A 32-year-old contact lens-wearing man with recent travel history to the Caribbean was referred for a corneal infiltrate in the left eye that worsened following 1-week of steroid-antibiotic therapy. Corneal cultures were obtained and sent to our facility's clinical microbiology laboratory for analysis. Same-day in vivo confocal microscopy revealed fungal elements. Nucleic acid sequencing performed on the isolated determined it to be a member of the entomopathogenic genus Metarhizium. Over the course of 3 months, the patient's corneal infiltrate ultimately resolved following topical natamycin 5

Ticks exert a significant economic impact on the livestock industry, particularly in Asian regions. Presently, chemical acaricides constitute the primary method employed to combat tick infestations in livestock, but their use carries adverse environmental consequences. Overreliance on acaricides has contaminated milk and meat products with chemical residues while fostering tick resistance to these agents due to improper and intensive application. Various alternative methods have been explored, including using vaccines to manage tick populations. However, the efficacy of these treatments varies

The current study examined the impact of ultraviolet (UV)-B radiation in Metarhizium pingshaense blastospores' photolyase expression and their virulence against Rhipicephalus microplus. Blastospores were exposed to UV under laboratory and field conditions. Ticks were treated topically with fungal suspension and exposed to UV-B in the laboratory for three consecutive days. The expression of cyclobutane pyrimidine dimmers (CPDs)-photolyase gene maphr1-2 in blastospores after UV exposure followed by white light exposure was accessed after 0, 8, 12, 24, 36, and 48 h. Malaria control efforts require implementation of new technologies that manage insecticide resistance. Metarhizium pingshaense provides an effective, mosquito-specific delivery system for potent insect-selective toxins. A semifield trial in a MosquitoSphere (a contained, near-natural environment) in Soumousso, a region of Burkina Faso where malaria is endemic, confirmed that the expression of an insect-specific toxin (Hybrid) increased fungal lethality and the likelihood that insecticide-resistant mosquitoes would be eliminated from a site. Also, as Hybrid-expressing M. pingshaense is effectiv

Salicylic acid (SA), a cell-signaling metabolite in plants, is involved in resistance of plants to pathogens and environmental stresses; however, there is little information available on the responses of fungi to SA. Conidia of Metarhizium robertsii (ARSEF 2575) (Hypocreales: Clavicipitaceae) were produced on potato dextrose agar medium plus yeast extract (PDAY) supplemented with 1, 2, 4, or 8 mM SA (pH adjusted to 6.9) and incubated under constant-dark conditions. Then the tolerance of conidia against wet heat (45 °C, 3 h) and UV-B radiation (7.0 kJ m(-2)) was tested. For comparison, conidia

Differential immunoproteomics was applied to identify proteins secreted by Metarhizium anisopliae induced by the Rhipicephalus microplus cuticle. In addition, IgG anti-spore surface proteins were used for searching for proteins possibly involved in early stages of fungus versus tick infection. LC-MS/MS of differentially secreted proteins led to the identification of proteases (carboxypeptidase and Pr1A), chitinase, carboxylic acid transport and proline-rich protein. Differential immunoproteomics strategy facilitated the detection and the identification of new proteins related to M. anisopliae

Metarhizium spp. are widely used as biocontrol agents against insect pests, but the chemical basis underlying their nematicidal activity remains largely unknown. Here, bioassay-guided fractionation and GC-MS identified dimethyl disulfide (DMDS) and dimethyl trisulfide (DMTS) as the major volatile nematicidal compounds produced by Metarhizium anisopliae. Both compounds exhibited concentration-dependent activity against three plant-parasitic nematodes (PPNs), with 24 h LC50 values of 0.19-1.38 μg/mL for DMDS and 0.04-0.07 μg/mL for DMTS. In tomato pot trials, the fermentation broth achieved 79.4

To date, there has been only one published report on the infectious sclerokeratitis caused by Metarhizium anisopliae, which is an entomopathogenic fungus. Regarding corneal infection, three reports have been published to date. Although the prognoses of the corneal infections are favourable, prognosis when scleral infection is involved is very poor. A 76-year-old patient presented with foreign body sensation in the left eye. Microscopic examination with Fungi Flora Y staining of the corneal scraping revealed fungal infection. The conjunctiva was melted by the infection over a wide area. Althoug

In the contemporary context of agricultural pest management, bioinsecticides have emerged as a sustainable alternative to conventional chemical insecticides, representing a growing trend of significance. Gryllus bimaculatus, a species of Orthoptera that serves as a valuable model for studying immune responses to fungal bioinsecticide infection, thereby contributing to a deeper understanding of insect immune mechanisms within this order. In this study, the expression patterns of immune responses in G. bimaculatus were analysed at three critical time points following infection with the bioinsect

Tick-borne diseases (TBDs) pose a major threat to global livestock production, particularly in tropical and subtropical regions, and their burden is projected to increase worldwide in the coming decades as a result of climate change. In Brazil, tick infestations are responsible for economic losses estimated at about USD 3.24 billion annually. Growing resistance to chemical acaricides necessitates alternative control strategies such as biological agents. This study assessed the pathogenicity of a local strain of Metarhizium anisopliae (PR1) against adult Rhipicephalus microplus ticks using the

Fungi are central to every terrestrial and many aquatic ecosystems, but the mechanisms underlying fungal tolerance to mercury, a global pollutant, remain unknown. Here, we show that the plant symbiotic fungus Metarhizium robertsii degrades methylmercury and reduces divalent mercury, decreasing mercury accumulation in plants and greatly increasing their growth in contaminated soils. M. robertsii does this by demethylating methylmercury via a methylmercury demethylase (MMD) and using a mercury ion reductase (MIR) to reduce divalent mercury to volatile elemental mercury. M. robertsii can also rem

The bovine tick, Rhipicephalus microplus, is the main ectoparasite of cattle and causes loss of billions of dollars worldwide in lost meat, milk, and leather production, as well as control expenses. In addition to systemically impacting the host during the parasitic act, this parasite is also an important disease vector. Traditionally, the main commercial control of the tick is achieved through application of chemical acaricides, which can leave residues in the meat and milk. Moreover, ticks can become resistant to these chemicals due to their massive and incorrect use. Many alternative method

Aedes aegypti is the vector of a wide range of diseases (e.g. yellow fever, dengue, Chikungunya and Zika) which impact on over half the world's population. Entomopathogenic fungi such as Metarhizium anisopliae and Beauveria bassiana have been found to be highly efficacious in killing mosquito larvae but only now are the underlying mechanisms for pathogenesis being elucidated. Recently it was shown that conidia of M. anisopliae caused stress induced mortality in Ae. aegypti larvae, a different mode of pathogenicity to that normally seen in terrestrial hosts. Blastospores constitute a different

Susceptibility of Phlebotomus papatasi Scopoli (Diptera: Psychodidae) larvae to the entomopathogenic fungus Metarhizium anisopliae (Metschinkoff) Sorokin (Ma79) (Hypocreales: Clavicipitaceae) was evaluated at two different temperatures. The ability of the fungus to reinfect healthy sand flies was followed up for approximately 20 wk and the effect of in vivo repassage on the enhancement of its virulence was assessed. The fungus reduced the adult emergence at 26 +/- 1 degrees C when applied to larval diet. Six spore concentrations were used in the bioassays ranging from 1 x 10(6) to 5 x 10(8) sp

Conidial germination and the formation of appressoria are important events in the interactions between entomopathogenic fungi and their arthropod hosts. In this study, we demonstrate the effects of lipids extracted from tick epicuticle and the surface of a mammalian host (calf) on conidial germination and the development of appressoria in two subspecies of Metarhizium anisopliae, M. anisopliae var. anisopliae (M.an.an.-7) and M. anisopliae var. acridum (M.an.ac.-5), which have different levels of virulence toward ticks. Pentane extracts of epicuticles of ticks susceptible and resistant to fung

Fungus-growing termites harbour diverse microbial communities in their combs and guts. However, culture-based surveys, cross-colony and cross-regional comparisons and assessments of their antimicrobial activities against entomopathogens and fungal weeds remain limited. Through culture-dependent isolation, we surveyed the bacterial and fungal diversity associated with the combs and guts of termites in the genera Macrotermes and Odontotermes, recovering 600 non-Termitomyces fungal isolates representing 79 genera, as well as 59 bacterial isolates. Pathogenicity tests on Macrotermes gilvus reveale

The environment in which a fungus grows directly influences fungal traits such as development, transmission, and pathogenic potential. The environmental niche encompasses factors such as nutrient availability, biotic and abiotic stressors, as well as host-derived chemical cues. In fungal pathogens, where conidia act as the infectious agents, the environmental niche impacts the quantity and quality of the produced conidia, thereby affecting the ability of conidia to infect and kill hosts. In the present study, we investigated the effect of host-derived medium types on various phenotypes, includ
