Trauma-specific insults to the cochlear nucleus in the rat.

The effect of acoustic overstimulation on the neuronal number of the cochlear nucleus (CN) was investigated by using unbiased stereological methods in rats. We found that, after 9 weeks of recovery, neurons in the anteroventral cochlear nucleus (AVCN) degenerated, whereas those in the posteroventral and dorsal cochlear nuclei (PVCN and DCN) were preserved. The noise trauma induced near complete loss of the outer hair cells throughout the cochlea, and the inner hair cells were preserved only in the more apical regions. This pattern of selective loss of AVCN neurons in this study was different f

[Chlorphentermine-induced lipidosis of the cochlea and the cochlear nucleus].

The anoretic drug chlorphentermine induces, after long-term feeding, an accumulation of phospholipids mainly stored in lysosomes. Inner hair cells and nerve running to these cells are especially affected, whereas the outer hair cells and their nerves are less changed. Supporting cells and ganglion cells of the spiral ganglion always exhibit inclusions. Changes in the perikarya of the cochlear nucleus are not so outstanding and are in the scope of the alterations found in other parts of the nervous system. The morphological alterations allow some conclusions about the phospholipid metabolism an

Nucleus 22 cochlear implantation results in postmeningitic deafness.

Cochlear implant surgery was performed on 13 patients with postmeningitic deafness (seven adults, six children). Two adults and two children (30.8%) had severe labyrinthitis ossificans requiring radical "drill-out." Five of 13 (38.5%) had some bone growth requiring partial drill-out, and four of 13 (30.8%) had normal insertion with no drill-out. Hearing results for patients with no bone growth were similar to nonmeningitic patients; three of four (75%) had open-set speech recognition. Performance of patients with total drill-out was poor; "auditory only" performance was limited to detection an

Somatosensory context alters auditory responses in the cochlear nucleus.

The cochlear nucleus, the first central auditory structure, performs initial stimulus processing and segregation of information into parallel ascending pathways. It also receives nonauditory inputs. Here we show in vivo that responses of dorsal cochlear nucleus (DCN) principal neurons to sounds can change significantly depending on the presence or absence of inputs from the somatosensory dorsal column nucleus occurring before the onset of auditory stimuli. The effects range from short-term suppression of spikes lasting a few milliseconds at the onset of the stimulus to long-term increases or d

Somatic (craniocervical) tinnitus and the dorsal cochlear nucleus hypothesis.

PURPOSE: Of all nonauditory sensory systems, only the somatosensory system seems to be related to tinnitus (eg, temporomandibular joint syndrome and whiplash). The purpose of this study is to describe the distinguishing characteristics of tinnitus associated with somatic events and to use these characteristics to develop a neurological model of somatic tinnitus. MATERIALS AND METHODS: Case series. RESULTS: Some patients with tinnitus, but no other hearing complaints, share several clinical features including (1) an associated somatic disorder of the head or upper neck, (2) localization of th

Avian cochlear nucleus neurons exhibit tonotopic specializations across development.

Tonotopy is a hallmark feature of auditory structures that describes the spatial separation of sound frequencies along a gradient from high to low. Identifying biophysical differences along the frequency axis is essential for understanding how tonotopic maps develop in auditory structures and how these maps serve specific functions. In the mammalian cochlear nucleus and the analogous avian nucleus magnocellularis (NM), neurons are arranged along a tonotopic gradient resulting from ordered synaptic connections from the auditory nerve. Although mature NM neurons exhibit some intrinsic (i.e., ion

[Changes of GABA immunoreactivity in aged rat cochlear nucleus].

OBJECTIVES: To detect the changes of gama-aminobutyric acid (GABA) immunoreactivity in the aged rat cochlear nucleus and to investigate its relationship with the initiation of presbycusis. METHODS: The cochlear nucleuses of adult (2-3 months) and aged (20-36 months) Wistar rats were stained with streptavidin/peroxidase (S-P) technique. GABA-positive neurons were counted with a computer-aid image analysis system. RESULTS: Most of the GABA-immunoreactive cell bodies were found in the superficial layers of the dorsal cochlear nucleus (DCN). The number of GABA-positive neurons decreased signific

Fitting the Nucleus-22 cochlear implant for Spanish speakers.

This work aimed to study the speech comprehension of 13 native Spanish-speaking patients implanted with the Nucleus-22 system. Experiments were performed under two conditions. First, tests of isolated vowel discrimination, bisyllable word recognition in a consonant-vowel-consonant context, and fluency speech comprehension were conducted with the speech processors (SPs) fitted with the frequency boundary distribution as specified by the diagnostic and programming system. This distribution was called default frequency boundary. For the second condition, the SP of each patient was refitted using

Baclofen reduces tone-evoked activity of cochlear nucleus neurons.

Recent evidence suggests that an excitant amino acid may be a neurotransmitter at acoustic nerve synapses in cochlear nucleus (CN). Release of excitant amino acids is reportedly reduced by baclofen, a lipophilic GABA-mimetic used to treat the spasticity of multiple sclerosis and spinal injury. Microiontophoresis of (-)baclofen suppressed spontaneous and tone-evoked activity in CN neurons. GABA inhibited the responses of most neurons responsive to (-)baclofen. However, iontophoresis of these two substances onto the same CN neuron resulted in dramatic differences in time course to maximum effect

TrkB levels in the cochlear nucleus after unilateral cochlear ablation: correlations with post-lesion plasticity.

Tyrosine kinase B (TrkB) levels in the adult guinea pig cochlear nucleus (CN) were determined from Western blots for up to 60 days after unilateral cochlear ablation (UCA). Compared to TrkB levels on the intact side, those on the lesioned side were elevated in the anteroventral CN (AVCN) at 7 and 60 days and in the posteroventral CN (PVCN) at 30 days. TrkB levels were depressed in the AVCN and the dorsal CN (DCN) at 30 days. Elevations in the AVCN on the lesioned side at 7 days coincided with a period of synaptogenesis. Other changes were temporally related to up- or downregulations of transmi

Age-related structural and functional changes in the cochlear nucleus.

Presbycusis - age-related hearing loss - is a key communication disorder and chronic medical condition of our aged population. The cochlear nucleus is the major site of projections from the auditory portion of the inner ear. Relative to other levels of the peripheral and central auditory systems, relatively few studies have been conducted examining age-related changes in the cochlear nucleus. The neurophysiological investigations suggest declines in glycine-mediated inhibition, reflected in increased firing rates in cochlear nucleus neurons from old animals relative to young adults. Biochemica

The role of Grn in the development of mouse cochlear nucleus synapses.

Progranulin (encoded by Grn) is a multifunctional glycoprotein implicated in neuronal survival and synapse function, yet its role in central auditory circuit development remains unclear. In this study, we investigated the role of Grn in mouse cochlear nucleus (CN) synapse development. Grn-/- and Grn+/+ mice were used in this study. Grn (Progranulin) expression was examined by reverse transcription PCR and immunofluorescence. Neuronal density and synaptic puncta expression within the CN were analyzed using immunofluorescence labeling and quantitative image analysis. Auditory function was assess

Effects of cochlear ablation on amino acid concentrations in the chinchilla posteroventral cochlear nucleus, as compared to rat.

Using a microchemical approach, we measured changes of amino acid concentrations in the chinchilla caudal posteroventral cochlear nucleus (PVCN) after cochlear ablation to determine to what extent slow decreases of glutamate and aspartate concentrations after carboplatin treatment resulted from slower effects of cochlear damage in chinchillas than in rats and guinea pigs, as opposed to effects of carboplatin treatment being slower than those of cochlear ablation. Our results indicate that both factors are involved: decreases of glutamate and aspartate concentrations after cochlear ablation are

Long-term degeneration in the cochlear nerve and cochlear nucleus of the adult chinchilla following acoustic overstimulation.

Adult chinchillas were exposed once to an octave-band noise, centered at 4 kHz, and allowed to survive for 16 days or for 1, 2, 4, and 8 months. Axonal degeneration was mapped in the cochlear nucleus, using the Nauta-Rasmussen silver method, and related to hair cell damage and to loss of myelinated nerve fibers in the osseous spiral lamina of the cochlea. Axonal degeneration in the dorsal cochlear nucleus had already reached a peak by 16 days and disappeared after 1 month. Meanwhile, myelinated nerve fiber degeneration in the cochlea extended basally, followed 2 weeks to 2 months later by spre

Ventral cochlear nucleus bushy cells encode hyperacusis in guinea pigs.

Psychophysical studies characterize hyperacusis as increased loudness growth over a wide-frequency range, decreased tolerance to loud sounds and reduced behavioral reaction time latencies to high-intensity sounds. While commonly associated with hearing loss, hyperacusis can also occur without hearing loss, implicating the central nervous system in the generation of hyperacusis. Previous studies suggest that ventral cochlear nucleus bushy cells may be putative neural contributors to hyperacusis. Compared to other ventral cochlear nucleus output neurons, bushy cells show high firing rates as wel

Wiener kernel analysis of responses from anteroventral cochlear nucleus neurons.

Responses to pseudo-random Gaussian white noise, tones and clics were recorded from neurons in the anteroventral cochlear nucleus (AVCN) of barbiturate anesthetized cats. The responses to white noise were used to calculate estimates of the zero-, first- and second-order Wiener kernels for these neurons. The Wiener kernels did contain useful information on the fundamental, DC and second harmonic components of the responses of AVCN neurons to tones, clicks and noise. However, they generally did not provide predictions of the difference tone distortion products found in the peripheral auditory sy
