Carbon monoxide dehydrogenase from Rhodospirillum rubrum produces formate.

Carbon monoxide dehydrogenase (CODH) from Rhodospirillum rubrum reversibly catalyzes the oxidation of CO to CO(2) at the active site C-cluster. In this article, the reduction of CO(2) to formate is reported as a slow side reaction catalyzed by both Ni-containing CODH and Ni-deficient CODH. Recently, the structures of R. rubrum CODH and its active site NiFeS cluster (the C-cluster) have been solved. The data in this manuscript describe the formate-producing capability of CODH with or without Ni in the active site.

Formate dehydrogenase from the methane oxidizer Methylosinus trichosporium OB3b.

Formate dehydrogenase (NAD+ dependent) was isolated from the obligate methanotroph Methylosinus trichosporium OB3b. When the enzyme was isolated anaerobically, two forms of the enzyme were seen on native polyacrylamide gels, DE-52 cellulose and Sephacryl S-300 columns; they were approximately 315,000 and 155,000 daltons. The enzyme showed two subunits on sodium dodecyl sulfate-polyacrylamide gels. The Mr of the alpha-subunit was 53,800 +/- 2,800, and that of the beta-subunit was 102,600 +/- 3,900. The enzyme (Mr 315,000) was composed of these subunits in an apparent alpha 2 beta 2 arrangement.

Formate from THF-C1 metabolism induces the AOX1 promoter in formate dehydrogenase-deficient Komagataella phaffii.

In Komagataella phaffii (Pichia pastoris), formate is a recognized alternative inducer to methanol for expression systems based on the AOX1 promoter (pAOX1). By disrupting the formate dehydrogenase encoding FDH1 gene, we converted such a system into a self-induced one, as adding any inducer in the culture medium is no longer requested for pAOX1 induction. In cells, formate is generated from serine through the THF-C1 metabolism, and it cannot be converted into carbon dioxide in a FdhKO strain. Under non-repressive culture conditions, such as on sorbitol, the intracellular formate generated from

The purification and properties of formate dehydrogenase and nitrate reductase from Escherichia coli.

The membrane-bound formate dehydrogenase of Escherichia coli grown anaerobically in the presence of nitrate was solubilized with deoxycholate and purified to near homogeneity. The purification procedure included ammonium sulfate fractionation and chromatography on Bio-Gel A-1.5m and DEAE Bio-Gel A in the presence of the nonionic detergent, Triton X-100. This detergent caused a significant decrease in the molecular weight of the soluble formate dehydrogenase complex and allowed the enzyme then to be resolved from other membrane components. Anaerobic conditions were required throughout due to th

NADH-dependent formate dehydrogenase mutants for efficient carbon dioxide fixation.

Bioconversion of CO2 to high-valuable products is a globally pursued sustainable technology for carbon neutrality. However, low CO2 activation with formate dehydrogenase (FDH) remains a major challenge for further upcycling due to the poor CO2 affinity, reduction activity and stability of currently used FDHs. Here, we present two recombined mutants, ΔFDHPa48 and ΔFDHPa4814, which exhibit high CO2 reduction activity and antioxidative activity. Compared to FDHPa, the reduction activity of ΔFDHPa48 was increased up to 743 % and the yield in the reduction of CO2 to methanol was increased by 3.16-f

Stabilization of Formate Dehydrogenase in a Metal-Organic Framework for Bioelectrocatalytic Reduction of CO2.

The efficient fixation of excess CO2 from the atmosphere to yield value-added chemicals remains crucial in response to the increasing levels of carbon emission. Coupling enzymatic reactions with electrochemical regeneration of cofactors is a promising technique for fixing CO2 , while producing biomass which can be further transformed into biofuels. Herein, a bioelectrocatalytic system was established by depositing crystallites of a mesoporous metal-organic framework (MOF), termed NU-1006, containing formate dehydrogenase, on a fluorine-doped tin oxide glass electrode modified with Cp*Rh(2,2'-b

Alcohol dehydrogenase activities of wine yeasts in relation to higher alcohol formation.

Alcohol dehydrogenase activities were examined in cell-free extracts of 10 representative wine yeast strains having various productivities of higher alcohols (fusel oil). The amount of fusel alcohols (n-propanol, isobutanol, active pentanol, and isopentanol) produced by the different yeasts and the specific alcohol dehydrogenase activities with the corresponding alcohols as substrates were found to be significantly related. No such relationship was found for ethanol. The amounts of higher alcohols formed during vinification could be predicted from the specific activities of the alcohol dehydro

Importance of tetramer formation by the nitrogen assimilation control protein for strong repression of glutamate dehydrogenase formation in Klebsiella pneumoniae.

The nitrogen assimilation control protein (NAC) from Klebsiella pneumoniae is a very versatile regulatory protein. NAC activates transcription of operons such as hut (histidine utilization) and ure (urea utilization), whose products generate ammonia. NAC also represses the transcription of genes such as gdhA, whose products use ammonia. NAC exerts a weak repression at gdhA by competing with the binding of a lysine-sensitive activator. NAC also strongly represses transcription of gdhA (about 20-fold) by binding to two separated sites, suggesting a model involving DNA looping. We have identified

Characterization of human alcohol dehydrogenase 4 and aldehyde dehydrogenase 2 as enzymes involved in the formation of 5-carboxylpirfenidone, a major metabolite of pirfenidone.

Pirfenidone (PIR) is used in the treatment of idiopathic pulmonary fibrosis. After oral administration, it is metabolized by cytochrome P450 1A2 to 5-hydroxylpirfenidone (5-OH PIR) and further oxidized to 5-carboxylpirfenidone (5-COOH PIR), a major metabolite excreted in the urine (90% of the dose). This study aimed to identify enzymes that catalyze the formation of 5-COOH PIR from 5-OH PIR in the human liver. 5-COOH PIR was formed from 5-OH PIR in the presence of NAD+ by human liver microsomes (HLMs) more than by human liver cytosol (HLC), with the concomitant formation of the aldehyde form (

Fast growth and high-titer bioproduction from renewable formate via metal-dependent formate dehydrogenase in Escherichia coli.

Microbial bioproduction using one-carbon (C1) feedstocks has the potential to decarbonize the manufacturing of materials, fuels, and chemicals. Formate is a promising C1 feedstock, and the realization of industrial, formatotrophic platform organisms is a key goal for C1-based bioproduction. So far, a major limitation for synthetic formatotrophy has been slow energy supply due to slow formate dehydrogenase activity. Here, we implement a fast, metal-dependent formate dehydrogenase complex in a synthetic formatotrophic Escherichia coli utilizing the reductive glycine pathway. After a short-term e

Mechanistic Insights Into the Formaldehyde Dehydrogenase-Catalyzed Reduction of Formate: A Quantum Chemical Study.

The multienzyme cascade conversion of CO2 to methanol is a promising route for sustainable chemical synthesis, yet the reduction of formate to formaldehyde remains its major bottleneck. Here, we use quantum chemical calculations to elucidate the catalytic mechanism of formaldehyde dehydrogenase from Burkholderia multivorans (BmFaldDH). The lowest-energy pathway initiates with formate activation via a proton-relay network involving His52, Ser49, and the NADH ribose, followed by a concerted yet asynchronous proton-hydride transfer to yield methanediol with a barrier of 16.0 kcal mol-1. Techno-economic analysis of industrial-scale fermentation for formate dehydrogenase (FDH) production.

The conversion of carbon dioxide (CO2) into formate offers a promising route to enable a circular, carbon-smart bioeconomy. Formate is increasingly recognized as a versatile and energy-dense platform molecule that can serve as a feedstock for microbial fermentation, energy storage, and sustainable chemical and fuel production. A key bottleneck in this value chain is the availability of cost-effective and scalable formate dehydrogenase (FDH), which catalyze the initial reduction of CO2 to formate. However, little is known about the economic feasibility of producing and purifying FDH at industri

Formate Reduces Ischemic Injury in Female Hearts Lacking Alcohol Dehydrogenase 5.

Ischemic heart disease is a primary cause of death for men and women in the United States. Recent epidemiologic findings, however, suggest that premenopausal women have inherent protection from many cardiovascular pathologies compared to age-matched men, which is lost with menopause. We and others have documented similar protective signaling in animal models, with females exhibiting protection from ischemic injury that is lost with ovariectomy (OVX). Furthermore, in recent studies, we demonstrated that the loss of alcohol dehydrogenase 5 (ADH5) blocked sex-specific cardioprotection in females,

Type 5 17beta-hydroxysteroid dehydrogenase: its role in the formation of androgens in women.

Type 5 17beta-HSD, one of the seven types of 17beta-hydroxysteroid dehydrogenase (17beta-HSD) so far characterized in humans, catalyzes the transformation of 4-androstenedione (4-dione) into testosterone (T). This reaction is also catalyzed by type 3 17beta-HSD which is responsible for pseudohermaphroditism in deficient man but is asymptomatic in deficient women. Since type 3 17beta-HSD is not found in the ovary, whereas type 5 is, it is suggested that the latter is involved in the conversion of 4-androstenedione to testosterone in the ovary. The comparison of type 5 17beta-HSD of different sp

An unexpected alternative viologen electron mediator site in tungsten-containing formate dehydrogenase.

The molecular mechanisms by which artificial electron mediators, such as ethyl viologen (EV), interact with tungsten-containing formate dehydrogenases (FDHs) during reversible CO2 reduction remain poorly understood. Here, we reveal an unexpected alternative mediator interaction site in FDH1 of Methylorubrum extorquens AM1. Removing the native flavin mononucleotide cofactor abolishes NAD+-dependent activity but preserves EV-driven catalysis. Through mutagenesis, kinetic analysis, and molecular docking, we identified a cooperative aromatic network-comprising residues F232, F471, and Y329-that st

Electrocatalysis of CO2 Reduction by Immobilized Formate Dehydrogenase without a Metal Redox Center.

Nicotinamide adenine dinucleotide-dependent formate dehydrogenase from Candida boidinii was immobilized in a 1,2-dimyristoyl-sn-glycero-3-phosphocholine/cholesterol floating lipid bilayer on the gold surface as a biocatalyst for electrochemical CO2 reduction. We report that, in contrast to common belief, the enzyme can catalyze the electrochemical reduction of CO2 to formate without the cofactor protonated nicotinamide adenine dinucleotide. The electrochemical data indicate that the enzyme-catalyzed reduction of CO2 is diffusion-controlled and is a reversible reaction. The orientation and conf

Bioelectrocatalytic carbon dioxide reduction by an engineered formate dehydrogenase from Thermoanaerobacter kivui.

Electrocatalytic carbon dioxide (CO2) reduction by CO2 reductases is a promising approach for biomanufacturing. Among all known biological or chemical catalysts, hydrogen-dependent carbon dioxide reductase from Thermoanaerobacter kivui (TkHDCR) possesses the highest activity toward CO2 reduction. Herein, we engineer TkHDCR to generate an electro-responsive carbon dioxide reductase considering the safety and convenience. To achieve this purpose, a recombinant Escherichia coli TkHDCR overexpression system is established. The formate dehydrogenase is obtained via subunit truncation and rational d

Direct reduction of CO2catalyzed by a formate dehydrogenase immobilized on carbon nanotubes without NADH cofactor.

This paper describes the electroenzymatic reduction of CO2to formate catalyzed by formate dehydrogenase fromCandida boidinii(CbFDH) immobilized on carbon nanotube (CNT)-modified gold electrodes. Cyclic voltammetry indicates thatCbFDH could catalyze CO2reduction to formate without protonated nicotinamide adenine dinucleotide (NADH) as a cofactor, exhibiting diffusion-controlled, quasi-reversible kinetics on both multi-walled CNT and single-walled CNT substrates. Surface-enhanced infrared absorption spectra indicate thatCbFDH adopts a near-parallel orientation on the CNT-modified gold surface, p

Semi-Rational Engineering of Formate Dehydrogenase Reveals Loop Dynamics as a Key Determinant of Thermostability.

Formate dehydrogenase (FDH) catalyzes the reversible formate oxidation using NAD+ to yield carbon dioxide and NADH. FDH is widely utilized for cofactor regeneration, enabling the continuous supplying of NADH for chiral chemical biosynthesis. Herein, we employed semi-rational engineering to improve the thermostability of highly catalytic FDH from Bacillus simplex (BsFDH). Through two rounds of mutational screening, BsFDHQ125F variant was identified to exhibit significantly improved thermostability with a 25-fold increase in half-life at 60°C. Additionally, the BsFDHQ125F variant revealed signif
