Squalene is a natural linear triterpene that can be found in high amounts in certain fish liver oils, especially from deep-sea sharks, and to a lesser extent in a wide variety of vegeTable oils. It is currently used for numerous vaccine and drug delivery emulsions due to its stability-enhancing properties and biocompatibility. Squalene-based vaccine adjuvants, such as MF59 (Novartis), AS03 (GlaxoSmithKline Biologicals), or AF03 (Sanofi) are included in seasonal vaccines against influenza viruses and are presently being considered for inclusion in several vaccines against SARS-CoV-2 and future

Squalene is a high-value triterpenoid. It is increasingly recognized for its regulatory roles in plant signaling. However, the functions of squalene in cotton remain unexplored. Here, we show that exogenous squalene can promote cotton developmental traits (root elongation and ovule development) and identify GhSQS1 as the predominant functional squalene synthase. Overexpression of GhSQS1 promotes root development, along with metabolic and hormonal reprogramming, including the activation of brassinosteroid (BR)-associated signaling. Furthermore, stable overexpression of GhSQS1 markedly elevates

A three-step synthesis of squalene (1), the universal precursor of sterols, an adjuvant for vaccines, and a component of cosmetics, has been achieved from plant-derived trans, trans-farnesol (2) and 2-nitrobenzenesulfonamide (3). The successful strategy described herein features an end-to-end pairing of farnesyl radicals formed in one laboratory operation from bis-farnesylamine (6) and Levin's anomeric amide (7). This concise synthesis provides squalene in 99.8% purity after purification of the derived thiourea clathrate, following the procedure of Nicolaides and Laves.

Metabolic disorders have been identified as one of the causes of drug-induced liver injury; however, the direct regulatory mechanism regarding this disorder has not yet been clarified. In this study, a single regulatory mechanism of small molecule kinase inhibitors, with crizotinib as the representative drug is elucidated. First, it is discovered that crizotinib induced aberrant lipid metabolism and apoptosis in the liver. A mechanistic study revealed that crizotinib treatment promoted the accumulation of squalene epoxidase (SQLE) by inhibiting autophagosome-lysosome fusion which blocked the a

This study describes the assessment of mucosal adjuvant activity of a squalene-based nanoemulsion (SQ@NE) following intravaginal delivery in mice. After immunization, a high level of recruitment of CD11b/c+ granulocytes and F4/80+ macrophages was observed in the vaginal mucosal tissues of the mice immunized with a model protein ovalbumin (OVA) formulated with SQ@NE, and then downstream regulated the expression of MHC II and costimulatory molecules CD40 and CD86 on CD11c+ cells harvested from the associated draining lymph node. With respect to cytotoxic T lymphocyte immunity, the mice immunized

The authors prepared experimental lipoid adjuvant of the oil-in-water (O/W) type, based on squalene (ESA: experimental squalene adjuvant), intended for liquid inactivated veterinary vaccines. The final concentration of squalene in the vaccine was 5% (w/v). Detergents Poloxamer 105 in the concentration of 4% (w/v) and Abil-Care in the concentration of 2% were used as emulsifiers. The water component was inactivated virus suspension. Emulsification at decreased temperatures (up to 37 degrees C) fails to denaturate the immunization antigen. The effectiveness of experimental squalene adjuvant was

Insights in molecular developmental biology in animals and humans are facilitating the understanding of pathophysiologic mechanisms in dysmorphogenesis or abnormalities in normal embryologic structural development. A milestone was recognition of the role of shh in morphogenesis of craniofacial structures, especially the development of holoprosencephaly. The dependence of hedgehog morphogens on cholesterol modification for normal hedgehog signaling function has particular relevance to disorders of cholesterol synthesis which manifest dysmorphogenesis. Four human disorders of morphogenesis (Smit

Cholesterol biosynthesis was evaluated in different parts of the human gastro-intestinal tract by incorporation of labelled acetic or mevalonic acid into non-saponifiable metabolites and also by measuring squalene epoxidase activity, oxido=squalene cyclase activity and dihydrolanosterol demethylation rate. With respect to rat liver, whole cholesterol biosynthesis was generally low and we found an anatomic localization of activity towards the end of the gastro-intestinal tract. The results of analysis of mevalonic acid metabolites implied the existence of post-HMG CoA regulation. A comparison w

Squalene is an antioxidant that plays an essential role in fat metabolism. The study aimed to assess the effect of squalene supplied in feed on the growth performance, health status, and fatty acid profiles of muscle and liver of Siberian sturgeon, rainbow trout, and Eurasian perch. The experimental feeds containing 0%, 0.5%, and 1.0% squalene were prepared for each fish species. Hematological and biochemical indices, liver histology, and fatty acid profiling of muscle and liver were analyzed. Squalene supplementation was safe for fish, and no negative influence on growth status was observed.

Immunosuppressive drugs are widely used for the treatment of autoimmune diseases and to prevent rejection in organ transplantation. Gusperimus is a relatively safe immunosuppressive drug with low cytotoxicity and reversible side effects. It is highly hydrophilic and unstable. Therefore, it requires administration in high doses which increases its side effects. To overcome this, here we encapsulated gusperimus as squalene-gusperimus nanoparticles (Sq-GusNPs). These nanoparticles (NPs) were obtained from nanoassembly of the squalene gusperimus (Sq-Gus) bioconjugate in water, which was synthesize

The skin is the largest organ in the integumentary system, protecting against various external threats, including ultraviolet exposure, heat, infections, dehydration and mechanical injuries. Skin disorders can arise from various causes, including allergic reactions or breaches in the skin barrier, which allow microorganisms or chemicals to penetrate the sweat ducts. These conditions encompass a wide range of issues, including acne, xerosis (dry skin), fungal infections, atopic dermatitis (eczema) and psoriasis. Collectively, these ailments affect a significant portion of the global population,

BACKGROUND: Lapaquistat acetate is a squalene synthase inhibitor investigated for the treatment of hypercholesterolemia.
METHODS AND RESULTS: This report summarizes the phase 2 and 3 results from the lapaquistat clinical program, which was halted at an advanced stage as a result of potential hepatic safety issues. Efficacy and safety data were pooled from 12 studies (n=6151). These were 6- to 96-week randomized, double-blind, parallel, placebo- or active-controlled trials with lapaquistat monotherapy or coadministration with other lipid-altering drugs in dyslipidemic patients, including a lar

Based on our previous finding (Biochem. Biophys. Res. Commun., 223, 578-582, 1996) of singlet oxygen generation from coproporphyrin excreted on the skin surface from Propionibacterium acnes, we hypothesized that singlet oxygen formed in this way under UV exposure would promote peroxidation of skin surface lipids. We found that squalene was oxidized efficiently by singlet oxygen derived from coproporphyrin under UV exposure, and that the rate constant of squalene peroxidation by singlet oxygen was ten-fold higher than that of other skin surface lipids examined. The reaction was promoted more ef

Overproduction of sebum, especially during adolescence, is causally related to acne and inflammation. As a way to reduce sebum and its interference with the process of follicular keratinization in the pilosebaceous unit leading to inflammatory acne lesions, antihistamines were investigated for their effect on sebocytes, the major cell of the sebaceous gland responsible for producing sebum. Reverse transcriptase-PCR analysis and immunofluorescence of an immortalized sebocyte cell line (SZ95) revealed the presence of histamine-1 receptor (H-1 receptor), and thus indicated that histamines and, co

Squalene, found in earlier studies in human atherosclerotic plaques, was measured in the serum of postmenopausal women with coronary artery disease (CAD) (n = 25) and randomly chosen age-matched healthy controls (n = 30). The squalene concentrations of the whole population ranged from 37.5 to 115.5 microg/dl, and were higher in serum of the CAD than healthy women (91.4+/-2.6 versus 65.2+/-2.6 microg/dl, P = 0.000), a finding observed also in relation to cholesterol (43.8+/-1.8 versus 32.9+/-1.1 10(2)x mmol/mol of cholesterol, P = 0.000). The squalene concentration was also increased in chylomi

Squalene is a triterpene with potent biological activities. Squalene (C30H50) is a linear polyunsaturated hydrocarbon composed of six isoprene units and six carbon-carbon double bonds. It serves as an essential precursor for sterols, steroid hormones, and vitamin D in humans and exhibits antioxidant, anti-tumor, and lipid-regulating properties. In plants, squalene is produced via the mevalonate (MVA) and 2-C-methyl-D-erythritol-4-phosphate (MEP) pathways. The key rate-limiting enzymes in these pathways include 3-hydroxy-3-methylglutaryl-CoA reductase (HMGR), farnesyl diphosphate synthase (FPS)

Bladder cancer (BC) is one of the most common cancers worldwide. Although the treatment and survival rate of BC are being improved, the risk factors and the underlying mechanisms causing BC are incompletely understood. Squalene monooxygenase (SQLE) has been associated with the occurrence and development of multiple cancers but whether it contributes to BC development is unclear. In this study, we performed bioinformatics analysis on paired BC and adjacent non-cancerous tissues and found that SQLE expression is significantly upregulated in BC samples. Knockdown of SQLE impairs viability, induce

Abstract The effects of indoor conditions (ozone concentration, temperature, relative humidity (RH), and the presence of NO(x)) on heterogeneous squalene oxidation were studied with Attenuated Total Reflectance-Fourier Transform Infrared spectroscopy. The heterogeneous kinetics of squalene-ozone reaction revealed a pseudo-first-order reaction rate constant of 1.22 x 10(-5)/s at [O(3)] = 40 ppb. Oxidation kinetics were insensitive to temperature over the range of 24-58 +/- 2 degrees C as well as to RH and presence of NO(x). Products, however, were affected by the environmental parameters. Metabolic reprogramming is a hallmark of malignancy. SQLE, a key regulated enzyme catalyzing the first oxygen-dependent step of cholesterol biosynthesis downstream of canonical HMG-CoA reductase, is aberrantly upregulated across multiple human malignancies and drives tumor progression and therapeutic resistance. This review examines the emerging role of SQLE as a potential metabolic-immune checkpoint, systematically integrating cutting-edge epitranscriptomic regulatory mechanisms published from 2024 to 2026, dissecting the full spectrum of SQLE-mediated immunosuppressive pathways, and conducti
