Non-Mendelian transmission in a human developmental disorder: split hand/split foot.

The study of Mendelian disorders that do not meet some Mendelian expectations has led to an increased understanding of such previously obscure genetic phenomena as anticipation. Split hand/split foot (SHSF), a human developmental malformation, demonstrates such distinctive genetic features as reduced penetrance and variable expressivity. In this study, new pedigrees with defined ascertainment confirm the existence of non-Mendelian transmission characterized by the overtransmission of SHSF from affected fathers to sons.

Paraganglioma of the tongue with SDHB gene mutation in a patient with Graves' disease.

We report a case of an apparently sporadic paraganglioma of the tongue with a germ-line mutation in a female patient with asymptomatic Graves' disease. The tongue is an unusual primary location. Genetic testing is mandatory in all cases. Thyroid gland dysfunction and autoimmune phenomena could be associated with some paragangliomas.

Genetic exchange in Trypanosoma brucei: evidence for meiosis from analysis of a cross between drug-resistant transformants.

Genetic exchange in Trypanosoma brucei spp. can occur when two strains are cotransmitted through the tsetse fly vector, but it is non-obligatory and a comparatively rare event. To increase recovery of hybrids, we crossed drug resistant parental strains and selected hybrids by double drug resistance [15]. Analysis of 29 hybrid clones from five separate genetic exchange events shows independent segregation of marker genes and a high frequency of triploidy, both of which phenomena have been observed previously for other trypanosome crosses. However, in addition we provide evidence of genetic reco

Senile dementia of the Alzheimer type: possibility of infectious etiology in genetically susceptible individuals.

The concept that SDAT is caused by an infectious agent acting in a genetically susceptible host was approached from a number of standpoints. The similarities between SDAT and the transmissible encephalopathies are discussed. One of the areas of similarity is the influence of genetic background on the development and expression of both conditions. Evidence is presented showing that genetics plays a role in many cases of SDAT and that there are known genetically controlled phenomena, the incidence of which is positively correlated with SDAT. For human encephalopathies the genetics of CJD and GSS

The autosomal dominant spinocerebellar ataxias: emerging mechanistic themes suggest pervasive Purkinje cell vulnerability.

The spinocerebellar ataxias are a genetically heterogeneous group of disorders with clinically overlapping phenotypes arising from Purkinje cell degeneration, cerebellar atrophy and varying degrees of degeneration of other grey matter regions. For 22 of the 32 subtypes, a genetic cause has been identified. While recurring themes are emerging, there is no clear correlation between the clinical phenotype or penetrance, the type of genetic defect or the category of the disease mechanism, or the neuronal types involved beyond Purkinje cells. These phenomena suggest that cerebellar Purkinje cells m

Down's syndrome and autoimmunity.

A patient with Down's syndrome (DS) with multiple autoimmune phenomena is described. She suffered from hypothroidism, a celiac-like enteropathy and hemolytic anemia, and displayed cellular immunity directed against peripheral nerve antigen and basic myeloprotein and serum autoantibodies to many other tissue antigens. Her mother did not suffer from any overt autoimmune disease, but similar autoantibodies were found in her serum. It is suggested that DS resembles other autoimmune diseases, especially that which occurs in the NZB mice. The resemblance is based on the assumption that in both cases

Hypotheses concerning the aetiology of Alzheimer's disease.

Theories on the aetiology and pathogenesis of Alzheimer's disease (AD) are revised. After discussing senile involution, the principal characteristic alterations of AD are presented. These pathological changes include involutive morphological phenomena (neurofibrillar tangles, senile neuritic plaques with varying = amyloid content) and functional phenomena (alterations in energy metabolism, in protein synthesis, and in the neurotransmitter metabolism). Currently it is assumed that the neurons most affected seem to be primarily those of the ascending cholinergic activating system, whose somas ar

Familial trends in palatal canines, anomalous lateral incisors, and related phenomena.

Our previous studies have established a strong link between small, peg-shaped and missing lateral incisors, and palatally-displaced maxillary canine teeth. Since these anomalies of lateral incisor development are known to be under strong genetic control, the hypothesis was that first degree relatives of patients with palatal canines would themselves show a raised prevalence of the anomalies. The present study also investigated a possible link between palatal canines and lateness in development of the dentition, and a relative absence of crowding, both of which are phenomena that have been ment

On the medical history of the doctrine of imagination.

In the early modern era the notion of imagination was made responsible for phenomena which were later explained in terms of embryology, genetics, psychology, bacteriology or other scientific disciplines. Images, often seated in the upper abdomen (hypochondriac region) or the womb (hysteria), were regarded as powerful influences on material reality. In the course of the seventeenth and eighteenth centuries the hypochondriac forms of imagination became mere whims and spleens, but they kept much of their original potency in respect of the uterus, accounting for monstrosities and the shaping of hu

Mitochondrial encephalomyopathies: clinical and molecular analysis.

The classification of mitochondrial encephalomyopathies relied upon clinical, biochemical, and histological features until the discovery of mitochondrial DNA defects in 1988. Since then, an outburst of molecular genetic information has aided our understanding of the pathogenesis and the classification of these heterogeneous disorders. Novel concepts of maternal inheritance, mitochondrial DNA (mtDNA) heteroplasmy, tissue distribution, and threshold have explained many of the clinical characteristics. The discovery of point mutations, large-scale mtDNA deletions, duplications, and autosomally in

Rare postoperative delayed malignant hyperthermia after off-pump coronary bypass surgery and brief review of literature.

Postoperative malignant hyperthermia (MH) is a very rare phenomena. It is generally observed within less than an hour after discontinuation of the anesthetic trigger. Present case describes rare delayed postoperative presentation of MH after off-pump coronary bypass surgery. Prompt recognition and immediate treatment with dantrolene can effectively treat the fatal syndrome. Family education and genetic counseling should be encouraged.

"Mikrob" Russian Research Anti-Plague Institute, Saratov Studies of the genomic evolution of pathogenic bacteria became a priority research trend of modern molecular genetics. Vibrio cholerae, whose pathogenic properties are conditioned by the presence of virulence blocks of differing phylogenetic origins in 2 chromosomes, turned out to be a unique model object for studies of evolutionary transformations of genomes that are causative agents of extra dangerous infections. The molecular-and-genetic mechanisms underlying the change of biovariants and the emergence of a cholera agent of a new sero

Understanding neonatal hyperbilirubinaemia in the era of genomics.

The genomics revolution offers novel approaches to scientific investigation. Application of genomics technologies including microarray gene chips will provide a more complete picture of biological phenomena and help define the genetic contribution to disease by monitoring changes in expression across thousands of genes in physiological and clinical contexts. We briefly summarize identified genetic components that contribute to the genesis of neonatal hyperbilirubinaemia with a focus on inborn errors of hepatic bilirubin conjugation and discuss the potential use of microarray gene expression pr

Molecular genetic advances in sleep research and their relevance to sleep medicine.

Neurobiology has been profoundly impacted by the availability of molecular genetic techniques within the past decade, and the sleep research field is no exception to this trend. Genetic influences on normal and pathologic sleep have long been recognized, but only recently have the contributions of discrete genetic loci to the regulation of sleep timing and sleep-related physiologic phenomena begun to be demonstrated, particularly through the use of genetically engineered strains of rodents. The ability to monitor the expression of a large number of genes has also accelerated in recent years, l

Altered T and B lymphocyte signaling pathways in lupus.

Systemic lupus erythematosus (SLE) has long been recognized to be characterized by dysregulated signaling pathways in T and B lymphocytes, beginning with observations of cellular hyperactivity and hyperresponsiveness, and evolving to recent studies focused upon the genetic and molecular bases of such phenomena. This review focuses on recently elucidated signaling abnormalities currently thought to be intrinsic to T and/or B cells in human SLE.

Etiology of maxillary canine impaction: a review.

This article is a review that enumerates the causes of impaction of the maxillary permanent canines, including hard tissue obstructions, soft tissue lesions, and anomalies of neighboring teeth, and discusses the much-argued relationship between environmental and genetic factors. These phenomena have been shown in many investigations to accompany the diagnosis of canine impaction and have been presented as unrelated anomalous features, each of which is etiologically construed as genetic, including the aberrant canine itself. While in general the influence of genetics pervades the wider picture,

Pathology of systemic lupus erythematosus: the challenges ahead.

Many studies have explored the pathology of systemic lupus erythematosus (SLE), an autoimmune rheumatic disorder with a striking female predominance. Numerous autoimmune phenomena are present in this disease, which ultimately result in organ damage. However, the specific cellular and humoral mechanisms underlying the immune dysfunction are not yet fully understood. It is postulated that autoimmunity is based on the interaction of genetic predisposition, hormonal and environmental triggers that result in reduced tolerance to self-tissues. These phenomena could occur because of altered antigen p

Mitochondrial encephalomyopathies: a review of routine morphological diagnostic methods with emphasis on the role of electron microscopy.

Mitochondrial encephalomyopathies (MEs) are a group of clinically and genetically heterogeneous diseases. They can be caused by defects in both mitochondrial or nuclear coded genes. Their phenotypic expression is governed by unique biological phenomena such as the dual genetic control, mitotic segregation, heteroplasmy and threshold effects. Currently, the correct diagnosis of ME relies on a multidisciplinary approach which includes clinical information as well as laboratory data from muscle morphology, biochemistry and molecular genetics. Among the morphological methods, histology, histochemi

[Similarities and differences between bipolar disorder and hypertension: may be mania is the hypertension of the mood?].

A possibly shared pathophysiological background between bipolar disorders and essential hypertension is suggested by the several similarities and overlaps between their genetic background, underlying biological disturbances (including partially shared neuroanatomical and neurochemical correlates), concomitant personality and temperamental characteristics, precipitating factors, comorbidity and treatment response. In this paper we outline and extend our previously published hypothesis discussing the similar origins of these two biphasic/bidirectional phenomena.
