INTRODUCTION: The Gram-positive actinobacterium Streptomyces scabies is the major causal agent of potato common scab. The main pathogenicity factor is thaxtomin A, a phytotoxin that causes atypical cell death, although other secondary metabolites have been described to play a role in S. scabies virulence. Despite this, many aspects of the interaction between S. scabies and its primary host Solanum tuberosum L. remain to be elucidated.
METHODS: Intracellular proteins of S. scabies EF-35 grown in the presence of in vitro produced tubers (microtubers) of the Russet Burbank and Yukon Gold potato

Tropinone reductases (TRs) are essential enzymes in the tropane alkaloid biosynthesis, providing either tropine for hyoscyamine and scopolamine formation or providing pseudotropine for calystegines. Two cDNAs coding for TRs were isolated from potato (Solanum tuberosum L.) tuber sprouts and expressed in E. coli. One reductase formed pseudotropine, the other formed tropine and showed kinetic properties typical for tropine-forming tropinone reductases (TRI) involved in hyoscyamine formation. Hyoscyamine and tropine are not found in S. tuberosum plants. Potatoes contain calystegines as the only pr

The toxicity of cadmium (Cd) has posed major public health concern in crops grown in the Cd-contaminated soils. The effects of five amendments, nano-hydroxyapatite (n-HA) and it combined with lime, zeolite, bone mill and fly ash on Cd immobilization in soils and uptake in potatoes, were investigated in a contaminated soil by pot experiments. The result showed that the applications of combined amendments significantly decreased the bioavailable Cd concentrations extracted by TCLP, DTPA-TEA and MgCl2 in the contaminated soils, and changed the soluble and exchangeable and specifically sorbed frac

To evaluate the role of salicylic acid (SA) in Nb-mediated hypersensitive resistance to Potato virus X (PVX) avirulent strain ROTH1 in Solanum tuberosum, we have constructed SA-deficient transgenic potato plant lines by overexpressing the bacterial enzyme salicylate hydroxylase (NahG), which degrades SA. Evaluation of these transgenic lines revealed hydrogen peroxide accumulation and spontaneous lesion formation in an age- and light-dependent manner. In concordance, NahG potato plants were more sensitive to treatment with methyl viologen, a reactive oxygen species-generating compound. In addit

An effective system for isolating and regenerating protoplasts is crucial for research in genome engineering. This study focused on refining a protocol for the isolation and regeneration of mesophyll protoplasts from the leaves of Solanum tuberosum cv. Kufri Jyoti. Key factors influencing protoplast yield and viability, such as dark pretreatment, pre-plasmolysis, enzyme concentrations, and osmoticum levels, were thoroughly assessed and optimized. The highest protoplast yield and viability were achieved with an enzyme mixture of 1.0% cellulase R-10 and 0.5% macerozyme R-10 after 16 h of incubat

A growing trend in plant protection is replacing chemical preparations with environmentally friendly biological compositions. Chitosan, due to its biocompatibility, biodegradability, and bioactivity, is an effective agent against plant diseases. The purpose of the study was to evaluate chitosan as a potential biopesticide for potato plants. Three variants of chitosan were tested: high (310-375 kDa, >75% deacetylated), medium (190-310 kDa, 75-85% deacetylated), and low (50-190 kDa, 75-85% deacetylated) molecular weight. The chitosan variants were dissolved in lactic and succinic acids and teste

Thaxtomin A (TA) is a phytotoxin secreted by Streptomyces scabies that causes common scab in potatoes. However, the mechanism of potato proteomic changes in response to TA is barely known. In this study, the proteomic changes in potato leaves treated with TA were determined using the Isobaric Tags for Relative and Absolute Quantitation (iTRAQ) technique. A total of 693 proteins were considered as differentially expressed proteins (DEPs) following a comparison of leaves treated with TA and sterile water (as a control). Among the identified DEPs, 460 and 233 were upregulated and downregulated, r

Drought stress is a primary abiotic constraint limiting potato productivity. While polyphenol oxidase (PPO) is known to participate in stress responses, the specific role of StuPPO9 in drought tolerance remains poorly understood. In this study, we generated StuPPO9-overexpressing (OE) and CRISPR/Cas9-mediated knockout (C4) lines in the potato cultivar 'Atlantic'. Under sustained drought stress, OE lines exhibited significantly superior growth phenotypes compared to wild-type (WT) and C4 plants, characterized by increased leaf and root relative water content, root number and enhanced photosynth

The valorization of agri-food by-products aligns with circular economy principles and offers sustainable sources of bioactive compounds. This study investigated the peels of the purple-fleshed Solanum tuberosum L. cv. Vitelotte Noire (VN), cultivated in Sardinia, as a potential resource for nutraceutical antioxidants. Extracts were obtained using solvents of different polarities (water, 80% and 96% ethanol) and characterized. Phytochemical screening revealed high concentrations of total phenolics, flavonoids, and anthocyanins, with the 96% ethanolic extract showing superior anthocyanin content

Karyotypic studies on cell suspensions and calli of an S-(2-aminoethyl)cysteine resistant cell line of the interdihaploid potato H(2)578 (2n=24) revealed a high degree of variation in the number of chromosomes (33-217) and dicentric chromosomes (0-8). The suspensions also exhibited megachromosomes and fused chromosomes. Differential staining showed that in suspensions dicentrics survived mitotic cycles mainly by parallel separation of the chromatids during anaphase and hardly by the breakage-fusion-bridge cycle. Two phenotypes of plantlets regenerated, each with 34 or 35 chromosomes with gross

Pectobacterium carotovorum (Pc) is a phytopathogenic strain that causes soft rot disease in potato (Solanum tuberosum L.), resulting in postharvest losses. Chemical control is effective for managing this disease, but overdoses cause adverse effects. Because farmers insist on using chemical agents for crop protection, it is necessary to develop more effective pesticides in which the active compound released can be regulated. In this context, we proposed the synthesis of ZnAl-NADS, in which nalidixic acid sodium salt (NADS) is linked to a ZnAl-NO3 layered double hydroxide (LDH) host as a nanocar

Yeast models are widely used to study molecular chaperones from diverse organisms, including plants, because of their well-characterized genetics and the conservation of the protein-folding machinery among eukaryotes. Cross-species complementation studies in yeast have yielded valuable insights into conserved biochemical activity and molecular functions that manage protein folding, assembly, and repair during stress. This study evaluated the functional capacity of three potato StBiP isoforms (StBiP1, StBiP2, and StBiP3) to complement the kar2 deletion (kar2Δ) strain under a range of environmen

AIMS: To study the antifungal effects of the potato secondary metabolites α-solanine, α-chaconine, solanidine and caffeic acid, alone or combined.
METHODS AND RESULTS: Resistance to glycoalkaloids varied among the fungal species tested, as derived from minimum inhibitory concentrations assays. Synergistic antifungal activity between glycoalkaloids and phenolic compounds was found. Changes in the fluidity of fungal membranes caused by potato secondary plant metabolites were determined by calculation of the generalized polarization values. The results partially explained the synergistic effect

Solanidine is a steroidal aglycon of potato (Solanum tuberosum L.) glycoalkaloids and a very important precursor for the synthesis of hormones and some pharmacologically active compounds. Glycoalkaloids are hydrolyzed by mineral acid, yielding solanidine. This paper deals with the kinetics of solanidine hydrolytic extraction in different solid-liquid-liquid systems. The dried and milled potato (S. tuberosum L.) vines were used as a source of glycoalkaloids and as the solid phase. The solutions of hydrochloric acid in 2 and 10% (w/v) aqueous acetic acid, in 50% (volume) aqueous methanol, and in

Mutants in the eukaryotic initiation factor 4E (eIF4E) gene family provide recessive resistance to plant viruses. Previously, we demonstrated that knocking out SteIF4E1 potato alleles (A, B, B, B1) resulted in limited resistance to a necrogenic PVY isolate (NTN). Expanding on this, we used a non-transgenic method employing Cas12a and a single-stranded oligodeoxynucleotide (ssODN) as a repair template to introduce amino acid changes in SteIF4E1, attempting to mimic the Capsicum PVY resistance allele pvr21. However, no plants that had been regenerated from transfected protoplast cells displayed

Malnutrition is one of the global issues of public health concern, and iron and zinc deficiencies are at the top of the list. Iron deficiency affects more than 2 billion people in the world and is a major cause of anemia. Potato has the potential to be an important source of iron and zinc. This study assessed the nature and magnitude of genetic variability in Fe and Zn concentrations, tuber yield, and quality traits among biofortified tetraploid potato clones and their relationships through correlation and path analysis. A total of 45 potato genotypes, including the variety Gudanie, were grown

Potato virus Y (PVY) is one of the most harmful phytopathogens. It causes big problems for potatoes and other important crops around the world. Nanoclays have been extensively studied for various biomedical applications. However, reports on their interactions with phytopathogens, particularly viral infections, are still limited. In this study, the protective activity of Egyptian nanoclay (CE) and standard nanoclay (CS) against PVY was evaluated on potato (Solanum tuberosum L.) plants. Their physicochemical and morphological properties were examined with scanning electron microscopy (SEM), tran

Potato viral infections are among the main factors contributing to reduced quality of planting material and decreased tuber productivity. Currently, no reliable chemical control methods are available for plant viral diseases. Therefore, the development of potato cultivars carrying virus resistance genes remains one of the most effective and comprehensive approaches to this problem. In this study, 29 Russian and foreign potato cultivars, as well as 31 Far Eastern potato hybrids were evaluated. Resistance genes were identified using PCR analysis. The following cultivars carrying target resistanc

Cellular RNA-dependent RNA polymerases (RDRs) catalyze synthesis of double-stranded RNAs that can serve to initiate or amplify RNA silencing. Arabidopsis thaliana has six RDR genes; RDRs 1, 2 and 6 have roles in anti-viral RNA silencing. RDR6 is constitutively expressed but RDR1 expression is elevated following plant treatment with defensive phytohormones. RDR1 also contributes to basal virus resistance. RDR1 has been studied in several species including A. thaliana, tobacco (Nicotiana tabacum), N. benthamiana, N. attenuata and tomato (Solanum lycopersicum) but not to our knowledge in potato (
