Prioritizing strategies for wheat biofortification: Inspiration from underutilized species.

The relationship between malnutrition and climate change is still poorly understood but a comprehensive knowledge of their interactions is needed to address the global public health agenda. Limited studies have been conducted to propose robust and economic-friendly strategies to augment the food basket with underutilized species and biofortify the staples for nutritional security. Sea-buckthorn is a known "superfood" rich in vitamin C and iron content. It is found naturally in northern hemispherical temperate Eurasia and can be utilized as a model species for genetic biofortification in cash c

Current Strategies for Selenium and Iodine Biofortification in Crop Plants.

Selenium and iodine are essential trace elements for both humans and animals. Among other things, they have an essential role in thyroid function and the production of important hormones by the thyroid gland. Unfortunately, in many areas, soils are deficient in selenium and iodine, and their amount is insufficient to produce crops with adequate contents to cover the recommended daily intake; thus, deficiencies have an endemic character. With the introduction of iodized table salt in the food industry, the thyroid status of the population has improved, but several areas remain iodine deficient.

Provitamin A Biofortification of Durum Wheat through a TILLING Approach.

Macro- and micronutrients, essential for the maintenance of human metabolism, are assimilated daily through the diet. Wheat and other major cereals are a good source of nutrients, such as carbohydrates and proteins, but cannot supply a sufficient amount of essential micronutrients, including provitamin A. As vitamin A deficiency (VAD) leads to several serious diseases throughout the world, the biofortification of a major staple crop, such as wheat, represents an effective way to preserve human health in developing countries. In the present work, a key enzyme involved in the branch of carotenoi

Chloroplast-to-chromoplast transition envisions provitamin A biofortification in green vegetables.

The carotenoids available in food are vital dietary micronutrients for human health. Plants synthesize and accumulate different carotenoids in plastids in a tissue-specific manner. The level of β-carotene (provitamin A) and other nutritionally important carotenoids is substantially low in the green tissues such as leaves compared to the fruits and roots. In photosynthetic tissues, chloroplasts can accumulate a moderate level of carotenoids, mainly to facilitate photosynthesis and environmental stress tolerance. However, chromoplasts from the storage tissues such as tomato fruit and carrot root

Combining ability and heterosis for grain iron biofortification in bread wheat.

BACKGROUND: Iron is one of the nutrients that is essential for the human body. Despite the abundance of iron on earth, about two billion people worldwide are affected by iron deficiency. Iron biofortification of wheat, instead of supplementation and food fortification, provides a pragmatic approach to solve the problem of iron deficiency. In this study, 144 diverse wheat genotypes were evaluated for grain iron and yield potential, to estimate the potential for the iron biofortification of high-yielding wheat varieties. RESULTS: Genotypes did not differ significantly across the species, but wi

Variability of carotenoids in a Musa germplasm collection and implications for provitamin A biofortification.

Bananas are important staples in tropical and sub-tropical regions and their potential as a source of provitamin A has recently attracted attention for biofortification. A collection of 189 banana genotypes (AAB-plantains, M. acuminata cultivars and bred hybrids) was screened to determine variability in fruit pulp provitamin A carotenoid (pVAC) content using high performance liquid chromatography. Total carotenoid content in tested genotypes varied from 1.45 µg/g for hybrid 25447-S7 R2P8 to 36.21 µg/g for M. acuminata cultivar ITC.0601 Hung Tu with a mean of 8.00 µg/g fresh weight. Seed Biofortification and Phytic Acid Reduction: A Conflict of Interest for the Plant?

Most of the phosphorus in seeds is accumulated in the form of phytic acid (myo-inositol-1,2,3,4,5,6-hexakisphosphate, InsP₆). This molecule is a strong chelator of cations important for nutrition, such as iron, zinc, magnesium, and calcium. For this reason, InsP₆ is considered an antinutritional factor. In recent years, efforts to biofortify seeds through the generation of low phytic acid (lpa) mutants have been noteworthy. Moreover, genes involved in the biosynthesis and accumulation of this molecule have been isolated and characterized in different species. Beyond its role in phosphorus stor

Zn and Fe biofortification: the right chemical environment for human bioavailability.

A considerable fraction of global disease burden and child mortality is attributed to Fe and Zn deficiencies. Biofortification, i.e. the development of plants with more bioavailable Zn and Fe, is widely seen as the most sustainable solution, provided suitable crops can be generated. In a cereal-dominated diet availability of Fe and Zn for absorption by the human gut is generally low and influenced by a highly complex chemistry. This complexity has mostly been attributed to the inhibitory effect of Fe and Zn binding by phytate, the principal phosphorus storage compound in cereal and legume seed

Natural genetic variation in lycopene epsilon cyclase tapped for maize biofortification.

Dietary vitamin A deficiency causes eye disease in 40 million children each year and places 140 to 250 million at risk for health disorders. Many children in sub-Saharan Africa subsist on maize-based diets. Maize displays considerable natural variation for carotenoid composition, including vitamin A precursors alpha-carotene, beta-carotene, and beta-cryptoxanthin. Through association analysis, linkage mapping, expression analysis, and mutagenesis, we show that variation at the lycopene epsilon cyclase (lcyE) locus alters flux down alpha-carotene versus beta-carotene branches of the carotenoid

Microbial fermentation for riboflavin biofortification in African traditional fermented foods.

Lactic Acid Bacteria (LAB) are commonly employed in the food industry as microbial starters, probiotics and for in situ biofortification of fermented foods. The use of vitamin-overproducing LAB represents a promising strategy for enhancing the nutritional value of staple home-made fermented foods. This approach could play a significant role in addressing deficiencies of B-group vitamins, especially vitamin B2, in underdeveloped regions.

Biofortification of tomatoes with beta-carotene through targeted gene editing.

Vitamin A deficiency is one of the most severe micronutrient-related health issues worldwide. Tomatoes, a widely cultivated crop for their adaptability, nutritional value, and lycopene content (a beta-carotene precursor), are ideal candidates for biofortification. In this study, CRISPR-mediated knockout mutants (cr-SlLCYe and cr-SlBCH) were generated to enhance the precursor supply to the β-carotene biosynthetic pathway and reduce its degradation. Carotenoids profiling showed that β-carotene levels in the mutants were 1.7 to 2.5-fold higher than in the wild-type, whereas lycopene levels remain

Toward Eradication of B-Vitamin Deficiencies: Considerations for Crop Biofortification.

'Hidden hunger' involves insufficient intake of micronutrients and is estimated to affect over two billion people on a global scale. Malnutrition of vitamins and minerals is known to cause an alarming number of casualties, even in the developed world. Many staple crops, although serving as the main dietary component for large population groups, deliver inadequate amounts of micronutrients. Biofortification, the augmentation of natural micronutrient levels in crop products through breeding or genetic engineering, is a pivotal tool in the fight against micronutrient malnutrition (MNM). Although

Biofortification of soy (Glycine max (L.) Merr.) with strontium ions.

Soy (Glycine max (L.) Merr.) is an annual plant cultivated worldwide mostly for food. Moreover, due to its pharmacological properties it is widely used in pharmacy for alleviating the symptoms of osteoporosis. The aim of the present study was to investigate the biofortification of soy treated with various concentrations of strontium. Soy was found to have a strong capacity to absorb Sr(2+) (bioconcentration factor higher than 1). A positive linear correlation (R(2) > 0.98) between the amount of strontium in the growth medium and its content in the plant was also observed. Moreover, at a concen

Unraveling transcriptomics of sorghum grain carotenoids: a step forward for biofortification.

BACKGROUND: Sorghum (Sorghum bicolor [L.] Moench) is a promising target for pro-vitamin A biofortification as it is a global staple crop, particularly in regions where vitamin A deficiency is prevalent. As with most cereal grains, carotenoid concentrations are low in sorghum, and breeding could be a feasible strategy to increase pro-vitamin A carotenoids to biologically relevant concentrations. However, there are knowledge gaps in the biosynthesis and regulation of sorghum grain carotenoids, which can limit breeding effectiveness. The aim of this research was to gain an understanding of the tr

CRISPR-mediated iron and folate biofortification in crops: advances and perspectives.

Micronutrient deficiency conditions, such as anemia, are the most prevalent global health problem due to inadequate iron and folate in dietary sources. Biofortification advancements can propel the rapid amelioration of nutritionally beneficial components in crops that are required to combat the adverse effects of micronutrient deficiencies on human health. To date, several strategies have been proposed to increase micronutrients in plants to improve food quality, but very few approaches have intrigued `clustered regularly interspaced short palindromic repeats' (CRISPR) modules for the enhancem

Provitamin A biofortification of crop plants: a gold rush with many miners.

Carotenoids are synthesized de novo by plants, where they play fundamental physiological roles as photosynthetic pigments and precursors for signaling molecules. They are also essential components of a healthy diet, as dietary antioxidants and vitamin A precursors. Vitamin A deficiency is a public health problem in developing countries, which has prompted a series of efforts toward the biofortification of plant-derived foods with provitamin A carotenoids (mainly β-carotene), giving rise to 'golden' crops. Since the 'golden rice' exploit, a number of biofortified crops have been generated, usin
