A novel linear B cell epitope of the canine coronavirus nucleocapsid protein identified by a monoclonal antibody.

The infection of canine coronavirus (CCoV) causes a highly contagious disease in dogs with acute gastroenteritis. The efficient serological diagnostics is critical for controlling the disease caused by CCoV. Nucleocapsid (N) protein of CCoV is an important target for developing serological approaches. However, little is known about the antigenic sites in the N protein of CCoV. In this study, we generated a monoclonal antibody (mAb) against the N protein of CCoV, designated as 13E8, through the fusion of the sp2/0 cells with the spleen cells from a mouse immunized with the purified recombinant

Phylogenetic characterization of partial spike gene sequences of canine coronavirus from shelter dogs in the Republic of Korea.

This study investigated the molecular characteristics of canine coronavirus (CCoV) circulating in animal shelters in Korea, focusing on phylogenetic characterization of the partial spike (S) gene. A total of 339 rectal swabs were analyzed to assess the prevalence and phylogenetic relationships of circulating strains. Among these, 20 and 13 samples were identified as CCoV-I and CCoV-IIa, respectively, indicating one co-infection case. Phylogenetic analysis demonstrated that Korean isolates exhibited distinct sequence variations relative to global reference strains and formed regionally differen

Development and validation of nanoplate-based RT-dPCR assay for canine respiratory coronavirus detection in various clinical samples.

BACKGROUND: Canine respiratory coronavirus (CRCoV) is a major contributor to the canine infectious respiratory disease complex (CIRDC). Despite its widespread prevalence, molecular assays for CRCoV detection remain limited. Additionally, the efficiency and accuracy of detection can vary depending on the type of clinical sample used, such as nasal swabs (NS), oropharyngeal swabs (OS), and rectal swabs (RS). To address these challenges, we developed a nanoplate-based reverse transcription digital polymerase chain reaction (RT-dPCR) method for detecting the spike gene of CRCoV in various clinical

Nucleotide sequence and expression of the spike (S) gene of canine coronavirus and comparison with the S proteins of feline and porcine coronaviruses.

We have cloned, sequenced and expressed the spike (S) gene of canine coronavirus (CCV; strain K378). Its deduced amino acid sequence has revealed features in common with other coronavirus S proteins: a stretch of hydrophobic amino acids at the amino terminus (the putative signal sequence), another hydrophobic region at the carboxy terminus (the membrane anchor), heptad repeats preceding the anchor, and a cysteine-rich region located just downstream from it. Like other representatives of the same antigenic cluster (CCV-Insavc-1 strain, feline infectious peritonitis and enteric coronaviruses, po

Design and preclinical evaluation of a multi-epitope circular RNA vaccine targeting the spike protein of canine coronavirus genotype II.

The rapid evolution and immune evasion of the Spike (S) protein in canine coronavirus type II (CCoV-II) necessitate the development of vaccines with enhanced durability and broad-spectrum efficacy. Circular RNA (circRNA) offers a superior platform to linear mRNA due to its covalently closed structure, which resists exonuclease degradation and ensures sustained antigen expression. In this study, we designed a multiepitope circRNA vaccine targeting conserved CTL, HTL, and LBL epitopes of the CCoV-II S protein, encapsulated in lipid nanoparticles (LNPs). The LNP-circRNA formulations exhibited hig

Imiquimod, a Promising Broad-Spectrum Antiviral, Prevents SARS-CoV-2 and Canine Coronavirus Multiplication Through the MAPK/ERK Signaling Pathway.

Respiratory viruses can cause life-threatening conditions such as sepsis and acute respiratory distress syndrome. However, vaccines and effective antivirals are available for only a limited number of infections. The majority of approved antivirals are direct-acting agents, which target viral proteins essential for infection. Unfortunately, mutations have already emerged that confer resistance to these antivirals. In addition, there is an urgent need for broad-spectrum antivirals to address the unpredictable emergence of new viruses with pandemic potential. One promising strategy involves modul

A multiplex PCR method for the simultaneous detection of three viruses associated with canine viral enteric infections.

The aim of this study was to establish a multiplex PCR (mPCR) method that can simultaneously detect canine parvovirus (CPV-2), canine coronavirus (CCoV) and canine adenovirus (CAV), thereby eliminating the need to detect these pathogens individually. Based on conserved regions in the genomes of these three viruses, the VP2 gene of CPV-2, the endoribonuclease nsp15 gene of CCoV, and the 52K gene of CAV were selected for primer design. The specificity of the mPCR results showed no amplification of canine distemper virus (CDV), canine parainfluenza virus (CPIV), or pseudorabies virus (PRV), indic

Canine distemper and related diseases: report of a severe outbreak in a kennel.

An outbreak of canine distemper in a kennel of German shepherds in the province of Bari is reported. Six 42-day-old pups developed typical signs of canine distemper (fever, conjunctivitis, respiratory distress and enteritis) and died within 7-10 days. Neurological symptoms were observed only in one pup. Four additional pups, which had shown no sign of illness, were separated and vaccinated, but two of these developed a severe, fatal nervous form 15 days later. Post-mortem examination, carried out on two pups which died without neurological signs, showed pneumonia and enteritis, more severe in

Identification of coronaviruses in farmed wild animals reveals their evolutionary origins in Guangdong, southern China.

Coronavirus infections cause diseases that range from mild to severe in mammals and birds. In this study, we detected coronavirus infections in 748 farmed wild animals of 23 species in Guangdong, southern China, by RT-PCR and metagenomic analysis. We identified four coronaviruses in these wild animals and analysed their evolutionary origins. Coronaviruses detected in Rhizomys sinensis were genetically grouped into canine and rodent coronaviruses, which were likely recombinants of canine and rodent coronaviruses. The coronavirus found in Phasianus colchicus was a recombinant pheasant coronaviru

Circulation and Genetic Characterizations of Coronaviruses From Companion Animals in Chengdu, Southwest China: One-Year Postpandemic.

Coronaviruses (CoVs) can cross species barriers and endanger public health. Despite reports on their circulation and evolution in companion animals during the pandemic, postpandemic surveillance remains crucial. Therefore, during the first postpandemic year, 309 samples from 263 companion animals (cats and dogs) in Chengdu, China, were detected for CoVs using a universal assay based on Rdrp genes combined with one-generation sequencing. Four kinds of CoVs, including feline CoV (FCoV), canine CoV (CCoV), CRCoV, and SARS-CoV-2 (the first reported case of SARS-CoV-2 in a dog in mainland China, co

Development of a recombinase polymerase amplification fluorescence assay to detect feline coronavirus.

Feline coronavirus (FCoV) is classified into two pathotypes: the avirulent feline enteric coronavirus (FECV), and the virulent feline infectious peritonitis virus (FIPV). Rapid pathogen detection, which is efficient and convenient, is the best approach for early confirmatory diagnosis. In this study, we first developed and evaluated a rapid recombinase polymerase amplification (RPA) detection method for FCoV that can detect FCoV within 15 min at 39 °C. The detection limit of that assay was 233 copies/μL DNA molecules per reaction. The specificity was high: it did not cross-react with canine di
