The Effects of Environmental Enrichment on the Behavioral Profile of an Attention-Deficit/Hyperactivity Disorder Animal Model: A Systematic Review and Meta-Analysis.

In preclinical research, animal models are a valid alternative for studying both the genetics and environmental factors related to human psychopathologies. Indeed, in the last few decades, a great emphasis has been placed on the spontaneously hypertensive rat (SHR) inbred strain as an attention-deficit/hyperactivity disorder (ADHD) animal model. Therefore, this systematic review and meta-analysis aim to evaluate the effects of environmental enrichment (EE) on the behavior profile of the SHRs. All methods used to conduct the review were registered a priori on the PROSPERO platform (CRD420234421

Role of preexisting inhibitory control deficits vs. drug use history in mediating insensitivity to aversive consequences in a rat model of polysubstance use.

RATIONALE: The nature and predictors of insensitivity to aversive consequences of heroin + cocaine polysubstance use are not well characterized. OBJECTIVES: Translational methods incorporating a tightly controlled animal model of drug self-administration and measures of inhibitory control and avoidance behavior might be helpful for clarifying this issue. METHODS: The key approach for distinguishing potential contributions of pre-existing inhibitory control deficits vs. drug use history in meditating insensitivity to aversive consequences was comparison of two rat strains: Wistar (WIS/Crl), a

Microphthalmia and cataract in rats with a novel point mutation in connexin 50 - L7Q.

PURPOSE: We isolated an autosomal semi-dominant cataract from our inbred SHR/OlaIpcv rat colony. Heterozygotes express pulverulent cataract with smaller eyes; homozygotes express marked microphthalmia with hypoplastic lens. We call this mutation Dca (for dominant cataract). In this study, we focus on the identification of the responsible gene. METHODS: We performed linkage mapping using 93 F2(SHR-Dca x PD) hybrids and a panel of microsatellite markers. In a separate group of animals with a SHR genetic background, we examined the lenses histologically using Epon semi-thin sections and toluidin

Influence of arterial hypertension on colonic healing in rats.

PURPOSE: Evaluation of colonic healing in spontaneously hypertensive rats. METHODS: Fifty male, young and inbred rats were used. Twenty-five Wistar Kyoto rats (WKY) as control and twenty-five spontaneously hypertensive rats (SHR) as an experimental group. Colotomy and bowel suture at 2.5 cm from the peritoneal reflection were performed. All animals were allocated randomly into sub-groups for review at the third, seventh and fourteenth days after surgery. We evaluated the concentration of angiotensin II, the burst pressure, epithelialization, the organization of the tunics of the bowel wall, i

Genetic Variation in Renal Expression of Folate Receptor 1 (Folr1) Gene Predisposes Spontaneously Hypertensive Rats to Metabolic Syndrome.

Metabolism of homocysteine and other sulfur amino acids is closely associated with metabolism of folates. In this study, we analyzed the possible role of folates and sulfur amino acids in the development of features of the metabolic syndrome in the BXH/HXB recombinant inbred strains derived from the spontaneously hypertensive rat (SHR) and Brown Norway progenitors. We mapped a quantitative trait locus for cysteine concentrations to a region of chromosome 1 that contains a cis-acting expression quantitative trait locus regulating mRNA levels of folate receptor 1 (Folr1) in the kidney. Liver copper content of rats hypo- or hyperresponsive to dietary cholesterol.

The question addressed is whether cholesterol intake reduces the hepatic copper content in rats. For this purpose we have compared the hepatic copper content of two selected rat inbred strains after feeding the animals a control or a high fat, high cholesterol diet. One strain was dietary cholesterol resistant (SHR/OlaIpcv), whereas the other strain was susceptible to dietary cholesterol (BN-Lx/Cub). Dietary cholesterol-susceptible rats have a lower baseline hepatic copper content when compared with their resistant counterparts. The consumption of a hypercholesterolemic diet decreased the live

The teratogenic action of retinoic acid in rat congenic and recombinant inbred strains.

The teratogenic effect of all-trans retinoic acid (RA) was tested in the system of congenic and recombinant inbred (RI) strains of the laboratory rat carrying the mutant Lx allele which determines the polydactyly-luxate syndrome. It was demonstrated that the teratogenic effect of RA is influenced by both the genotype at the Lx locus and the modifying genes. The dose of 100 mg/kg administered by gavage on day 11 of pregnancy induced a statistically significant incidence of preaxial polydactyly in hind limbs of LEW/BN, +/Lx foetuses heterozygous in the mutant allele, whereas no specific limb def

Genetic analysis of the rat hypodactylous mutation.

Autosomal recessive rat hypodactylous mutation Hd leads in homozygous condition to reductive changes of the digital arch of all feet. There is a variable preaxial reduction of the number of fingers in both sexes. Moreover, homozygous males are sterile. Testes of homozygous Hd/Hd and +/Hd adult rats were examined in the light and electron microscopes. Spatial organization of stages of the spermatogenetic cycle was not confirmed in Hd/Hd testes comparing with +/Hd males. Significant decrease in the number of germ cells in seminiferous tubules of Hd/Hd testes was accompanied with loosening and va

Hypothalamic-pituitary-adrenal response to chronic stress in five inbred rat strains: differential responses are mainly located at the adrenocortical level.

The effects of chronic stress on the hypothalamic-pituaitary-adrenocortical (HPA) axis were studied in five inbred rat strains, i.e. Brown Norway (BN), Fischer (FIS), Lewis (LEW), Spontaneously Hypertensive (SHR) and Wistar Kyoto (WKY). Previously, these rat strains had been shown to display clear behavioral differences in the forced swimming test that presumably measures depression-like behavior, BN and WKY being more passive than the other strains. Here we test the hypothesis that the differences in behavioral immobility might be associated with an abnormal HPA response to chronic immobiliza

Spontaneous nonsense mutation in the tuftelin 1 gene is associated with abnormal hair appearance and amelioration of glucose and lipid metabolism in the rat.

Recently, we have identified a recessive mutation, an abnormal coat appearance in the BXH6 strain, a member of the HXB/BXH set of recombinant inbred (RI) strains. The RI strains were derived from the spontaneously hypertensive rat (SHR) and Brown Norway rat (BN-Lx) progenitors. Whole genome sequencing of the mutant rats identified the 195875980 G/A mutation in the tuftelin 1 (Tuft1) gene on chromosome 2, which resulted in a premature stop codon. Compared with wild-type BXH6 rats, BXH6-Tuft1 mutant rats exhibited lower body weight due to reduced visceral fat and ectopic fat accumulation in the

Cardiac hypertrophy and remodelling: pathophysiological consequences and protective effects of melatonin.

This survey summarizes the limited findings related to the ability of endogenously produced and exogenously administered melatonin to modify the response of the heart musculature to pathological situations that are normally accompanied by hypertrophy of the cardiomyocytes and alterations in cardiovascular physiology. Whereas melatonin did not reduce left ventricular hypertrophy in spontaneously hypertensive rats or in nitric oxide-deficient hypertension, it did have other beneficial effects, e.g. it curtailed oxidative damage to the heart that resulted in an attenuation of left ventricular fib

Is there a common link between the mechanisms of mineralocorticoid-induced and genetic hypertension?

We have shown that (1) mineralocorticoids increase vascular smooth muscle (VSM) membrane permeability to Na+ through a receptor-mediated mechanism as mineralocorticoid-induced hypertension develops, and (2) prehypertensive spontaneously hypertensive rats (SHR) have elevated plasma aldosterone and expanded interstitial fluid volumes, both of which normalize as the rats reach the early hypertensive phase. Others have reported elevation in other mineralocorticoids, VSM membrane permeability changes to Na+ and exaggerated adrenocorticotrophic hormone (ACTH) response to stress in SHR. We hypothesiz

Humoral factors in primary hypertension.

In 56 spontaneously hypertensive rats of the Muenster strain, either parabiosis or cross circulation was performed with normotensive Wistar Kyoto rats. Crossed circulation was made through the common carotid arteries and external jugular veins using a peristaltic pump. In parabiosis and in cross-circulation experiments, hypertension was transmitted from spontaneously hypertensive to normotensive rats. Nephrectomy or adrenalectomy in the spontaneously hypertensive rat before cross circulation abolished this effect. After volume depletion in the hypertensive animals, hypertension was not transmi

Influence of prazosin on systemic iron levels and the associated iron metabolic alterations in spontaneously hypertensive rats.

The relationship between cardiovascular diseases and iron disorders has gained increasing attention; however, the effects of hypotensive drugs on iron metabolic alterations in hypertension are not well understood. The purpose of this study was to investigate iron metabolic changes after prazosin treatment of spontaneously hypertensive rats (SHRs) and Wistar-Kyoto (WKY) rats. Our second objective was to examine the effects of hypertension and anti-hypertensive drugs on bone formation and resorption. SHRs and WKY rats were randomized into either prazosin-treated groups (WKY + PZ and SHR + PZ) or

New Cortical Neurodegenerative Pathways in the Hypertensive Rat Brain.

Hypertension is a risk factor for neurodegenerative diseases. We hypothesized that chronic hypertension underlies neurodegeneration. In this study, we examined the expression of brain cortical proteins involved in homeostasis, apoptosis, and brain functions in Spontaneously Hypertensive Rats (SHR) compared with normotensive Wistar-Kyoto (WKY) rats. We used paraffin-embedded brain sections of 8-month-old SHR and WKY rats, immunohistochemically stained and analyzed by image processing. In SHR, cytochrome c oxidase subunit 7A increased, indicative of hypoxia; heat shock protein 40, the chaperon f

A comparison of the blood supply of the carotid body in rats and rabbits.

Light-microscopic findings regarding the arterial blood supply of the carotid bodies in 145 Wistar rats, in 175 spontaneously hypertensive rats (SHR) and in 14 rabbits were compared. In rats, only one glomic artery was found in each of 637 carotid bifurcations. In 2 bifurcations 2 carotid body arteries supplied the glomus caroticum, the glomic artery being of the muscular type. At the origin of the carotid body artery almost regularly intraarterial cushions were observed in rats. They were also demonstrable at the origin of first or second order branches of the glomic artery. Concerning the or

Significance of 19-noraldosterone, a new mineralocorticoid, in clinical and experimental hypertension.

19-Noraldosterone, which was recently shown to be synthesized and produced in the human adrenal gland, possesses potent mineralocorticoid and hypertensinogenic activities. 18,19-Dihydroxycorticosterone (18,19-(OH)2-B) and 18-hydroxy-19-norcorticosterone (18-OH-19-nor-B), a possible precursor of 19-noraldosterone, have been identified in human urine. These mineralocorticoid hormones are regulated by the renin-angiotensin system and synthesized in adrenal glomerulosa cells. Urinary 19-noraldosterone correlated with urinary 18,19-(OH)2-B, 18-OH-19-nor-B, 18-hydroxycorticosterone (18-OH-B), and al

Splicing mutation in Sbf1 causes nonsyndromic male infertility in the rat.

In the inbred SHR/OlaIpcv rat colony, we identified males with small testicles and inability to reproduce. By selectively breeding their parents, we revealed the infertility to segregate as an autosomal recessive Mendelian character. No other phenotype was observed in males, and females were completely normal. By linkage using a backcross with Brown Norway strain, we mapped the locus to a 1.2Mbp segment on chromosome 7, harboring 35 genes. Sequencing of candidate genes revealed a G to A substitution in a canonical 'AG' splice site of intron 37 in Sbf1 (SET binding factor 1, alias myotubularin-

Short-term effect of ligature-induced periodontitis on cardiovascular variability and inflammatory response in spontaneously hypertensive rats.

BACKGROUND: We previously reported that periodontal disease (PD) induces high arterial pressure variability (APV) consistent with sympathetic overactivity and elicits myocardial inflammation in Balb/c mice. However, it is unknown whether PD can change APV and heart rate variability (HRV) in spontaneously hypertensive (SHR) and normotensive Wistar-Kyoto (WKY) rats. This study aimed to evaluate the hemodynamic level, HRV, and APV associating with myocardial inflammation and plasma concentrations of oxide nitric (NO) in SHR and WKY rats with PD. METHODS: Three weeks after bilateral ligation of t
