Industrial applications of Phanerochaete chrysosporium lignin-degrading enzymes: current status, production challenges, and future directions.

Phanerochaete chrysosporium (Pc) is a white-rot fungus recognized for its highly efficient lignin-degrading enzymes (LDEs), including lignin peroxidase (LiP), manganese peroxidase (MnP), and laccase. These oxidative enzymes possess transformative capabilities across multiple industrial applications, such as biopulping, biofuel production, bioremediation and for the treatment of industrial wastewater. However, the commercial use of these enzymes is limited due to time-consuming scale-up procedures in native hosts, instability in industrial environments, and high production costs. The recent dev

Manganese deficiency can replace high oxygen levels needed for lignin peroxidase formation by Phanerochaete chrysosporium.

The combined effects of Mn and oxygen on lignin peroxidase (LIP) activity and isozyme composition in Phanerochaete chrysosporium were studied by using shallow stationary cultures grown in the presence of limited or excess N. When no Mn was added, LIP was formed in both N-limited and N-excess cultures exposed to air, but no LIP activity was observed at Mn concentrations greater than 13 mg/liter. In oxygen-flushed, N-excess cultures, LIP was formed at all Mn concentrations, and the peak LIP activity values in the extracellular fluid were nearly identical in the presence of Mn concentrations rang

Polyvinyl alcohol-immobilized Phanerochaete chrysosporium and its application in the bioremediation of composite-polluted wastewater.

A novel biosorbent, polyvinyl alcohol (PVA)-immobilized Phanerochaete chrysosporium, was applied to the bioremediation of composite-polluted wastewater, containing both cadmium and 2,4-dichlorophenol (2,4-DCP). The optimum removal efficiency achieved was 78% for Cd(II) and 95.4% for 2,4-DCP at initial concentrations of 20 mg/L Cd(II) and 40 mg/L 2,4-DCP. PPBs had significantly enhanced the resistance of P. chrysosporium to 2,4-DCP, leading to the degradation rates of 2,4-DCP beyond 90% with varying initial 2,4-DCP concentrations. This research demonstrated that 2,4-DCP and secreted proteins mi

Transcriptional effect of a calmodulin inhibitor, W-7, on the ligninolytic enzyme genes in Phanerochaete chrysosporium.

We investigated the effects of a calmodulin (CaM) inhibitor, W-7, on the expression of lignin peroxidase (LiP) and manganese peroxidase (MnP) genes in Phanerochaete chrysosporium to consider the role of cam gene, which was upregulated in parallel with the total activities of LiP and MnP in our previous transcriptomic analysis. The addition of 100 μM W-7 to the fungal cultures repressed the total activities of LiP and MnP, whereas the addition of 100 μM W-5, which is a control drug of W-7, retained approximately half of them, indicating that the effect of W-7 was attributable to CaM inhibition.

Changes in the gene expression of the white rot fungus Phanerochaete chrysosporium due to the addition of atropine.

We constructed a LongSAGE (Long Serial Analysis of Gene Expression) library from a 3-d culture of Phanerochaete chrysosporium supplemented with atropine, which inhibits the production of lignin-degrading enzymes. The library (the atropine library) contains 13,108 LongSAGE tags and 6,783 unique tags. The gene expression profile represented by the tags was compared with those of two previously constructed libraries, one of which was constructed using 2-d cultures in which the fungus had not yet produced ligninolytic enzymes (the 2-d library) and the other was constructed using 3-d cultures in wh

Hydrogen sulfide and calcium effects on cadmium removal and resistance in the white-rot fungus Phanerochaete chrysosporium.

Hydrogen sulfide (H2S), an emerging gas transmitter, has been shown to be involved in multiple intracellular physiological and biochemical processes. In this study, the effects of hydrogen sulfide coupled with calcium on cadmium removal and resistance in Phanerochaete chrysosporium were examined. The results revealed that H2S enhanced the uptake of calcium by P. chrysosporium to resist cadmium stress. The removal and accumulation of cadmium by the mycelium was reduced by H2S and Ca2+ pretreatment. Moreover, oxidative damage and membrane integrity were alleviated by H2S and Ca2+. Corresponding

Alleviation of heavy metal and silver nanoparticle toxicity and enhancement of their removal by hydrogen sulfide in Phanerochaete chrysosporium.

Hydrogen sulfide (H2S), an important cellular signaling molecule, plays vital roles in mediating responses to biotic/abiotic stresses. Influences of H2S on metal removal, cell viability, and antioxidant response of Phanerochaete chrysosporium upon exposure to heavy metals and silver nanoparticles (AgNPs) in the present study were investigated. An enhancement in Pb(ΙΙ) removal with an increase in concentration of the H2S donor sodium hydrosulfide (NaHS) was observed, and the maximum removal efficiencies increased by 31% and 17% under 100 and 200 mg/L Pb(ΙΙ) exposure, respectively, in the presen

Porous Carbon Sponge from White-Rot Fungus Phanerochaete chrysosporium for the Removal of Oils and Organic Solvents.

Oil leakage incidentally occurs and leads to environmental disasters. Because of their porous and hydrophobic characteristics, graphene sponges are often studied as an oil adsorbent to repair oil spills at sea. Graphene materials are very expensive, and their biological toxicity has been given serious concerns; however, the easier preparation and eco-friendly, biomass-derived porous carbon materials can be used as an alternative to graphene materials. In this study, we prepared a porous carbon sponge (PCS) for oil and organic solvent removal by carbonizing white-rot fungus Phanerochaete chryso

Mechanisms of gradient enzymatic conversion of lignin by a dual-fungal system consisting of Phanerochaete chrysosporium and Pleurotus eryngii.

Chinese Distillers' grains (CDGs), a typical agro-industrial byproduct, contain a highly recalcitrant lignin that limits the utilization. In this study, a dual-fungal solid-state fermentation system consisting of Phanerochaete chrysosporium and Pleurotus eryngii was constructed for lignin conversion in CDGs. The lignin degradation ratio reached 58.66%, indicating that the system can effectively convert lignin. Laccase (Lac) and manganese peroxidase (MnP) activities peaked at 10 d, reaching 38.54 and 50.36 U/g, respectively, whereas lignin peroxidase (LiP) reached its maximum activity (51.67 U/

Elucidation of a novel fungal biotransformation pathway of ibuprofen by Phanerochaete sordida YK-624 under non-ligninolytic conditions.

Ibuprofen, a widely used non-steroidal anti-inflammatory drug (NSAID), has been detected in the environment and is known to contaminate waterbodies. Here, we investigated the biotransformation of ibuprofen using the hyper-lignin-degrading fungus Phanerochaete sordida YK-624. This fungus exhibited excellent transformation capacity, removing 75% of ibuprofen within 5 h of incubation at a final concentration of 100 μM. Three novel metabolites of this transformation were isolated, and their structures were determined by nuclear magnetic resonance and high-resolution electrospray ionization mass sp

Synergistic lignin degradation between Phanerochaete chrysosporium and Fenton chemistry is mediated through iron cycling and ligninolytic enzyme induction.

Removal of recalcitrant lignin from wastewater remains a critical bottleneck in multiple aspects relating to microbial carbon cycling ranging from incomplete treatment of biosolids during wastewater treatment to limited conversion of biomass feedstock to biofuels. Based on previous studies showing that the white rot fungus Phanerochaete chrysosporium and Fenton chemistry synergistically degrade lignin, we sought to determine optimum levels of Fenton addition and the mechanisms underlying this synergy. We tested the extent of degradation of lignin under different ratios of Fenton reagents and f

Exogenous microbial antagonism affects the bioaugmentation of humus formation under different inoculation using Trichoderma reesei and Phanerochaete chrysosporium.

This study was aimed at exploring the effect of antagonism of Trichoderma reesei (T.r) and Phanerochaete chrysosporium (P.c) on humification during fermentation of rice (RS) and canola straw (CS). Results showed that exogeneous fungi accelerated straw degradation and enzyme activities of CMCase, xylanase and LiP. P.c inhibited the activity of LiP when co-existing with T.r beginning, it promoted the degradation of lignin and further increased the production of humus-like substances (HLS) and humic-like acid (HLA) in later fermentation when nutrients were insufficient. The HLS of RTP was 54.9 g/

Studies on the treatment of anaerobically digested sludge by white-rot fungi: evaluation of the effect of Phanerochaete chrysosporium and Trametes versicolor.

BACKGROUND: The composition of anaerobically digested sludge is inherently complex, enriched with structurally complex organic compounds and nitrogenous constituents, which are refractory to biodegradation. These characteristics limit the subsequent rational utilization of resources from anaerobically digested sludge. White-rot fungi (WRF) have garnered significant research interest due to their exceptional capacity to degrade complex and recalcitrant organic pollutants. However, the exploration of WRF in the context of sludge treatment remains an under-investigated area within the scientific

The 1,4-beta-D-glucan cellobiohydrolases from Phanerochaete chrysosporium. I. A system of synergistically acting enzymes homologous to Trichoderma reesei.

A physico-chemical and structural characterization of three 1,4-beta-D-glucan cellobiohydrolases (EC. 3.2.1.91), isolated from a culture filtrate of the white-rot fungus Phanerochaete chrysosporium, reveals that the cellulolytic enzyme secretion pattern and thus the general degradation strategy for P. chrysosporium is similar to that of Trichoderma reesei. Partial sequence data show that two of the isolated enzymes, i.e., CBHI, pI 3.82 and CBH62, pI 4.85, are homologous with CBHI and EGI from T. reesei; while, the third, i.e., CBH50, pI 4.87, is homologous to T. reesei CBHII.
