Radioimmunotherapy (RIT) has long been pursued to improve outcomes in acute leukemia and higher-risk myelodysplastic syndrome (MDS). Of increasing interest are alpha-particle-emitting radionuclides such as astatine-211 (211At) as they deliver large amounts of radiation over just a few cell diameters, enabling efficient and selective target cell kill. Here, we developed 211At-based RIT targeting CD123, an antigen widely displayed on acute leukemia and MDS cells including underlying neoplastic stem cells. We generated and characterized new murine monoclonal antibodies (mAbs) specific for human C

In this work, we study the chemical bond in molecules containing heavy and super-heavy elements according to the current state-of-the-art bonding models. An Energy Decomposition Analysis in combination with Natural Orbital for Chemical Valence (EDA-NOCV) within the relativistic four-component Dirac-Kohn-Sham (DKS) framework is employed, which allows to successfully include the spin-orbit coupling (SOC) effects on the chemical bond description. Simple halogen-bonded adducts ClX⋯L (X=At, Ts; L=NH3, Br-, H2O, CO) of astatine and tennessine have been selected to assess a trend on descending along

Access to 211At- and 125I-radiolabeled compounds in excellent RCCs and RCYs was achieved in just 10 min at room temperature using a Cu catalyst. The reaction conditions are applicable to a broad class of aryl and heteroaryl boronic reagents with varying steric and electronic properties as well as late-stage astatination and iodination of anticancer PARP inhibitors. This protocol eliminates the traditional need for toxic organotin reagents, elevated temperatures, and extended reaction times, providing a more practical and environmentally friendly approach to developing α-emitting radiotherapeut

Successful transplantation of autologous gene-modified hematopoietic stem and progenitor cells (HSPCs) requires efficient ablation of resident hematopoietic stem cells. Considering that conventional myeloablative conditioning regimens are associated with nonhematologic toxicities, we evaluated CD45-directed radioimmunotherapy (RIT) using the α-emitter astatine 211 (211At) before the transplantation of ex vivo gene-edited autologous HSPCs in nonhuman primates as an alternative. We humanized the CD45 antibody, BC8, and labeled it with 211At. As a model, mobilized CD34+ HSPCs were multiplex gene-

Radioimmunotherapy (RIT) is used to treat patients with hematologic malignancies known to infiltrate the bone marrow (BM) microenvironment. RIT uses target-specific monoclonal antibodies stably conjugated to radionuclides to deliver cytotoxic radiation to cells of interest. Although RIT is effective at delivering radiation to cancer cells, normal tissue is also exposed to radiation during RIT, the consequences of which are largely unknown. Here, we studied the cellular and molecular effects of CD45-targeted astatine-211 (211At) RIT, immunoglobulin G (IgG) nontargeted 211At RIT, and Cesium-137

We present a systematic study of electron-correlation and relativistic effects in diatomic molecular species of the heaviest halogen astatine (At) within relativistic single- and multireference coupled-cluster approaches and relativistic density functional theory. We establish revised reference ab initio data for the ground states of At2, HAt, AtAu, and AtO+ using a highly accurate relativistic effective core potential model and in-house basis sets developed for accurate modeling of molecules with large spin-orbit effects. Spin-dependent relativistic effects on chemical bonding in the ground s

BACKGROUND: Astatine-211 (211At) is a promising radionuclide for targeted alpha therapy (TAT), yet its clinical translation is hindered by poor in vivo stability and pronounced non-target organ accumulation. The fundamental radiochemical mechanisms governing the biodistribution of free 211At remain insufficiently understood, limiting rational radiopharmaceutical design.
RESULTS: Comparative in vivo studies revealed that free 211At and free iodine-131 (131I) initially share similar distribution patterns but diverge markedly thereafter, with 211At exhibiting pronounced accumulation in the stoma

Recently, 211At-related endoradiotherapy has emerged as an important oncotherapy strategy. Conjugating 211At with a nanocarrier provides a vital candidate for radionuclide therapy of various malignant tumors. In this study, we proposed utilizing the intrinsically high affinity of heavy halogens and sulfhydryl compounds for metallic silver to achieve highly efficient conjugation between 211At and Ag-based nanoparticles in a simple way. 211At@Ag-PEG-FA was obtained via a one-pot assembly of 211At, Ag, and SH-PEG-FA in extremely high radiolabeling yield (>95%) within 15 min and maintained excelle

UNLABELLED: The alpha emitter astatine-211 (211At) is a promising candidate for cancer treatment based on Targeted Alpha (α) Therapy (TAT). A small number of facilities, distributed across the United States, are capable of accelerating α-particle beams to produce 211At. However, challenges remain regarding strategic methods for shipping 211At in a form adaptable to advanced radiochemistry reactions and other uses of the radioisotope.
PURPOSE: Our method allows shipment of 211At in various quantities in a form convenient for further radiochemistry.
PROCEDURES: For this study, a 3-octanone imp

The 211At-labeled compound, 4-[211At]astato-l-phenylalanine, is one of the most promising amino acid derivatives for use in targeted alpha therapy (TAT) for various cancers. Electrophilic demetallation of a stannyl precursor is the most widely used approach for labeling biomolecules with 211At. However, the low acid-resistance of the stannyl precursor necessitates the use of an N- and C-terminus-protected precursor, which results in a low overall radiochemical yield (RCY) due to the multiple synthetic steps involved. In this study, a deprotected organosilyl compound, 4-triethylsilyl-l-phenylal

PURPOSE: Targeted α-therapy (TAT) for prostate-specific membrane antigen (PSMA) is a promising treatment for metastatic castration-resistant prostate cancer (CRPC). Astatine is an α-emitter (half-life=7.2 h) that can be produced by a 30-MeV cyclotron. This study evaluated the treatment effect of 211At-labeled PSMA compounds in mouse xenograft models.
METHODS: Tumor xenograft models were established by subcutaneous transplantation of human prostate cancer cells (LNCaP) in NOD/SCID mouse. [211At]PSMA1, [211At]PSMA5, or [211At]PSMA6 was administered to LNCaP xenograft mice to evaluate biodistrib

Astatine-211 (211At) is a promising alpha emitter for cancer treatment, wherein tumor-selective accumulation is pivotal due to its short path length. While sodium ascorbate (SA) successfully protects radioactive antibodies from reactive oxygen species (ROS)-induced denaturation, it does not reduce 211At distribution in normal organs or mitigate body weight loss. Here, we aimed to attenuate this normal organ uptake. We demonstrated that sodium perchlorate (SP), a competitive inhibitor of the sodium iodide symporter (NIS) expressed in thyroid and gastric mucosal cells, significantly reduced 211A

PURPOSE: Astatine-211 (At-211) is a promising alpha emitter for radionuclide therapy. High-resolution in vivo imaging of At-211 in small animals is needed for the development of At-211 radiopharmaceuticals. For this purpose, we developed a low-energy x-ray camera using a thin YAlO3 :Ce (YAP(Ce)) plate to image the low-energy x rays (73-87 keV) from the daughter radionuclide of At-211 (Po-211).
METHOD: We optically coupled a 38 × 38 × 1-mm YAP (Ce) plate to a position-sensitive photomultiplier (PSPMT) to develop an imaging detector. A pinhole or a parallel hole collimator was attached to the i

BackgroundGlioblastoma (GB), the most aggressive brain cancer, remains a critical clinical challenge due to its resistance to conventional treatments. Here, we introduce a locoregional targeted-α-therapy (TAT) with the rat monoclonal antibody 9E7.4 targeting murine syndecan-1 (SDC1) coupled to the α-emitter radionuclide astatine-211 (211At-9E7.4).MethodsWe orthotopically transplanted 50,000 GL261 cells of murine GB into the right striatum of syngeneic female C57BL/6JRj mice using stereotaxis. After MRI validation of tumour presence at day 11, TAT was injected at the same coordinates. Biodistri

Fibroblast activation proteins (FAP) are overexpressed in the tumor stroma and have received attention as target molecules for radionuclide therapy. The FAP inhibitor (FAPI) is used as a probe to deliver nuclides to cancer tissues. In this study, we designed and synthesized four novel 211At-FAPI(s) possessing polyethylene glycol (PEG) linkers between the FAP-targeting and 211At-attaching moieties. 211At-FAPI(s) and piperazine (PIP) linker FAPI exhibited distinct FAP selectivity and uptake in FAPII-overexpressing HEK293 cells and the lung cancer cell line A549. The complexity of the PEG linker

The recent accurate and precise determination of the electron affinity (EA) of the astatine atom At0 warrants a re-investigation of the estimated thermodynamic properties of At0 and astatine containing molecules as this EA was found to be much lower (by 0.4 eV) than previous estimated values. In this contribution we estimate, from available data sources, the following thermodynamic and physicochemical properties of the alkali astatides (MAt, M = Li, Na, K, Rb, Cs): their solid and gaseous heats of formation, lattice and gas-phase binding enthalpies, sublimation energies and melting temperature

PURPOSE: Radiopharmaceutical therapy (RPT) aims to irradiate tumors using antibodies or small molecules chelated with radioisotopes that target tumor cells. The biological response resulting from the complex interplay between radioisotope decay and cell binding processes is not yet fully understood. Because dose, including its spatiotemporal pattern, strongly correlates with ionizing radiation effects, detailed dosimetry is essential to predict biological responses. This study introduces TOol for PArticle Simulation (TOPAS)-RPT, a Monte Carlo platform for stochastic and spatiotemporal heteroge

Monoclonal antibodies C215, reactive with colorectal carcinomas, and MOv18, reactive with most of the ovarian carcinomas, were radiohalogenated with [211At]astatine. The radiohalogen was conjugate coupled to antibodies via the intermediate labelling reagent N-succinimidyl-3-(trimethylstannyl)benzoate (m-MeATE) in a two-step, single-pot reaction. Optimisation of the labelling of the reagent was achieved using N-iodosuccinimide, NIS, as the oxidising agent. The yields ranged from 69-95% in the labelling of 0.1-1.0 nmole of the m-MeATE precursor. Subsequent conjugation to antibodies resulted in y
