A modified vaccinia Ankara vaccine vector expressing a mosaic H5 hemagglutinin reduces viral shedding in rhesus macaques.

The rapid antigenic evolution of influenza viruses requires frequent vaccine reformulations. Due to the economic burden of continuous vaccine reformulation and the threat of new pandemics, there is intense interest in developing vaccines capable of eliciting broadly cross-reactive immunity to influenza viruses. We recently constructed a "mosaic" hemagglutinin (HA) based on subtype 5 HA (H5) and designed to stimulate cellular and humoral immunity to multiple influenza virus subtypes. Modified vaccinia Ankara (MVA) expressing this H5 mosaic (MVA-H5M) protected mice against multiple homosubtypic

Argonaute 5-mediated antiviral defense and viral counter-defense in Nicotiana benthamiana.

The argonaute (AGO) family proteins play a crucial role in preventing viral invasions through the plant antiviral RNA silencing pathway, with distinct AGO proteins recruited for specific antiviral mechanisms. Our previous study revealed that Nicotiana benthamiana AGO5 (NbAGO5) expression was significantly upregulated in response to bamboo mosaic virus (BaMV) infection. However, the roles of NbAGO5 in antiviral mechanisms remained to be explored. In this research, we examined the antiviral functions of NbAGO5 in the infections of different viruses. It was found that the accumulation of NbAGO5 w

Construction and Characterization of an Attenuated Vaccine Vector Based on Cucumber Mosaic Virus RNA2.

Plant viruses are major plant pathogens and account for nearly half of emerging plant diseases worldwide. To date, few effective management measures are available for the efficient control of plant viral diseases. Cross-protection based on attenuated vaccine is an effective strategy to prevent plant viral diseases. The primary requirement for the development of attenuated vaccines is a vector with excellent characteristics, such as low pathogenicity, genetic stability, and the capacity for stable insertion of exogenous fragments. In this study, the cucumber mosaic virus (Fny strain) was geneti

Whole genome sequencing of recombinant viruses obtained from co-infection and superinfection of Vero cells with modified vaccinia virus ankara vectored influenza vaccine and a naturally occurring cowpox virus.

Modified vaccinia virus Ankara (MVA) has been widely tested in clinical trials as recombinant vector vaccine against infectious diseases and cancers in humans and animals. However, one biosafety concern about the use of MVA vectored vaccine is the potential for MVA to recombine with naturally occurring orthopoxviruses in cells and hosts in which it multiplies poorly and, therefore, producing viruses with mosaic genomes with altered genetic and phenotypic properties. We previously conducted co-infection and superinfection experiments with MVA vectored influenza vaccine (MVA-HANP) and a feline C

Emerging Viruses Associated with Chayote (Sechium edule): Current Knowledge, Perspectives, Insights, Challenges, and Future Directions.

Chayote (Sechium edule) is an important cucurbit crop grown in tropical and subtropical climates for its edible fruits, shoots, and roots. Its production faces growing threats from various viral pathogens, including Tomato leaf curl New Delhi virus, Squash leaf curl Philippines virus, Zucchini yellow mosaic virus, and Tomato spotted wilt virus which are in emerging trend. The viruses spread by vectors such as whiteflies, aphids, and thrips are predominant in agroecosystems where chayote is grown. Viral infection can cause symptoms such as mosaic patterns, leaf curling, stunting, and necrosis,

Occurrence and Distribution of Major Viruses Infecting Passion Fruit in Guizhou Province, China, and Molecular Characterization of Two Potyviruses.

The planting of passion fruit (Passiflora edulis) in Guizhou Province has gradually increased, and the area under cultivation ranks third in China. However, the cultivation and production of passion fruit is severely affected by viral diseases. In 2021 and 2022, we investigated the occurrence of multiple viral diseases in major cultivation areas, identified the main viruses and conducted field surveys in different growing areas of passion fruit in Guizhou Province, China. In total, 308 samples were randomly collected from 10 different passion fruit cultivation areas, and seven viral diseases w

A Polysaccharide Derived from a Trichosporon sp. Culture Strongly Primes Plant Resistance to Viruses.

Plant viruses cause devastating diseases in plants, yet no effective viricide is available for agricultural application. We screened cultured filtrates derived from various soil microorganisms cultured in vegetable broth that enhanced plant viral resistance. A cultured filtrate, designated F8 culture filtrate, derived from a fungus belonging to the genus Trichosporon, induced strong resistance to various viruses on different plants. Our inoculation assay found the infection rate of Tobacco mosaic virus (TMV)-inoculated Nicotiana benthamiana with F8 culture filtrate pretreatment may decrease to

Pigeonpea sterility mosaic virus: a legume-infecting Emaravirus from South Asia.

UNLABELLED: Pigeonpea sterility mosaic virus (PPSMV), a species of the genus Emaravirus, is the causal agent of sterility mosaic disease (SMD) of pigeonpea [Cajanus cajan (L.) Millsp]. This disease, dubbed the 'green plague', as the infected plants remain in the vegetative state without flower production, has been reported from India and a few other South-East Asian countries. SMD is estimated to result in an annual yield loss of over US$300 million in India alone. The aetiology of SMD, which remained a mystery for over 70 years, was resolved with the discovery of PPSMV in 2000 and its complet

Development of a highly efficient virus-free regeneration system of Salvia miltiorrhiza from Sichuan using apical meristem as explants.

BCAKGROUND: The dry root and rhizome of Salvia miltiorrhiza are used to treat cardiovascular diseases, chronic pain, and thoracic obstruction over 2000 years in Asian countries. For high quality, Sichuan Zhongjiang is regarded as the genuine producing area of S. miltiorrhiza. Given its abnormal pollen development, S. miltiorrhiza from Sichuan (S.m.-SC) relies on root reproduction and zymad accumulation; part of diseased plants present typical viral disease symptoms and seed quality degeneration. This study aim to detected unknown viruses from mosaic-diseased plants and establish a highly effic

Genomic surveillance of an imported recombinant Clade Ib and IIb monkeypox virus case in India.

Double-stranded DNA viruses can recombine, such as the monkeypox virus (MPXV), but only one recombinant MPXV has been reported previously in the UK. Here we characterize a recombinant Clade Ib and IIb MPXV isolated from a travel-associated mpox case in India. The mosaic genome contains clade-defining mutations from both Ib and IIb and clusters with the UK recombinant. Both recombinant viruses show 34 recombinant tracts and 21 recombinant lineage-associated mutations. This has implications for genomic surveillance and diagnostics.

Sec62 restricts ER-replicating positive-strand RNA virus infections via UPR-dependent ER-phagy.

Positive-strand RNA [(+)RNA] viruses induce endomembrane remodeling to form viral replication organelles (VROs), which disrupt organelle homeostasis. How hosts restore organelle homeostasis and how these responses influence viral replication remain elusive. Using beet black scorch virus (BBSV), a (+)RNA virus that replicates on the endoplasmic-reticulum (ER) and induces severe deformation of ER membranes, as a model in Nicotiana benthamiana, we demonstrated that BBSV induces ER-phagy, primarily mediated by its auxiliary replication protein p23. p23 interacts with the ER-phagy receptor NbSec62,

Evolutionary dynamics of HIV-1 recombinants: analysis of contemporary and historical viral populations in East Africa.

BACKGROUND: Understanding the genetic evolution of HIV-1 Transmitted/Founder (T/F) virus is crucial for developing effective treatment and prevention strategies due to its rapid mutation and recombination rates. METHODS: This study compared the genetic diversity of 24 contemporary T/F viruses collected between 2016 and 2021 in Uganda and Kenya with 29 historical T/F sequences sampled between 2006 and 2011. RESULTS: Subtype analysis based on near-full-length (NFL) HIV-1 T/F genomes revealed that 57.1% (12/21) of contemporary viruses were recombinants, predominantly involving Subtype A1, D, an

Facing the future of respiratory virus surveillance: "The mosaic surveillance framework".

It is impossible to address the many complex needs of respiratory virus surveillance with a single system. Therefore, multiple surveillance systems and complementary studies must fit together as tiles in a "mosaic" to provide a complete picture of the risk, transmission, severity, and impact of respiratory viruses of epidemic and pandemic potential. Below we present a framework (WHO Mosaic Respiratory Surveillance Framework) to assist national authorities to identify priority respiratory virus surveillance objectives and the best approaches to meet them; to develop implementation plans accordi

Dahlia Virome Analysis Reveals Multiple Novel Viruses in Mixed Infections.

Dahlia is an economically important ornamental crop in the United States and worldwide. Dahlia is primarily propagated vegetatively through bulbs, which can result in vertical transmission of and constant accumulation of viruses. In this study, mosaic and yellowing symptoms were observed in Dahlia plants in a residential garden in California. Dahlia samples were collected and sequenced using high-throughput sequencing (HTS). HTS analysis was used to explore the Dahlia virome in the symptomatic plants, which revealed the presence of several contigs with homology to known Dahlia-infecting viruse

CRISPR-Cas13-based amplification-free detection of three quarantine-significant sugarcane viruses.

Crop losses from viral pathogens cost billions of dollars annually, and sugarcane is particularly vulnerable. Sugarcane is the most cultivated crop in the world by volume; therefore, significant yield losses result in large dollar losses for farmers and trade economies. In some cases, viruses contribute to more than half of the yield reduction. Three quarantine-significant pathogens-Sugarcane mosaic virus (ScMV), Sugarcane streak mosaic virus (ScSMV), and Sugarcane yellow leaf virus (ScYLV)- pose major threats to global sugarcane production because of their rapid spread, asymptomatic infection

Mosaic Inverted Hemagglutinin Extracellular Vesicle Vaccines Elicit Protective Systemic and Mucosal Immunity against Heterosubtypic Influenza Infection.

Developing innovative vaccine platforms and delivery strategies to induce broad, protective immunity in the respiratory tract is crucial for preventing influenza infection and transmission in potential epidemics and pandemics. In this study, we used cell-derived extracellular vesicles (EVs) as a vaccine platform to display mosaic human and avian influenza hemagglutinins (HAs) concurrently, including HA1/HA9/HA12 or HA3/HA4/HA10, on the EV surfaces. Immunization with the mosaic HA-EV vaccine elicited cross-reactive antibodies against influenza HA stalks and viruses, robust virus-specific cellul

Mutants of sindbis virus. IV. Heterotypic complementation and phenotypic mixing between temperature-sensitive mutants and wild-type Sindbis and Western equine encephalitis viruses.

Heterotypic complementation between temperature-sensitive (ts) mutants of Sindbis (SIN) and Western equine encephalitis (WEE) viruses occurs under appropriate conditions. One heterotypic pair, SIN ts153 X WEE ts39 showed efficient complementation, and four other combinations gave detectable complementation, indicating that these two viruses, which are closely related serologically and biochemically, are sufficiently closely related to complement each other functionally. Cells mixedly infected with ts mutants or wild-type strains of both SIN and WEE viruses produced phenotypically mixed virions

One-Step Supramolecular Multifunctional Coating on Plant Virus Nanoparticles for Bioimaging and Therapeutic Applications.

Plant viral nanoparticles (plant VNPs) are promising biogenetic nanosystems for the delivery of therapeutic, immunotherapeutic, and diagnostic agents. The production of plant VNPs is simple and highly scalable through molecular farming in plants. Some of the important advances in VNP nanotechnology include genetic modification, disassembly/reassembly, and bioconjugation. Although effective, these methods often involve complex and time-consuming multi-step protocols. Here, we report a simple and versatile supramolecular coating strategy for designing functional plant VNPs via metal-phenolic net
