Decarboxylation of alpha-ketoisovaleric acid after oral administration in man.

The keto analogues of essential amino acids represent a promising therapeutic modality in hereditary and acquired disorders of nitrogen metabolism. The utilization of these substances in humans has been assayed primarily by nitrogen balance studies. A simple and accurate breath excretion test for 14CO2 enabled us to measure the decarboxylation of 1-14C-alpha-ketoisovaleric acid (KIV, the keto analogue of valine) in two normal and six diseased subjects. Normal volunteers as well as patients with gastrectomy, hepatic failure, renal failure, and myotonic dystrophy were tested in 5-g protein diets

Electrochemical N‑Propargylation of N‑Heterocycles via Decarboxylation of Allenoic Acids.

Broadly applicable and selective C-N bond formation remains a cornerstone challenge in organic synthesis. Although electrochemical methods have recently emerged as an efficient way for achieving decarboxylative C-N coupling, transforming allenoic acids into N-functionalized products remains elusive. Herein, we report the development of an electrochemical strategy for the decarboxylative N-propargylation of N-heterocycles using readily available 2,3-allenoic acids. By correlating the oxidation potentials of allenoic acids and nucleophilic coupling partners, we derived predictive criteria for an

Transient adducts of CO2 with nucleophiles enhance rates of enzymic decarboxylation.

Benzoylformate decarboxylase (BFDC) forms an adduct of thiamin diphosphate (ThDP), and benzoylformate, following the mechanism proposed by Breslow for related enzymes. Structural studies by Hasson and coworkers revealed inactivation of the enzyme by reaction with benzoylphosphonate, a substrate analogue that produces a phosphate ester of the active site serine (Ser26). This suggests that the side chain of Ser26 can trap CO2 as a transient hemicarbonate that prevents the reverse reaction that captures CO2, accelerating the forward reaction toward the ultimate release of benzaldehyde. Through en

Amino acid decarboxylation preserves Salmonella fitness during phagocyte-derived oxidative stress.

UNLABELLED: Successful establishment of infection by non-typhoidal Salmonella depends upon its ability to resist the antimicrobial defenses of the host innate immune response. To withstand the membrane depolarization that potentiates the killing activity of reactive oxygen species (ROS) produced by the phagocyte NADPH oxidase, Salmonella employs metabolic adaptations that maintain intracellular pH homeostasis and membrane energetics. Here, we identify amino acid decarboxylation as a critical determinant of Salmonella virulence and resistance to the oxidative pressures within the host environme

Chloroform as a hydrogen atom donor in Barton reductive decarboxylation reactions.

The utility of chloroform as both a solvent and a hydrogen atom donor in Barton reductive decarboxylation of a range of carboxylic acids was recently demonstrated (Ko, E. J. et al. Org. Lett. 2011, 13, 1944). In the present work, a combination of electronic structure calculations, direct dynamics calculations, and experimental studies was carried out to investigate how chloroform acts as a hydrogen atom donor in Barton reductive decarboxylations and to determine the scope of this process. The results from this study show that hydrogen atom transfer from chloroform occurs directly under kinetic

Photochemical Iron-Catalyzed Decarboxylative S-Glycosylation.

Alkyl thioglycosides represent key structural motifs in pharmaceuticals and are highly valued for their role as carbohydrate mimics in drug design. Herein, we report the first protocol for the direct construction of alkyl thioglycosides via photoinduced iron-catalyzed decarboxylative coupling of glycosyl thiosulfonates with primary, secondary, and tertiary alkyl carboxylic acids. This novel strategy circumvents the intractable anomeric control challenge, enabling the synthesis of a diverse range of alkyl thioglycosides with complete stereoretention. Furthermore, they are applicable to the late

Synthesis and decarboxylative Wittig reaction of difluoromethylene phosphobetaine.

A key intermediate, difluoromethylene phosphobetaine, in the Wittig reaction of ClCF2CO2Na-Ph3P with aldehydes was synthesized and characterized, which confirmed the reaction mechanism. The decarboxylation of this stable intermediate was a convenient approach for Wittig difluroolefination. Its reactivity could be adjusted by the modification of the substituent on the phosphorus.

Dopa-decarboxylation in the striata of rats with unilateral substantia nigra lesions.

The source and site of the DOPA decarboxylation to dopamine in Parkinson's disease (PD) and animal models of PD are controversial. Since most of aromatic L-amino acid decarboxylase (AADC) are lost along with the degenerating dopaminergic neurons, we addressed the possibility that other decarboxylases or a novel protein that is structurally different from AADC decarboxylate L-DOPA in the denervated striatum. Immunotitration of the extracts from the denervated striatum with AADC antibody showed that all activity can be attributed to AADC-immunoreactive protein. We then investigated if there are

Pyridoxal 5'-Phosphate-Dependent Enzymatic Decarboxylative Annulation.

Pyridoxal 5'-phosphate (PLP)-dependent enzymes are among the most versatile biocatalysts, yet transformations involving the Cγ-nucleophilic vinylglycine quinonoid (VGQ) intermediate remain exceptionally rare. Understanding the untapped reactivity of VGQ could open new avenues for developing PLP-dependent biocatalysts. Here, we establish a biocatalytic platform that artificially accesses and exploits the reactivity of this high-energy intermediate. By reprogramming SphA, a PLP-dependent enzyme that natively catalyzes decarboxylative Claisen condensation, to generate VGQ in situ through facile d

Iron-Catalyzed Decarboxylative Sulfinylation of Alkyl Carboxylic Acids.

Sulfinamides, sulfonamides, and sulfonimidamides are valuable motifs in medicinal chemistry, yet methods to synthesize alkyl variants from simple, readily available feedstocks remain scarce. In this report, we detail the synthesis of these three distinct sulfur functional groups, using readily available and structurally diverse alkyl carboxylic acids as the starting materials. The method harnesses alkyl radical generation from carboxylic acids using commercial iron salts and visible light irradiation, in combination with commercial sulfinylamine reagents, to deliver alkyl sulfinamide products.

Cobalt-Catalyzed Decarboxylative Allylations: Development and Mechanistic Studies.

In recent years, there has been a concerted drive to develop methods that are greener and more sustainable. Being an earth-abundant transition metal, cobalt offers an attractive substitute for commonly employed precious metal catalysts, though reactions engaging cobalt are still less developed. Herein, we report a method to achieve the decarboxylative allylation of nitrophenyl alkanes, nitroalkanes, and ketones employing cobalt. The reaction allows for the formation of various substituted allylated products in moderate-excellent yields with a broad scope. Additionally, the synthetic potential

Visible-Light-Enabled Direct Decarboxylative N-Alkylation.

The development of efficient and selective C-N bond-forming reactions from abundant feedstock chemicals remains a central theme in organic chemistry owing to the key roles of amines in synthesis, drug discovery, and materials science. Herein, we present a dual catalytic system for the N-alkylation of diverse aromatic carbocyclic and heterocyclic amines directly with carboxylic acids, by-passing their preactivation as redox-active esters. The reaction, which is enabled by visible-light-driven, acridine-catalyzed decarboxylation, provides access to N-alkylated secondary and tertiary anilines and

Decarboxylative Cross-Coupling of Acyl Fluorides with Potassium Perfluorobenzoates.

We report the transition metal-free decarboxylative cross-coupling reactions of acyl fluorides with potassium perfluorobenzoates. Compared with traditional transition metal-catalyzed cross-couplings, this protocol presents an extremely environmentally benign pathway to afford unsymmetrical diaryl ketones. To install perfluorophenyl groups, this method highlights highly selective, inexpensive, and nontoxic conditions. The reaction system tolerates various functional groups in acyl fluorides. Remarkably, all of the starting materials can be prepared from abundant carboxylic acids and the reactio

UbiB proteins mediate an ATP-dependent decarboxylation step in bacterial ubiquinone biosynthesis.

Polyisoprenoid quinones such as ubiquinone (UQ) play an essential role in cellular physiology, acting as membrane-bound electron and proton carriers in respiratory chains and other biological processes across all domains of life. In Escherichia coli, the canonical UQ biosynthesis pathway is well characterized. It involves 12 proteins (UbiA-UbiK and UbiX), most of which catalyze one of the eight modifications of the aromatic ring derived from 4-hydroxybenzoic acid (4-HB), while others (UbiB, UbiJ, UbiK) act as accessory factors ensuring efficient UQ production. Following prenylation by UbiA and

Phosphatidylethanolamine homeostasis under conditions of impaired CDP-ethanolamine pathway or phosphatidylserine decarboxylation.

Phosphatidylethanolamine is the major inner-membrane lipid in the plasma and mitochondrial membranes. It is synthesized in the endoplasmic reticulum from ethanolamine and diacylglycerol (DAG) by the CDP-ethanolamine pathway and from phosphatidylserine by decarboxylation in the mitochondria. Recently, multiple genetic disorders that impact these pathways have been identified, including hereditary spastic paraplegia 81 and 82, Liberfarb syndrome, and a new type of childhood-onset neurodegeneration-CONATOC. Individuals with these diseases suffer from multisystem disorders mainly affecting neurona

Alanine umbilical uptake, disposal rate, and decarboxylation rate in the fetal lamb.

Fetal plasma alanine disposal rate (DR) and decarboxylation rate were measured at 132 +/- 1 days gestation in nine fetal lambs infused with L-[1-14C]alanine via a brachial vein. In five experiments, L-[1-13C]alanine was added to the infusate. Using L-[1-14C]alanine, we found mean DR to be 15.5 +/- 1.8 mumol.min-1 . kg-1. DR was significantly correlated to both arterial plasma alanine and whole blood lactate concentrations. Placental uptake of fetal plasma alanine accounted for 19 +/- 4% of DR. Fetal and placental production of CO2 from the first carbon of alanine were 61 +/- 2 and 16 +/- 2% of

Effect of copper salts on hydrothermal oxidative decarboxylation: a study of phenylacetic acid.

Decarboxylation of carboxylic acids is favored under hydrothermal conditions, and can be influenced by dissolved metals. Here, we use phenylacetic acid as a model compound to study its hydrothermal decarboxylation in the presence of copper(ii) salts but no O2. Our results showed a strong oxidizing role of copper in facilitating oxidative decarboxylation.

Decarboxylation without CO2: why bicarbonate forms directly as trichloroacetate is converted to chloroform.

Patterns in the observed catalysis of decarboxylation reactions required us to conclude that these reactions involve initial hydration of the carboxylate and subsequent loss of bicarbonate. This raises the important and general question of why CO2 is not formed directly. Reaction profiles for the direct decarboxylation of trichloroacetate were generated with DFT calculations and show no significant barrier to the recombination of the incipient trichloromethide and CO2. In contrast, cleavage of the C-C bond from the hydrated intermediate shows a substantial barrier to recombination that allows

Growth yield increase and ATP formation linked to succinate decarboxylation in Veillonella parvula.

Veillonella parvula strain 259 (= DSM 2007) was able to grow on a mineral salts medium supplemented with (per litre) 1 g yeast extract, 1 g Tween-80, and 3 mg putrescine.2 HCl, with 6 mM thioglycolate as reductant and lactate as growth substrate. Succinate did not serve as a growth substrate, but when added in conjunction with lactate, it was decarboxylated to propionate and resulted in a measurable increase in growth yield, corresponding to the formation of 2.4 g cell dry mass per mol succinate. A growth yield increase linked to succinate metabolism occurred only while lactate was also being
