Demonstration of two distinct tachykinin receptors in rat brain cortex.

Eledoisin and substance P are members of a class of peptides termed tachykinins. They share a similar spectrum of biological activities but their relative potencies in various pharmacological assays differ. We have investigated whether there is more than one receptor for these tachykinins in rat brain cortex membranes. 125I-Bolton Hunter-conjugated eledoisin specifically binds to rat brain cortex membranes with high affinity. The binding is inhibited over 95% by unlabeled eledoisin (6.6 microM). Scatchard analysis of the binding of this ligand is curvilinear suggesting that there are two bindi

Isolation and characterization of neurokinin A, neurokinin A(3-10) and neurokinin A(4-10) from a neutral water extract of a metastatic ileal carcinoid tumour.

A metastasis to the right liver lobe of an argyrophil/argentaffin midgut carcinoid tumour in a patient with the classical carcinoid syndrome was examined for the presence of tachykinins other than substance P, using a specific antiserum. The extract was initially purified using SepPak cartridges, and subsequently subjected to cation-exchange chromatography on SP Sephadex C-25 which separated the immunoreactive material into two main components (components I and II). Both were further purified by anion-exchange chromatography on DEAE-Sephadex A-25, and by reverse-phase fast protein liquid chrom

Pharmacological characterisation of two tachykinin binding sites in the rat cerebral cortex.

The pharmacological properties of two types of tachykinin receptor were characterised on rat cortical synaptosomes using 125I-Bolton Hunter substance P (125I-BHSP) or with 125I-Bolton Hunter eledoisin (125I-BHE). Shorter SP C-terminal fragments, such as SP (6-11) or (pGlu)-SP (6-11), were more potent than SP itself or longer SP C-terminal fragments in competing for 125I-BHE binding; their efficacy was comparable to that of eledoisin. In contrast, longer SP C-terminal fragments exhibited a higher affinity than shorter ones for the 125I-BHSP binding sites as previously reported. SP N-terminal fr

The mechanism of peptidergic miosis. I. The structural basis of miotic potency among biologically active polypeptides.

The miotic potency of eleven naturally occurring polypeptides and poly-DL-alanine was tested on the isolated anterior segment of hooded rat and golden hamster eyes in order to gain information on the chemical nature of the peptides or classes of peptides which may be regarded as possible mediators of the miotic response of the mammalian eye to chemical irritants or mechanical or surgical trauma. Substance P, P-octapeptide, physalaemin and eledoisin, and eledoisin related peptide were found to be potent miotics on these preparations, yielding ED50 values in the range of 8 to 95 nM and complete

Tachykinins: effects on gastric secretion and emptying in rats.

To test the effects on gastric acid secretion and gastric emptying of non-mammalian (eledoisin, physalaemin, kassinin), mammalian (substance P. neurokinin A, neurokinin B) and synthetic tachykinins (septide, senktide), intracerebroventricular injections of these peptides were given to conscious pylorus-ligated rats (gastric secretory study) or to animals fed with a phenol red meal (gastric emptying study). The tachykinins able to cause central inhibition of acid output were those active on NK2 and NK3 receptors (eledoisin, kassinin, neurokinin A, neurokinin B and senktide). Tachykinins active

Comparison of the responses to the sensory neuropeptides, substance P, neurokinin A, neurokinin B and calcitonin gene-related peptide and to trigeminal nerve stimulation in the iris sphincter muscle of the rabbit.

Three mammalian tachykinins (substance P, neurokinin A and B) and two non-mammalian ones (eledoisin and physalaemin) produced potent contractions of the isolated rabbit iris sphincter muscle. The rank order of potencies was eledoisin greater than neurokinin B = physalaemin greater than substance P greater than neurokinin A. The maximum efficacy was much the same. The contractile responses to neurokinin A and eledoisin developed more rapidly than did those to the other tachykinins used and were selectively attenuated by [D-Arg1, D-Pro2, D-Trp7,9, Leu11]-SP. Tachykinin binding sites in the interpeduncular nucleus of the rat: normal distribution, postnatal development and the effects of lesions.

Tachykinin binding sites in the basal midbrain were labeled in adult and neonatal rats using 125I-Bolton Hunter (BH) substance P (SP) and 125I-BH eledoisin as ligands. In the adult, binding was very low in the tegmentum and raphe adjacent to the interpeduncular nucleus (IPN). Within the IPN, no binding with either ligand was seen in the target subnuclei of the habenular SP and substance K projections, the lateral subnuclei and the cap of the rostral subnucleus. Labeling with 125I-BH-SP was very light and was restricted primarily to the central subnucleus of the IPN while 125I-BH-eledoisin labe

Characterization and autoradiographic localization of multiple tachykinin binding sites in gastrointestinal tract and bladder.

Binding sites for the [125I]Bolton-Hunter-labeled tachykinins substance K (BHSK), eledoisin (BHE) and substance P (BHSP) were investigated using crude membrane suspensions and autoradiography. In smooth muscle membranes from guinea-pig small intestine and rat duodenum, specific binding of BHSK was saturable and reversible, showing a single class of sites with a KD of 1 to 3 nM and maximum number of specific binding sites of 1 to 2 fmol/mg of wet weight tissue. Pharmacological characterization of this binding revealed a novel receptor site (K) with affinity for substance K greater than kassinin

Characterization of receptors for two Xenopus gastrointestinal tachykinin peptides in their species of origin.

Two tachykinin peptides, bufokinin and Xenopus neurokinin A (X-NKA) were recently isolated from Xenopus laevis. In this study we investigated the tachykinin receptors in the Xenopus gastrointestinal tract. In functional studies using stomach circular muscle strips, all peptides had similar potencies (EC50 values 1-7 nM). The rank order of potency to contract the intestine was physalaemin (EC50 1 nM)> or =bufokinin (EC50 3 nM)>substance P (SP)> or =cod SP>NKA>>X-NKA (EC50 1,900 nM). Stimulation of rabbit lacrimal gland secretion with biologically active peptides.

To determine whether biologically active peptides can stimulate lacrimal gland secretion, we measured fluid and protein secretion from the cannulated lacrimal gland excretory duct of anesthetized rabbits after arterial injection of various peptides. Vasoactive intestinal peptide (VIP, 0.003-3 nmol) and porcine histidine isoleucine-containing peptide (PHI, 0.01-3 nmol) stimulated fluid and protein secretion in a dose-dependent manner. Effect of somatostatin on basal and stimulated gastric secretion in the cod, Gadus morhua.

In vivo secretion of gastric acid and pepsin has been studied in pylorus-ligated cod. Basal acid output amounted to 100-150 mumol H+.kg-1.h-1 and pepsin secretion to 1 mg.kg-1.h-1. In response to bombesin nonapeptide (2.4 nmol.kg-1.h-1) and histamine (81 nmol.kg-1.h-1), acid secretion increased to approximately 200 and 600% of the basal level, respectively. Pepsin output was marginally affected by histamine but increased to approximately 3 and 15 times the basal level during treatment with bombesin and eledoisin (3.27 nmol.kg-1.h-1). Somatostatin (SS-14, 15 nmol.kg-1.h-1) inhibited basal acid

Undeca- and octa-peptide antagonists for substance P, a study on the guinea pig trachea.

Undeca - and octa-peptide analogues of substance P (SP), acting as antagonists in the guinea pig ileum, have been tested on the guinea pig trachea. The antagonistic properties of several octapeptides, particularly of [pro4, trp7 ,9,10,Phe11]SP-(4-11) have been confirmed, while the undecapeptides have been found to be potent stimulants of the trachea. The contractions of the guinea pig trachea in response several undecapeptides , particularly [pro2, trp7 ,9, Leu11 ]SP, undergo rapid tachyphylaxis, are significantly reduced in the presence of diphenhydramine and are not influenced by octapeptide

Effects of tachykinins on the secretion of fluid and glycoproteins from the submandibular glands of rat, mouse, hamster and guinea pig.

The effects of substance P, neurokinin A, physalamine, and eledoisin on the secretion of fluid and glycoproteins from the submandibular glands of various rodents were investigated. Following i.v. injection of each peptide at a dose of 20 micrograms/kg, the major glycoprotein species secreted from rats and guinea pigs were shown to be electrophoretically identical with those found in the acini. However, saliva was not elicited from the mice and hamsters. These results suggest that in both rats and guinea pigs, tachykinins act on the acinar cells of the submandibular gland only.

Densities of BHSP binding sites were low in fish brain, increased in lower vertebrates, were high in birds and rodents, and relatively constant in cat, monkey and human. In contrast, BHELE binding site densities were moderate in fish brain and high in frog, snake, chick, pigeon, mouse and rat brain. Low and very low densities were localized in guinea pig and cat, while no significant BHELE spec

[Special indications for the use of soft contact lenses as a drug-release-system (author's transl)].

Research has been performed, both experimentally and clinically, to establish the value of the association of soft contact lenses and some types of eye drops. The use of soft contact lenses with eye drops may be useful in some special cases: a) more prolonged and more sustained effect compared with the usual way of administration of eye drops (especially antiglaucomatous substances, antimetabolites, mydriatics); b) possibility of reducing the concentration to avoid local discomfort or systemic side-effects, without loss of their effectiveness on the eye conditions to be treated. The action of ranatensin, a new polypeptide from amphibian skin, on the blood pressure of experimental animals.

1. The blood pressure response to ranatensin, an undecapeptide from the skin of the frog, Rana pipiens, has been studied in various experimental animals.2. Ranatensin raised blood pressure in the dog and rabbit. The response was not altered by atropine, phentolamine, propranolol or hexamethonium, suggesting a direct peripheral vasoconstrictor action. In both animals ranatensin was about one-tenth as potent as angiotensin. Tachyphylaxis to ranatensin did occur, but there was no cross-tachyphylaxis with angiotensin, bradykinin, or noradrenaline.3. The peptide lowered blood pressure in the monkey

Characterization of the substance P receptor in rat brain cortex membranes and the inhibition of radioligand binding by guanine nucleotides.

Rat brain cortex membranes bind to a conjugate of substance P and 125I-labeled Bolton-Hunter reagent, and this binding can be inhibited by a low concentration of substance P (Kd = 1.2 +/- 0.4 X 10(-8) M). This binding is reversible and saturable (0.5 +/- 0.1 pmol of binding sites/mg of protein). Fragments of substance P as small as the carboxyl-terminal hexapeptide can inhibit the binding although their potency decreases with the decrease in the length of the peptides. The binding affinities of smaller peptides or peptides in which the carboxyl-terminal amide or amino acids are removed are dra

Influence of secretagogues on volume and protein pattern in rabbit lacrimal fluid.

After certain tear proteins had been found to be markers of duct cell activity in the rabbit lacrimal gland, different secretagogues were tested to increase volume and protein secretion. Carbamyl-choline and the peptides eledoisin and vasoactive intestinal peptide (VIP) stimulated volume flux as well as secretion of the marker proteins indicating duct cell activation. Beta-adrenergic stimulation also increased secretion of a further tear protein. This might indicate that water is secreted by the duct cells. These are stimulated by the secretagogues mentioned above. Beta-adrenergic stimulation

Somatostatin and calcitonin stimulate neurite regeneration of molluscan neurons in vitro.

Isolated neurons from the gastropod Physella heterostropha fail to regenerate their neurites if cultured in defined medium in the absence of growth factors. A significant number of cultured neurons extend neurites if they are cultured in brain-conditioned or hemocyte-conditioned medium. Synthetic somatostatin and salmon calcitonin also stimulate neurite regeneration in a dose-dependent manner. Thyrotropin-releasing hormone, arginine vasotocin, and eledoisin fail to promote neurite outgrowth. Histamine in carcinoid syndrome.

The exact etiology of carcinoid flushing remains unknown, but the symptoms are probably mediated through release of one or several humoral substances. Flushing seen in fore-gut carcinoids (gastric carcinoids) has been ascribed to excessive histamine release, whereas flushing seen in mid-gut carcinoids (ileal carcinoids) tentatively has been ascribed to excessive release of serotonin, bradykinin, substance P, substance K or eledoisin. In this study plasma histamine was measured in 8 patients with mid-gut carcinoids and carcinoid syndrome using an enzymatic isotopic method in order to evaluate h
