[Snake venom Kunitz/BPTI family: Structure, classification and pharmacological potential].

Snake venoms are rich sources of serine proteinase inhibitors that are members of the KunitzBPTI (bovine pancreatic trypsin inhibitor) family. Generally, these inhibitors are formed by 60 amino acids approximately. Their folding is characterised by a canonical loop that binds in a complementary manner to the active site of serine protease. Some variants from snake venoms show only weak inhibitory activity against proteases while others are neurotoxic. Moreover, proteases inhibitors are involved in various physiological prdcesses, such as blood coagulation, fibrinolysis, and inflammation. Also,

Structure and regulation of the Asr gene family in banana.

Abscisic acid, stress, ripening proteins (ASR) are a family of plant-specific small hydrophilic proteins. Studies in various plant species have highlighted their role in increased resistance to abiotic stress, including drought, but their specific function remains unknown. As a first step toward their potential use in crop improvement, we investigated the structure and regulation of the Asr gene family in Musa species (bananas and plantains). We determined that the Musa Asr gene family contained at least four members, all of which exhibited the typical two exons, one intron structure of Asr ge

Residual structure, backbone dynamics, and interactions within the synuclein family.

The human synuclein protein family includes alpha-synuclein, which has been linked to both familial and sporadic Parkinson's disease, and the highly homologous beta and gamma-synuclein. Mutations in alpha-synuclein cause autosomal dominant early onset Parkinson's, and the protein is found deposited in a fibrillar form in hereditary and idiopathic forms of the disease. No genetic link between beta and gamma-synuclein, and any neurodegenerative disease has been established, and it is generally considered that these proteins are not highly pathogenic. In addition, beta and gamma-synuclein are rep

Structure, assembly, and mechanism of the MmpL family of transporters.

SUMMARYThe mycobacterial cell envelope, one of the most complex membranes found in bacteria, plays a major role in bacterial pathogenesis, virulence, and antimicrobial resistance. Biogenesis and modeling of this cell envelope are heavily influenced by the mycobacterial membrane protein large (MmpL) family of transporters due to their ability to export fatty acids and lipid components. Select MmpL transporters can also function as siderophore exporters to help regulate the acquisition of iron, which is critical for mycobacterial survival. Additionally, certain MmpLs can participate in active ef

Structure, Function and Pharmacology of SLC7 Family Members and Homologues.

Amino acids are essential components of all living cells serving as building blocks of proteins, as energy source, and as precursors of metabolites and signaling molecules. Amino acid transporters are membrane proteins that mediate the transfer of amino acids across the plasma membrane, and between compartments in cells, different cells and organs. The absence, overexpression or malfunction of specific amino acid transporters have been associated with human disease. One of the projects within the Swiss National Centre of Competence in Research (NCCR) TransCure was directed at SLC7 family amino

Family structure and childhood mental disorders: new findings from Australia.

PURPOSE: Many children now live in non-traditional families-including one-parent, blended, and step families. While a substantial body of international evidence indicates that these children display poorer cognitive and socio-emotional outcomes than children living in traditional families, research on childhood mental disorders is scarce. This report provides new evidence of the relationships between family structure and childhood mental disorders in an under-researched context, Australia. METHODS: We use recent, nationally representative data on children aged 4-17 from Young Minds Matter, th

Regulation of osteoclast structure and function by FAK family kinases.

Osteoclasts are highly specialized cells that resorb bone and contribute to bone remodeling. Diseases such as osteoporosis and osteolytic bone metastasis occur when osteoclast-mediated bone resorption takes place in the absence of concurrent bone synthesis. Considerable effort has been placed on identifying molecules that regulate the bone resorption activity of osteoclasts. To this end, we investigated unique and overlapping functions of members of the FAK family (FAK and Pyk2) in osteoclast functions. With the use of a conditional knockout mouse model, in which FAK is selectively targeted fo

[Population structure analysis of Mycobacterium tuberculosis Beijing family in Japan].

The Beijing family is a genotypic lineage of M. tuberculosis that reportedly predominates throughout eastern Asia and in parts of Russia yet dispersed worldwide. About 80% of clinical isolates in Japan are belonging to this family. The predominance of a narrow range of genotypes, in this case predominance by Beijing family strains, hypothesized that these strains may have a selective advantage either with virulence or transmissibility that led to clonal expansion. The strains are monophyletic within the M. tuberculosis species and also reveal further sublineages within this family. It is belie

Structural studies of the doublecortin family of MAPs.

Doublecortin (DCX) is a microtubule (MT)-stabilizing protein essential for neuronal migration during human brain development. Missense mutations in DCX cause severe brain defects. This implies that the many other MT-stabilizing proteins in neurons cannot compensate for DCX function. To understand the unusual properties of DCX, we expressed the recombinant human DCX in Sf9 cells and undertook structural characterization of its interaction with MTs using cryo-electron microscopy. DCX specifically nucleates 13-protofilament (13-pf) MTs, the architecture of human MTs in vivo. Cryo-electron tomogra

[Function and structure of families with an epileptic child].

Although the impact of psycho-social factors on the individual patient with epilepsy has been widely investigated, the influence of the illness on the family as a whole is still underestimated. By means of the Family Assessment Measure (FAM III), a well-validated instrument, we investigated which measurable influence the epilepsy of one child had on the functioning of the families. Data from a group of 72 families with a child suffering from epilepsy (EG), but without any other handicap were analysed and compared with those of 75 families with a child with severe mental retardation (SMG) and 7

Two Family Gradients in Children's Physical Activity and Fitness: Socioeconomic Position, Family Structure, and Where They Diverge.

BACKGROUND/OBJECTIVES: Family circumstances are related to children's physical activity and fitness, yet the two are rarely examined together in the same children. We examined how socioeconomic position and family structure (only-child status) relate to domain-specific reported activity and measured fitness. Socioeconomic position was the mean of maternal and paternal educational attainment, with higher values indicating higher attainment. METHODS: We designed and conducted a cross-sectional survey of 8228 children in grades 1 to 9 from 27 schools. The questionnaire covered the preceding six

Is Family Structure Associated with Deviance Propensity during Adolescence? The Role of Family Climate and Anger Dysregulation.

Transgressive conduct and opposition towards the rules often characterize adolescence. During the development, antisocial and aggressive behavior could be a way to grow personally and to be independent. According to previous studies results, the family has a high impact on teens' aggressive behaviors and moral disengagement. Our research involved 2328 Italian adolescents (13-19 years old) who have filled in the following questionnaires: deviant behavior questionnaire; aggression questionnaire; family communication scale; moral disengagement scale; the multidimensional scale of perceived social

The evolution and structure/function of bHLH-PAS transcription factor family.

Proteins that contain basic helix-loop-helix (bHLH) and Per-Arnt-Sim motifs (PAS) function as transcription factors. bHLH-PAS proteins exhibit essential and diverse functions throughout the body, from cell specification and differentiation in embryonic development to the proper function of organs like the brain and liver in adulthood. bHLH-PAS proteins are divided into two classes, which form heterodimers to regulate transcription. Class I bHLH-PAS proteins are typically activated in response to specific stimuli, while class II proteins are expressed more ubiquitously. Here, we discuss the gen

Remodeling of HIV-1 Nef Structure by Src-Family Kinase Binding.

The HIV-1 accessory protein Nef controls multiple aspects of the viral life cycle and host immune response, making it an attractive therapeutic target. Previous X-ray crystal structures of Nef in complex with key host cell binding partners have shed light on protein-protein interactions critical to Nef function. Crystal structures of Nef in complex with either the SH3 or tandem SH3-SH2 domains of Src-family kinases reveal distinct dimer conformations of Nef. However, the existence of these Nef dimer complexes in solution has not been established. Here we used hydrogen exchange mass spectrometr

Crystal structure of CsCrAs2O7, a new member of the diarsenate family.

Caesium chromium(III) diarsenate(V), CsCrAs2O7, was prepared by solid-state reactions. The title structure consists of isolated CrO6 octa-hedra and As2O7 diarsenate groups, sharing corners to build up a three-dimensional [CrAs2O7](-) anionic framework. In this framework, channels extending parallel to [001] are present in which the ten-coordinate Cs(+) ions reside. CsCrAs2O7 is isotypic with the monoclinic A (I) M (III) X 2O7 (A (I) = alkali metal; M (III) = Al, Cr, Fe; X = As, P) type I family of compounds crystallizing in the space group P21/c.

The Ce2Li0.39Ni1.61Si2 structure as a new derivative of the AlB2 family.

A new quaternary dicerium lithium/nickel disilicide, Ce2Li0.39Ni1.61Si2, crystallizes as a new structure type of intermetallic compounds closely related to the AlB2 family. The crystal-chemical interrelationships between parent AlB2-type, BaLiSi, ZrBeSi and the title compound are discussed using the Bärnighausen formalism. Two Ce atoms occupy sites of 3m. symmetry. The remainder, i.e. Ni, mixed Ni/Li and Si atoms, occupy sites of -6m2 symmetry. The environment of the Ce atom is an 18-vertex polyhedron and the Ni, Ni/Li and Si atoms are enclosed in tricapped trigonal prisms. The title structure

Family history of Alzheimer's disease and hippocampal structure in healthy people.

OBJECTIVE: Structural brain changes appear years before the onset of Alzheimer's disease, the leading cause of dementia late in life. Determining risk factors for such presymptomatic brain changes may assist in identifying candidates for future prevention treatment trials. In addition to the e4 allele of the apolipoprotein E gene (APOE-4), the major known genetic risk factor, a family history of Alzheimer's disease also increases the risk to develop the disease, reflecting yet unidentified genetic and, perhaps, nongenetic risks. The authors investigated the influence of APOE-4 genotype and fam
