Influences of Coagulant Polarity on the Modulus of the Chitin Hydrogel.

The chitin hydrogel draws great attention in biomedical fields owing to its high similarity and good affinity for peptides. The conversion of raw chitin to the designed hydrogel through a sol-gel process prevails, while the modulus of the chitin hydrogel is significantly influenced by the factors of gelation technology (i.e., coagulation, involving polymer chain rearrangement and the reconstruction of multiple interactions). Water and several organic solvents such as ethanol, DMAc, and DMSO are effective coagulants for aqueous chitin/KOH/urea solutions. In particular, the concentration of aque

Evidence of chitin in the ampullae of Lorenzini of chondrichthyan fishes.

We previously reported that the polysaccharide chitin, a key component of arthropod exoskeletons and fungal cell walls, is endogenously produced by fishes and amphibians in spite of the widely held view that it was not synthesized by vertebrates [1]. Genes encoding chitin synthase enzymes were found in the genomes of a number of fishes and amphibians and shown to be correspondingly expressed at the sites where chitin was localized [1,2]. In this report, we present evidence suggesting that chitin is prevalent within the specialized electrosensory organs of cartilaginous fishes (Chondrichthyes).

Application of chitin and chitosan derivatives in the pharmaceutical field.

Chitin and chitosan derivatives are used as excipients and drug carriers in the pharmaceutical field. Their derivatization contributed to expansion of application and decrease toxicity. Chitosan is used as an excipient in oral dosage form. Chitosan tablet can exhibit a sustained drug release compared to commercial products. Films prepared using chitin or chitosan have been developed as wound dressings, oral mucoadhesive and water-resisting adhesive by virtue of their release characteristics and adhesion. Intratumoral administration of gadopentetic acid-chitosan complex nanoparticles (approxima

A novel family of chitin-binding proteins from insect type 2 peritrophic matrix. cDNA sequences, chitin binding activity, and cellular localization.

The peritrophic matrix is a prominent feature of the digestive tract of most insects, but its function, formation, and even its composition remain contentious. This matrix is a molecular sieve whose toughness and elasticity are generated by glycoproteins, proteoglycans, and chitin fibrils. We now describe a small, highly conserved protein, peritrophin-15, which is an abundant component of the larval peritrophic matrices of the Old World screwworm fly, Chrysomya bezziana, and sheep blowfly, Lucilia cuprina. Their deduced amino acid sequences code for a 8-kDa secreted protein characterized by a

Functional impact of dietary chitin upon Listeria monocytogenes pathogenesis.

Chitin, the second most abundant polymer in nature, is recognized for biocompatibility and diverse effects on microbes and the host. However, the role of dietary chitin against foodborne pathogens such as Listeria monocytogenes remains underexplored. Here, we show that chitin functions as an environmental signal that attenuates L. monocytogenes virulence and mitigates infection outcomes. Consistent with previous studies, exposure of L. monocytogenes to chitin resulted in downregulation of key virulence genes and we demonstrate that this is accompanied by reduced pathogenesis in intestinal epit

Chitin nanofibrils modulate mechanical response in tympanic membrane replacements.

The tympanic membrane (TM), is a thin tissue lying at the intersection of the outer and the middle ear. TM perforations caused by traumas and infections often result in a conductive hearing loss. Tissue engineering has emerged as a promising approach for reconstructing the damaged TM by replicating the native material characteristics. In this regard, chitin nanofibrils (CN), a polysaccharide-derived nanomaterial, is known to exhibit excellent biocompatibility, immunomodulation and antimicrobial activity, thereby imparting essential qualities for an optimal TM regeneration. Morintides: cargo-free chitin-binding peptides from Moringa oleifera.

BACKGROUND: Hevein-like peptides are a family of cysteine-rich and chitin-binding peptides consisting of 29-45 amino acids. Their chitin-binding property is essential for plant defense against fungi. Based on the number of cysteine residues in their sequences, they are divided into three sub-families: 6C-, 8C- and 10C-hevein-like peptides. All three subfamilies contain a three-domain precursor comprising a signal peptide, a mature hevein-like peptide and a C-terminal domain comprising a hinge region with protein cargo in 8C- and 10C-hevein-like peptides. RESULTS: Here we report the isolation

Characteristics of corneal lens chitin in dragonfly compound eyes.

Chitin in the compound eyes of arthropods serves as a part of the visual system. The quality of chitin in such highly specialised body parts deserves more detailed examination. Chitin in the corneal (ommatidial) lenses of dragonfly (Sympetrum fonscolombii) compound eyes was isolated by using the classical chemical method. The chitin content of the corneal lenses was determined to be quite high (20.3±0.85%). The FT-IR analysis showed that corneal lens chitin was in the α-form as found in all arthropod species where mechanical strength is required. The surface morphology analysis by scanning ele

Crystallization of calcium carbonate salts into beta-chitin scaffold.

Composites of beta-chitin with calcium carbonate polymorphs were prepared by precipitation of the mineral into a chitin scaffold by means of a double diffusion system. The beta-chitin was obtained from the pen of the Loligo sp. squid. The three main polymorphs of calcium carbonate: aragonite, calcite and vaterite, were observed. Their location within the matrix is a function of the polymorph. The supersaturation inside the compartmentalized space in the chitin governs the location and polymorphism of the crystals.

Efficient labeling of pathogenic fungal chitin with bioorthogonal probes.

The fungal cell wall plays a large role in pathogenicity and is essential to the survival and virulence of fungi such as Candida albicans. Filamentous growth and its causes have been studied, however specific cell wall features that contribute to this morphology have yet to be investigated thoroughly. Understanding how the makeup of cell wall polymers changes during hyphal growth is therefore critical to developing novel targets for therapeutics. Bio-orthogonal chemistry has emerged as a powerful tool to profile critical cellular processes without interfering with canonical machinery. This cha

Biocompatible chitin-based Janus hydrogel membranes for periodontal repair.

Periodontal defects caused by severe periodontitis are a widespread issue globally. Guided tissue regeneration (GTR) using barrier membranes for alveolar bone repair is a common clinical treatment. However, most commercially available collagen barrier membranes are expensive and lack the antibacterial properties essential for effective bone regeneration. Herein, we report a natural polysaccharide chitin hydrogel barrier membrane with a Janus structure (ChT-PDA-p-HAP), featuring high antibacterial and protein-repelling activity on the outer side and good osteogenesis ability on the inner side.

Carrageenan/Chitin Nanowhiskers Cryogels for Vaginal Delivery of Metronidazole.

The development of polymeric carriers based on partially deacetylated chitin nanowhiskers (CNWs) and anionic sulfated polysaccharides is an attractive strategy for improved vaginal delivery with modified drug release profiles. This study focuses on the development of metronidazole (MET)-containing cryogels based on carrageenan (CRG) and CNWs. The desired cryogels were obtained by electrostatic interactions between the amino groups of CNWs and the sulfate groups of CRG and by the formation of additional hydrogen bonds, as well as by entanglement of carrageenan macrochains. It was shown that the

Preparation of Composite Materials from Self-Assembled Chitin Nanofibers.

Although chitin is a representative abundant polysaccharide, it is mostly unutilized as a material source because of its poor solubility and processability. Certain specific properties, such as biodegradability, biocompatibility, and renewability, make nanofibrillation an efficient approach for providing chitin-based functional nanomaterials. The composition of nanochitins with other polymeric components has been efficiently conducted at the nanoscale to fabricate nanostructured composite materials. Disentanglement of chitin microfibrils in natural sources upon the top-down approach and regene

Fungal Chitin Reduces Platelet Activation Mediated via TLR8 Stimulation.

Platelets play an important role in the innate immune response. During candidaemia, circulating fungal polysaccharides, including chitin, are released into the bloodstream and can interact with platelets and induce modulation of platelet activities. However, the role of circulating chitin in platelet modulation has not been investigated. The aims of the present study were to assess the effect of fungal chitin on activation, adhesion, aggregation and receptor expression of platelets and their impact on the host defense against Candida albicans. Platelets pre-treated with different concentration

Protective Effect of Chitin Urocanate Nanofibers against Ultraviolet Radiation.

Urocanic acid is a major ultraviolet (UV)-absorbing chromophore. Chitins are highly crystalline structures that are found predominantly in crustacean shells. Alpha-chitin consists of microfibers that contain nanofibrils embedded in a protein matrix. Acid hydrolysis is a common method used to prepare chitin nanofibrils (NFs). We typically obtain NFs by hydrolyzing chitin with acetic acid. However, in the present study, we used urocanic acid to prepare urocanic acid chitin NFs (UNFs) and examined its protective effect against UVB radiation. Hos: HR-1 mice coated with UNFs were UVB irradiated (30

Construction of Flexible Kaolin/Chitin Composite Aerogels and Their Properties.

In this work, kaolin/chitin (K/Ch) composite aerogels with different mass ratios were successfully fabricated via a freeze-drying approach. The influence of kaolin content on the microstructure, properties and hemostatic performance of the composite aerogels was systematically investigated. The results demonstrated that the incorporation of kaolin endowed the chitin-based aerogels with tunable porous structures, excellent water absorption capacity (up to 4282% for K0.25/Ch2), and enhanced water retention (73.7% for K2/Ch2 at 60 min). Moreover, the K/Ch composite aerogels exhibited good biodegr

Chitin synthase, a fungal glycosyltransferase that is a valuable antifungal target.

During the last 30 past years, more life-threatening fungal infections have appeared due to the increasing frequency of patients with weakened immune systems. Inhibition of fungal enzymes involved in the biosynthesis of sterols is considered to be a safe and effective option for antifungal therapy. However, the intensive use of sterol-biosynthesis inhibitors for years has resulted in resistance development. Consequently, the search for alternative therapeutics must be intensified. In this context, the biosynthesis of chitin, an essential component of the fungal cell wall that is absent in mamm

Use of Chitin for Controlling Heterodera avenae and Tylenchulus semipenetrans.

The nematicidal effect of chitin, relative to other pesticides, was evaluated against two plant-parasitic nematodes, Heterodera avenae and Tylenchulus semipenetrans. Wheat seedlings, grown in soils artificially or naturally infested with H. avenae, were treated with 0.4% (w/w) ClandoSan (CLA) prepared from crustacean chitin, aldicarb (Temik 15G), or ethylene dibromide (EDB 90EC). The CLA treatment significantly increased wheat straw, ear, and average grain dry weights of nematode-infected plants, compared with the other two treatments. Adsorption of enzymes on krill chitin modified with carbon disulfide.

Krill chitin modified with carbon disulfide was used as a support of enzymes. The dithiocarbamino groups created after reaction of CS(2) with chitinous NH(2) can participate in binding the enzymes without denaturation and loss in activity. The stability of these bonds depends on pH and decreases gradually at ionic strengths higher than 0.01. Chitin modified with CS(2) can be used as a support for enzymes having high activity at mild conditions of pH and ionic strength.
