Modified Jiles-Atherton Model for Dynamic Magnetization in X-Space Magnetic Particle Imaging.

OBJECTIVE: Magnetic particle imaging (MPI) is a promising medical modality that can image superparamagnetic iron-oxide nanoparticle (SPIO) concentration distributions safely and with high sensitivity. In the x-space reconstruction algorithm, the Langevin function is inaccurate in modeling the dynamic magnetization of SPIOs. This problem prevents the x-space algorithm from achieving a high spatial resolution reconstruction. METHODS: We propose a more accurate model to describe the dynamic magnetization of SPIOs, named the modified Jiles-Atherton (MJA) model, and apply it to the x-space algorit

Field-Orientation Effects in Amplitude-Modulation Magnetic Particle Imaging for High-Resolution Navigation.

Amplitude-modulation magnetic particle imaging (AM-MPI) has strong potential for real-time navigation of magnetic nanoparticles because it provides tracer-specific spatial feedback using a narrowband acquisition scheme with relatively low excitation power and simplified signal detection. However, improving spatial resolution by increasing the selection-field gradient becomes increasingly difficult as scanner dimensions increase. This study investigates whether field orientation can be used to improve and control the spatial resolution of three-dimensional AM-MPI with field-free-point (FFP) and

Ultrasensitive detection of lung inflammation in male mice using immuno-magnetic particle imaging.

Timely and sensitive detection of inflammatory and infectious lung diseases remains a major clinical challenge. Current diagnostic approaches often identify pathology only at advanced stages, leaving gaps in acute triage, fibrotic activity assessment, transplant rejection monitoring and inflammation follow-up. Magnetic Particle Imaging (MPI) offers a sensitive, background-free, whole-body imaging modality without ionizing radiation. Here, we present immuno-Magnetic Particle Imaging (immuno-MPI) using micron-sized iron oxide particles targeted to VCAM-1, a key marker of endothelial inflammation

iMPI: portable human-sized magnetic particle imaging scanner for real-time endovascular interventions.

Minimally invasive endovascular interventions have become an important tool for the treatment of cardiovascular diseases such as ischemic heart disease, peripheral artery disease, and stroke. X-ray fluoroscopy and digital subtraction angiography are used to precisely guide these procedures, but they are associated with radiation exposure for patients and clinical staff. Magnetic Particle Imaging (MPI) is an emerging imaging technology using time-varying magnetic fields combined with magnetic nanoparticle tracers for fast and highly sensitive imaging. In recent years, basic experiments have sho

Magnetic Particle Imaging (MPI): Experimental Quantification of Vascular Stenosis Using Stationary Stenosis Phantoms.

Magnetic Particle Imaging (MPI) is able to provide high temporal and good spatial resolution, high signal-to-noise ratio and sensitivity. Furthermore, it is a truly quantitative method as its signal strength is proportional to the concentration of its tracer, superparamagnetic iron oxide nanoparticles (SPIOs). Because of that, MPI is proposed to be a promising future method for cardiovascular imaging. Here, an interesting application may be the quantification of vascular pathologies like stenosis by utilizing the proportionality of the SPIO concentration and the MPI signal strength. In this st

From Magnetic Moment to Magnetic Particle Imaging: A Comprehensive Review on MPI Technology, Tracer Design and Biological Applications.

Background/Objectives: Magnetic nanoparticles have emerged as powerful tools for biomedical imaging, targeted drug delivery, and hyperthermia therapy. Magnetic particle imaging (MPI) is among the most promising technologies built around its properties: a radiation-free, quantitative tomographic modality that detects superparamagnetic iron oxide nanoparticles (SPIONs) directly against a biologically silent background. This review synthesizes MPI's physical principles, nanoparticle design strategies, and preclinical applications within the broader landscape of magnetic material engineering for b

Magnetic Particle Imaging of Macrophage Phagocytosis Evaluates Plaque Vulnerability and Therapeutic Efficacy in Atherosclerosis.

BACKGROUND: Developing new atherosclerosis therapeutics remains challenging. A noninvasive imaging method to assess plaque vulnerability is needed to provide early efficacy readouts and help select promising candidates for clinical trials. We aimed to identify macrophage phagocytosis markers associated with plaque vulnerability and evaluate whether magnetic particle imaging (MPI) of phagocytic activity can detect therapeutic responses and changes in plaque instability. METHODS: Integrated transcriptomic and immunofluorescence staining analyses of human carotid plaques were conducted to identi

Towards robot-assisted magnetic particle imaging for mobile point-of-care (neuro)vascular applications.

Rapid diagnosis, timely treatment, and continuous monitoring are essential for improving outcomes in neurovascular diseases (time=brain). Such point-of-care applications, however, require imaging to be conducted in dynamic and often confined spaces. While magnetic particle imaging (MPI) shows preclinical promise for fast (neuro)vascular imaging, it relies on complex and restricted imaging gantries. Here, we present a unique gantry-free robot-assisted MPI strategy (Robotic MPI) that is 'portable'. Using permanent magnets, Hall sensors and correction coils, we developed a single-sided detector w

Quantitative Drug Release Monitoring in Tumors of Living Subjects by Magnetic Particle Imaging Nanocomposite.

In vivo drug release monitoring provides accurate and reliable information to guide drug dosing. Image-based strategies for in vivo monitoring are advantageous because they are non-invasive and provide visualization of the spatial distribution of drug, but those imaging modalities in use (e.g., fluorescence imaging (FI) and magnetic resonance imaging (MRI)) remain inadequate because of the low tissue penetration depth (for FI) or difficulty with quantification of release rate and signal convolution with noise sources (for MRI). Magnetic particle imaging (MPI), employing superparamagnetic nanop

Fluorescence imaging and targeted distribution of bacterial magnetic particles in nude mice.

Bacterial magnetic particles (BMPs) are of interest as potential carriers of bioactive macromolecules, drugs, or liposomes. In this study, a high-pressure homogenizer was used to disrupt Magnetospirillum gryphiswaldense strain MSR-1 cells, and BMPs were purified. BMPs were labeled with fluorescence reagent 1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocianin perchlorate (DiI) and injected into the tail vein of BALB/c nude mice. Distribution of fluorescence signals of DiI-BMPs in vivo was examined using a whole-body fluorescence imaging system. The result showed that fluorescence signals were

Unleashing the Power of Magnetic Particle Imaging: Tailored Magnetic Nanoparticles for Ultrasensitive Detection of Bone Metastases in Prostate Cancer.

Magnetic particle imaging (MPI) is a groundbreaking imaging technique hindered by suboptimal tracers, as current magnetic nanoparticles (MNPs) are primarily designed for magnetic resonance imaging and are not specifically tailored for the distinct physics of MPI. To address this issue, optimizing the magnetic diameter of MNPs is focused on, a pivotal parameter influenced by both physical size and magnetic disorder, yet underexplored in the development of MPI tracers. By employing a precise temperature-controlled synthesis strategy, this study successfully modulates these parameters synergistic

Harmonic-aware transformer for real-time catheter localization in interventional procedures of magnetic particle imaging.

Objective.Magnetic particle imaging (MPI) enables real-time, radiation-free tracking of magnetic nanoparticle-coated instruments, making it highly suitable for interventional procedures. This study proposes a harmonic-aware transformer framework that directly predicts catheter tip positions from raw MPI voltage signals, eliminating the need for image reconstruction and reducing computational latency.Approach.The framework incorporates frequency-domain preprocessing to isolate the 2nd-8th drive-field harmonics, enhancing the signal-to-noise ratio while preserving motion-relevant features. TSMPI-Net: a time-series generative adversarial network for short-frame-interval magnetic particle imaging.

Objective.Magnetic particle imaging (MPI) is an emerging imaging modality that offers high sensitivity and the potential for high-speed imaging. In many MPI applications, the imaged object moves over time, requiring reconstruction of temporally coherent image sequences at short frame intervals. However, conventional MPI reconstruction methods, such as the system matrix method and X-space method, typically reconstruct frames in isolation and fail to fully exploit the temporal correlations essential for dynamic imaging, resulting in degraded temporal reconstruction quality. Recently developed de

Towards clinical magnetic particle imaging: safety measurements of medical implants in a human cadaver model.

Magnetic Particle Imaging (MPI) is a preclinical imaging modality with potential for future clinical usage. The radiation-free guidance of endovascular interventions with MPI is especially promising. Here, we present a safety study on the heating of metallic medical implants during MPI measurements under realistic conditions in an extracorporeally-perfused cadaver model. The measurements were conducted by fiberoptic thermometers and showed no detectable heating of the tested endovascular devices in the cadaver model. A temperature increase of no more than 0.11 K was observed on the surface of

Development of Iron Oxide Nanochains as a Sensitive Magnetic Particle Imaging Tracer for Cancer Detection.

The advancement of imaging technologies plays a crucial role in improving the diagnosis and monitoring of diseases, including cancer. This study introduces a new design of iron oxide-based nanoparticles specifically developed for magnetic particle imaging (MPI), aimed at tracking and diagnosing breast cancer more effectively. By precisely controlling the size, shape, and magnetic properties of these nanoparticles, we enhance the responsiveness of MPI, resulting in an increased signal. In our research, we established a novel synthetic route for fabricating iron oxide nanochains (FeONCs) charact

Magnetic particle imaging enables nonradioactive quantitative sentinel lymph node identification: feasibility proof in murine models.

BACKGROUND: Sentinel lymph node biopsy (SLNB) is an important cancer diagnostic staging procedure. Conventional SLNB procedures with 99mTc radiotracers and scintigraphy are constrained by tracer half-life and, in some cases, insufficient image resolution. Here, we explore an alternative magnetic (nonradioactive) image-guided SLNB procedure. PURPOSE: To demonstrate that magnetic particle imaging (MPI) lymphography can sensitively, specifically, and quantitatively identify and map sentinel lymph modes (SLNs) in murine models in multiple regional lymphatic basins. Distance measurements between 5 nanometer diamonds - single particle magnetic resonance or optical super-resolution imaging?

5 nanometer sized detonation nanodiamonds (DNDs) are studied as potential single-particle labels for distance measurements in biomolecules. Nitrogen-vacancy (NV) defects in the crystal lattice can be addressed through their fluorescence and optically-detected magnetic resonance (ODMR) of a single particle can be recorded. To achieve single-particle distance measurements, we propose two complementary approaches based on spin-spin coupling or optical super-resolution imaging. As a first approach, we try to measure the mutual magnetic dipole-dipole coupling between two NV centers in close DNDs us

Quantitative Magnetic Particle Imaging Monitors the Transplantation, Biodistribution, and Clearance of Stem Cells In Vivo.

Stem cell therapies have enormous potential for treating many debilitating diseases, including heart failure, stroke and traumatic brain injury. For maximal efficacy, these therapies require targeted cell delivery to specific tissues followed by successful cell engraftment. However, targeted delivery remains an open challenge. As one example, it is common for intravenous deliveries of mesenchymal stem cells (MSCs) to become entrapped in lung microvasculature instead of the target tissue. Hence, a robust, quantitative imaging method would be essential for developing efficacious cell therapies.

Detection of brain pathology by magnetic resonance imaging of iron oxide micro-particles.

Contrast agents are widely used with magnetic resonance imaging (MRI) to increase the contrast between regions of interest and the background signal, thus providing better quality information. Such agents can work in one of two ways, either to specifically enhance the signal that is produced or to localize in a specific cell type of tissue. Commonly used image contrast agents are typically based on gadolinium complexes or super-paramagnetic iron oxide, the latter of which is used for imaging lymph nodes. When blood-brain barrier (BBB) breakdown is a feature of central nervous system (CNS) path
