INTRODUCTION: Pine wilt disease caused by Bursaphelenchus xylophilus constitutes a severe threat to global pine forests. The UDP-glycosyltransferase (UGT) gene family is involved in xenobiotic detoxification, environmental adaptation, and other biological processes, but systematic identification of this family in B. xylophilus has not yet been performed. This study aimed to comprehensively identify UGT family members in B. xylophilus and clarify their molecular roles in xenobiotic detoxification and host adaptation regulation.
METHODS: Based on genome-wide data, we identified 47 UGT genes in

The pine wood nematode, Bursaphelenchus xylophilus, causes severe damage to pine trees. Current chemical control methods have raised environmental concerns and the resistance issue of the nematode. Due to its high specificity, RNA interference (RNAi) has emerged as a promising approach for controlling pests, including plant-parasitic nematodes, through the application of double-stranded RNA (dsRNA). This study has developed an effective dsRNA-based nematicide for controlling B. xylophilus under field conditions via trunk injection. First, the core RNAi machinery of B. xylophilus was identified

BACKGROUND: Bursaphelenchus xylophilus is one of the most destructive invasive alien species in global forestry, causing widespread pine mortality and severe ecological damage. Clarifying its evolutionary patterns, invasion routes and adaptive signatures helps refine the evolutionary theory of plant-parasitic nematodes, and provides practical references for invasion prevention and targeted management.
RESULTS: The first chromosome-level gap-free reference genome of B. xylophilus was assembled. The genome spans 78.02 Mb with a contig N50 of 12.60 Mb. All sequences were anchored to six chromoso

Twenty-three lignans, including three new compounds (1-3), were isolated from Dendrobium chrysotoxum. Compound 1 exhibited significant contact and oral toxicities against Acyrthosiphon pisum, with LC50 values of 88.7 and 43.1 μg/mL, respectively, along with favorable repellent activity. Its aphicidal mechanism was associated with shrunken and wrinkled aphid cuticles, as well as altered activities of chitinase and digestive and detoxification enzymes. Meanwhile, compound 1 showed optimal nematicidal activity against Bursaphelenchus xylophilus, with an LC50 of 32.9 μg/mL. SEM, TEM and physiologi

The pine-wood invasive species nematode Bursaphelenchus xylophilus causes great forestry damage globally, particularly in Eurasia. B. xylophilus can hybridize with its native sibling, Bursaphelenchus mucronatus, with whom it shares an interestingly asymmetric mating behavior. However, the molecular mechanism underlying interspecific asymmetric mating has yet to be clarified. ntr-1, a nematocin receptor gene, is involved in an oxytocin/vasopressin-like signaling system that can regulate reproduction. Structural analysis using bioinformatics revealed that both Bxy- and Bmu-ntr-1 encode 7TM-GPCR,

Pinewood nematode (PWN) (Bursaphelenchus xylophilus) is a destructive pest of Pinus trees worldwide which depends for transmission on the cerambycid insects of the genus Monochamus. EFSA was asked to assess the potential role of young host plants for afforestation or reforestation planting as a pathway for PWN. According to legislation (EU COM/2023/415 final), these seedlings are not older than 2 years. Based on communication with SNPF (Syndicat National des Pépiniéristes Forestiers), such pine seedlings are assumed to have a collar diameter no more than 12 mm. It was evaluated that seedlings,

BACKGROUND: Pinus sylvestris var. mongolica is a naturally susceptible host for the pine wood nematodes (PWNs, Bursaphelenchus xylophilus) and a dominant afforestation tree species in northeastern China. With the continuous northward expansion of PWNs, P. sylvestris var. mongolica may become a critical conduit for their further expansion. To prevent pine wilt disease (PWD) from spreading via P. sylvestris var. mongolica in northeastern China, it is essential to elucidate its defense mechanisms against PWN and enhance its resistance.
RESULTS: Transcriptome analysis of P. sylvestris var. mongol

BACKGROUND: Pine wilt disease caused by Bursaphelenchus xylophilus can devastate the pine forest ecosystem. Lignification is a vital strategy to help hosts resist pathogens, but how B. xylophilus effectors regulate the lignin biosynthesis of pine trees are unknown.
RESULTS: In this study, we identified a B. xylophilus neprilysin effector BxNEP1, which rose in the three B. xylophilus transcriptomes of different infection stages. We demonstrated that BxNEP1 is required for B. xylophilus reproduction and virulence during parasitic stages. Silencing of the BxNEP1 gene resulted in the upregulation

The pinewood nematode (PWN) Bursaphelenchus xylophilus (Steiner & Buhrer 1934) Nickle 1970 is the causal agent of pine wilt disease. It is especially damaging in East Asian countries, including Japan, China, and Korea. In China, the nematode has been found in Anhui, Guangdoung, Guizhou, Chongqing, and Zhejiang Provinces since its discovery in Jiangsu Province in 1982 (1). China is confronted with an enormous threat to its pine forests. B. xylophilus is transmitted by the insect vector pine sawyer beetle (Monochamus alternatus). The main host trees are Pinus massoniana, P. thunbergii, and P. de

Pine wilt disease, caused by Bursaphelenchus xylophilus and associated bacterial communities, represents a serious biosecurity threat to global forestry. Phytosanitary management of infested wood requires agents that combine rapid, broad-spectrum bactericidal activity with direct nematicidal efficacy. We characterized the antimicrobial and nematicidal properties of AQAS, a multi-component disinfectant comprising a ternary blend of short-, long-, and double-chain quaternary ammonium compounds (QACs) together with ethanol and a dual-chelation buffering system. Against representative bacterial te

Widespread concern for the occurrence of resistant strains, along with the avoidance of the use of highly toxic insecticides and their wide environmental dispersal, highlights the need for the development of new and safer pest control agents. Natural products provide inspiration for new chemical entities with biological activities, and their analogues are good lead compounds for the development of new pest control agents. For this purpose, we evaluated the larvicidal and nematicidal activities of 48 3-acylbarbituric acid analogues against the Asian tiger mosquito, Aedes albopictus and the pine

The spatial distribution of vine plants contaminated by Xylophilus ampelinus, the agent responsible for bacterial necrosis, was studied over a 5-year period within two vineyards in the Cognac area. Both vineyards were planted with Vitis vinifera cv. Ugni blanc but were different in age and agronomic location. The emission of X. ampelinus in contaminated bleeding sap was observed during vine sprouting. Contaminated bleeding sap is an important source of inoculum for external contamination due to the high susceptibility of young merging shoots to the pathogen. X. ampelinus emission by bleeding s

Plant-parasitic nematodes cause substantial economic losses in agriculture globally. Chemical nematicides with cost-effectiveness and immediate availability continue to be widely used; however, their environmental and health risks have driven the discovery of eco-friendly alternatives. In this study, 60 natural quinones were screened for nematicidal activity to discover new nematicides that are environmentally friendly. Therefore, by evaluating the activities of these compounds against B. xylophilus, A. besseyi, and D. destructor, and find that 5,8-dihydroxy-1,4-naphthoquinone (DHNQ) exhibited

BACKGROUND: The pine wilt disease (PWD) caused by Bursaphelenchus xylophilus is considered the 'cancer' of pine trees, and protecting pine forests from the destructive impact of the nematode remains a significant challenge. Chemical control is one of the most effective methods for managing plant nematodes. Developing new nematicides is an effective strategy and an urgent need for Bursaphelenchus xylophilus management.
RESULTS: Amide derivatives containing 1,2,4-oxadiazole were designed and synthesized through a scaffold-hopping strategy. Some target compounds exhibited significant nematicidal

Bursaphelenchus xylophilus is a parasitic nematode that causes serious ecological and economic harm to pine trees. Taking ZSZLT-240 (LC50 = 0.889 μg·mL-1) as the lead compound, 26 azoxy compounds were designed and synthesized. Most of the azoxy compounds showed excellent nematicidal activity, especially Ib, IIa, and IIIh, with LC50 values of 0.424, 0.267, and 0.803 μg·mL-1, respectively, significantly better than avermectin (LC50 = 16.4 μg·mL-1) and emamectin benzoate (LC50 = 13.3 μg·mL-1). Scanning electron microscopy (SEM) showed that the nematodes treated with compounds Ib and IIIh were sev

Plant-parasitic nematodes are primary pathogen that cause global crop yield losses, the economic cost to global agriculture is $157 billion a year. With the prohibition of highly toxic chemical nematicides, there are few nematicides available for prevention and control of nematodes. Tioxazafen (3-phenyl-5-thiophen-2-yl-1,2,4-oxadiazole) is a new broad-spectrum nematicide with good control effects against various nematodes. In order to discover new and environmentally friendly chemical nematicides, five series of 43 novel compounds containing 1,2,4-oxadiazole pharmacophore were designed and syn

Climate change may redistribute forest pests, pathogens, natural enemies, and host trees, yet these components are still often projected independently. We used ensemble species distribution models (SDMs) to map potential climatic suitability for Monochamus alternatus, Dastarcus helophoroides, Scleroderma guani, Bursaphelenchus xylophilus, and a genus-level Pinus host layer, and then summarized niche overlap, range overlap, and multispecies co-suitability under SSP1-2.6, SSP3-7.0, and SSP5-8.5 for the 2050s and 2090s. A structural equation model (SEM) fitted to thresholded binary layers was ret

BACKGROUND: Plant parasitic nematodes severely impair agricultural and forestry production due to the lack of effective nematicides, causing significant yield losses and economic damage.
RESULTS: For discovery of new and efficient nematicidal agents, we developed a series of double 1,2,4-oxadiazole derivatives from our previous studies. Bioassays revealed that compounds B10 and B12 displayed 100% mortality against Bursaphelenchus xylophilus, Aphelenchoides besseyi and Ditylenchus destructor at 50 μg mL-1. In particular, compound B10 was the most potent against B. xylophilus, showing a median

Pine wilt disease is one of the most important forest tree diseases, especially in the East Asian countries of Japan, China, and Korea. The causal agent, Bursaphelenchus xylophilus (3), is transmitted by the insect vectors, pine sawyer beetles (Monochamus spp.). The vectors mainly attack coniferous trees and the infected trees die within several weeks. In Korea, pine wilt disease was first reported in Busan City in 1988, and now, the damaged area covers 7,820 ha and more than 60 cities in Korea. The main host trees are Pinus densiflora and P. thunbergii, which are the most common pines distrib
