Bioengineering of elastic cartilage with aggregated porcine and human auricular chondrocytes and hydrogels containing alginate, collagen, and kappa-elastin.

Transplantation of isolated chondrocytes has long been acknowledged as a potential method for rebuilding small defects in damaged or deformed cartilages. Recent advances in tissue engineering permit us to focus on production of larger amounts of cartilaginous tissue, such as might be needed for reconstructive surgery of the entire auricle. In this report we describe modification of the basic techniques that lead to production of a large amount of elastic cartilage originated from porcine and human isolated chondrocytes. Small fragments of auricular cartilage were harvested from children underg

Remaining microtia tissue as a source for 3D bioprinted elastic cartilage tissue constructs, potential use for surgical microtia reconstruction.

The absence of ears in children is a global problem. An implant made of costal cartilage is the standard procedure for ear reconstruction; however, side effects such as pneumothorax, loss of thoracic cage shape, and respiratory complications have been documented. Three-dimensional (3D) printing allows the generation of biocompatible scaffolds that mimic the shape, mechanical strength, and architecture of the native extracellular matrix necessary to promote new elastic cartilage formation. We report the potential use of a 3D-bioprinted poly-ε-caprolactone (3D-PCL) auricle-shaped framework seede

Unconventional auricular reconstruction using controlled scaffold buckling and chondrogenic cocktail containing muscle-derived cells.

Tissue engineered auricles face challenges such as subpar graft mechanical robustness, insufficient stem/progenitor cells, and unidentified factors that specifically promote elastic cartilage. To tackle these, we developed resilient and bioactive 3D-printed scaffolds using two unconventional concepts, including controlled buckling compliant lattices for mechanical resilience and muscle-derived stem/progenitor cells (MDSCs) with defined biochemical factors for facile elastic cartilage-like regeneration. Our functionally graded design reduced stress by 63.1% relative to other graded designs, whi

Tissue engineering by cocultivating human elastic chondrocytes and keratinocytes.

To date, there is no optimal way to reconstruct an external ear in cases of microtia or after trauma or burns damaging the external ear. However, success in the area of tissue engineering has indicated that autologous elastic cartilage produced in vitro might be of great importance in the future treatment of these patients. In the present study we have engineered human, elastic cartilage in vitro by culturing chondrocytes in fibrin glue. Furthermore, the engineered elastic cartilage was seeded with human keratinocytes to investigate the possibility of combining these two tissues into one integ

Comparison of the compressive mechanical properties of auricular and costal cartilage from patients with microtia.

Children born with a small or absent ears undergo surgical reconstruction to restore their auricle. Currently, rib (costal) cartilage is used to carve the auricle. However as alternative, tissue engineered and synthetic materials are being developed to restore the auricle shape to overcome donor site morbidity and limited availability of rib cartilage. However, to date there is limited knowledge regarding the mechanical properties of the auricular and costal cartilage to optimise the required compressive properties of the graft. The remnant auricular and costal cartilage from 20 patients under

The short-term compressive properties of adult human articular cartilage.

An apparatus and test method were developed to determine the elastic stiffness and damping coefficient of human cartilage in compression. An underdamped, counterbalanced beam applied a sudden compressive force to a full-thickness cylindrical specimen of articular cartilage. The initial oscillatory response decayed to steady state creep after approximately 10 cycles of oscillation. The results were consistent with a Voigt phenomenological model with linear stiffness and damping terms. Standard dynamics analysis of the transient oscillatory response enabled the elastic stiffness to be determined

A pilot study comparing mechanical properties of tissue-engineered cartilages and various endogenous cartilages.

BACKGROUND: Mechanical properties of tissue-engineered cartilage and a variety of endogenous cartilage were measured. The main goal was to evaluate if the tissue-engineered cartilage have similar mechanical characteristics to be replaced with rib cartilage in microtia reconstruction. Such study lays the foundation for future human clinical trials for microtia reconstruction. METHOD: Atomic force microscopy and compression testing were used to measure the viscoelasticity of tissue-engineered cartilage (stem cell seeded on Poly lactic co-glycolytic acid nanofibers and Pellet) and endogenous car

Relapsing polychondritis. Electron microscopic study of ear cartilage.

Light microscopic and for the first time electron microscopic (EM) studies of involved elastic ear cartilage in a patient with relapsing polychondritis confirm the superficial location of the inflammatory reaction. EM studies of deep cartilage were identical to those on controls except for a rare necrotic cell. Superficial cartilage showed a finely granular electron-dense material. This material, which may be protein, possibly of enzyme or immunoglobulin origin, also surrounded some elastic fibrils. Findings of predominantly superficial changes and the multisystem disease in the present patien

Cell-engineered human elastic chondrocytes regenerate natural scaffold in vitro and neocartilage with neoperichondrium in the human body post-transplantation.

We have developed a unique method that allows us to culture large volumes of chondrocyte expansion from a small piece of human elastic cartilage. The characteristic features of our culturing method are that fibroblast growth factor-2 (FGF2), which promotes proliferation of elastic chondrocytes, is added to a culture medium, and that cell-engineering techniques are adopted in the multilayered culture system that we have developed. We have subsequently discovered that once multilayered chondrocytes are transplanted into a human body, differentiation induction that makes use of surrounding tissue

An analytical model to study blunt impact response of the rabbit P-F joint.

While mechanisms of post-traumatic osteoarthrosis are largely unknown, excessive stresses and strains generated in articular cartilage and the underlying bone may play a role. In this manuscript a technique is described for studying the impact response of a diarthrodial joint. A mathematical model of the rabbit PF joint indicated that contact pressures predicted by a quasi-static plane strain linear elastic model compared well with experimental data when Poisson's ratio and Young's modulus of the cartilage were 0.49 and 2 MPa, respectively. This value for the elastic modulus compared well with

Characterization of pediatric microtia cartilage: a reservoir of chondrocytes for auricular reconstruction using tissue engineering strategies.

The external ear is composed of elastic cartilage. Microtia is a congenital malformation of the external ear that involves a small reduction in size or a complete absence. The aim of tissue engineering is to regenerate tissues and organs clinically implantable based on the utilization of cells and biomaterials. Remnants from microtia represent a source of cells for auricular reconstruction using tissue engineering. To examine the macromolecular architecture of microtia cartilage and behavior of chondrocytes, in order to enrich the knowledge of this type of cartilage as a cell reservoir. Auricu

[Relapsing polychondritis and cartilage].

Atrophic polychondritis (APC) results from elective lesions of the components of elastic cartilage. Clinical criteria have been established in terms of the incidence of auricular, nasal, ocular, tracheobronchial or cochlear involvement. However, recent clinical and pathological reviews have shown the frequency and severity of vascular lesions, aortic as well as peripheral. Several pathological mechanisms probably participate together in the destruction of elastic cartilage. A direct enzyme abnormality of glycoaminoglycanes and of elastic tissue are believed to be interlinked with an auto-immun

Effects of ageing on the insertion zones of the human vocal fold.

The vocal ligaments insert at the anterior and posterior commissures of the larynx. These structures fulfil biomechanical functions, balancing the different elastic moduli of tendon, cartilage or bone and undergo age-related changes that may be responsible for voice changes with increasing age. The aim of this study was to analyse the insertion structures of the vocal ligaments by means of macroscopic, histological, immunohistochemical and electron-microscopic methods and to draw conclusions from age-related structural changes on a functional basis. Investigations were carried out on the laryn

A viscoelastic constitutive model can accurately represent entire creep indentation tests of human patella cartilage.

Cartilage material properties provide important insights into joint health, and cartilage material models are used in whole-joint finite element models. Although the biphasic model representing experimental creep indentation tests is commonly used to characterize cartilage, cartilage short-term response to loading is generally not characterized using the biphasic model. The purpose of this study was to determine the short-term and equilibrium material properties of human patella cartilage using a viscoelastic model representation of creep indentation tests. We performed 24 experimental creep i

Age-related distribution of elastic fibers in the rabbit knee.

The distribution and histochemical properties of elastic fibers in the knee of newborn, young, adult, and old New Zealand White rabbits were studied. In the growing rabbit, elastic fibers, especially oxytalan fibers, are abundant in the perichondrium and the fibrous layer of the periosteum. They are homogeneously distributed in tendons, ligaments, and menisci, and are absent in the epiphyseal cartilage and growth plate and at the enthesis. In mature rabbits, the articular capsule, the synovial membrane, the peritenon, the periosteum, the perimysium, and the perivascular connective tissue are r

Biomechanical characterisation of the human nasal cartilages; implications for tissue engineering.

Nasal reconstruction is currently performed using autologous grafts provides but is limited by donor site morbidity, tissue availability and potentially graft failure. Additionally, current alternative alloplastic materials are limited by their high extrusion and infection rates. Matching mechanical properties of synthetic materials to the native tissue they are replacing has shown to be important in the biocompatibility of implants. To date the mechanical properties of the human nasal cartilages has not been studied in depth to be able to create tissue-engineered replacements with similar mec

Comparison of Gene Expression Patterns in Articular Cartilage and Xiphoid Cartilage.

Cartilage is a resilient and smooth connective tissue that is found throughout the body. Among the three major types of cartilage, namely hyaline cartilage, elastic cartilage, and fibrocartilage, hyaline cartilage is the most widespread type of cartilage predominantly located in the joint surfaces (articular cartilage, AC). It remains a huge challenge for orthopedic surgeons to deal with AC damage since it has limited capacity for self-repair. Xiphoid cartilage (XC) is a vestigial cartilage located in the distal end of the sternum. XC-derived chondrocytes exhibit strong chondrogenic differenti

Third eyelid in the small Indian mongoose (Herpestes javanicus): a morphological and histological study.

The Indian mongoose (Herpestes javanicus) is native to parts of Asia, Iran. The purpose of this study was to describe the gross anatomy of the cartilage and histology of the superficial gland of the third eyelid of two adult mongooses. The animals, in terminal stages of disease and near death due to aging or unknown reasons, were referred from Park Zoo (Shiraz, Iran) to our center. By using a modified maceration technique, the morphological characteristics of the cartilage were examined. For histological examinations of the superficial gland of the third eyelid, the samples were stained with h

Biochemical properties of tissue-engineered cartilage.

OBJECTIVE: Microtia is treated with rib cartilage sculpting and staged procedures; though aesthetically pleasing, these constructs lack native ear flexibility. Tissue-engineered (TE) elastic cartilage may bridge this gap; however, TE cartilage implants lead to hypertrophic changes with calcification and loss of flexibility. Retaining flexibility in TE cartilage must focus on increased elastin, maintained collagen II, decreased collagen X, with prevention of calcification. This study compares biochemical properties of human cartilage to TE cartilage from umbilical cord mesenchymal stem cells (U
