Metabolic markers for the yield of lipophilic indole alkaloids in dried woad leaves (Isatis tinctoria L.).

The pharmacologically active dichloromethane extracts of dried woad leaves (Isatis tinctoria L.), and the methanol extracts of comparable fresh leaves of the same plants, were analyzed by LC-MSn. The fresh leaf metabolite profile revealed a complex pattern of indolic compounds. Besides the known indigo precursors, isatan A, isatan B and indican, seven previously unreported indole derivatives were characterized: acetylindican, malonylindican, two dioxindole glucosides, dioxindole malonylglucoside, 6-hydroxyindole-3-carboxylic acid 6-O-glucoside and 6-hydroxyindole-3-carboxylic acid glucose este

Acid Degradation, Structure Characterization of a Novel Polysaccharide from Leaves of Isatis indigotica Fort. with Immunomodulatory Activity.

The immunomodulatory potential of natural polysaccharides is often limited by their structural complexity and high molecular weight. In this study, DFIP-A3-1 (Mw = 8.68 × 103 g/mol) was obtained from the leaves of Isatis indigotica Fort. by ultrafiltration, DEAE-650M, and Sephadex G-100 chromatography, followed by acid degradation. Fortunately, DFIP-A3-1 exhibited the most potent immunostimulatory activity in vitro. HPGPC, HPSEC-MALLS-RID, GC-MS, FT-IR, Congo red tests, SEM, and AFM were used to characterize their structure, and 1D/2D NMR was used for further investigation of DFIP-A3-1 for in-

IiWRKY34 Regulates Plant Root Growth Through a Direct Interaction With NAC1 and Its Interactor NAC18 in Isatis indigotica.

WRKY34 transcription factor has a substantial role in inducing the accumulation of bioactive lignan compounds, improving the biomass of hairy roots, and increasing stress tolerance in Isatis indigotica. However, the precise role of the IiWRKY34 transcriptional network in modulating root growth remains to be elucidated. Here, we observed that heterogeneous expression of IiWRKY34 increased the primary root length in Arabidopsis thaliana. IiWRKY34 bound directly to the promoter of IiNAC1, which itself bound to the promoter of an auxin biosynthesis gene, IiYUCCA6. Molecular docking displayed the c

(-)-Lariciresinol Isolated from the Roots of Isatis indigotica Fortune ex Lindl. Inhibits Hepatitis B Virus by Regulating Viral Transcription.

Chronic hepatitis induced by hepatitis B virus (HBV) infection is a serious public health problem, leading to hepatic cirrhosis and liver cancer. Although the currently approved medications can reliably decrease the virus load and prevent the development of hepatic diseases, they fail to induce durable off-drug control of HBV replication in the majority of patients. The roots of Isatis indigotica Fortune ex Lindl., a traditional Chinese medicine, were frequently used for the prevention of viral disease in China. In the present study, (-)-lariciresinol ((-)-LRSL), isolated from the roots of Isa

A synthetic derivative of bioactive constituents from Isatis indigotica ameliorates hypersensitivity and arthritis by inhibiting JAK2-STAT3 pathway in mice.

The JAK-STAT pathway plays a crucial role in the signaling cascade associated with various cytokines that have been implicated in the pathogenesis of inflammatory diseases and myeloproliferative neoplasms (MPN). Among the isoforms of JAKs, the JAK2 subtype is primarily responsible for the function of hematopoietic system cells, making it a significant target in the treatment of MPN. However, the precise regulatory role of JAK2 in inflammatory diseases requires further investigation and confirmation. The current study employed a selective JAK2 inhibitor, ZT55, derived from Isatis indigotica roo

Prevention of experimentally induced irritant contact dermatitis by extracts of Isatis tinctoria compared to pure tryptanthrin and its impact on UVB-induced erythema.

Lipophilic extracts of Isatis tinctoria L. exhibit significant activity against several clinically relevant targets of inflammation. The alkaloid tryptanthrin was identified as one of the active principles in woad and characterised as a potent dual inhibitor of COX-2 and 5-LOX. Here, the anti-inflammatory efficacy of topical application of three different Isatis extracts and tryptanthrin was investigated in human volunteers. Two different models were used, namely the sodium lauryl sulphate (SLS)-induced irritant contact dermatitis (ICD) and UVB-induced erythema. Twenty healthy volunteers witho

Hydrogen sulfide decreases Cd translocation from root to shoot through increasing Cd accumulation in cell wall and decreasing Cd2+ influx in Isatis indigotica.

Hydrogen sulfide (H2S), a small gaseous signalling molecule, plays a pivotal role in the plant response to heavy metal stress. Here, we revealed a novel mechanism of Isatis indigotica resistance to cadmium (Cd) stress, in which H2S promotes Cd accumulation in the root and decreases the long-distance transport of Cd from the root to shoot. Cd significantly inhibited Isatis indigotica growth and induced the endogenous H2S level. Application of NaHS (a H2S donor) alleviated the effects of Cd. NaHS restriction of the translocation factor of Cd, elevated the Cd content in roots and depressed the Cd
