Baseline Susceptibility of Helicoverpa armigera (Lepidoptera: Noctuidae) to Indoxacarb, Emamectin Benzoate, and Chlorantraniliprole in Australia.

Baseline susceptibility of Helicoverpa armigera (Hübner) to emamectin benzoate, chlorantraniliprole, and indoxacarb was determined in feeding assays on insecticide-incorporated artificial diet in the laboratory. The intraspecific variation of H. armigera was established from field populations collected between September 2012 and March 2013, primarily from commercial farms across eastern Australia. Emamectin benzoate had the highest toxicity with a median lethal concentration (LC50) of 0.01 µg/ml diet (n=20 strains). The LC50 for chlorantraniliprole was 0.03 µg/ml diet (n=21 strains), while ind

Sublethal effects of matrine and neemarin on the demography of Habrobracon hebetor developing on treated Helicoverpa armigera larvae.

Integrated pest management (IPM) relies on the compatibility of multiple control strategies. In controlling Helicoverpa armigera (Hübner) with the braconid parasitoid Habrobracon hebetor (Say), evaluating insecticide efficacy and compatibility is essential. Botanical insecticides may provide a safer alternative to synthetic chemicals due to reduced non-target effects. This study investigated the effects of sublethal concentrations of matrine and neemarin ingested by H. armigera larvae on H. hebetor developing on them, to assess their compatibility for potential integration into IPM programs. Interaction of plant cell signaling molecules, salicylic acid and jasmonic acid, with the mitochondria of Helicoverpa armigera.

The cotton bollworm, Helicoverpa armigera is a polyphagous pest in Asia, Africa, and the Mediterranean Europe. Salicylic acid (SA) and jasmonic acid (JA) are the cell signaling molecules produced in response to insect attack in plants. The effect of these signaling molecules was investigated on the oxidative phosphorylation and oxidative stress of H. armigera. SA significantly inhibited the state III and state IV respiration, respiratory control index (RCI), respiratory complexes I and II, induced mitochondrial swelling, and cytochrome c release in vitro. Under in vivo conditions, SA induced s

Complexity in specificities and expression of Helicoverpa armigera gut proteinases explains polyphagous nature of the insect pest.

Helicoverpa armigera is a devastating pest of cotton and other important crop plants all over the world. A detailed biochemical investigation of H. armigera gut proteinases is essential for planning effective proteinase inhibitor (PI)-based strategies to counter the insect infestation. In this study, we report the complexity of gut proteinase composition of H. armigera fed on four different host plants, viz. chickpea, pigeonpea, cotton and okra, and during larval development. H. armigera fed on chickpea showed more than 2.5- to 3-fold proteinase activity than those fed on the other host plants

miR-252a-5p targets OVOL to regulate ovary development and egg viability (hatchability) in Helicoverpa armigera.

Disruption of reproduction is an important pest control tactics for management of high-fecundity pests like Helicoverpa armigera (Hübner). In this study, we investigated the role of the testis-biased miR-252a-5p in regulating gonad development and reproduction. Through a combination of bioinformatics prediction, dual luciferase reporter assays, functional injections (agomir, antagomir, and siRNA), and phenotypic evaluations, we identified OVOL as a key target gene of miR-252a-5p. We found that miR-252a-5p negatively regulates OVOL expression. Injection of miR-252a-5p agomir or OVOL siRNA into

Juvenile hormone controls trehalose metabolism by regulating trehalase 2 activity in ovarian development of Helicoverpa armigera.

Trehalase (Treh) is crucial for ovarian development as it directly regulates the energy supply by hydrolyzing trehalose into glucose. Juvenile hormone (JH) is also essential for ovarian development, but how it affects Treh2 activity remains unclear. This study, which employed Helicoverpa armigera as a model, showed that HaTreh2 transcription and enzymatic activity peaks coincided with the peak of JH titers (the 2 and 3 days after emergence). Compared to the dsGFP control, knockdown of HaTreh2 transcription severely impaired ovarian development. LC-MS/MS and site mutation experiments demonstrat

Harnessing next-generation molecular technologies for precision control of Tuta absoluta and Helicoverpa armigera in tomato.

Tomato (Solanum lycopersicum) is one of the most significant vegetable crops cultivated around the world. Among biotic stresses, production of tomato is severely affected by insect pests mainly by tomato fruit borer (Helicoverpa armigera) and tomato leaf miner (Tuta absoluta). Traditionally to manage the destruction caused by insect pests, farmers mostly used chemical insecticides and by different cultural practices. Use of chemical pesticides is of great concern as it is harmful for the environment and other living beings and with the passage of time, insects develop resistance against pestic

Transcriptome Analysis of the Brain and Gnathal Ganglion Reveals Feeding-Mediated Genes in Helicoverpa armigera Larvae.

Despite the central role of the gustatory nervous system in regulating feeding behavior in the larvae of the major agricultural pest Helicoverpa armigera, the systemic molecular basis of this process is largely unknown. To investigate the molecular basis, we performed RNA-seq on dissected brains and gnathal ganglion (GNG) from fifth-instar larvae, revealing 944 differentially expressed genes (DEGs) that are potentially involved in feeding behaviors. Bioinformatic analyses revealed significant enrichment of these DEGs in pathways including "taste transduction", "neuroactive ligand-receptor inte

AMPK regulates pupal diapause by modulating glucose metabolism in the brain of the cotton bollworm, Helicoverpa armigera.

BACKGROUND: Diapause is a vital adaptive strategy enabling insects to endure adverse environmental conditions and significantly extend their lifespan, by suspending development and conserving energy. During diapause, profound physiological and metabolic changes occur. Key genes involved in diapause have long been considered a potential target for pest control. RESULTS: In this study, we observed elevated glucose levels in the brains of Helicoverpa armigera diapause-destined pupae. Concurrently, the expression of glucose transporter 1 (Glut1), critical for glucose uptake, was upregulated, disp

Molecular characterization and elucidation of the function of Hap38 MAPK in the response of Helicoverpa armigera (Hübner) to UV-A stress.

The cotton bollworm Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae), an important pest of cotton, is detrimental to cotton production. Light from UV-A ultraviolet lamps is regarded as a form of environmental stress for insects. In order to investigate the response of H. armigera exposed to UV-A, we explored Hap38 MAPK expression and functions. We hope that the findings of this study will lay the foundation for future investigations into the insect's phototaxis mechanism. A p38 MAPK was cloned and named Hap38 MAPK. A phylogenetic tree showed that Hap38 MAPK was highly conserved. The gene

Dynamic transcriptome analysis and volatile profiling of Gossypium hirsutum in response to the cotton bollworm Helicoverpa armigera.

In response to insect herbivory, plants emit elevated levels of volatile organic compounds for direct and indirect resistance. However, little is known about the molecular and genomic basis of defense response that insect herbivory trigger in cotton plants and how defense mechanisms are orchestrated in the context of other biological processes. Here we monitored the transcriptome changes and volatile characteristics of cotton plants in response to cotton bollworm (CBW; Helicoverpa armigera) larvae infestation. Analysis of samples revealed that 1,969 transcripts were differentially expressed (l

The molecular characterization of antioxidant enzyme genes in Helicoverpa armigera adults and their involvement in response to ultraviolet-A stress.

Ultraviolet (UV) radiation can cause oxidative stress in insects. To gain insight into the roles of different antioxidant enzymes of Helicoverpa armigera adults in response to oxidative stress caused by UV-A at a molecular level, three antioxidant enzyme genes [encoding copper zinc superoxide dismutase (Cu/ZnSOD), catalase (CAT) and glutathione peroxidase (GPX)] were cloned. The deduced amino acid sequences of these genes are similar to that of other insects. In addition, the expression profiles of genes from a classic antioxidant defense system, comprising Cu/ZnSOD, manganese superoxide dismu

Putative phosphorylation sites on WCA domain of HA2 is essential for Helicoverpa armigera single nucleopolyhedrovirus replication.

Protein phosphorylation is one of the most common post-translational modification processes that play an essential role in regulating protein functionality. The Helicoverpa armigera single nucleopolyhedrovirus (HearNPV) orf2-encoded nucleocapsid protein HA2 participates in orchestration of virus-induced actin polymerization through its WCA domain, in which phosphorylation status are supposed to be critical in respect to actin polymerization. In the present study, two putative phosphorylation sites ((232)Thr and (250)Ser) and a highly conserved Serine ((245)Ser) on the WCA domain of HA2 were mu
