Easy Come, Easy Go: Capillary Forces Enable Rapid Refilling of Embolized Primary Xylem Vessels.

Protoxylem plays an important role in the hydraulic function of vascular systems of both herbaceous and woody plants, but relatively little is known about the processes underlying the maintenance of protoxylem function in long-lived tissues. In this study, embolism repair was investigated in relation to xylem structure in two cushion plant species, Azorella macquariensis and Colobanthus muscoides, in which vascular water transport depends on protoxylem. Their protoxylem vessels consisted of a primary wall with helical thickenings that effectively formed a pit channel, with the primary wall bei

Testing the 'microbubble effect' using the Cavitron technique to measure xylem water extraction curves.

Plant resistance to xylem cavitation is a major drought adaptation trait and is essential to characterizing vulnerability to climate change. Cavitation resistance can be determined with vulnerability curves. In the past decade, new techniques have increased the ease and speed at which vulnerability curves are produced. However, these new techniques are also subject to new artefacts, especially as related to long-vesselled species. We tested the reliability of the 'flow rotor' centrifuge technique, the so-called Cavitron, and investigated one potential mechanism behind the open vessel artefact

Identification of Flavonoids in the Xylem of Castanopsis hystrix and Their Content Variation Patterns During Heartwood Formation.

BACKGROUND: Flavonoid secondary metabolites accumulate in xylem during heartwood formation and critically influence wood color and economic value. However, the distribution patterns of individual flavonoids across sapwood, transition zone, and heartwood remain poorly characterized in Castanopsis hystrix, limiting mechanistic understanding of heartwood formation and rational resource utilization. METHODS: Sapwood, transition zone, and heartwood samples were collected and analyzed using ultra-performance liquid chromatography coupled with Q-Exactive Orbitrap mass spectrometry (UPLC-QE-MS) for q

The effects of copper deficiency on lignification, xylem vessel structure, and hydraulic traits in hybrid poplar.

Copper (Cu) homeostasis is integral to many plant physiological processes, including lignification of plant cell walls. This link occurs through Cu's role as a cofactor in the apoplastic laccase enzymes that oxidize monolignols that then polymerize to form the hydrophobic lignin polymer, which provides rigidity and strength to the water transport system. In this study, we investigated the effect of Cu deficiency on lignin content and chemistry in poplar stems. We also examined the effect of Cu deficiency on the stiffness of stem wood and the hydraulic properties of leaves. Cu deficiency result

Influence of xylem fluid chemistry on planktonic growth, biofilm formation and aggregation of Xylella fastidiosa.

Xylella fastidiosa is the causal agent of Pierce's disease in grapevines. The mechanisms of pathogenicity are largely due to occlusion of xylem vessels by aggregation of X. fastidiosa and biofilm formation. Xylella fastidiosa was subjected to xylem fluids with varying chemistries to examine the effects of nutritional components on bacterial growth in vitro. The exposure of X. fastidiosa to xylem fluids collected from different Vitis genotypes resulted in highly significant differences in both planktonic growth and biofilm formation. Planktonic growth of X. fastidiosa in Vitis xylem fluid was c

Evidence for a trade-off between growth rate and xylem cavitation resistance in Callitris rhomboidea.

The ideal plant water transport system is one that features high efficiency and resistance to drought-induced damage (xylem cavitation), however, species rarely possess both. This may be explained by trade-offs between traits, yet thus far, no proposed trade-off has offered a universal explanation for the lack of water transport systems that are both highly drought-resistant and highly efficient. Here, we find evidence for a new trade-off, between growth rate and resistance to xylem cavitation, in the canopies of a drought-resistant tree species (Callitris rhomboidea). Wide variation in cavita

Seasonal Xylem Sap Acidification Is Governed by Tree Phenology, Temperature and Elevation of Growing Site.

pH of xylem sap (pHx) was determined in three trees (Malus domestica (apple tree), Picea abies and Pinus cembra) in response to seasonal changes. Conifer trees from lowland (600 m) were compared to trees growing at the alpine timberline (1950 m a.s.l.). Xylem sap was extracted with a Scholander pressure bomb and pHx was measured with a pH microsensor. In all species, pHx changed markedly with season. In spring, pHx was acidic; during winter, the pHx was more alkaline. In apple trees, the pHx did not show a significant correlation with temperature but was rather affected by developmental stage.

Salicylic acid-mediated competitive regulation of xylem vessel formation and defense responses in Arabidopsis thaliana.

In vascular plants, xylem vessels transport water and contribute to structural integrity. As part of vessel formation, xylem cells deposit secondary cell walls (SCWs), which are composed of cellulose, hemicellulose, and lignin polymers. Under environmental challenges such as pathogen attack, a growth-defense trade-off limits xylem vessel development. Understanding the mechanism regulating this trade-off has implications for understanding of plant strategy to utilize their carbon resources because SCWs contain large amounts of densely packed high-carbon compounds. Here, we investigated the effe

Gene expression in vessel-associated cells upon xylem embolism repair in Vitis vinifera L. petioles.

In this work, the involvement of vessel-associated cells in embolism recovery was investigated by studying leaf petiole hydraulics and expression profiles of aquaporins and genes related to sugar metabolism. Two different stress treatments were imposed onto grapevines to induce xylem embolism: one involved a pressure collar applied to the stems, while the other consisted of water deprivation (drought). Embolism formation and repair were monitored during stress application and release (recovery). At the same time, stomatal conductance (g(s)), leaf water potential (Ψ(leaf)) and leaf abscisic aci

Integrated Analysis of Grapevine Responses to Winter Pruning Reveals Temperature-Associated, Polyphenol-Rich Xylem Occlusions.

Perennial crops are continually wounded throughout cultivation. To limit pathogen infection and tissue dehydration, plants have evolved a range of wound responses that protect damaged tissues. These responses include changes in gene expression and the synthesis of protective compounds such as polyphenols. In addition, plants occlude damaged xylem vessels following wounding and pathogen infection, forming structures such as gels, gums, and tyloses. We explored grapevine responses to pruning. First, we examined the dynamics of xylem vessel occlusion formation at different time points and depths

Effect of long-term forest fertilization on Scots pine xylem quality and wood borer performance.

We tested whether changes in long-term nutrient availability would affect the xylem quality and characteristics of Scots pine trees as a food source for the larvae of the xylophagous wood borer Hylotrupes bajulus L. (Cerambycidae). We looked for an effect of host plant growth and xylem structural traits on H. bajulus larval performance, and looked for delayed effects of long-term forest fertilization on xylem chemical quality. In general, larval performance was dependent on larval developmental stage. However, the growth of larvae also varied with host plant quality (increases in the concentra

Assimilation dynamics of xylem-transported CO2 by woody tissue photosynthesis revealed with 11C- and 13C-labelling.

A considerable quantity of locally respired CO2 in stems can be assimilated via woody tissue photosynthesis (Pwt). Hence, Pwt locally provides carbon fuelling radial stem growth and therefore plays a major role in the stem carbon balance. It is, however, not straightforward to quantify or visualize its dynamic behaviour, especially on small spatial scales. To assess Pwt we traced xylem-transported 11CO2 in detached branches of Populus tremula L. and used positron autoradiography to visualize and quantify its dynamics on a submillimetre scale, while manipulating light exposure to the branch. Xylem anatomy and hydraulic traits in Vitis grafted cuttings in view of their impact on the young grapevine decline.

Grapevine grafting is an essential practice in viticulture and over the years, various bench grafting techniques have been developed to mechanize the nursery process and to increase the yield in number of viable cuttings. Bench grafting is a fundamental nursery practice that can potentially affect the quality of propagation material also in young decline associated to grapevine trunk diseases and has been recently reported to influence leaf symptoms development associated with diseases of Esca complex. The study aimed to investigate how three bench grafting methods [i.e., (i) Omega graft as me

CsNAC083 and CsVND1 are associated with tea plant leaf angle through xylem development and lignin deposition.

Tea (Camellia sinensis) is an important perennial horticultural crop, but the genetic control of leaf angle, a key trait affecting plant architecture and harvesting efficiency, remains unclear. In this study, we measured leaf angles at different leaf positions and examined the anatomy of the bending region between the leaf midrib and the main stem across 12 tea cultivars. Hongyan 1(HY 1) with large leaf angles and Hongyan 4(HY 4) with small leaf angles were selected for transcriptome analysis. Differentially expressed genes were mainly enriched in hormone signaling, MAPK signaling, and phenylp

Toward scalable wood anatomy: a toolkit for automated xylem cell identification and quantification in woody angiosperms.

Xylem is essential for water and nutrient transport, mechanical support, and carbohydrate storage. Identification and quantification of vascular cell types remain manual, time-consuming, and prone to observer bias, limiting throughput and reproducibility. Automated, integrated tools are critical for scaling wood anatomical studies and enabling comparative analyses across taxa. We assembled a poplar xylem dataset of 1,790 microscopy images with 173,434 annotated instances. Using this dataset, we evaluated seven semantic segmentation models and five YOLOv8 detection models across section types f

Invasion of Clavibacter michiganensis into the Xylem and Surrounding Tissues Drives Histological Alterations and Disease Development in Tomato.

Clavibacter michiganensis is a xylem-colonizing and seed-borne phytopathogen that causes bacterial canker disease in tomato plants. In this study, we aimed to determine the colonization patterns of C. michiganensis, as well as the histological changes in tomato plants during infection. To achieve these, GFP-tagged C. michiganensis was inoculated into tomato plants using three methods: stem injection, soil drenching with or without prior root cutting, and cotton-pad inoculation. After inoculation, bacterial colonization was observed using GFP signals, and histological changes were examined usin

A member of the heavy metal P-type ATPase OsHMA5 is involved in xylem loading of copper in rice.

Heavy metal-transporting P-type ATPase (HMA) has been implicated in the transport of heavy metals in plants. Here, we report the function and role of an uncharacterized member of HMA, OsHMA5 in rice (Oryza sativa). Knockout of OsHMA5 resulted in a decreased copper (Cu) concentration in the shoots but an increased Cu concentration in the roots at the vegetative stage. At the reproductive stage, the concentration of Cu in the brown rice was significantly lower in the mutants than in the wild-type rice; however, there was no difference in the concentrations of iron, manganese, and zinc between tw

Salicylic acid and salicylic acid glucoside in xylem sap of Brassica napus infected with Verticillium longisporum.

Salicylic acid (SA) and its glucoside (SAG) were detected in xylem sap of Brassica napus by HPLC-MS. Concentrations of SA and SAG in xylem sap from the root and hypocotyl of the plant, and in extracts of shoots above the hypocotyl, increased after infection with the vascular pathogen Verticillium longisporum. Both concentrations were correlated with disease severity assessed as the reduction in shoot length. Furthermore, SAG levels in shoot extracts were correlated with the amount of V. longisporum DNA in the hypocotyls. Although the concentration of SAG (but not SA) in xylem sap of infected p

Nitrate transiently rewires transcription in developing xylem of Populus wood to promote cell expansion via CRF4.

Nitrate functions both as a nutrient and a signaling molecule, but how nitrate is perceived and translated into developmental outputs within woody tissues is unknown. Here, we show that developing xylem in hybrid aspen (Populus tremula × P. tremuloides) mounts a rapid, cell-type-specific transcriptional response to nitrate that promotes xylem cell expansion. To isolate local nitrate signaling in wood-forming tissues, we delivered nitrate directly into the xylem sap and profiled transcriptomic responses from 2 to 48 h after treatment. Bulk RNA sequencing revealed a transient response peaking wi

Two Clones, Two Paths: divergent intrinsic xylem traits and hydraulic responses to prolonged drought in poplar.

BACKGROUND AND AIMS: Climate change is increasing the frequency and intensity of droughts, challenging the growth, hydraulic function and survival of woody plants. Poplar is a fast-growing species widely cultivated for biomass and cellulose production, but it is also highly sensitive to water deficit, with substantial variation among species and clones. METHODS: This study compared the drought responses of two Populus×canadensis clones, 'Tucano' and 'San Martino', under gradually imposed, prolonged water stress. Physiological traits and intrinsic wood anatomical characteristics revealed diffe
