Computational network toxicology of non-nutritive sweeteners in ulcerative colitis: From Aspartame-MMP9 interaction to mechanism-guided intervention.

BACKGROUND: Non-nutritive sweeteners (NNS), including aspartame, sucralose, and saccharin, are widely consumed globally. Ulcerative colitis (UC) manifests as epithelial barrier dysfunction accompanied by complex metabolic and immune system remodeling. However, the mechanistic link between NNS exposure and UC, along with clinically interpretable biomarkers, remains unclear. METHODS: We integrated NNS ADMET properties and aggregated targets from six chemogenomic resources. Subsequently we scored 114 metabolic pathways using ssGSEA and classified metabolic reprogramming subtypes via consensus cl

Environmental impact ramifications of replacing added sugar with non-nutritive sweeteners in a high-calorie Dutch diet: the SWEET project.

PURPOSE: Growing concern about excess added sugar intake and associated adverse health effects has increased interest in non-nutritive sweeteners (NNS) as low-calorie alternatives that preserve sweetness. Previous studies focus on health effects of replacing added sugar, or environmental impacts of NNS alone. However, none evaluate the environmental impact ramifications of replacing added sugar with NNS in a whole dietary setting. This study addresses that gap. METHODS: The baseline diet reflected an average diet of Dutch adults consuming > 2200 kcal/day (mean 2866 kcal/day). Added sugar (54

Effects of non-nutritive sweeteners on growth and intestinal health by regulating hypothalamic RNA profile and ileum microbiota in guinea pigs.

BACKGROUND: Non-nutritive sweeteners (NNS) are commonly used in sweetened foods and beverages; however their role in metabolic regulation is still not clear. In this experiment, we used guinea pigs as an animal model to study the effect of NNS on body growth and intestinal health by modifying gut microbiota and hypothalamus-related proteins. RESULTS: For a 28-day feeding experiment a total of 40 guinea pigs were randomly divided into four groups, one control (CN) group and three treatments, in which three NNS were added to the diet: rebaudioside A (RA, 330 mg kg-1), sodium saccharin (SS, 800

The effects of non-nutritive sweeteners on lipid profile in adults: a systematic review and meta-analysis of randomized controlled clinical trials.

Non-nutritive sweeteners (NNS) have been suggested as a viable solution to reduce sugar consumption. However, conflicting and uncertain evidence regarding their impact on lipid profile exists. This meta-analysis assesses the effects of different types of NNS on lipid profile markers. PubMed, Web of Science ISI, Scopus, and Google Scholar were searched for relevant articles up to September 2024. The Cochrane Collaboration tool was used for the quality assessment, and the evidence's trustworthiness was assessed using the Grading of Assessment, Development, and Evaluation of Recommendations (GRAD

The non-nutritive sweetener erythritol adversely affects brain microvascular endothelial cell function.

The experimental aim of this study was to determine, in vitro, the effect of the non-nutritive sweetener erythritol on brain microvascular endothelial cell oxidative stress, nitric oxide (NO) and endothelin (ET)-1 production, as well as tissue-type plasminogen activator (t-PA) release. Human cerebral microvascular endothelial cells (hCMECs) were cultured and treated with 6 mM erythritol, equivalent to a typical amount of erythritol (30 g) in an artificially sweetened beverage, for 3 h. Intracellular reactive oxygen species (ROS) production was significantly higher in hCMECs treated with erythr

Consumption of Non-Nutritive Sweetener during Pregnancy and Weight Gain in Offspring: Evidence from Human Studies.

The relationship between the consumption of maternal non-nutritive sweeteners (NNS) during pregnancy and the risk of obesity in offspring remains inconsistent. We aimed to systematically evaluate and clarify the relationship between NNS intake during pregnancy and weight gain in offspring based on evidence from population and clinical research. Databases including PubMed (via Medline), EMBASE, and the Cochrane Library were systematically searched for eligible human studies. The primary outcome was the differences in body mass index (BMI) z-scores between offspring at 1 year of age who were wit

Non-nutritive sweeteners and body weight in children with type 1 and type 2 diabetes: a critical narrative review of current evidence, methodological challenges, biological mechanisms, and clinical implications.

INTRODUCTION: Body weight management is an important component of pediatric diabetes care, although its clinical relevance differs between type 1 diabetes (T1D) and type 2 diabetes (T2D) because of distinct pathophysiology, insulin physiology, and therapeutic goals. Non-nutritive sweeteners (NNSs) are widely used to replace added sugars, reducing glycaemic load and, in some settings, energy intake. However, their long-term effects on body weight in children with diabetes remain controversial. This narrative review evaluates current evidence on the association between NNS consumption and body w

Differential fructose and glucose appetition in DBA/2, 129P3 and C57BL/6 × 129P3 hybrid mice revealed by sugar versus non-nutritive sweetener tests.

Inbred mouse strains differ in their postoral appetite stimulating response (appetition) to fructose as demonstrated in intragastric (IG) sugar conditioning and oral sugar vs. nonnutritive conditioning experiments. For example, FVB and SWR strains show experience-induced preferences for 8% fructose over a 0.1% sucralose + 0.1% saccharin (S + S) solution, whereas C57BL/6 (B6) and BALB/c strains do not. All strains, however, learn to prefer 8% glucose to S + S after experience, which is attributed to the potent appetition actions of this sugar. The present study extended this analysis to DBA/2 (

Bioinspired chitosan-xanthan gum self-assembled nanosponge incorporating natural deep eutectic solvent: batch adsorption, mechanism, sustainability, and selective non-nutritive sweetener elimination.

A bio-inspired self-assembled nanosponge, constructed from a binary chitosan-xanthan gum (CX) gelling network incorporating a [betaine]:[tannic acid] natural deep eutectic solvent (NADES), is introduced as an innovative platform for the selective tracing of emerging non-nutritive sweeteners (NNSs) in waters and non-alcoholic beverages, due to their growing environmental relevance. The molar ratio of NADES precursors, CS-to-XG weight proportion, and NADES fraction within the CX network were optimized. Physicochemical characterization corroborated that the nanosponge possesses a porous, channel-

Autoimmune Thyroiditis with Hypothyroidism Induced by Sugar Substitutes.

The use of sugar substitutes (artificial sweeteners or non-nutritive sweeteners) has increased dramatically in the past few decades. They have been used as a substitute for sucrose (table sugar) in various diet-related disorders. Their excessive use has been linked to hyperphagia and obesity-related disorders. Hashimoto's thyroiditis (chronic autoimmune thyroiditis) is a disease that involves the immune-mediated destruction of the thyroid gland, gradually leading to its failure. Animal studies report that artificial sweeteners affect the immune system. Moreover, animal studies show that sucral

Low- and no-calorie sweeteners and health: unravelling the evidence and controversy.

Low- and no-calorie sweeteners (LNCS) (also referred to as non-nutritive sweeteners) have been the subject of controversy, especially considering recent conditional recommendations by the World Health Organization (WHO) not to use LNCS as a means of controlling weight and disease risk reduction. We summarize the data presented and discussions held between a panel of international nutrition research experts at a high-level Canadian Nutrition Society Thematic conference on "Non-nutritive sweeteners and health: unravelling the evidence and controversy", which took place in Toronto, Canada on 27 J

Sweeteners Maintain Epithelial Barrier Function Through the miR-15b/RECK/MMP-9 Axis, Remodel Microbial Homeostasis, and Attenuate Dextran Sodium Sulfate-Induced Colitis in Mice.

Non-nutritive sweeteners are the most widely used food additives designed to provide sweetness and reduce caloric intake. Studies have confirmed a link between sweeteners and colitis, yet supporting scientific data remain exiguous and controversial. In this study, three common sweeteners (Saccharin sodium, Stevioside, and Sucralose) in acceptable daily intake dosage were added to water in order to determine their effects on dextran sodium sulfate-induced colitis in mice. Our results show that the three sweeteners meliorate colitis to varying degrees─Saccharin exerts the most pronounced effect,

Endocrine and Metabolic Impact of Oral Ingestion of a Carob-Pod-Derived Natural-Syrup-Containing D-Pinitol: Potential Use as a Novel Sweetener in Diabetes.

The widespread use of added sugars or non-nutritive sweeteners in processed foods is a challenge for addressing the therapeutics of obesity and diabetes. Both types of sweeteners generate health problems, and both are being blamed for multiple complications associated with these prevalent diseases. As an example, fructose is proven to contribute to obesity and liver steatosis, while non-nutritive sweeteners generate gut dysbiosis that complicates the metabolic control exerted by the liver. The present work explores an alternative approach for sweetening through the use of a simple carob-pod-de

Rationale, Design and Participants Baseline Characteristics of a Crossover Randomized Controlled Trial of the Effect of Replacing SSBs with NSBs versus Water on Glucose Tolerance, Gut Microbiome and Cardiometabolic Risk in Overweight or Obese Adult SSB Consumer: Strategies to Oppose SUGARS with Non-Nutritive Sweeteners or Water (STOP Sugars NOW) Trial and Ectopic Fat Sub-Study.

BACKGROUND: Health authorities are near universal in their recommendation to replace sugar-sweetened beverages (SSBs) with water. Non-nutritive sweetened beverages (NSBs) are not as widely recommended as a replacement strategy due to a lack of established benefits and concerns they may induce glucose intolerance through changes in the gut microbiome. The STOP Sugars NOW trial aims to assess the effect of the substitution of NSBs (the "intended substitution") versus water (the "standard of care substitution") for SSBs on glucose tolerance and microbiota diversity.
