Pharmacological Potential and Molecular Targets of Tetrahydrofurofuranoid Lignans From Magnoliae Flos.

Tetrahydrofurofuranoid lignans (TFLs) are lignans extracted from the dried flower buds of Magnoliae Flos (MF). The chemical structures of the seven major constituents, magnolin, fargesin, aschantin, lirirresinol B dimethyl ether, epimognolin, eudesmin, and kobusin, are strikingly similar. Despite their potential therapeutic effects against various pathological conditions, their underlying mechanisms have not been systematically analyzed. The primary objective of this review was to examine the mechanisms by which TFLs exert therapeutic effects on inflammation, allergies, cancer, cardiovascular

Karst tiankengs preserve but constrain evolutionary potential in the endangered tree Magnolia aromatica.

Karst tiankengs-giant enclosed sinkholes-harbor humid, shaded forests that may buffer climate stress yet isolate populations. Here, we generated a near telomere-to-telomere genome assembly for the endangered karst tree Magnolia aromatica and resequenced 26 populations across tiankeng interiors and surrounding habitats in southwest China. Population genomics resolved four lineages and indicated a history of divergence with gene flow constrained by strong dispersal barriers. At the lineage scale, tiankeng-associated populations exhibited intermediate genomic diversity and mutation load relative

hERG channel blockade and additive interactions of magnolol and honokiol from Magnolia species.

The Magnolia officinalis cortex (the dried bark known as "Houpo") is widely used in traditional Chinese medicine (TCM). In this study, we discovered some potential off-target effects of M. officinalis due to the blocking effects on hERG (human Ether-a-go-go-Related Gene) channels by magnolol and honokiol, two main active chemicals of M. officialis. This research investigated the modulatory effects of two neolignans and their cooperative effects on hERG channels using manual whole-cell patch-clamp techniques. Results showed that magnolol and honokiol blocked hERG currents with IC50s of 5.2 ± 0.

Bioactive Neolignans and Other Compounds from Magnolia grandiflora L.: Isolation and Antiplasmodial Activity.

Bioassay-guided fractionation of a methanol extract of Magnolia grandiflora against Plasmodium falciparum yielded two new (1 and 2) and six known (3 - 8) bioactive compounds. The structures of the new compounds were assigned by mass spectrometric and 1D- and 2D-NMR data. Known compounds were identified by comparison of 1 H-NMR and MS data with literature data. The two known neolignans 3 and 4 showed moderate antiplasmodial activity with the IC50 values of 2.8 ± 0.1 and 3.4 ± 0.1 μm, respectively. Weak antiplasmodial activity was recorded for compounds 1, 2, 5, 6, 7, and 8, with the IC50 values

[The therapeutic effect of magnolia officinalis on blood stasis in Sheng Nong's Herbal Classic].

The efficacy of Hou po in treating blood stasis can be traced back to Sheng Nong's Herbal Classic. However, its relative explanations of medical principles in later generations are scarcely seen and most of them were based on its pungent and warm nature. This paper examined the descriptions of the therapeutic effects of Magnolia officinalis(Hou po) in traditional Chinese materia medica literature throughout its history, analysed the symptoms and pathogenesis of blood stasis in traditional Chinese medicine and referred to other herbs listed in Sheng Nong's Herbal Classic used for treating blood

[Geographical origin discrimination of Magnoliae Officinalis Cortex based on hyperspectral imaging technology].

As a commonly used bark medicinal material in clinical practice, Magnoliae Officinalis Cortex(MOC) is widely distributed across different production areas, with noticeable quality differences among origins. However, it is difficult to determine its origin based solely on macroscopic characteristics. In this study, MOC samples(stem bark collected above 1 m from trees aged 15-25 years, shade-dried) from 26 production areas in 8 provinces were selected as research objects. Hyperspectral data of the outer surface, cross-section, and inner surface were collected, and six preprocessing algorithms we

Phased chromosome-level genomes reveal young evolutionary origin of sex determination in Magnolia kwangsiensis.

While sex determination has been extensively studied in animals, the origin and early evolution of sex chromosomes in plants remain poorly understood. Magnolia kwangsiensis, an early-diverging dioecious flowering plant, provides a unique opportunity to investigate sex chromosome evolution at early stages. Here we generated phased chromosome-level genome assemblies for male and female M. kwangsiensis individuals (~2.4-2.5 Gb of 19 chromosomes) and uncovered a nascent XY sex chromosome system. The sex-determining region (SDR) is located on chromosome 4 and spans ~10.4 Mb on the X chromosome and

Apoptosis induced by Magnolia Grandiflora extract in chlorambucil-resistant B-chronic lymphocytic leukemia cells.

BACKGROUND: B-cell chronic lymphocytic leukemia (B-CLL) still remains as an uncurable disease. Even the newest antineoplastic agents have demonstrated limitations in their efficacy. For this reason, further research of new compounds must be done. New pharmacological properties can be obtained from a great diversity botanical species. Among these products, Magnolia Grandiflora receives our attention since it mainly contains Honokiol which had demonstrated effect against B-CLL cells activating different cell death pathways. AIM: To test the ability of Magnolia Grandiflora extracts to induce apo

Inhibitory effect of obovatol from Magnolia obovata on the Salmonella type III secretion system.

In many pathogenic Gram-negative bacteria, such as Salmonella, Escherichia coli, Yersinia and Chlamydia spp., which cause diseases in humans, the type III secretion system (TTSS) is an important virulence factor that translocates effector proteins into the cytosol of host cells. Thus, the TTSS is a good target for antibacterial agents. Here we used a hemolysis assay to search for TTSS inhibitors and found that a compound from Magnolia obovata called obovatol blocks the TTSS of Salmonella. Obovatol showed potent inhibitory activity (IC50=19.8 μM) against the TTSS-related hemolysis of Salmonella

Magnolia officinalis (L.) Bark Extract Counteracts Oxidative Brain Injury: A Proteomic Investigation into Neuroprotective Mechanisms.

Neurodegenerative diseases involve progressive neuronal loss associated with oxidative stress (OS) and inflammation. Given the limited efficacy of current therapies, natural compounds with multitarget neuroprotective potential are of growing interest. In this study, we investigated the neuroprotective effects of a standardized Magnolia officinalis (L.) bark extract (MOE) in rat brain cortical slices exposed to hydrogen peroxide-induced OS. MOE significantly recovered tissue viability and reduced ROS and malondialdehyde levels caused by OS while attenuating caspase-3, -8, and -9 activation, sug

Anti-biofilm and bactericidal effects of magnolia bark-derived magnolol and honokiol on Streptococcus mutans.

Dental caries affects people of all ages and is a worldwide health concern. Streptococcus mutans is a major cariogenic bacterium because of its ability to form biofilm and induce an acidic environment. In this study, the antibacterial activities of magnolol and honokiol, the main constituents of the bark of magnolia plants, toward planktonic cell and biofilm of S. mutans were examined and compared with those of chlorhexidine. The minimal inhibitory concentrations of magnolol, honokiol and chlorhexidine for S. mutans were 10, 10 and 0.25 µg/mL, respectively. In addition, each agent showed bacte

Anti-inflammatory effect of magnolol, isolated from Magnolia officinalis, on A23187-induced pleurisy in mice.

In the present study, A23187-induced pleurisy in mice was used to investigate the anti-inflammatory effect of magnolol, a phenolic compound isolated from Chinese medicine Hou p'u (cortex of Magnolia officinalis). A23187-induced protein leakage was reduced by magnolol (10 mg kg-1, i.p.), indomethacin (10 mg kg-1, i.p.) and BW755C (30 mg kg-1, i.p.). A23187-induced polymorphonuclear (PMN) leucocyte infiltration in the pleural cavity was suppressed by magnolol and BW755C, while enhanced by indomethacin.
