Yarrowia lipolytica morphological mutant enables lasting in situ immobilization in bioreactor.

In the present study, we have isolated and characterized a Yarrowia lipolytica morphological mutant growing exclusively in the pseudohyphal morphology. The gene responsible for this phenotype, YALI0E06519g, was identified as homologous to the mitosis regulation gene HSL1 from Saccharomyces cerevisiae. Taking advantage of its morphology, we achieved the immobilization of the Δhsl1 mutant on the metallic structured packing of immobilized-cell bioreactors. We obtained significant cell retention and growth on the support during shake flask and bioreactor experiments without an attachment step prio

Molecular characterization of Yarrowia lipolytica and Candida zeylanoides isolated from poultry.

Yeast isolates from raw and processed poultry products were characterized using PCR amplification of the internally transcribed spacer (ITS) 5.8S ribosomal DNA region (ITS-PCR), restriction analysis of amplified products, randomly amplified polymorphic DNA (RAPD) analysis, and pulsed-field gel electrophoresis (PFGE). ITS-PCR resulted in single fragments of 350 and 650 bp, respectively, from eight strains of Yarrowia lipolytica and seven strains of Candida zeylanoides. Digestion of amplicons with HinfI and HaeIII produced two fragments of 200 and 150 bp from Y. lipolytica and three fragments of

Evaluation of 1,2-diacyl-3-acetyl triacylglycerol production in Yarrowia lipolytica.

Plants produce many high-value oleochemical molecules. While oil-crop agriculture is performed at industrial scales, suitable land is not available to meet global oleochemical demand. Worse, establishing new oil-crop farms often comes with the environmental cost of tropical deforestation. The field of metabolic engineering offers tools to transplant oleochemical metabolism into tractable hosts while simultaneously providing access to molecules produced by non-agricultural plants. Here, we evaluate strategies for rewiring metabolism in the oleaginous yeast Yarrowia lipolytica to synthesize a fo

Investigating Proteome and Transcriptome Defense Response of Apples Induced by Yarrowia lipolytica.

A better understanding of the mode of action of postharvest biocontrol agents on fruit surfaces is critical for the advancement of successful implementation of postharvest biocontrol products. This is due to the increasing importance of biological control of postharvest diseases over chemical and other control methods. However, most of the mechanisms involved in biological control remain unknown and need to be explored. Yarrowia lipolytica significantly inhibited blue mold decay of apples caused by Penicillium expansum. The findings also demonstrated that Y. lipolytica stimulated the activitie

Biotransformation with cellulase, hemicellulase and Yarrowia lipolytica boosts health benefits of okara.

Okara (soybean residue) is a highly perishable food processing by-product from soymilk and tofu manufacture. It contains a large proportion of insoluble dietary fibre (40-60% on a dry basis), as well as digestion-resistant proteins, trypsin inhibitors and phytic acid. These factors contribute lead to the under-utilisation of okara. To improve the overall nutritional quality of okara, sequential saccharification of okara by Celluclast® 1.5L (cellulase) or Viscozyme® L (cellulase and hemicellulase) and fermentation by the yeast Yarrowia lipolytica were performed. The changes in the antioxidant c

[Genetic system for maintaining the mitochondrial human genome in yeast Yarrowia lipolytica].

For the first time, the possibility of maintaining an intact human mitochondrial genome in a heterologous system in the mitochondria of yeast Yarrowia lipolytica is shown. A method for introducing directional changes into the structure of the mitochondrial human genome replicating in Y. lipolytica by an artificially induced ability of yeast mitochondria for homologous recombination is proposed. A method of introducing and using phenotypic selection markers for the presence or absence of defects in genes tRNA-Lys and tRNA-Leu of the mitochondrial genome is developed. The proposed system can be

Enhanced Expression of CsCCD2 Enables Highly Efficient Crocetin Production in Yarrowia lipolytica.

Crocetin, a bioactive carotenoid derivative from saffron, exhibits antioxidant, anti-inflammatory, and metabolic regulatory properties. Its low natural abundance and limited extraction production hinder large-scale production. Microbial synthesis offers a promising alternative but is constrained by low carotenoid cleavage dioxygenase (CCD2) expression. Here, we engineered a zeaxanthin-producing Yarrowia lipolytica strain to enhance CsCCD2 expression via fermentation temperature optimization, SUMOstar fusion, subcellular localization, and precursor supply improvement. The engineered strain achi

Microbial oil production by Yarrowia lipolytica strains using glycerol as carbon source.

Yeasts can produce microbial oil from hydrophobic substrates such as glycerol, a byproduct of the biodiesel industry that is often undervalued in the market. Yarrowia lipolytica is capable of degrading glycerol and is considered a model organism for studying lipid accumulation. Different yeast strains and culture conditions can result in varying lipid yields with distinct fatty acid profiles. Thus, in this study, five strains of Y. lipolytica were tested for microbial oil production using pure glycerol at temperatures of 15 °C and 25 °C. None of the isolates were considered oleaginous at 15 °C

A combined TLC-GC approach for the regiospecific analysis of triacylglycerols in Yarrowia lipolytica.

This study developed a robust, cost-effective thin-layer chromatography (TLC) combined with gas chromatography (GC) protocol for the regiospecific analysis of triacylglycerols (TAGs), with application to microbial oils from engineered Yarrowia lipolytica. Key steps, such as sn-1,3-specific lipase hydrolysis, TLC separation, staining, and fatty acid methyl ester (FAME) derivatization, were systematically optimized. Porcine pancreatic lipase was identified as the most efficient and economical enzyme for hydrolysis. A petroleum ether/diethyl ether/acetic acid (32:8:0.8, v/v/v) TLC solvent system

[Respiratory activity of yeast Yarrowia lipolytica under oxidative stress and heat shock].

Heat shock (45 degrees C) and the effect of oxidants (H2O2) resulted in a decrease of the respiratory activity of yeast cells and their survival rate. Increased resistance to stress effects after mild heat treatment (37 degrees C) or treatment with a nonlethal dose of oxidants (0.5 mM H2O2 for 60 min) was accompanied by appearance of an alternative (cyanide-resistant) oxidative pathway in the mitochondria, which promotes survival due to retention of the capacity for ATP synthesis in the first coupling point at the level of endogenous NADH dehydrogenase. The alternative oxidative pathway is mor

Sustainable microbial production of retinol in Yarrowia lipolytica: From strain engineering to formulation.

Retinol (vitamin A) is traditionally produced by chemical synthesis, but growing demand for sustainable alternatives has increased interest in microbial production. However, the intrinsic instability of retinol complicates both biosynthesis and downstream processing, requiring integrated strategies for product recovery and stabilization. In this study, we report the first purification and formulation of microbially produced retinol. We previously engineered Yarrowia lipolytica for retinol production and improved retinol recovery by supplementing the fermentation with Tween 80 and the antioxida

Expanding Yarrowia lipolytica's metabolic potential for detoxification of cyanogenic glycosides in edible plants.

Cyanides are highly toxic chemicals in several edible plants that threaten food safety and human health. A phenotypically distinct Yarrowia lipolytica strain that efficiently detoxifies multiple cyanogenic glycosides from edible plants was constructed using a family 1 glycosyl-hydrolase (GH1). The strain displayed higher growth rates and metabolic activities when exposed to high concentrations of cyanides than the wild-type. It overexpressed genes that promoted the binding of molecular oxygen to the cytochrome iv complex. The engineered strain repressed fatty acid production to optimize energy

Systematic enzyme and cofactor engineering for efficient ursolic acid biosynthesis in Yarrowia lipolytica.

Ursolic acid (UA) is a pharmaceutically valuable pentacyclic triterpenoid, but its microbial production is constrained by inefficient cytochrome P450 catalysis and limited cofactor availability. Here, we engineered an efficient α-amyrin-producing Yarrowia lipolytica chassis for UA biosynthesis through integrated enzyme engineering, cofactor optimization, and metabolic flux balancing. Screening of heterologous plant cytochrome P450 monooxygenases identified CYP716A48 from Olea europaea as the most efficient α-amyrin oxidase in Y. lipolytica. Fusion of OeCYP716A48 with its redox partner AtCPR1 u

Bioleaching of fly ash by the tropical marine yeast, Yarrowia lipolytica NCIM 3589.

Fly ash collected from an Indian thermal power plant was characterised by scanning electron microscope (SEM)-energy dispersive spectrometer, X-ray diffraction and energy dispersive X-ray fluorescence analysis. The effect of fly ash on the growth and morphology of a metal-tolerant tropical marine yeast, Yarrowia lipolytica NCIM 3589, was studied. The growth of the yeast was unaffected by the presence 0.1, 0.2 or 0.3 % fly ash although the surface-to-volume ratio decreased. The yeast formed biofilms on immobilized fly ash as evidenced by SEM observations. The organism produced citric acid and ad

Harnessing Yarrowia lipolytica as a platform for animal-free and secretory production of human lactoferrin.

Human lactoferrin (hLF) is an iron-binding glycoprotein with important nutritional and therapeutic functions. Microbial production of recombinant hLF offers a scalable and animal-free alternative to extraction from milk, but efficient secretion remains a major challenge. Here, we developed Yarrowia lipolytica as a microbial platform for high-level production and secretion of recombinant hLF. By systematically improving protein expression, secretion, and protein-folding factors, we identified a key limitation in hLF production: simply increasing gene copy number did not proportionally increase

A Novel Strategy for Highly Efficient Heterologous Expression of Carbonic Anhydrase in Yarrowia lipolytica.

Carbonic anhydrases (CAs) efficiently catalyze CO2 reversible hydration, critical for carbon capture and sequestration (CCS), but naturally low yield limits industrial use. Yarrowia lipolytica, an unconventional yeast, is an ideal heterologous expression host with robust adaptability, post-translational modification capacity, and versatile genetic tools. In this study, 10 α-, β-, and γ-class CAs were successfully expressed in Y. lipolytica, and two top-performing candidates were identified: Methanosarcina mazei γ-CA (MmaCA) and Sulfurihydrogenibium azorense α-CA (SazCA). Their production was f

Proteomics Readjustment of the Yarrowia lipolytica Yeast in Response to Increased Temperature and Alkaline Stress.

Yeasts cope with a wide range of environmental challenges using different adaptive mechanisms. They can prosper at extreme ambient pH and high temperatures; however, their adaptation mechanisms have not been entirely investigated. Previously, we showed the pivotal role and flexibility of the sugar and lipid composition of Yarrowia lipolytica W 29 upon adaptation to unfavorable conditions. In this study, we showed that extreme pH provoked significant changes in the cell wall proteins expression, with an increase in both the chaperones of heat shock protein HSP60 and some other proteins with cha

Yarrowia lipolytica and Komagataella phaffii (Pichia pastoris) as Hosts for Heterologous Expression of Lipases.

The market for industrial lipases is growing due to their wide range of applications. To produce these recombinant enzymes to increase the productivity of the bioprocess, various cell factories, including yeast, have been selected. In the last two decades, the study and improvement of two nonconventional yeasts: K. phaffii and Y. lipolytica as cell factories for heterologous lipase production have increased. However, it is important to consider their differences when choosing between the two cell factories. This chapter compares the characteristics of these yeasts and explores options for enha

Engineering of CO2 recycling and formate metabolism for succinic acid production in Yarrowia lipolytica.

Succinic acid (SA), a key platform chemical in biorefineries, faces industrial production challenges stemming from inefficient CO2 fixation and redox imbalances in microbial cell factories. Here, we report a novel strategy to enhance SA biosynthesis in unconventional yeast Yarrowia lipolytica by cooperatively integrating carbonic anhydrase (CA)-mediated CO2 recycling with formate metabolism. Initially, the expression of CA improved the carboxylation efficiency of reductive TCA (rTCA) pathway, resulting in the strain Hi-SA-YlCA, which produced 89.75 g/L of SA with a yield of 1.01 g/g glucose un

Understanding and Eliminating the Detrimental Effect of Thiamine Deficiency on the Oleaginous Yeast Yarrowia lipolytica.

Thiamine is a vitamin that functions as a cofactor for key enzymes in carbon and energy metabolism in all living cells. While most plants, fungi, and bacteria can synthesize thiamine de novo, the oleaginous yeast Yarrowia lipolytica cannot. In this study, we used proteomics together with physiological characterization to elucidate key metabolic processes influenced and regulated by thiamine availability and to identify the genetic basis of thiamine auxotrophy in Y. lipolytica Specifically, we found that thiamine depletion results in decreased protein abundance for the lipid biosynthesis pathwa
