This study investigates whether pregnant women and their fetuses are exposed to environmental tobacco smoke (ETS) as a result of living in apartments. We measured cotinine concentrations in serum, a biomarker of exposure to ETS, in non-smoking women's umbilical cord blood collected at delivery and in maternal blood drawn shortly after delivering a baby. Concurrently, information was collected regarding the women's housing situation, whether family members or co-workers smoked, and other potential exposure factors. Newborns whose non-smoking mothers lived in an apartment during pregnancy were m

The qualitative direct barbituric acid (DBA) method of detecting urine nicotine metabolites was modified to make it quantitative. The performance of the quantitative DBA method was compared with the qualitative method and an established cotinine radio-immunoassay (RIA), using a panel of urines from 128 reported smokers and 383 reported non-smokers. The quantitative DBA method results were highly correlated with the cotinine RIA results, r = 0.85. The coefficients of variation for the two methods were 6% and 10%, respectively. Assuming that the reported smoking history was correct the qualitati

Public awareness of the adverse effects of tobacco use, tobacco regulations, and alternative forms of tobacco consumption have all increased over time. While these advances may influence perception of tobacco consumption, its temporal consistency remains unclear. We define this temporal consistency of tobacco use perception as the stability of biomarker thresholds for self-identification as a tobacco user. We use data from ten waves of the United States National Health and Nutrition Examination Survey (NHANES), covering 1999-2000 through 2017-2018 (N = 36,624). We compare survey responses rega

Cancer epidemiology, biomarkers & prevention : a publication of the American Association for Cancer Research, cosponsored by the American Society of Preventive Oncology2003eng
Epidemiologic data have shown increased risks of lung cancer in nonsmokers exposed to environmental tobacco smoke (ETS). We measured biomarkers in urine samples from nonsmokers before and after a 4-h visit to a casino where smoking is allowed. The tobacco-specific lung carcinogen, NNK [4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone] is a constituent of ETS. Urinary metabolites of NNK, 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol (NNAL) and its glucuronides (NNAL-Gluc), are excellent biomarkers of human uptake of NNK and NNAL. NNAL, as with NNK, is a potent pulmonary carcinogen. Subjects colle

BACKGROUND: Prenatal or early childhood secondhand tobacco smoke (SHS) exposure increases obesity risk. However, the potential mechanisms underlying this association are unclear, but obesogenic eating behaviors are one pathway that components of SHS could perturb. Our aim was to assess associations of prenatal and early childhood SHS exposure with adolescent eating behaviors.
METHODS: Data came from a prospective pregnancy and birth cohort (N = 207, Cincinnati, OH). With multiple informant models, we estimated associations of prenatal (mean of 16 and 26 weeks of gestation maternal serum cotin

BACKGROUND: Nicotine lozenges help reduce nicotine gradually. They are a discreet NRT alternative. This study compared nicotine dependence, cotinine levels, quit rates, and physical effects before and after one month of NRT lozenges, laser therapy, and counselling.
METHODS: This randomised controlled trial tested nicotine lozenges, laser therapy, and counselling. This trial included 75 participants (25 per group) over one month. Seventy-five participants were divided into three groups of 25. The "Jus-CheckTM Rapid Nicotine Test" was used. Laser therapy targeted both ears weekly for four weeks

Smoking is associated with increased plasma homocysteine levels, and both are associated with an increased risk of cardiovascular disease. However, little information is available on the effects of passive smoking on the level of homocysteine in nonsmokers. We analyzed the data of self-reported never-smokers (aged > or = 20 years, n = 3,232), who were from the Third National Health and Nutrition Examination Survey. We quantified the passive nicotine exposure by dividing the never-smokers into quartiles as based on the serum cotinine values. Multiple linear and logistic regression models were u

OBJECTIVES: Using saliva cotinine as a quantitative marker, we examined the contribution of factors other than parental smoking to children's passive exposure to tobacco smoke.
METHODS: Saliva specimens from a random sample of 734 7-year-old schoolchildren in Edinburgh, Scotland, were analyzed for cotinine. Their parents completed a questionnaire covering smoking habits and conditions in the home.
RESULTS: A number of independent predictors of cotinine were identified in addition to the main one of smoking by household members. These predictors included home ownership, social class, day of t

BACKGROUND: Depression is a common mental illness among middle-aged and elderly people. However, few studies have explored the relationship between smoke exposure and depression in those aged ≥ 45 years. This study aims to determine the association between the level of smoke exposure and depression in middle-aged and elderly individuals.
METHODS: We extracted the data from the National Health and Nutrition Examination Survey (NHANES) 2005-2018, focusing on U.S. participants aged 45 years or older with complete data. Depression was assessed using the Patient Health Questionnaire-9 (PHQ-9), bas

Rationale: Protective effects of a high-quality diet on respiratory health, particularly among youths exposed to environmental tobacco smoke (ETS), are unknown. Objectives: To assess if a higher-quality diet is associated with improved respiratory symptoms and lung function among adolescents and if these associations are modified by ETS exposure. Methods: This was a cross-sectional study on 7,026 nonsmoking adolescents of the 2003-2012 National Health and Nutrition Examination Survey. Diet quality was assessed using the Healthy Eating Index-2010 score (HEI-2010), categorized into quintiles. ET

Urinary determination of nicotine metabolites provides an ideal tool for the quantitative assessment of the tobacco use-related nicotine dose, provided that the considered metabolites comprise a large share of the amount taken up. A method based on liquid chromatography-tandem mass spectrometry (LC-MS/MS) was developed for the sensitive, fast and robust analysis of nicotine and 10 major nicotine metabolites ("Nic+10"), including cotinine, trans-3'-hydroxy-cotinine, nicotine-N-glucuronide, cotinine-N-glucuronide, trans-3'-hydroxy-cotinine-O-glucuronide, nornicotine, norcotinine, nicotine-N'-oxi

OBJECTIVES: Passive smoking has been demonstrated in many situations. We designed an experimental protocol to measure passive smoking in the coaches of the French high-speed train (TGV) and to attempt to identify interindividual variability in sensitivity.
METHODS: Ten healthy non-smokers (5 males, 5 females) volunteered to avoid exposure to tobacco smoke for the duration of the study. On three separate occasion they were subjected to a 5-hour trip in the smoking coaches of the French TGV (south-east line). Twelve-hour urine samples were collected before each trip and over the following 72 ho

We examined the relationship of paternal smoking habits to birthweight in a cohort of infants enrolled at birth (n = 1219). To assess validity of parental smoking reports, cord serum levels of cotinine were measured in 175 newborns. A mean birthweight deficit of 88 g was found in newborns of nonsmoking mothers whose fathers smoked more than 20 cigarettes/day. Only 2 of 138 newborns whose mothers said they were nonsmokers had cotinine levels indicating that their mothers were active smokers. Among infants of nonsmoking mothers, detectability of cotinine in cord blood was significantly correlate

A high pressure liquid chromatographic procedure has been developed for the determination of the two principal N- and C-oxidation products of nicotine in hamster liver subcellular fractions. Advantage was taken of the fact that cyanide ion forms a stable adduct with the microsomal metabolite that is the precursor of cotinine. The rate and extent as well as the sensitivity of inhibition were similar for cotinine, the 5'-cyanonicotine adduct, and an as yet unidentified microsomal metabolite which is presumed to be the initial microsomal metabolite on the pathway to cotinine formation. The rates

BACKGROUND: The underreporting of environmental tobacco smoke (ETS) exposure by parents of study children may depend on the instrument used and population studied, underlining the need for questionnaire validation in specific study settings. This study explores the validity of parent-reported ETS exposure in a French multicenter study on asthma.
METHODS: The study population was composed of 313 children ages 4 to 14 years. Exposure to ETS was evaluated both by questionnaires on recent ETS exposure and by assessment of urinary cotinine by an enzyme immunoassay.
RESULTS: According to parents'

BACKGROUND: Tobacco smoking affects both the fraction of exhaled nitric oxide (FeNO) and blood eosinophil (B-Eos) count, two clinically useful biomarkers in respiratory disease that represent local and systemic type-2 inflammation, respectively.
OBJECTIVE: We aimed to study the influence of objectively measured smoke exposure on FeNO and B-Eos in a large population of subjects with and without asthma.
METHODS: We utilized the US National Health and Nutrition Examination Surveys 2007-2012 and included 10 669 subjects aged 6-80 years: 9869 controls and 800 asthmatics. Controls were defined as

We characterize nicotine exposure in the U.S. population by measuring urinary nicotine and its major (cotinine, trans-3′-hydroxycotinine) and minor (nicotine 1′-oxide, cotinine N-oxide, and 1-(3-pyridyl)-1-butanol-4-carboxylic acid, nornicotine) metabolites in participants from the 2015−2016 National Health and Nutrition Examination Survey. This is one of the first U.S. population-based urinary nicotine biomarker reports using the derived total nicotine equivalents (i.e., TNEs) to characterize exposure. Serum cotinine data is used to stratify tobacco non-users with no detectable serum cotinine
