Black Hoof Medicinal Mushroom Phellinus linteus (Agaricomycetes) Extracts Protect Against Radiation-Induced Hematopoietic Abnormality in Mice.

We investigated the effects of Phellinus linteus extracts (PLEs) against radiation damage in mice. First, BALB/c mice were irradiated once with γ-rays at 4, 5, 6, or 8 Gy and allowed to recover for 20 days. Results reveal that 8-Gy radiation caused death in 100% of mice on day 13, and 6-Gy radiation caused death in 86.7% of mice (13/15) at the end of the experiment, whereas 4- and 5-Gy radiation did not result in any death. We then used 5-Gy γ-ray radiation to examine the protective effects of PLEs. Mice were orally administered a PLE (500, 1000, and 1500 mg/kg) daily for 2 weeks before radiat

Protoilludane-derived sesquiterpenoids from edible mushroom Phellinus tremulae: Anti-inflammatory evaluation in vitro and in vivo.

Phellinus tremulae is a valued mushroom with dual medicinal and edible properties. A chemical investigation on P. tremulae led to the isolation and structural elucidation of eight previously undescribed sesquiterpenoids, named phellitremulins A-H (1, 2, and 5-10), along with six known analogues. Compound 5 features a unique dimeric scaffold linked by two fomannosane units, while 6 should be a key intermediate in the biosynthetic pathway leading to protoilludane sesquiterpenoids. Compounds 1-5 notably inhibited NO production in LPS-stimulated RAW264.7 cells, with IC50 ranging from 1.1 to 6.6 μM

Enhancement hispolon production from Phellinus linteus via epigenetic-modified culture to inhibit human breast cancer cells.

Phellinus linteus (PL) is a medicinal fungus known for producing hispolon, a bioactive compound with antioxidant, anti-inflammatory, and anticancer properties. However, the natural scarcity of PL and the unsuccessful cultivation of its fruiting bodies have led to the exploration of alternative methods for enhancing its bioactive compound production. In this study, static fermentation was employed, and Valproic acid (VPA), a histone deacetylase inhibitor (HDACi), was added to the culture medium to induce epigenetic modifications and enhance hispolon production. After 30 days of fermentation, th

Receptor-ligand affinity-based screening and isolation of water-soluble 5-lipoxygenase inhibitors from Phellinus igniarius.

We developed an efficient combination method for extraction, biological activity screening, and preparation of 5-lipoxygenase inhibitors from Phellinus igniarius. 5-Lipoxygenase inhibitors were rapidly screened using ultrafiltration-liquid chromatography based on the receptor-ligand affinity. Parameters such as extraction time, extraction times, and temperature as well as liquid-solid ratio were optimized using response surface methodology to maximize the total yield of the three target compounds. Next, bioactive ingredients were isolated using high-speed countercurrent chromatography and semi

Hepatoprotective and Antioxidant Effects of Ethanol Extract from Phellinus merrillii on carbon tetrachloride-induced liver damage.

In the present study, we investigated the hepatoprotective and antioxidant capacities of ethanol extract of Phellinus merrillii (PM) on carbon tetrachloride-induced hepatotoxicity. In high-performance liquid chromatography (HPLC) analysis, the finger print chromatogram of PM was established. Both hispolon and PM showed a similar peak at the retention time of 6 min. This implied that PM did contain the active ingredient of hispolon. Treatment with PM (0.5, 1 and 2 g/kg) prior to the administration of carbon tetrachloride (1.5 ml/kg in olive oil, 20%) significantly prevented the increased serum

An alkali-extracted neutral heteropolysaccharide from Phellinus nigricans used as an immunopotentiator in immunosuppressed mice by activating macrophages.

A neutral heteropolysaccharide (PNANb) was isolated with alkali (0.1 M NaOH) from mycelia of Phellinus nigricans, and the structure, immunostimulating activity and some of the underlying molecular mechanisms of action of PNANb were explored in the current study. PNANb (14.95 kDa) predominantly consisted of Gal, Glc, and Man with minor Fuc. GC-MS and NMR analyses indicated that the backbone of PNANb was mainly composed of 6-α-Galp, 2,6-α-Galp with minor 3,6-β-Glcp, which was substituted with complex side chains at C-2 of 2,6-α-Galp and C-3 of 3,6-β-Glcp.
