In the rat, paradoxical sleep (PS) is preceded by a short-lasting stage characterized by high amplitude cortical spindles and low frequency hippocampal theta rhythm. This intermediate stage (IS) is massively extended at the expense of PS by barbiturates and benzodiazepines. To further study the relationship between the IS and PS, six rats were PS deprived for 48 h by sitting on one of three small platforms surrounded by water. A control group of six other rats remained in dry cages with shavings. After 24 h PS deprivation the latency of IS occurrence increased, was unchanged after 48 h depriva

At the turn of the 19(th) century, trypanosomes were identified as the causative agent of sleeping sickness and their presence within the cerebrospinal fluid of late stage sleeping sickness patients was described. However, no definitive proof of how the parasites reach the brain has been presented so far. Analyzing electron micrographs prepared from rodent brains more than 20 days after infection, we present here conclusive evidence that the parasites first enter the brain via the choroid plexus from where they penetrate the epithelial cell layer to reach the ventricular system. Adversely, no

OBJECTIVE: The purpose of this study was to identify prognostic indicators that would lead to stratification of patients likely to have successful surgery for sleep-disordered breathing (SDB) versus those destined to fail.
STUDY DESIGN: We retrospectively reviewed 134 patients to correlate palate position and tonsil size to the success of the UPPP as based on postoperative polysomnography results. Similar to our previously published data on the Friedman Score as a predictor of the presence and severity of SDB, the palate position was determined on physical examination of the oral cavity and w

The accuracy of actigraphy for sleep staging is assumed to be poor, but examination is limited. This systematic review aimed to assess the performance of actigraphy in sleep stage classification of adults. A systematic search was performed using MEDLINE, Web of Science, Google Scholar, and Embase databases. We identified eight studies that compared sleep architecture estimates between wrist-worn actigraphy and polysomnography. Large heterogeneity was found with respect to how sleep stages were grouped, and the choice of metrics used to evaluate performance. Quantitative synthesis was not possi

STUDY OBJECTIVES: Sleep is an important biological process that is perturbed in numerous diseases, and assessment of its substages currently requires implantation of electrodes to carry out electroencephalogram/electromyogram (EEG/EMG) analysis. Although accurate, this method comes at a high cost of invasive surgery and experts trained to score EEG/EMG data. Here, we leverage modern computer vision methods to directly classify sleep substages from video data. This bypasses the need for surgery and expert scoring, provides a path to high-throughput studies of sleep in mice.
METHODS: We collect

In 10 peptic ulcer patients, 5 duodenal ulcer and 5 gastric ulcer patients, the relation of nocturnal intragastric pH to EEG sleep stages was examined. The nocturnal intragastric pH was monitored by pH-measuring telemeter and sleep stages were recorded simultaneously by electroencephalography throughout the night. The EEG sleep stages were classified after the criteria of Dement and Kleitman. The average of intragastric pH level at each sleep stage was higher in gastric ulcer than the duodenal ulcer. The intragastric pH level became low as the patients' sleep advanced to deep stage. This tende

INTRODUCTION: Alzheimer's disease (AD) is characterized by progressive cognitive decline and brain network dysfunction. EEG microstates offer a sight into rapid network dynamics. However, microstate alterations in AD during waking and different sleep stages remain unexplored.
METHODS: Overnight EEG was recorded from 15 AD patients and 15 healthy controls. Sleep stages (Wake, N1, N2, N3, REM) were manually scored. Microstate analysis extracted temporal parameters and transition probabilities separately for each sleep stage using a common set of grand-mean template maps derived from all stages.

Sleep stages exhibit differing patterns of cardiac, respiratory and nervous system activation. Rapid eye movement (REM) sleep is associated with deeper oxygen desaturation events in obstructive sleep apnea. However, no studies have looked to the effect of sleep staging on re-saturation characteristics. Polysomnographic data from the Sleep Heart Health Study were utilised to derive oxygen saturation parameters from events having both desaturation and re-saturation parts. Sleep stages were described as non-REM (NREM) 1 or 2 (combined), 3 and REM. Sleep stage effects on desaturation and re-satura

BACKGROUND: Proteostasis, in particular the impairment of autophagic activity, is linked to sleep dysregulation and is an early sign of dementias including Alzheimer's disease (AD). This coupling of events may be a critical alteration driving proteinopathy and AD progression. In the present study, we investigated sleep-wake and memory regulating neurons for vulnerability to autophagic impediment, and related these findings to progression of the sleep and cognitive phenotype. METHODS: Using the double knock-in AD mouse model, AppNL-G-FxMAPT, we examined phenotypic and pathological alterations

OBJECTIVE: Sleep dysregulation in Parkinson's disease (PD) has been hypothesized to occur, in part, from dysfunction in the basal ganglia-cortical circuit. Assessment of this relationship requires accurate sleep stage determination, a known challenge in this clinical population. Our objective was to optimize the consensus on the sleep staging process and reduce interrater variability in a cohort of advanced PD subjects.
METHODS: Fifteen PD subjects were enrolled from three sites in a clinical trial that involved recordings from subthalamic nucleus (STN) deep brain stimulation (DBS) leads (NCT

BACKGROUND: The Multiple Sleep Latency Test (MSLT) remains an important diagnostic tool in the diagnosis of hypersomnias. However, a positive MSLT may be found in other sleep disorders, such as behaviourally induced inadequate sleep syndrome (BIISS). It has been demonstrated that in sleep onset rapid eye movement (SOREM) periods in BIISS, REM sleep tends to arise from stage 2 sleep (non-REM (NREM) 2), rather than stage 1 sleep (NREM1), as in narcolepsy.
METHODS: We performed sleep stage sequence analysis on 127 patients with nocturnal polysomnography and MSLT, including 25 with narcolepsy wit

BACKGROUND: Isolated REM sleep behavior disorder (iRBD) is a prodromal stage of α-synucleinopathies, including Parkinson's disease (PD). Conventional sleep staging assigns each epoch to a single dominant stage, potentially obscuring within-epoch probabilistic features and stage mixing. We aimed to characterize alternation of probabilistic sleep-stage patterns in iRBD and to examine their clinical and neurobiological correlations.
METHODS: In this case-control study, 104 patients with iRBD, 28 patients with PD and RBD (PD + RBD), and 50 controls underwent video polysomnography. Sleep-stage unc

BACKGROUND/OBJECTIVE: Sound-based AI models for sleep staging face challenges in shared sleeping environments due to acoustic interference from bed partners. This study aimed to evaluate the performance of a sound-based model in two-person polysomnography (PSG) scenarios, with independently recorded sound data for each participant.
METHODS: Eighty-eight participants (37 males, 51 females) were recruited, including 74 from mixed-gender pairs and 14 from all-female pairs. Bed partners underwent simultaneous PSG in a shared room, with sound recorded separately for each participant using MEMS mic

OBJECTIVES: Sleep apnea (SA) is common in patients with end-stage renal disease (ESRD) and such patients are likely to suffer additional sleep disruption associated with restless legs syndrome (RLS) and periodic leg movements (PLM). Our objective was to evaluate sleep quality in ESRD patients who are newly diagnosed with SAand determine the additional contribution of PLM to sleep disruption.
METHODS: Two groups of patients with SA (apnea-hypopnea index (AHI) > 15) were compared, one with ESRD (n = 12) and the other with normal renal function (n = 18), using a sleep history questionnaire, slee

Human African trypanosomiasis (HAT), due to the transmission of Trypanosoma brucei (T. b.) gambiense and T. b. rhodesiense by tsetse flies, is re-emerging in inter-tropical Africa. It evolves from the hemolymphatic Stage I to the meningo-encephalitic Stage II. The latter is generally treated with melarsoprol, an arseniate provoking often a deadly encephalopathy. A precise determination of the HAT evolution stage is therefore crucial. Stage II patients show: (i) a deregulation of the 24-h distribution of the sleep-wake alternation; (ii) an alteration of the sleep structure, with frequent sleep

Insomnia increases the risk of Alzheimer's disease and is associated with increased amyloid-β (Aβ) levels, but assessments using rigorous objective measures are lacking. The purpose of this cross-sectional study was to determine the association between sleep characteristics and Aβ burden using polysomnography and positron emission tomography (PET). Cognitively normal older adults (ages 60-85) with symptoms of insomnia underwent PET and overnight polysomnography. Pearson correlations determined the association between sleep variables and the standard uptake values ratio (SUVR) within global cer
