Stereoselective α-Deuteration of Serine, Cysteine, Selenocysteine, and 2,3-Diaminopropanoic Acid Derivatives.

Efficient methodologies for synthesizing enantiopure α-deuterated derivatives of serine, cysteine, selenocysteine, and 2,3-diaminopropanoic acid have been developed. H/D exchange was achieved by deprotonation of a chiral bicyclic serine equivalent followed by selective deuteration. Additionally, diastereoselective additions of thiols, selenols, and amines to a chiral bicyclic dehydroalanine in deuterated alcohols allowed site-selective deuteration at the Cα atom of cysteine, selenocysteine, and 2,3-diaminopropanoic acid derivatives. A deuterated analogue of carbocysteine, a drug for the treatm

Inherited Selenocysteine Transfer RNA Mutation: Clinical and Hormonal Evaluation of 2 Patients.

INTRODUCTION: Iodothyronine deiodinases are selenoproteins with the amino acid selenocysteine (Sec) introduced into the position of a TGA stop codon by a complex machinery involving tRNA[Ser]Sec when a cis-acting Sec-insertion sequence element is present in the 3' end of the mRNA. Recently, a variant in the TRU-TCA1-1 gene encoding for tRNA[Ser]Sec was reported, which resulted in reduced expression of stress-related selenoproteins. The proband presented with multisystem symptoms, euthyroid hyperthyroxinemia, and selenium deficiency. Here, we describe 2 new members of a family harboring the sam

Selenocysteine vs Cysteine: Tuning the Derivatization on Benzenesulfonyl Moiety of a Triazole Linked Dansyl Connected Glycoconjugate for Selective Recognition of Selenocysteine and the Applicability of the Conjugate in Buffer, in Serum, on Silica Gel, and in HepG2 Cells.

A dansyl derivatized triazole linked glucopyranosyl conjugate ((NO2)L) has been synthesized and characterized and was used in the present study. The conjugate (NO2)L releases a fluorescent product upon reaction by Cys-SeH in aqueous PBS buffer by exhibiting a ∼210-fold fluorescence enhancement even in the presence of 20 other amino acids with a minimum detection limit of (1.5 ± 0.2) × 10(-7) M. The selectivity of the Cys-SeH to (NO2)L was further proven by extending the fluorescence study to different other selenium compounds. The role of para-nitrobenzenesulfonyl (pNBS) center in (NO2)L in th

Global loss of selenocysteine lyase in mice drives lipid accumulation in brown adipocytes.

The enzyme selenocysteine (Sec) lyase (SCLY) decomposes Sec, releasing selenide for the synthesis of selenoproteins, which contain Sec in their primary structure and participate in strong redox reactions, maintaining redox balance. We previously showed that global disruption of the Scly gene (Scly KO) in mice leads to obesity. Targeted deletion of Scly in agouti-related peptide neurons enhances energy expenditure and brown adipose tissue (BAT) activation, augmenting leanness. We hypothesized that Scly KO mice develop obesity due to failure of BAT-controlled mechanisms of energy expenditure due

Anthocyanin-functionalized selenocysteine nanotherapeutics alleviate cisplatin nephrotoxicity by inhibiting oxidative stress and ferroptosis.

Although cisplatin (CDDP) remains a widely used chemotherapeutic agent in clinical cancer treatment, severe oxidative stress and inflammatory responses induced by CDDP are still major causes of acute kidney injury (AKI) in patients. Our previous studies have demonstrated that anthocyanins and selenocysteine (SeC) exhibit potent antioxidant and anti-inflammatory properties. Here, we successfully synthesized a synergistic nanotherapeutic, cyanidin-3-O-glucoside-functionalized SeC nanoparticles (C3G@SeC NPs), which significantly alleviated CDDP-induced damage in HK-2 cells by inhibiting oxidative

Selenocysteine containing analogues of Atx1-based peptides protect cells from copper ion toxicity.

Seleno-substituted model peptides of copper metallochaperone proteins were analyzed for the metal affinity and in vitro anti-oxidative reactivity. An acyclic MTCXXC (X is any amino acid) reference peptide previously analyzed as a potent inhibitor of ROS production underwent substitution of the cysteine residues with selenocysteine to give two singly substituted derivatives C3U and C6U and the doubly substituted analogue C3U/C6U. Presumably due to the softer nature of Se vs. S, all selenocysteine containing peptides demonstrated high affinity to Cu(i), higher than that of the reference peptide,

SerRS-tRNASec complex structures reveal mechanism of the first step in selenocysteine biosynthesis.

Selenocysteine (Sec) is found in the catalytic centers of many selenoproteins and plays important roles in living organisms. Malfunctions of selenoproteins lead to various human disorders including cancer. Known as the 21st amino acid, the biosynthesis of Sec involves unusual pathways consisting of several stages. While the later stages of the pathways are well elucidated, the molecular basis of the first stage-the serylation of Sec-specific tRNA (tRNA(Sec)) catalyzed by seryl-tRNA synthetase (SerRS)-is unclear. Here we present two cocrystal structures of human SerRS bound with tRNA(Sec) in di

Replacing of Cysteine with Selenocysteine: Biochemical Considerations, Computational Modeling, and Protein Engineering Applications.

Selenocysteine (Sec), the 21st amino acid, combines the structural compatibility of cysteine with the unique chemical properties of selenium, enabling exceptional redox and catalytic performance in both natural and engineered systems. Its low pKa, high nucleophilicity, and increased polarizability underpin the activity of selenoproteins involved in antioxidant defence, redox regulation, and metabolic processes. This work integrates biochemical, structural, and evolutionary perspectives on Sec with advances in protein engineering and synthetic biology, where strategic Sec incorporation in place

Impairment in selenocysteine synthesis as a candidate mechanism of inducible coagulopathy in COVID-19 patients.

Coagulopathy has recently been recognized as a recurring complication of COVID-19, most typically associated with critical illness. There are epidemiological, mechanistic and transcriptomic evidence that link Selenium with SARS-CoV-2's intracellular latency. Taking into consideration the vital role of selenoproteins in maintaining an adequate immune response, endothelial homeostasis and a non-prothrombotic platelet activation status, we propose that impairment in selenocysteine synthesis, via perturbations in the aforementioned physiological functions, potentially constitutes a mechanism of co

Structural basis for early-onset neurological disorders caused by mutations in human selenocysteine synthase.

Selenocysteine synthase (SepSecS) catalyzes the terminal reaction of selenocysteine, and is vital for human selenoproteome integrity. Autosomal recessive inheritance of mutations in SepSecS-Ala239Thr, Thr325Ser, Tyr334Cys and Tyr429*-induced severe, early-onset, neurological disorders in distinct human populations. Although harboring different mutant alleles, patients presented remarkably similar phenotypes typified by cerebellar and cerebral atrophy, seizures, irritability, ataxia, and extreme spasticity. However, it has remained unclear how these genetic alterations affected the structure of

Targeting oncogenic FLT3 uncovers a ferroptosis vulnerability through selenocysteine recoding in acute myeloid leukaemia.

Ferroptosis, an iron-dependent form of cell death driven by lipid peroxidation, has emerged as a potential therapeutic strategy for therapy-resistant cancers. Glutathione peroxidase 4 and the selenoprotein biosynthesis pathway essential for its translation are key regulators of ferroptosis but lack effective therapeutic targeting. In a drug screening using a selenoprotein translation reporter, here we identify FMS-like tyrosine kinase 3 (FLT3) inhibitors as suppressors of selenoprotein translation that induce ferroptosis in FLT3-mutant acute myeloid leukaemia. Mechanistically, FLT3 inhibition

Selenocysteine-Activatable Near-Infrared Fluorescent Probe for Screening of Anti-inflammatory Components in Herbs.

Inflammation, a central process in numerous diseases, plays a crucial role in hepatic disorders, arthritis, cardiac conditions, and neurodegenerative ailments. Given the lack of effective anti-inflammatory drugs, it is imperative to assess inflammation severity and explore novel therapeutics. Selenocysteine (Sec), a key mediator of selenium's biological function, is closely involved in anti-inflammatory responses. We have synthesized a novel near-infrared fluorescent probe, Sec-BDP, which can image Sec dynamics in vivo with high selectivity and sensitivity. Sec-BDP detects Sec at concentration

Oxidation of S-e-carboxymethyl-selenocysteine by L-aminoacid oxidase and by D-aspartate oxidase.

Se-Carboxymethyl-DL-selnocysteine (CMSeC) has been prepared in a pure crystalline form from selenocysteine and monochloracetic acid. It has been shown that CMSeC is a substrate for the L-aminoacid oxidase form snake venom and for the D-aspartate oxidase from beef kidney. Oxygen consumption and ammonia production indicate that only the L or the D form of CMSeC ar acted upon respectively by one or the other of the above enzymes. No noticeable differences were shown in the oxidation rate of CMSeC and S-carboxymethylcysteine, an indication that the substitution of a selenium for a sulfur atom in t

Reassigning an amber codon to selenocysteine using a functional cysteine suppression pair in Escherichia coli.

Selenoproteins, characterized by the presence of selenocysteine (Sec) residues, are widely distributed across all domains of life. The unique attributes of selenoproteins are conferred by Sec, which is genetically encoded into nascent polypeptide chains through the recoding of the UGA codon. Human glutathione peroxidase 1 (GPx1) is a selenoprotein, which plays a crucial role in maintaining redox homeostasis. However, its complex recoding mechanism presents a major challenge for efficient selenoprotein expression in Escherichia coli. In this study, we engineered a functional amber suppression s

Molecular Aspects of Methylcadmium Toxicity: Effects on the H2O2 Reduction by Cysteine and Selenocysteine Disclosed In Silico.

Cadmium (Cd), like the other group 12 elements (Zn and Hg), has a high affinity for sulfur (S) and selenium (Se), a property that strongly influences its adverse biological effects. Although the symptoms of Cd toxicity are diverse, a common denominator is found in oxidative stress, resulting in the disruption of redox balance in cells and the proliferation of reactive oxygen species (ROS) and harmful radicals. Methylcadmium (CH3Cd+) is a convenient model to study Cd pro-oxidant activity in silico. In this work, the effect of CH3Cd+ on the peroxy-reducing potential of cysteine (Cys) and selenoc

Asgard archaeal selenoproteome reveals a roadmap for the archaea-to-eukaryote transition of selenocysteine incorporation machinery.

Selenocysteine (Sec) is encoded by the UGA codon that normally functions as a stop signal and is specifically incorporated into selenoproteins via a unique recoding mechanism. The translational recoding of UGA as Sec is directed by an unusual RNA structure, the SECIS element. Although archaea and eukaryotes adopt similar Sec encoding machinery, the SECIS elements have no similarities to each other with regard to sequence and structure. We analyzed >400 Asgard archaeal genomes to examine the occurrence of both Sec encoding system and selenoproteins in this archaeal superphylum, the closest prok

Advances on the Bioactivities, Safety, Preparation, and Applications of L-Se-Methyl-selenocysteine: Emphasis on Mechanistic Insights.

As a critical public health threat, selenium inadequacy augments the risk of life-threatening disorders across multiple population subgroups. Conventional selenium supplements, classified chemically as inorganic salts or organic selenocompounds or physically as selenium nanoparticles based on particle size, are limited by suboptimal bioavailability, unresolved safety concerns, high production costs, and poor stability. L-Se-methyl-selenocysteine (L-Se-MSC), a distinct naturally occurring organoselenium, represents a promising alternative due to its superior stability, low toxicity, and versati

Analysis of Novel Interactions between Components of the Selenocysteine Biosynthesis Pathway, SEPHS1, SEPHS2, SEPSECS, and SECp43.

In mammalian cells, the incorporation of the 21st amino acid, selenocysteine, into proteins is guided by the Sec machinery. The function of this protein complex requires several protein-protein and protein-RNA interactions, leading to the incorporation of selenocysteine at UGA codons. It is guided by stem-loop structures localized in the 3' untranslated regions of the selenoprotein-encoding genes. Here, we conducted a global analysis of interactions between the Sec biosynthesis and incorporation components using a bioluminescence resonance energy transfer assay in mammalian cells that showed t
