Clinical and biological impact of the exposome on the skin.

The skin exposome is defined as the totality of environmental exposures over the life course that can induce or modify various skin conditions. Here, we review the impact on the skin of solar exposure, air pollution, hormones, nutrition and psychological factors. Photoageing, photocarcinogenesis and pigmentary changes are well-established consequences of chronic exposure of the skin to solar radiation. Exposure to traffic-related air pollution contributes to skin ageing. Particulate matter and nitrogen dioxide cause skin pigmentation/lentigines, while ozone causes wrinkles and has an impact on

Influence of the Urban Exposome on Birth Weight.

BACKGROUND: The exposome is defined as the totality of environmental exposures from conception onwards. It calls for providing a holistic view of environmental exposures and their effects on human health by evaluating multiple environmental exposures simultaneously during critical periods of life. OBJECTIVE: We evaluated the association of the urban exposome with birth weight. METHODS: We estimated exposure to the urban exposome, including the built environment, air pollution, road traffic noise, meteorology, natural space, and road traffic (corresponding to 24 environmental indicators and 6

Modeling the Exposome in Murine Cardiovascular Disease.

Our exposome, encompassing our environmental and lifestyle exposures across the life span, is a critical determinant of cardiovascular health. Clinical and epidemiological data demonstrate that sleep, stress, exercise, smoking, viral respiratory infections, and environmental pollution each modulate cardiovascular disease (CVD) risk, yet the pathobiological mechanisms underlying these associations remain poorly understood. Fundamental research into how the exposome and the physiological responses to it shape cardiovascular pathophysiology holds significant promise for expanding biologic underst

Exposome Profiles and Asthma among French Adults.

Rationale: Although previous studies in environmental epidemiology focused on single or a few exposures, a holistic approach combining multiple preventable risk factors is needed to tackle the etiology of multifactorial diseases such as asthma. Objectives: To investigate the association between combined socioeconomic, external environment, early-life environment, and lifestyle-anthropometric factors and asthma phenotypes. Methods: A total of 20,833 adults from the French NutriNet-Santé cohort were included (mean age, 56.2 yr; SD, 13.2; 72% women). The validated asthma symptom score (continuous

Exposome involvement in the development of acne vulgaris.

Acne vulgaris is one of the most prevalent chronic inflammatory skin diseases worldwide, characterized by marked clinical heterogeneity, fluctuating disease course, and strong sensitivity to environmental and lifestyle factors. The exposome, encompassing lifelong environmental, lifestyle, psychosocial, microbial, and intrinsic exposures, offers an integrative framework for re-conceptualizing acne as an environmentally modulated inflammatory disease. This review synthesizes external and internal exposomal drivers of acne, including pollution, radiation, climate and occupational factors, diet, s

Climate change, the exposome, and early-onset carcinogenesis.

The incidence of early-onset cancers (diagnosis before 50 years of age) has been steadily increasing globally, presenting a significant public health challenge. While genetic predispositions play a role, most cancers cannot be traced to heritable components. Furthermore, the rapid rise in the incidence rates of these cancers strongly suggests the influence of environmental factors that constitute the "exposome." This perspective article integrates recent findings linking climate change and broader planetary health challenges to pervasive inflammation in the development of chronic diseases, esp

The exposome of brain aging across 34 countries.

The physical and social exposome affects human aging, and brain clocks may track its effects. However, most studies neglect multidomain exposures (physical, social and political) across diverse settings globally and their associations with brain aging. In this study, we characterized the associations between 73 country-level physical and social exposomal factors and multimodal brain age in 18,701 participants from 34 countries (healthy individuals and those with Alzheimer's disease, frontotemporal lobar degeneration or mild cognitive impairment). Exposome effects were assessed using generalize

Spaceflight exposome/microgravity effects on the psychoimmunoneuroendocrine system.

Spaceflight missions represent a new exposome, unpredicted by evolution. Life-threatening agents activate non-specific stress responses involving the psycho-immune-neuroendocrine network. The main molecular pathways regulating this network and those affected by spaceflight exposome/microgravity are reported in various experimental setups. They include gut microbiota-derived metabolites, potentially valuable for countermeasure development. Meaningfully, the application of neuromodulation techniques, like the non-invasive transcutaneous vagal stimulation, for the prevention of pathophysiological

Exposome on skeletal muscle system: a mini-review.

Exposomics is an ever-expanding field which captures the cumulative exposures to chemical, biological, physical, lifestyle, and social factors associated with biological responses. Since skeletal muscle is currently considered as the largest secretory organ and shows substantial plasticity over the life course, this reviews addresses the topic of exposome and skeletal muscle by reviewing the state-of-the-art evidence and the most intriguing perspectives. Muscle stem cells react to stressors via phosphorylated eukaryotic initiation factor 2α and tuberous sclerosis 1, and are sensible to hormeti

Reconstruction of the radiological component of the exposome in the CONSTANCES cohort.

As an innovative approach to contribute to the study of the health effects of ionizing radiation (IR) exposure, we developed a method to characterize the radiological component of the exposome as exhaustively as possible. We estimated annual equivalent IR doses in millisieverts (mSv) received by six target organs (brain, breast, lung, colon, prostate and thyroid) from the largest possible set of sources, for 21,720 adult participants in the French general population cohort CONSTANCES from birth (age 0 year) until the age they reached in 2020. We considered the following environmental sources o

Proteomics/diagnosis of chorioamnionitis and of relationships with the fetal exposome.

Proteomics, a relatively young science, originally emerged as a complement to genomics research. By definition, the goal of proteomics is to provide a snapshot of all the proteins within an organism, tissue or biological sample at a given moment. Proteomics has the ability to single out one or more proteins (biomarkers) that change consistently in affected subjects as compared to those disease-free. From a proteomics perspective, chorioamnionitis poses both challenges and opportunities. Challenges relate to the dynamic course of the inflammatory process, and compartmentalization of the gestati

Modular, Scalable, and Customizable LC-HRMS for Exposomics.

In this chapter, we describe a multi-purpose, reversed-phase liquid chromatography-high-resolution mass spectrometry (LC-HRMS) workflow for acquiring high-quality, non-targeted exposomics data utilizing data-dependent acquisition (DDA) combined with the use of toxicant inclusion lists for semi-targeted analysis. In addition, we describe expected retention times for >160 highly diverse xenobiotics in human plasma and serum samples. The method described is intended to serve as a generic LC-HRMS exposomics workflow for research and educational purposes. Moreover, it may be employed as a primer, a

Exposome Risk Factors for Vitiligo: A Systematic Evidence Review.

BACKGROUND: Vitiligo is a multi-factorial autoimmune skin disorder often triggered by environmental exposures. Although the exposome has gained attention, no systematic review has fully assessed its role in vitiligo. OBJECTIVE: We aimed to evaluate evidence linking exposomal factors to vitiligo onset and progression, focusing on quantifiable associations and study quality. METHODS: A systematic search of PubMed and Embase (inception to 25 August, 2024) followed PRISMA (Preferred Reporting Items for Systematic reviews and Meta-Analyses) 2020 guidelines and was registered in PROSPERO (CRD42024

Associations between the chemical exposome and pregnancy induced hypertension.

Exposure to environmental chemicals has been linked to an increased risk of pregnancy-induced hypertension (PIH). This prospective cohort study examined the associations between PIH and maternal chemical exposure to four classes of chemicals (i.e., phthalates, bisphenols, perfluoroalkyl acids, non-essential metals and trace minerals). Participants included 420 pregnant women from the Alberta Pregnancy Outcomes and Nutrition (APrON) cohort who had data available on diagnosed PIH and environmental chemical exposure. Twelve phthalate metabolites, two bisphenols, eight perfluoroalkyl acids and ele

Multi-omics signatures of the human early life exposome.

Environmental exposures during early life play a critical role in life-course health, yet the molecular phenotypes underlying environmental effects on health are poorly understood. In the Human Early Life Exposome (HELIX) project, a multi-centre cohort of 1301 mother-child pairs, we associate individual exposomes consisting of >100 chemical, outdoor, social and lifestyle exposures assessed in pregnancy and childhood, with multi-omics profiles (methylome, transcriptome, proteins and metabolites) in childhood. We identify 1170 associations, 249 in pregnancy and 921 in childhood, which reveal pot

Toxicants, Exposome, and Hantavirus Disease: A One Health Perspective.

Although hantaviruses have traditionally been considered geographically restricted rodent-borne pathogens, globalization, climate change, ecosystem disruption, and environmental contamination may collectively favor novel transmission scenarios and altered epidemiological patterns. The experience gained during the SARS-CoV-2 pandemic showed the importance of environmental determinants, airborne exposure, and host susceptibility factors in emerging viral diseases. In this context, increasing but still indirect evidence suggests that environmental toxicants and the exposome may modulate susceptib

Exposome and Skin. Part 2. The Influential Role of the Exposome, Beyond UVR, in Actinic Keratosis, Bowen's Disease and Squamous Cell Carcinoma: A Proposal.

Actinic keratosis (AK) is the main risk factor for the development of cutaneous invasive squamous cell carcinoma (SCC). It represents the first sign of severe chronic ultraviolet radiation exposure, which has a clear significant effect. Nevertheless, the skin is exposed to many other exposome factors which should be thoroughly considered. Our aim was to assess the impact of exposome factors other than ultraviolet radiation (UVR) on the etiopathology of AK and Bowen's disease (BD) and progression of AK to SCC and to design tailored prevention strategies. We performed an exhaustive literature se

The diabetes exposome: interplay of environmental and genetic determinants in diabetes.

Diabetes mellitus arises from complex interactions between biological susceptibility and diverse environmental influences that extend beyond traditional "genes versus lifestyle" models. The exposome is defined as the aggregate of non-genetic environmental exposures and their biological consequences across the life course. It provides a systems-level framework to evaluate this interaction, spanning chemical, physical, social, and behavioral domains while integrating them with internal molecular responses to decode how environments shape metabolic health. Emerging assessment strategies, includin

[The cardiovascular exposome: areas of action for prevention and practical cardiology].

Environmental risk factors are recognized as essential determinants of the cardiovascular disease burden but are still poorly represented in clinical guidelines. External environmental exposures such as air pollution, traffic noise, heat, light pollution, chemical contamination of soil and water as well as urban planning, our social environment and lifestyle contribute significantly to the global disease burden and premature deaths. In particular, cardiovascular morbidity and mortality are significantly influenced by these environmental stressors. The exposome concept provides an overarching f
