Multidrug antibiotic resistance is an urgent public health concern. Multiple strategies have been suggested to alleviate this problem, including the use of antibiotic combinations and cyclic therapies. We examine how adaptation to (1) combinations of drugs affects resistance to individual drugs, and to (2) individual drugs alters responses to drug combinations. To evaluate this, we evolved multiple strains of drug resistant Staphylococcus epidermidis in the lab. We show that evolving resistance to four highly synergistic combinations does not result in cross-resistance to all of their componen

The cell envelope is essential for survival and adaptation of bacteria. Bacterial membrane proteins include the major porins that mediate the influx of nutrients and several classes of antimicrobial drugs. Consequently, membrane remodelling is closely linked to antimicrobial resistance (AMR). Knowledge of bacterial membrane protein biogenesis and turnover underpins our understanding of bacterial membrane remodelling and the consequences that this process have in the evolution of AMR phenotypes. At the population level, the evolution of phenotypes is a reversible process, and we can use these i

The causes of antibiotic resistance are often complex and it is difficult to identify strategies to prevent or delay its emergence. One strategy has been to use less active members of a drug class, so that when resistance develops the more active members will still prevail. This strategy may often fail because this resistance may form the basis of resistance to the whole class. Often, less active drugs are the first to be discovered and more active versions follow, so we have had no choice; however, increasingly less active drugs are available to deal with specific infections and this may have

Accelerating antibiotic development from natural products. Natural products with antimicrobial activity can act through multiple mechanisms to overcome antibiotic resistance. These include directly inhibiting bacterial growth, reversing resistance by blocking efflux pumps and disrupting biofilms, and strengthening host defenses. Together, these strategies help restore antibiotic effectiveness, overcome resistance, and support more sustainable approaches to treating infections. Image created by Dr Ankita Nag in BioRender. Multidrug resistant bacteria infections are associated with an increase in attributable mortality and morbidity in ICU patients. Unfortunately, an emerging resistance to novel antibiotics used in the therapy of gram negative and gram positive bacteria infections is often reported in literature. Old antibiotics have been reintroduced in clinical practice. In this review we report the efficacy and safety use of older antimicrobial agents in critically ill patients. Polymyxins are used for nosocomial infection caused by Pseudomonas aeruginosa and Acinetobacter baumannii resistant strains. Patient

Antibiotics, the full-stop of invasive bacteria, have been used in clinical setups from unthreatening fever to massive challenging therapies. Constant dependency on medication upsurges the evasion of microbes from antibiotics contemporarily along with ecological footprint. Thus, the infested pathogen became resilient to antibiotics, disguised as multidrug-resistant bacteria (MDR), pandrug-resistant bacteria (PDR), and extensively drug-resistant bacteria (XDR). The etymology of genetic modifications and horizontal gene transfer played an external influence on the arising resurgence. Also, intri

BACKGROUND: Antimicrobial resistance (AMR) must be predicted to combat antibiotic-resistant illnesses. Based on high-priority AMR genomes, it is possible to track resistance and focus treatment to stop global outbreaks. Large language models (LLMs) are essential for identifying Porhyromonas gingivalis multiresistant efflux genes to prevent resistance. Antibiotic resistance is a serious problem; however, by studying specific bacterial genomes, we can predict how resistance develops and find better kinds of treatment.
OBJECTIVES: This paper explores using advanced models to predict the sequence

Spherical heterojunction nanocomposite materials are utilized to treat wound infections caused by drug-resistant bacteria by generating reactive oxygen species (ROS) and multiple cations (multiple inorganic or organic ions with positive points). However, there is an ongoing debate on the relative contributions of ROS and multiple cations toward antibacterial activity. In this study, the CuFe2O4/Cu@PEI/Ag (ZPA) nanocomposites were synthesized for releasing abundant•O2-,•OH, Fe3+, Cu2+, Ag+, and polyethylenimine (PEI), and studied the contribution of the released ions to the bacteriostatic activ

Hydrogen sulfide (H2S) is fundamentally a gaseous signaling molecule in bacteria. Recent studies have reported that H2S is a key mediator of bacterial antibiotic tolerance, making it a promising strategic target for overcoming tolerance, eliminating persisters, and restoring antibiotic efficacy. We systematically review recent advances in H2S-mediated multidrug tolerance mechanisms, highlighting three core pathways: iron chelation for antioxidant defense, reactive sulfur species-mediated antibiotic inactivation and efflux pump activation, and metabolic dormancy induced by respiratory inhibitio

Over the past 10 years, microbial resistance has seriously threatened human life and health, and the treatment of multidrug-resistant bacteria remains a challenge for clinicians, pharmacists, and infectious disease physicians. Bacterial resistance is affected by a variety of factors, such as the environment, economy, and drug abuse. This review compared the differences in bacterial resistance rates between high- and low-altitude areas and explored the relevant mechanisms of bacterial resistance in the low-oxygen environment of plateaus, providing new clinical research ideas for curbing the occ

Antimicrobial use in food-producing animals such as pigs is a significant issue due to its association with antimicrobial resistance. Florfenicol is a broad-spectrum phenicol antibiotic used in swine for various indications; however, its effect on the swine microbiome and resistome is largely unknown. This study investigated these effects in piglets treated intramuscularly with florfenicol at 1 and 7 days of age. Fecal samples were collected from treated (n = 30) and untreated (n = 30) pigs at nine different time points up until 140 days of age, and the fecal metagenomes were sequenced. The fe

Diabetic foot wounds and infections pose a significant and evolving challenge in diabetes care. Diabetic wound healing has become a major global concern for a very long time. Continuous research has been conducted to increase the healing process in diabetic ulcers to the rate of amputation. Wound healing is prolonged in diabetic patients due to various conditions, such as high glucose levels, neuropathy, poor blood circulation, and prolonged inflammation around the limbs, which causes the healing to be delayed compared to normal patients. Understanding the complexity of chronic foot wounds and

A comprehensive analysis of the clinical antibacterial pipeline demonstrates that there is a limited range of strategies that are primarily focused on modified versions of widely used chemical classes. These modifications aim to circumvent class-specific resistance mechanisms and reduce resistance rates in certain multidrug-resistant pathogens. Owing to the great variation in resistance rates and mechanisms, the clinical success of current approaches varies substantially across different countries, regions, and economic and environmental conditions, which affects the global societal value of t

Resistance to antibiotics in target bacterial populations has long complicated antibacterial chemotherapy. First described in the early 1980s (1, 2), efflux mechanism of resistance, whereby the antibiotic is actively (i.e., in an energy-dependent fashion) pumped from the bacterial cell, are being described with increasing frequency in recent years. Initial examples of bacterial antibiotic efflux systems were agent-specific, providing for export of and resistance to single agents (e.g., tetracyclines, chloramphenicol, macrolides) (3). More recently, bacterial drug efflux systems of broad substr

BACKGROUND: Antimicrobial resistance (AMR) develops into a worldwide health emergency through genetic and biochemical adaptations which enable microorganisms to resist antimicrobial treatment. β-lactamases (blaNDM, blaKPC) and efflux pumps (MexAB-OprM) working with mobile genetic elements facilitate fast proliferation of multidrug-resistant (MDR) and exttreme drug-resistant (XDR) phenotypes thus creating major concerns for healthcare systems and community health as well as the agricultural sector.
OBJECTIVES: The review dissimilarly unifies molecular resistance pathways with public health imp
