyde (GA)-derived Advanced Glycation End-Products (AGEs), also known as Toxic AGEs (TAGE). Therefore, the anti-glycation effect of CTP may be one of the mechanisms contributing to its ability to improve vascular elasticity. This study aims to investigate the antiglycation effects of the tripeptide, glycine-proline-hydroxyproline (Gly-Pro-Hyp; GPHyp), the main component of CTP, using in vitro GA glycation model

Delayed Intervention With Pyridoxamine Improves Metabolic Function and Prevents Adipose Tissue Inflammation and Insulin Resistance in High-Fat Diet-Induced Obese Mice.

Obesity is associated with an increased risk for the development of type 2 diabetes and vascular complications. Advanced glycation end products are increased in adipose tissue and have been associated with insulin resistance, vascular dysfunction, and inflammation of adipose tissue. Here, we report that delayed intervention with pyridoxamine (PM), a vitamin B6 analog that has been identified as an antiglycating agent, protected against high-fat diet (HFD)-induced body weight gain, hyperglycemia, and hypercholesterolemia, compared with mice that were not treated. In both HFD-induced and db/db o

Superior antiglycation efficacy of silymarin from Silybum marianum in human αA-crystallin: Implications for diabetic cataract prevention.

Hyperglycemia activates the polyol pathway, producing fructose, which promotes glycation and denatures α-crystallin, ultimately leading to diabetic cataracts. This study compared the antiglycation effects of SMR and SBN, using a fructose-induced human αA-crystallin glycation model. Through fluorescence analysis, SDS-PAGE, and Western blotting methods, we found that glycation caused αA-crystallin to form fluorescent advanced glycation end products (AGEs), cross-linking AGEs, and Nε-carboxymethyllysine (CML). Results show that SMR (> 20 μg/mL) and SBN (> 100 μg/mL) effectively inhibited cross-li

Recent advances in the potential of Phyllanthus emblica L. and its related foods for combating metabolic diseases through methylglyoxal trapping.

Methylglyoxal (MG) serves as the primary precursor for the nonenzymatic glycation of proteins and DNA, leading to advanced glycation end products (AGEs). Regular intake of dietary MG is strongly correlated with low-grade inflammation, potentially accelerating the pathogenesis of metabolic diseases, including obesity, diabetes, cancers, liver diseases, Alzheimer's disease, cardiovascular diseases, aging, and bone loss. Although pharmaceutical agents (pimagedine and candesartan) have been developed to inhibit MG formation, they often come with serious side effects (nausea, diarrhea, headache, ga

Urolithins Modulate the Viability, Autophagy, Apoptosis, and Nephrin Turnover in Podocytes Exposed to High Glucose.

Urolithins are bioactive compounds generated in human and animal intestines because of the bacterial metabolism of dietary ellagitannins (and their constituent, ellagic acid). Due to their multidirectional effects, including anti-inflammatory, antioxidant, anti-cancer, neuroprotective, and antiglycative properties, urolithins are potential novel therapeutic agents. In this study, while considering the future possibility of using urolithins to improve podocyte function in diabetes, we assessed the results of exposing mouse podocytes cultured in normal (NG, 5.5 mM) and high (HG, 25 mM) glucose c

Multi-mode ultrasonic-driven self-assembly of banana-derived resistant starch nanoparticles for epigallocatechin gallate encapsulation and in vitro antiglycation evaluation.

Starch nanoparticles, as a type of nanomaterial, have been increasingly applied in food and biomedical fields due to their excellent biocompatibility, safety, non-toxicity, and resource sustainability. In this study, banana resistant starch nanoparticles (RSNPs) were prepared by ultrasonic-assisted enzymatic hydrolysis self-assembly method. Compared with RSNPs prepared by traditional enzymatic hydrolysis, the particle size was significantly reduced to 58.48 ± 0.53 nm under low-high-frequency alternating treatment (MG), and the crystallinity was increased simultaneously. Elucidating the Antiglycation Potential of Plumbagin Against Methylglyoxal-Induced Glycation of Bovine Liver Catalase.

Non-enzymatic glycosylation of proteins leads to the formation of advanced glycation end products (AGEs), implicated in oxidative stress and the progression of several chronic diseases. Among the reactive carbonyl compounds, methylglyoxal (MGO) is highly potent in inducing glycoxidative damage by modifying proteins. Herein, we investigated the protective effect of plumbagin, a naphthoquinone derived from Plumbago zeylanica, to counteract MGO-induced glycation of catalase, employing integrated biophysical and computational approaches. Polyphenols in Alzheimer's Disease and in the Gut-Brain Axis.

Polyphenolic antioxidants, including dietary plant lignans, modulate the gut-brain axis, which involves transformation of these polyphenolic compounds into physiologically active and neuroprotector compounds (called human lignans) through gut bacterial metabolism. These gut bacterial metabolites exert their neuroprotective effects in various neurodegenerative diseases, such as Alzheimer's disease (AD) and Parkinson's disease (PD), and also have protective effects against other diseases, such as cardiovascular diseases, cancer, and diabetes. For example, enterolactone and enterodiol, the therap
