Here we report for the first time of a new Kunitz-type trypsin inhibitor (termed BvvTI) from seeds of the Camel's foot tree, Bauhinia variegata var. variegata. BvvTI shares the same reactive site residues (Arg, Ser) and exhibits a homology of N-terminal amino acid sequence to other Bauhinia protease inhibitors. The trypsin inhibitory activity (K(i), 0.1 x 10(-9)M) of BvvTI ranks the highest among them. Besides anti-HIV-1 reverse transcriptase activity, BvvTI could significantly inhibit the proliferation of nasopharyngeal cancer CNE-1 cells in a selective way. This may partially be contributed

Neuroinflammation is pivotal in the development of numerous neurodegenerative disorders, such as Alzheimer's disease (AD), Parkinson's disease (PD), and multiple sclerosis (MS). Microglial cells, the principal immune cells of the central nervous system (CNS), are essential mediators of this process. Upon exposure to pathogenic stimuli such as lipopolysaccharide (LPS), microglia activate and release pro-inflammatory mediators, leading to heightened oxidative stress and neuronal damage. Therefore, targeting microglial activation is a promising therapeutic approach to prevent or slow neurodegener

Dengue virus (DENV) is the most prevalent mosquito-borne viral pathogen and made the disease a major health concern worldwide. However, specific antiviral drugs against this arbovirose or vaccines are not yet available for treatment or prevention. Thus, here we aimed to study the antiviral activity of hydroethanolic extract, fraction ethyl acetate and subfractions of the leaves of Bauhinia holophylla (Fabaceae:Cercideae), a native plant of the Brazilian Cerrado, against DENV-2 by methylthiazolyldiphenyl-tetrazolium bromide (MTT) method in mammalian cells culture. As results, the hydroethanolic

BACKGROUND: The search for new active compounds from the Brazilian flora has intensified in recent years, especially for new drugs with antibiotic potential. Accordingly, the aim of this study was to determine whether riachin has antibiotic activity in itself or is able to modulate the activity of conventional antibiotics.
METHODS: A non-cyanogenic cyanoglycoside known as riachin was isolated from Bauhinia pentandra, and was tested alone and in combination with three antibiotics (clindamycin, amikacin, and gentamicin) against multiresistant bacterial strains (Escherichia coli, Pseudomonas aer

Scorpion envenoming causes an intense autonomic discharge, leading to a massive release of neurotransmitters, giving rise to several pathophysiological effects. In this work we report the effects of a Bauhinia forficata aqueous extract (BfAE) upon hyperglycemia, glycogenolysis, increase of plasma catecholamines, lethality and changes in serum insulin and plasma electrolytes induced by Tityus serrulatus scorpion venom (TSV). We compare them with the effects of the regular insulin therapy. The following treatments were performed: TSV (500 microg/kg, i.p.); BfAE (1g/kg, p.o.), 24, 12 and 1 h befo

Plants have served as the foundation of traditional medicine since the earliest civilisations. In light of this, it is crucial to study the phytochemical and physicochemical properties of Bauhinia variegata seed oil to discover novel secondary metabolites. Seed extracts were obtained through successive Soxhlet extraction using methanol and petroleum ether. The particle size of 0.25 mm achieved the highest bio-oil yield at 24.8%. The bio-oil was characterised using various techniques, including FTIR, GC-MS, and 1H & 13C NMR. Elemental analysis revealed the presence of Pd (0.03 ± 0.01 ppm), Cd (

In this study, a phytochemical investigation of Bauhinia brachycarpa led to the isolation of 23 compounds, including three new ones: bausesquitate C (1), 5-(benzofuran-2-yl)-2,3-dimethoxyphenol (2), and its β-D-glucoside (3). In addition, 14 known compounds were reported from this species for the first time. Compound 1 exhibited moderate α-glucosidase inhibitory activity (IC50 = 5.39 ± 0.30 μM) and acted as a mixed-type inhibitor (Ki = 0.29, Kis = 21.3), with stronger affinity for the free enzyme. Its binding mode and conformational stability were further elucidated by molecular docking and mo

Proteinase inhibitors extracted form medicinal plants are an interesting source of new drugs. Modifications in the structure of some of this kind of macromolecules could also lead to compounds of interesting biological properties. In this work, we synthesized and tested one fragment containing the reactive site of the Bauhinia bauhinioides kallikrein inhibitor (BbKI), denoted BbKI51-62 , and a related analog (P2 ) in which a proline residue was inserted in order to mimic the N-terminal region of the bradykinin molecule. The related retro-inverso counterparts Ri-BbKI51-62 and Ri-P2 were also in
