Skin-innervating glutamatergic neurons modulate aging.

Peripheral nerves regulate skin homeostasis by secreting neurotransmitters, but their role during skin aging remains incompletely understood. Here, we report that cutaneous denervation accelerates skin aging, as evidenced by collagen reduction. Neurofilament heavy chain (Nefh) is decreased in aged skin and is predominantly expressed in vesicular glutamate transporter 2-positive (Vglut2+) skin-innervating glutamatergic neurons. Notably, dermal fibroblasts, the primary producers of collagen, frequently contact Nefh+ nerve fibers. Moreover, Nefh deletion in Vglut2+ glutamatergic neurons drives sk

The microbiome in human skin aging.

Microbiome dysbiosis correlates with aging-associated pathological skin conditions, and our understanding of how the microbiome regulates skin aging at a molecular level is rapidly advancing. Classical hallmarks of skin aging, including genomic instability and telomere attrition, loss of proteostasis, epigenetic alterations, and altered intercellular communication, are critically regulated by the microbiome. Oxidative stress represents a key factor implicated in virtually all hallmarks of skin aging. Meanwhile, recent data underscore the role of metabolism in intercellular communication and ag

Human Skin Aging: Physiology and Pathophysiology.

Skin is the outermost layer which covers all organs of the body. Skin can be divided into three layers, including epidermis, dermis and hypodermis. Similarly to many organs of the body, skin also experiences biological process that could not resist aging. There are the following two types of human skin aging: endogenous and exogenous. Endogenous aging involves a series of physiological changes that gradually occur over time and are influenced by genes and hormones. In contrast, extrinsic aging (or exogenous aging), also known as photoaging, involves significant structural and functional change

Atmospheric skin aging-Contributors and inhibitors.

Cutaneous aging is a complex biological process consisting of 2 elements: intrinsic aging, which is primarily determined by genetics, and extrinsic aging, which is largely caused by atmospheric factors, such as exposure to sunlight and air pollution, and lifestyle choices, such as diet and smoking. The role of the solar spectrum, comprised of ultraviolet light, specifically UVB (290-320 nm) and UVA (320-400) in causing skin damage, including skin cancers, has been well documented. In recent years, the contribution of visible light (400-700 nm) and infrared radiation (above 800 nm) in causing s

[Skin of the aging human being].

Cutaneous aging includes true aging and photo-aging due to sun exposure. A decrease in epidermal turnover rate results in epidermal atrophy and delayed wound-healing. A reduction in the number of epidermal Langerhans' cells is responsible for a decrease in delayed immune responsiveness in skin observed in the elderly. Reduced numbers of fibroblasts and mast cells are typical histologic findings in aging human dermis. Collagen bundles become fragmented, less elastic and more brittle. Telomere shortening at the end of chromosomes is probably the major mechanism of cellular senescence in skin. Co

Aging and the skin immune system.

OBJECTIVE: To give a brief summary of age-related alterations that occur in the immune system (immunosenescence), with special regard to the skin immune system. DATA SOURCES: MEDLINE and institutional libraries were searched for relevant articles on immunosenescence and corresponding key words. DATA SYNTHESIS: The immune system of aged animals and humans undergoes alterations that may account for an increased susceptibility to certain infections, autoimmune diseases, or malignancies. However, some data on the parameters of immunosenescence are controversial. This appears to be caused by vari

New Insights Into the Skin Microbial Communities and Skin Aging.

Although it is well-known that human skin aging is accompanied by an alteration in the skin microbiota, we know little about how the composition of these changes during the course of aging and the effects of age-related skin microbes on aging. Using 16S ribosomal DNA and internal transcribed spacer ribosomal DNA sequencing to profile the microbiomes of 160 skin samples from two anatomical sites, the cheek and the abdomen, on 80 individuals of varying ages, we developed age-related microbiota profiles for both intrinsic skin aging and photoaging to provide an improved understanding of the age-d

Skin Pigmentation Abnormalities and Their Possible Relationship with Skin Aging.

Skin disorders showing abnormal pigmentation are often difficult to manage because of their uncertain etiology or pathogenesis. Abnormal pigmentation is a common symptom accompanying aging skin. The association between skin aging and skin pigmentation abnormalities can be attributed to certain inherited disorders characterized by premature aging and abnormal pigmentation in the skin and some therapeutic modalities effective for both. Several molecular mechanisms, including oxidative stress, mitochondrial DNA mutations, DNA damage, telomere shortening, hormonal changes, and autophagy impairment

Aging-related IgG accumulation promotes skin inflammation.

Aging skin is characterized by wrinkles, loss of elasticity, and impaired barrier function, largely driven by alterations in dermal fibroblasts and extracellular matrix remodeling. Chronic, age-related inflammation, called inflammaging, is a central activity underlying these changes. Immunoglobulin G (IgG) has recently been shown to accumulate in multiple organs with aging, where it activates macrophages to promote tissue inflammation. However, whether IgG contributes to skin aging remains unknown. Here, using mouse and human tissues we show that IgG accumulates in the dermis with age and prom

3D bioprinting-a model for skin aging.

Human lifespan continues to extend as an unprecedented number of people reach their seventh and eighth decades of life, unveiling chronic conditions that affect the older adult. Age-related skin conditions include senile purpura, seborrheic keratoses, pemphigus vulgaris, bullous pemphigoid, diabetic foot wounds and skin cancer. Current methods of drug testing prior to clinical trials require the use of pre-clinical animal models, which are often unable to adequately replicate human skin response. Therefore, a reliable model for aged human skin is needed. The current challenges in developing an

Asymmetric facial skin viscoelasticity during climacteric aging.

BACKGROUND: Climacteric skin aging affects certain biophysical characteristics of facial skin. The purpose of the present study was to assess the symmetric involvement of the cheeks in this stage of the aging process. METHODS: Skin viscoelasticity was compared on both cheeks in premenopausal and post-menopausal women with indoor occupational activities somewhat limiting the influence of chronic sun exposure. Eighty-four healthy women comprising 36 premenopausal women and 48 early post-menopausal women off hormone replacement therapy were enrolled in two groups. The tensile characteristics of

Airborne particle exposure and extrinsic skin aging.

For decades, extrinsic skin aging has been known to result from chronic exposure to solar radiation and, more recently, to tobacco smoke. In this study, we have assessed the influence of air pollution on skin aging in 400 Caucasian women aged 70-80 years. Skin aging was clinically assessed by means of SCINEXA (score of intrinsic and extrinsic skin aging), a validated skin aging score. Traffic-related exposure at the place of residence was determined by traffic particle emissions and by estimation of soot in fine dust. Exposure to background particle concentration was determined by measurements

Clinical and photographic scoring of skin aging.

Chronic exposure to sunlight induces clinical, histological and physiological changes that are described as photoaging. To assess the resulting skin changes different clinical and photographic scores have been evaluated. Regarding different scoring systems a standardized grading system would be useful in a variety of indications, in particular to improve the quality of epidemiologic and clinical studies of photodamage. Photonumeric grading is considered superior to descriptive scales; however, the combination of both may suggest an advanced assessment of photodamage.

Mechanisms of photoaging and chronological skin aging.

Human skin, like all other organs, undergoes chronological aging. In addition, unlike other organs, skin is in direct contact with the environment and therefore undergoes aging as a consequence of environmental damage. The primary environmental factor that causes human skin aging is UV irradiation from the sun. This sun-induced skin aging (photoaging), like chronological aging, is a cumulative process. However, unlike chronological aging, which depends on the passage of time per se, photoaging depends primarily on the degree of sun exposure and skin pigment. Individuals who have outdoor lifest

[Changes in cutaneous zinc during skin aging].

Variations of epidermal zinc concentration with aging were studied by measuring zinc levels in the epidermis of two groups of healthy subjects: group A aged less than 35 years; group B aged more than 65 years. The epidermis was obtained from a suction blister, and zinc levels measurements were performed by absorption spectrophotometry. There was a significant decrease (p less than 0.01) of epidermal zinc concentration with aging, and zinc contents of the epidermis were the same in men and women. On the other hand, no significant difference was found in zinc blister fluid levels between the two

Free radicals and aging of the skin.

Cutaneous aging is the result of genetically determined or intrinsic aging superimposed by degenerative changes due to actinic irradiation, also called photoaging. The manifestations of cutaneous aging, as it relates to the perception of age, is caused by ultraviolet light, in particular in those parts of the body exposed daily to solar radiation. Free radical generation in the skin by UV light and from other sources, such as cellular infiltrations or the xanthine oxidase reaction, may be detected by direct and indirect methods. The decrease in antioxidant enzymes and small molecular weight an

Glyoxalase System in the Progression of Skin Aging and Skin Malignancies.

Dicarbonyl compounds, including methylglyoxal (MGO) and glyoxal (GO), are mainly formed as byproducts of glucose metabolism. The main glyoxalase system consists of glyoxalase I and II (Glo1 and Glo2) and is the main enzyme involved in the detoxification of dicarbonyl stress, which occurs as an accumulation of MGO or GO due to decreased activity or expression of Glo1. Dicarbonyl stress is a major cause of cellular and tissue dysfunction that causes various health issues, including diabetes, aging, and cancer. The skin is the largest organ in the body. In this review, we discuss the role of the
