Cadmium sulfide nanoparticle biomineralization and biofilm formation mediate cadmium resistance of the deep-sea bacterium Pseudoalteromonas sp. MT33b.

Cadmium (Cd) is a common toxic heavy metal in the environment, and bacteria have evolved different strategies to deal with Cd toxicity. Here, a bacterium designated Pseudoalteromonas sp. MT33b possessing strong Cd resistance was isolated from the Mariana Trench sediments. Supplement of cysteine significantly increased bacterial Cd resistance and removal rate. Biofilm formation was demonstrated to play a positive role toward bacterial Cd resistance. Transcriptome analysis showed the supplement of cysteine effectively prevented Cd2+ from entering bacterial cells, promoted saccharide metabolism a

Cloning, characterization and anion inhibition studies of a new γ-carbonic anhydrase from the Antarctic bacterium Pseudoalteromonas haloplanktis.

A new γ-class carbonic anhydrase (CA, EC 4.2.1.1) was cloned, purified and characterized from the Antarctic bacterium Pseudoalteromonas haloplanktis, PhaCAγ. The enzyme has a medium-low catalytic activity for the physiologic reaction of CO2 hydration to bicarbonate and protons, with a kcat of 1.4×10(5)s(-1) and a kcat/Km of 1.9×10(6)M(-1)s(-1). An anion inhibition study of PhaCAγ with inorganic anions and small molecule inhibitors is also reported. Many anions present in sea water, such as chloride, fluoride, sulfate, iodide, but also others such as azide, perchlorate and tetrafluoroborate did

Antibacterial Activity of Volatile Organic Compounds Produced by the Octocoral-Associated Bacteria Bacillus sp. BO53 and Pseudoalteromonas sp. GA327.

The present research aimed to evaluate the antibacterial activity of volatile organic compounds (VOCs) produced by octocoral-associated bacteria Bacillus sp. BO53 and Pseudoalteromonas sp. GA327. The volatilome bioactivity of both bacteria species was evaluated against human pathogenic antibiotic-resistant bacteria, methicillin-resistant Staphylococcus aureus, Acinetobacter baumanni, and Pseudomonas aeruginosa. In this regard, the in vitro tests showed that Bacillus sp. BO53 VOCs inhibited the growth of P. aeruginosa and reduced the growth of S. aureus and A. baumanni. Furthermore, Pseudoalter

On-board saline black water treatment by bioaugmentation original marine bacteria with Pseudoalteromonas sp. SCSE709-6 and the associated microbial community.

To reduce fresh water load on ships, seawater can be used for toilet flushing on-board. And saline black water was treated on-site by bioaugmentation original marine bacteria with Pseudoalteromonas sp. SCSE709-6 (P. sp. SCSE709-6) to prevent marine pollution. In the batch experiments, P. sp. SCSE709-6 was effective in nutrient removal, which was not closely related to the amount of inoculation. In the on-board continuous experiments, the systems inoculated with P. sp. SCSE709-6 possessed excellent TP removal ability (removal rate: 80.93% for T3 and 88.39% for T4). The inoculum of P. sp. Stringent Starvation Protein SspA and Iron Starvation Sigma Factor PvdS Coordinately Regulate Iron Uptake and Prodiginine Biosynthesis in Pseudoalteromonas sp. R3.

Organisms need sufficient intracellular iron to maintain biological processes. However, cells can be damaged by excessive iron-induced oxidation stress. Therefore, iron homeostasis must be strictly regulated. In general, bacteria have evolved complex mechanisms to maintain iron homeostasis. In this study, we showed that Pseudoalteromonas sp. R3 has four sets of iron uptake systems. Among these, the siderophore pyoverdine-dependent iron uptake system and the ferrous iron transporter Feo system are more important for iron uptake and prodiginine biosynthesis. Stringent starvation protein SspA pos

Structural Determinants of the Specific Activities of an L-Amino Acid Oxidase from Pseudoalteromonas luteoviolacea CPMOR-1 with Broad Substrate Specificity.

The Pseudoalteromonas luteoviolacea strain CPMOR-1 expresses a flavin adenine dinucleotide (FAD)-dependent L-amino acid oxidase (LAAO) with broad substrate specificity. Steady-state kinetic analysis of its reactivity towards the 20 proteinogenic amino acids showed some activity to all except proline. The relative specific activity for amino acid substrates was not correlated only with Km or kcat values, since the two parameters often varied independently of each other. Variation in Km was attributed to the differential binding affinity. Variation in kcat was attributed to differential position

Production of the antimicrobial compound tetrabromopyrrole and the Pseudomonas quinolone system precursor, 2-heptyl-4-quinolone, by a novel marine species Pseudoalteromonas galatheae sp. nov.

Novel antimicrobials are urgently needed due to the rapid spread of antibiotic resistant bacteria. In a genome-wide analysis of Pseudoalteromonas strains, one strain (S4498) was noticed due to its potent antibiotic activity. It did not produce the yellow antimicrobial pigment bromoalterochromide, which was produced by several related type strains with which it shared less than 95% average nucleotide identity. Also, it produced a sweet-smelling volatile not observed from other strains. Mining the genome of strain S4498 using the secondary metabolite prediction tool antiSMASH led to eight biosyn

Assessment of multiple probiotic strains that protect Montipora capitata coral from infection by Vibrio coralliilyticus.

Coral disease outbreaks threaten reef ecosystems, often leading to widespread mortality and declines in coral cover. Outbreaks of tissue loss diseases like acute Montipora white syndrome (aMWS) have impacted coral populations that include the Hawaiian rice coral (Montipora capitata). Multiple strains of Vibrio coralliilyticus are known pathogens, and strain OCN008 has been demonstrated as an etiological agent of aMWS in Hawai'i. Recent work has demonstrated that probiotic bacterial strains can be used to directly treat or prevent transmission (prophylaxis) of coral diseases. Based on their pro

Extracellular proteases and possible disease related virulence mechanisms of two marine bacteria implicated in an opportunistic bacterial infection of Nephrops norvegicus.

The extracellular products (ECP) secreted by two strains of gram-negative bacteria isolated from Nephrops norvegicus exhibiting signs of an opportunistic bacterial infection were investigated with the objective of understanding their role in the spoilage of host muscle tissue and identifying disease related virulence mechanisms. ECP from Vibrio sp. demonstrated no proteolytic activity. ECP from Pseudoalteromonas sp. (isolate N10) degraded several substrates, including azocasein and host muscle tissue. Proteolytic activity increased with temperature. Substrate-impregnated sodium dodecyl sulfate

Crown ether dopant to reduce ion suppression and improve detection in the liquid microjunction surface sampling probe.

RATIONALE: Sodium and potassium are required in agar media for the growth of some microorganisms (e.g., marine bacteria). However, alkali cations are a significant source of contamination for mass spectrometry causing ion suppression and adduct formation. Conventionally, salts can be removed before mass spectrometric analysis with appropriate and often lengthy sample preparation. The direct mass spectrometric sampling of bacterial colonies grown on agar media seeks to minimize or eliminate sample preparation to improve workflow. However, this may exacerbate ion suppression and contamination si

Re-investigation of the HExxH enzyme DarF reveals a dehydrogenation-epoxidation reaction.

Darobactin A is a potent Gram-negative antibiotic, but the biosynthesis of its derivative, dehydrodarobactin A, remains unclear. Here, we re-investigated the HExxH enzyme DarF from Pseudoalteromonas luteoviolacea H33 through in vitro characterization and found that it catalyzes a dehydrogenation-epoxidation reaction and detected these darobactin A derivatives in native strains.

Unlocking Marine Microbial Metabolites of Sponges and Sediments as a Source of Anti-Trypanosoma cruzi Compounds.

Marine microbial metabolites have long served as valuable sources for drug discovery, exhibiting diverse biological activities, including antiparasitic, antimicrobial, and antiviral effects. Chagas disease (CD), a neglected tropical disease caused by Trypanosoma cruzi, affects approximately 8 million people worldwide. Currently, only two drugs are available for treatment, both associated with significant toxicity, underscoring the urgent need for new therapeutic alternatives. In this context, and considering the remarkable chemical diversity of marine bacteria, the present study investigated t

[Gram-negative eubacteria isolated from the water, molluscs and algae of the Black Sea].

Microorganisms strains (384) have been isolated from water, algae and molluscs collected in the Karadag reserve and the village of Katsiveli (Black Sea shore of the Crimea); 81 of the above strains are gram-negative oxidase-positive rods which require sodium ions for their growth. A group of nonfermenting mobile bacteria deprived of arginine dihydrolase have been distinguished among the latter; as to their physiological and biochemical properties and sensitivity to some antibiotics these bacteria were referred to Alteromonas, Marinomonas, Pseudoalteromonas genera.

Cooperative Chemical Biosynthesis of Tetrodotoxin: Evidence from Marine Microbial Contributors.

Tetrodotoxin (TTX) is a potent neurotoxin widely distributed in pufferfish and other marine organisms, yet its microbial biosynthetic basis remains unresolved because no definitive bacterial TTX gene cluster has been identified. This study develops a hypothesis-generating comparative-genomic framework to prioritize bacterial genomes for experimental investigation of TTX-related metabolism; it does not demonstrate bacterial TTX biosynthesis. Thirteen genomes representing Pseudoalteromonas, Cytobacillus, Shewanella, and Vibrio were analyzed using whole-genome comparison, average amino acid ident

Tunicate-associated bacteria show a great potential for the discovery of antimicrobial compounds.

Tunicates (Ascidians, sea squirts) are marine protochordates, which live sedentary or sessile in colonial or solitary forms. These invertebrates have to protect themselves against predators and invaders. A most successful strategy, to not being eaten by predators and prevent pathogenic microorganisms to settle, is the usage of chemical molecules for defence. To accomplish this, tunicates take advantage of the specialized metabolites produced by the bacteria associated with them. Therefore, the microbiome of the tunicates can be regarded as a promising bioresource for bacterial strains producin

Reconciling the Conflict between Optical Transparency and Fouling Resistance with a Nanowrinkled Surface Inspired by Zebrafish's Cornea.

Surface topography has been demonstrated as an effective nonchemical strategy for controlling the fouling resistance of a surface, but its impact on optical transparency remains a barrier to the application of this strategy in optical materials. To reconcile the conflicting effects of surface topography on optical transparency and fouling resistance, here we study the optical properties and antifouling performance of nanowrinkled surfaces inspired by the corneal surface of zebrafish (Danio rerio). Experimental and numerical analyses demonstrate that a good compromise between optical transparen

Effects of atmospheric cold plasma treatment mode on muscle quality and bacterial community of red shrimp during cold storage.

Atmospheric cold plasma (ACP), known for its safety, non-thermal processing, and energy efficiency, is especially effective for preserving perishable items such as seafood. However, excessive use of ACP may cause oxidation and sensory quality deterioration, which limits its application in the seafood industry. Thus, optimizing ACP treatment modes to balance sterilization efficiency with minimal oxidative effects is essential. This study aims to explore the impact of different ACP treatment modes (cyclical and one-time treatment) on the muscle quality and bacterial community of red shrimp durin
