CD31 regulates metabolic switch in Treg migration attenuates rheumatoid arthritis.

CD31 (PECAM-1) plays a critical role in T cell migration, whilst its immunoreceptor tyrosine inhibitory motifs (ITIMs), Y663 and Y686, are recognised for their roles in endothelial function, the precise mechanism in regulating immune cell remains elusive. Here, we demonstrate that CD31 is essential for Treg migration. Upon ITIM engagement, CD31 activates and interacts with the protein tyrosine phosphatase SHP2. In vivo, CD31 Y663F gene transfer recapitulates the wild-type migration phenotype, driven by a metabolic switch to fructose utilisation under the regulation of the PFKFB3 gene. Canonical and Non-Canonical Roles of PFKFB3 in Brain Tumors.

PFKFB3 is a bifunctional enzyme that modulates and maintains the intracellular concentrations of fructose-2,6-bisphosphate (F2,6-P2), essentially controlling the rate of glycolysis. PFKFB3 is a known activator of glycolytic rewiring in neoplastic cells, including central nervous system (CNS) neoplastic cells. The pathologic regulation of PFKFB3 is invoked via various microenvironmental stimuli and oncogenic signals. Hypoxia is a primary inducer of PFKFB3 transcription via HIF-1alpha. In addition, translational modifications of PFKFB3 are driven by various intracellular signaling pathways that

Regulation of phosphofructokinase-1 on submandibular salivary glands of rats after isoproterenol administration.

The purpose of this investigation was to study the effect of isoproterenol (IPR) treatment on the regulation of phosphofructokinase-1 of submandibular salivary glands of rats. The animals were divided into control and experimental groups. In the first set of experiments, the rats received 5 mg of IPR/kg b.w. and were sacrificed at 24 hours after 1, 2, 3 and 4 doses. The content of fructose-2,6-bisphosphate (Fru-2,6-P(2)) and the activity of 6-phosphofructo-2-kinase (PFK-2) (active and total) were determined. The Fru-2,6-P(2) content was found to be reduced and the activity of PFK-2 (active and

Targeting PFKFB3 to restore glucose metabolism in acute pancreatitis via nanovesicle delivery.

BACKGROUND: Acute pancreatitis (AP) is a severe inflammatory disease frequently accompanied by disturbances in glucose metabolism, which further complicate the disease prognosis. This study aims to explore the role of PFKFB3, a key glycolytic enzyme, in regulating glucose metabolism in AP and assess the potential of PFKFB3 inhibition via nanovesicle delivery to mitigate metabolic dysfunction. METHODS: Transcriptomic data from Gene Expression Omnibus (GEO), including single-cell RNA sequencing (scRNA-seq) and bulk RNA sequencing, were analyzed to investigate the molecular mechanisms involved i

Neospora caninum hijacks host PFKFB3-driven glycolysis to facilitate intracellular propagation of parasites.

Infection with Neospora caninum leads to reproductive failure in ruminants, such as cattle and goats; however, no effective vaccines or treatments are currently available to control this infection. Carefully regulating the glycolysis of host cells is essential for the intracellular survival of pathogens. Nonetheless, the impact of N. caninum infection on host cell glycolysis and the effects and mechanisms of host cell glycolysis on the intracellular survival of this parasite remains unclear. In this study, the analysis of metabolomics and transcriptomics revealed that N. caninum infection incr

The small molecule inhibitor 3PO is a modulator of neutrophil metabolism, ROS production, and NET release.

Neutrophils are key effector leukocytes of the innate immune system and play a pivotal role in defending the host against microbial infections. Recent studies have identified a crucial link between glycolysis and neutrophil cellular functions. Using human neutrophils, we have investigated the intricate relationship between glycolysis, extracellular glucose availability, and the enzyme 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase 3 (PFKFB3), in the regulation of reactive oxygen species (ROS) and neutrophil extracellular trap (NET) production. We have identified that PFKFB3 is elevated i

Meclizine-induced enhanced glycolysis is neuroprotective in Parkinson disease cell models.

Meclizine is a well-tolerated drug routinely used as an anti-histamine agent in the management of disequilibrium. Recently, meclizine has been assessed for its neuroprotective properties in ischemic stroke and Huntington disease models. We found that meclizine protected against 6-hydroxydopamine-induced apoptosis and cell death in both SH-SY5Y cells and rat primary cortical cultures. Meclizine increases the level of 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3 (PFKFB3), which activates phosphofructokinase, a rate-determining enzyme of glycolysis. This protection is therefore mediated

PFKFB3-mediated Pro-glycolytic Shift in Hepatocellular Carcinoma Proliferation.

BACKGROUND & AIMS: Metabolic reprogramming, in particular, glycolytic regulation, supports abnormal survival and growth of hepatocellular carcinoma (HCC) and could serve as a therapeutic target. In this study, we sought to identify glycolytic regulators in HCC that could be inhibited to prevent tumor progression and could also be monitored in vivo, with the goal of providing a theragnostic alternative to existing therapies. METHODS: An orthotopic HCC rat model was used. Tumors were stimulated into a high-proliferation state by use of off-target liver ablation and were compared with lower-prol

Carbohydrate utilization and digestibility by tilapia, Oreochromis niloticus x O. aureus, are affected by chromic oxide inclusion in the diet.

A 12-wk feeding trial was conducted to study the influence of chromic oxide (Cr2O3) on carbohydrate utilization and digestibility by tilapia, Oreochromis niloticus x O. aureus. Two levels of chromic oxide (0.5 and 2%) were each incorporated into diets containing glucose or starch. Chromic oxide was added at 0 or 8 wk. The diets were fed to triplicate groups of fish weighing 1.11 +/- 0.05 g. Fish fed the starch diet had greater (P < 0.05) weight gain, feed efficiency ratio, protein efficiency ratio, protein deposition and digestibility of protein, lipid, carbohydrate and dry matter than fish fe

No effect of triheptanoin in patients with phosphofructokinase deficiency.

Phosphofructokinase deficiency (PFKD) is a rare disorder of glycogen metabolism. The lack of phosphofructokinase activity blocks the oxidative pathway from glucose and glycogen to pyruvate. Patients suffer from myopathy, exercise intolerance, and myoglobinuria. Currently, there is no specific treatment for PFKD. We hypothesized that 2 weeks treatment with triheptanoin could improve oxidative metabolism during exercise by bypassing the blocked pyruvate generation in PFKD. The study was a randomized, double-blind, placebo-controlled crossover study. Three genetically verified patients completed

Combined defects of muscle phosphofructokinase and AMP deaminase in a child with myoglobinuria.

A 14-year-old boy with exercise-related myalgia and cramps had several episodes of myoglobinuria since early childhood. An episode at 2 years of age caused acute renal failure. Histochemical and biochemical analysis of muscle showed a combined defect of phosphofructokinase (PFK) and adenosine monophosphate (AMP) deaminase. DNA analysis showed that the patient was homozygous for a G-to-C substitution at codon 39 of the PFK gene (previously described in an Italian patient) and for the common mutation found in AMP deaminase deficiency.

Enzyme trafficking and coclustering precede and accurately predict human breast cancer recurrences: an interdisciplinary review.

Although great effort has been expended to understand cancer's origins, less attention has been given to the primary cause of cancer deaths-cancer recurrences and their sequelae. This interdisciplinary review addresses mechanistic features of aggressive cancer by studying metabolic enzyme patterns within ductal carcinoma in situ (DCIS) of the breast lesions. DCIS lesions from patients who did or did not experience a breast cancer recurrence were compared. Several proteins, including phospho-Ser226-glucose transporter type 1, phosphofructokinase type L and phosphofructokinase/fructose 2,6-bisph

The dual effects of Ca2+ on binding of the glycolytic enzymes, phosphofructokinase and aldolase, to muscle cytoskeleton.

We show here that in rat diaphragm muscle, a short time of incubation with the Ca(2+)-ionophore A23187 induced an increase in cytoskeleton-bound phosphofructokinase (EC 2.7.1.11) and aldolase (EC 4.1.2.13), whereas a longer period of incubation, which causes a pathological rise in intracellular Ca2+, induced a decrease in bound enzymes. Lactate concentration correlated with both phases of Ca2+ action on the binding of the enzymes. The increase in cytoskeleton-bound enzymes could be prevented by treatment with the calmodulin antagonists trifluoperazine or CGS 9343B (a novel, potent, and selecti

Metabolic flexibility secures skeletal progenitor function upon reduced glycolysis driven by PFKFB3 loss.

Glycolysis is widely considered as a major metabolic pathway in several bone-residing cell types, including skeletal stem and progenitor cells (SSPCs) that are essential for bone development, maintenance, and regeneration. However, the contribution of glycolysis to the in vivo function of SSPCs remains unknown. To examine how reduced glycolysis affects SSPC biology, we conditionally deleted phosphofructokinase-2/fructose-2,6-bisphosphatase 3 (PFKFB3), a key regulator of glycolytic flux. PFKFB3 deletion decreased glycolytic flux by at least 30%, but also reduced glucose‑carbon incorporation int

Ubiquitination of PFKFB4 by CHIP regulates glycolysis and progression in endometriosis†.

Endometriosis is a common estrogen-dependent gynecological disorder characterized by chronic inflammatory responses and frequently associated with clinical infertility. Recent studies have demonstrated that the pathogenesis of this disease is closely linked to dysregulated post-translational modifications. This study focuses on the regulatory role of the ubiquitin-proteasome system (UPS) in endometriosis and, for the first time, reveals the molecular mechanism by which the E3 ubiquitin ligase STIP1 homology and U-box containing protein 1 (CHIP) specifically binds to and ubiquitinates the glyco

Intracellular BAPTA directly inhibits PFKFB3, thereby impeding mTORC1-driven Mcl-1 translation and killing MCL-1-addicted cancer cells.

Intracellular Ca2+ signals control several physiological and pathophysiological processes. The main tool to chelate intracellular Ca2+ is intracellular BAPTA (BAPTAi), usually introduced into cells as a membrane-permeant acetoxymethyl ester (BAPTA-AM). Previously, we demonstrated that BAPTAi enhanced apoptosis induced by venetoclax, a BCL-2 antagonist, in diffuse large B-cell lymphoma (DLBCL). This finding implied a novel interplay between intracellular Ca2+ signaling and anti-apoptotic BCL-2 function. Hence, we set out to identify the underlying mechanisms by which BAPTAi enhances cell death

Fulfilling the Krebs and Beavo criteria for studying protein phosphorylation in the era of mass spectrometry-driven kinome research.

The reversible phosphorylation of proteins controls virtually all aspects of cell function. However, in order to establish that the phosphorylation of a protein by a particular protein kinase is of physiological relevance, a series of criteria (proposed by Krebs & Beavo, 1979 ) should be satisfied. Surprisingly, amongst the thousands of protein kinase targets that have been reported in the literature, there are not so many for which there is good evidence for phosphorylation having functional consequences in vivo. Here we review the approaches that can be used to establish physiologically impo
