The Antifungal Activity of a Polygalacturonic and Caprylic Acid Ointment in an In Vitro, Three-Dimensional Wound Biofilm Model.

Candida colonization and biofilms are significant contributors to impaired wound healing. Consequently, improved treatments are needed to eradicate Candida biofilms in wounds. Wounds present complex biofilm extracellular matrix environments, with microbial cells frequently enmeshed in matrices comprising wound exudate macromolecular gels. We evaluated the ability of a polygalacturonic and caprylic acid (PG + CAP) ointment to eradicate Candida albicans, C. parapsilosis, C. glabrata, C. tropicalis, and C. auris biofilms in a fibrin gel wound biofilm model of the complex wound biofilm environment

A rapid colorimetric assay for the extracellular lipase of Pseudomonas fluorescens B52 using beta-naphthyl caprylate.

Conditions necessary for the determination of crude extracellular lipase activity from Pseudomonas fluorescens B52 using beta-naphthyl caprylate (beta-NC), an 8-carbon ester, as the substrate were examined. Maximum enzyme synthesis occurred at 20 degrees C in pyruvate mineral salts medium containing 1 mM-CaCl2. Bile salts were necessary for enzyme activity; 6 mM-Na taurocholate or Na deoxycholate gave maximum activity but the latter compound was inhibitory at higher concentrations. Activity was optimal in N-Tris[hydroxymethyl]methyl-2-aminoethane sulphonic acid buffer pH 8.0 at 40 degrees C. A

Anticonvulsant profile of caprylic acid, a main constituent of the medium-chain triglyceride (MCT) ketogenic diet, in mice.

The purpose of the present study was to evaluate the acute anticonvulsant effects of caprylic acid (CA), the main constituent of the medium-chain triglyceride ketogenic diet (MCT KD), in seizure tests typically used in screening for potential antiepileptic drugs in mice. Pharmacodynamic and pharmacokinetic interactions between CA and valproate (VPA) were also investigated. CA (p.o.) and VPA (i.p.) were administered 30 min before testing. Acute effects on motor coordination were assessed in the chimney test. Total plasma and brain concentrations of CA and VPA, when administered alone or in comb

Influence of Carbohydrate Intake on Caprylic Acid (C8:0)-Induced Ketogenesis-A Systematic Review and Meta-Analysis.

The ketogenic diet is used worldwide to treat various diseases, especially drug-resistant epilepsies. Medium-chain triglycerides or medium-chain fatty acids, primarily the major ketogenic compound caprylic acid (C8; C8:0), can significantly support ketogenesis. This review examines the effects of concurrent carbohydrate intake on C8-induced ketogenesis. A systematic literature search (PubMed and Web of Science) with subsequent data extraction was performed according to PRISMA guidelines and the Cochrane Handbook. Studies investigating the metabolic response to C8-containing MCT interventions w

Caprylic Acid Inhibits High Mobility Group Box-1-Induced Mitochondrial Damage in Myocardial Tubes.

Myocardial damage significantly impacts the prognosis of patients with cancer; however, the mechanisms of myocardial damage induced by cancer and its treatment remain unknown. We previously reported that medium-chain fatty acids (MCFAs) improve cancer-induced myocardial damage but did not evaluate the differences in effect according to MCFA type. Therefore, this study investigated the role of inflammatory cytokines in cancer-induced myocardial damage and the effects of three types of MCFAs (caprylic acid [C8], capric acid [C10], and lauric acid [C12]). In a mouse model, the C8 diet showed a gr

Sodium caprylate improves intestinal mucosal barrier function and antioxidant capacity by altering gut microbial metabolism.

Short-chain fatty acids (SCFA) produced by gut microbial metabolism have been reported to regulate the immunological response and intestinal health of the host by activating G protein-coupled receptors (GPR). It is unclear whether medium-chain fatty acids (MCFA) improve the intestinal barrier function by activating GPR. This study was conducted to investigate the effects of sodium caprylate and sodium butyrate on antioxidant capacity and intestinal barrier function in IPEC-J2 treated with H2O2 and C57/BL6 mouse models. For in vitro and in vivo assays, mice and oxidation-damaged IPEC-J2 (NC) we

Borneol-caprylic acid deep eutectic solvent (DES) based microemulsion gel for improved topical delivery of ivermectin.

Ivermectin (IVM), a broad-spectrum and hydrophobic antiparasitic drug, is used topically for the treatment of inflammatory skin conditions such as rosacea. The hydrophobicity of IVM and its limited solubility in lipid vehicles necessitate the development of new topical delivery approaches. Here, we evaluate the potential of deep eutectic solvent (DES) to improve the solubility of IVM, followed by the development of a DES-in-water microemulsion gel to improve the topical delivery of IVM. We demonstrate that borneol, a hydrophobic bicyclic terpene alcohol with penetration-enhancing capabilities,

Hepatic-portal oleic acid inhibits feeding more potently than hepatic-portal caprylic acid in rats.

In several human and animal studies, medium-chain triglycerides decreased food intake more than did long-chain triglycerides. It is possible that faster uptake and metabolism of medium-chain fatty acids in the liver is responsible for this difference. To test this hypothesis we compared the feeding effects of hepatic portal vein (HPV) infusion of the medium-chain fatty acid caprylic acid (CA) with those of the long-chain fatty acid oleic acid (OA). Contrary to our expectation, six-h HPV infusion of 14 microg/min (50 nmol/min) OA robustly inhibited feeding, whereas infusion of 22 or 220 microg/

Site-specific immunosuppression using a new formulation of topical cyclosporine A with polyethylene glycol-8 glyceryl caprylate/caprate.

PURPOSE: Dermal application of immunosuppressants can be an effective means of achieving site-specific immunosuppression (SITE) on skin allografts in burn wound management and in the treatment of various immune skin disorders. We have previously reported success with topical cyclosporine A (tCsA) in the treatment of skin allograft rejection in rats. Using a new tCsA formulation with a penetration enhancer (PE), polyethylene glycol-8 (PEG-8) glyceryl caprylate/caprate (Labrasol, Gattefossé, St. Priest, France), in a trinary drug delivery system, we hypothesized that we would induce SITE and sig

Inclusion of trans-cinnamaldehyde and caprylic acid in feed results in detectable concentrations in the chicken gut and reduces foodborne pathogen carriage.

Poultry act as a major reservoir host for Salmonella and Campylobacter spp., the 2 leading causes of foodborne illnesses globally and in the United States. Preharvest stage interventions to reduce foodborne pathogen carriage in poultry are increasingly informed by consumer preference for antibiotic-free poultry production. The in-feed inclusion of plant-derived antimicrobial compounds is a promising antibiotic alternative strategy to reduce foodborne pathogen load in the broiler chicken gut. Yet, the fate of these phytochemicals through the broiler chicken gastrointestinal tract is unknown. Li

Propanediol (and) Caprylic Acid (and) Xylitol as a New Single Topical Active Ingredient against Acne: In Vitro and In Vivo Efficacy Assays.

In addition to dermatological complications, acne can affect the quality of life of individuals in numerous ways, such as employment, social habits and body dissatisfaction. According to our expertise, caprylic acid and propanediol would not have a direct action on Cutibacterium acnes. Despite this, we investigated the existence of a synergistic effect among xylitol, caprylic acid and propanediol as a mixture of compounds representing a single topical active ingredient that could benefit the treatment against acne. In vitro and in vivo assays were performed to challenge and to prove the effica

Application of a caprylate/chromatography purification process for production of a high potency rabies immune globulin from pooled human plasma.

BACKGROUND: Human rabies immunoglobulin (RIG) is an integral part of post-exposure prophylactic treatment of rabies (along with rabies vaccination). Infiltration of most, if not all, of the RIG dose at the wound site is recommended. RIG produced by a caprylate/chromatography manufacturing process (RIG-C; HyperRAB) increased the potency and purity of this product over the existing licensed RIG from a solvent/detergent process (RIG-S/D; HyperRAB-S/D). METHODS: A series of studies were conducted to characterize the content and purity of RIG-C. A single-dose pharmacokinetic study in rabbits was p

Single-reagent one-step procedures for the purification of ovine IgG, F(ab')2 and Fab antivenoms by caprylic acid.

Antivenoms are typically produced in horses or sheep and often purified using salt precipitation of immunoglobulins or F(ab')2 fragments. Caprylic (octanoic) acid fractionation of antiserum has the advantage of not precipitating the desired antibodies, thereby avoiding potential degradation that can lead to the formation of aggregates, which may be the cause of some adverse reactions to antivenoms. Here we report that when optimising the purification of immunoglobulins from ovine antiserum raised against snake venom, caprylic acid was found to have no effect on the activity of the enzymes peps

Fungicidal efficacy of caprylic acid using three different bovine milk types against Candida albicans: An in vitro study.

BACKGROUND: The success of endodontic therapy depends on thorough biomechanical preparation and the use of effective intracanal irrigants to achieve a microbe-free canal before obturation. Candida albicans is the most commonly isolated fungus from infected root canals and has a strong dentin affinity. Owing to its resistance to several intracanal medicaments, including calcium hydroxide, alternative antifungal agents are required. Bovine milk, which contains caprylic acid - a medium-chain fatty acid with known antimicrobial properties, may provide a natural and biocompatible adjunct for root c

Efficacy of combined caprylic acid and thymol treatments for inactivation of Listeria monocytogenes on enoki mushrooms in household and food-service establishments.

Raw enoki mushroom is a high-risk vector for listeriosis, which led to foodborne outbreaks resulting in four deaths in the United States in 2020. This study aimed to investigate the washing method for the inactivation of L. monocytogenes in enoki mushrooms for household and food service establishments. Five methods of washing fresh agricultural products without using disinfectants were selected: (1) rinsing under running water (2 L/min, 10 min), (2-3) dipping in water (200 ml/20 g) at 22 or 40 °C for 10 min, and using (4) 10% NaCl or (5) 5% vinegar at 22 °C for 10 min. Minipool caprylic acid fractionation of plasma using disposable equipment: a practical method to enhance immunoglobulin supply in developing countries.

BACKGROUND: Immunoglobulin G (IgG) is an essential plasma-derived medicine that is lacking in developing countries. IgG shortages leave immunodeficient patients without treatment, exposing them to devastating recurrent infections from local pathogens. A simple and practical method for producing IgG from normal or convalescent plasma collected in developing countries is needed to provide better, faster access to IgG for patients in need. METHODOLOGY/PRINCIPAL FINDINGS: IgG was purified from 10 consecutive minipools of 20 plasma donations collected in Egypt using single-use equipment. Plasma do

Combination of organic acids benzoate, butyrate, caprylate, and sorbate provides a novel antibiotics-independent treatment option in the combat of acute campylobacteriosis.

INTRODUCTION: The food-borne Gram-negative bacterial pathogen Campylobacter jejuni may cause the acute enterocolitis syndrome campylobacteriosis in infected humans. Given that human C. jejuni infections are rising globally which hold also true for resistance rates against antibiotic compounds such as macrolides and fluoroquinolones frequently prescribed for the treatment of severe infectious enteritis, novel antibiotics-independent therapeutic strategies are needed. Distinct organic acids are well known for their health-beneficial including anti-microbial and immunomodulatory properties. In ou
