Loxoscelism, the term used to describe lesions and clinical manifestations induced by brown spider's venom (Loxosceles genus), has attracted much attention over the last years. Brown spider bites have been reported to cause a local and acute inflammatory reaction that may evolve to dermonecrosis (a hallmark of envenomation) and hemorrhage at the bite site, besides systemic manifestations such as thrombocytopenia, disseminated intravascular coagulation, hemolysis, and renal failure. The molecular mechanisms by which Loxosceles venoms induce injury are currently under investigation. In this revi

BACKGROUND: The pig is the donor animal of choice for human xenotransplantation. In the most relevant pig-to-baboon model, pig organs transplanted into baboons are hyperacutely rejected by natural xenoantibodies, which mainly bind to alpha-galactosyl (alphaGal) epitopes expressed at the surface of endothelial cells. Recent advances in controlling hyperacute rejection have led to improved survival of these xenografts, and it is now important to identify alphaGal binding sites in other cells and tissues that may be subject to immunologic attack. To this end, we have studied whether alphaGal anti

Occlusive coronary artery disease is an important factor of cardiovascular morbidity and mortality. The rupture of the thin fibrous cap of the atheroma may be one of the causes of acute coronary syndrome, however, the mechanism of formation of fibrous plaque are poorly understood. Elevation of plasma homocysteine, hyperhomocystinemia, H(e), has emerged as an independent risk factor for hypertension and fibrotic heart disease. The extracellular matrix (ECM) components, particularly fibrillar collagen, are elevated in the atherosclerotic lesions and are the essential integral element in holding

In previous work, we compared the steady-state levels of specific matrix components in human bone cells derived from patients with osteogenesis imperfecta (OI) to those of age-matched controls. A remarkable finding was the observation that there was a reduction not only in the total levels of collagen, but also in osteonectin and three proteoglycans (a large chondroitin sulfate proteoglycan, biglycan, and decorin). This pattern was observed in patients with and without detectable collagen defects. More recent analysis of extracellular matrix composition have yielded that, compared with age-mat

Results of experimental and histopathological studies of reactive production of extracellular matrix by corneal endothelial cells are summarized and evaluated. In anterior synechiae, epithelial ingrowth and a chronic corneal ulcer extracellular matrix was produced not only toward Descemet's membrane but also on the normally free endothelial cell surface. In some cases of anterior synechiae and cystic epithelial ingrowth, after partial separation of the new layer of extracellular matrix, recovery and regeneration of endothelial cells was possible. In diffuse epithelial invasion, extracellular m

Extracellular matrix proteins synthesized by bone cells isolated from 16 patients with different forms of osteogenesis imperfecta (OI) were analyzed in vitro. Specific components of the extracellular matrix by OI and age-matched cultures were investigated by steady-state radiolabeling followed by quantitation of label into specific proteins and comparison of OI cultures to those of age-matched controls. The in vitro proliferation of OI bone cells was found to be lower than that of control cells. In seven patients, abnormalities of the alpha 1(I) and/or alpha 2(I) chains of type I collagen were

Epigenetic alterations, particularly DNA methylation, play a crucial role in colorectal cancer (CRC) development and progression. We aimed to investigate DNA methylation changes across normal colon samples, adenomas, and adenocarcinomas to identify critical gene networks associated with disease progression. We conducted a comprehensive DNA methylation analysis on 2,346 samples. We extracted CRC methylation data of normal colon mucosa, adenomas, and adenocarcinomas from the genomic data commons data portal and the gene expression omnibus. We ranked differentially methylated genes by their area

Micronized extracellular matrix (mECM) comprises a class of micron-scale particulate biomaterials derived from native extracellular matrix or decellularized extracellular matrix. Compared with conventional bulk ECM/dECM scaffolds, mECM offers distinct advantages in injectability, local conformability, and integration into composite systems, making it increasingly attractive for tissue repair and regenerative medicine. This review systematically summarizes the fundamental concept, material characteristics, and major mechanisms through which mECM facilitates tissue repair, including the regulati

Psychosocial stress has been identified to increase the development and severity of cardiovascular disease, particularly heart failure. An underlying structural factor for the development and progression of certain forms of heart failure is the accumulation of extracellular matrix-generically termed myocardial fibrosis. However, it remains unclear what pathways and mechanisms by which psychosocial stress intersects with the development of myocardial fibrosis. This review will focus upon animal models of psychosocial stress and specific signaling pathways which may be relevant to the developmen

Chronic liver diseases, including metabolic dysfunction-associated steatotic liver disease (MASLD), frequently progress to liver fibrosis, yet effective antifibrotic therapies remain limited. Here, we investigated the therapeutic potential of the commensal bacterium Phocaeicola coprocola using Western diet (WD)-induced MASLD and DDC + TAA-induced fibrosis murine models. In the WD model, P. coprocola administration attenuated hepatic steatosis, reduced lipogenic gene expression, and improved metabolic and histological parameters. In contrast, in the DDC + TAA model of advanced fibrosis, P. copr

Endometrial disorders impact the uterine lining and can promote ectopic endometrial tissue growth. A key feature of endometrial disorders is unresponsiveness to hormonal treatments due to progesterone resistance. Because endometrial lesions are found within fibrotic extracellular matrix (ECM), we hypothesized that ECM stiffness and composition regulate progesterone responses. We used hydrogels to mimic physiological stiffnesses and examine how mechanical and biochemical ECM cues influence progesterone-induced differentiation (decidualization) in healthy fibroblasts. We determined that ECM liga

Collagens make up a major part of extracellular matrix (ECM) proteins, and their mechanical and chemical properties are regulated by collagen-modifying enzymes. The expression and activity of these enzymes are influenced by hypoxia through hypoxia-inducible factor (HIF), which plays a role in both physiological and pathological conditions. This chapter presents protocols for studying the effects of hypoxia on ECM, cell behavior, and collagen production, utilizing specialized hypoxia workstations for controlled cell culture conditions.

Animal studies and human tissue experiments have demonstrated that traumatic brain injury (TBI) causes damage to the extracellular matrix (ECM). To test the hypothesis that TBI causes disruption of sulfated glycosaminoglycan (sGAG) in the ECM, we measured levels of sGAG in the cerebrospinal fluid (CSF), blood, and urine, in patients with severe TBI in the acute postinjury period. Samples of CSF, blood, and urine were obtained within 72 h of injury in patients who received external ventricular drains as part of their treatment of severe TBI. Levels of chondroitin and heparan sGAGs were measured

It remains a significant challenge to construct a tracheal substitute with both a native-like structure and multiple essential physiological functions. In this study, a combination of 3D printing techniques and a modular strategy is employed to fabricate an engineered trachea, in which the decellularized extracellular matrix particles (DEPs) from diverse sources determined specific regenerative environments in different spatial regions. Costal cartilage-derived DEPs are integrated within the cartilage rings of the engineered trachea. They effectively activated chondrocytes to secrete specific

Organ function depends on the three-dimensional integrity of the extracellular matrix (ECM). The structure resulting from the location and association of ECM components is a central regulator of cell behavior, but a dearth of matrix-specific analysis keeps it unresolved. Here, we deploy a high-resolution, 3D ECM mapping method and design a machine-learning powered pipeline to detect and characterize ECM architecture during health and disease. We deploy these tools in the human lung, an organ heavily dependent on ECM structure that can host diseases with different histopathologies. We analyzed

The extracellular matrix (ECM) is a noncellular structure that plays an indispensable role in a series of cell life activities. Accumulating studies have demonstrated that ECM stiffness, a type of mechanical forces, exerts a pivotal influence on regulating organogenesis, tissue homeostasis, and the occurrence and development of miscellaneous diseases. Nevertheless, the role of ECM stiffness in ophthalmology is rarely discussed. In this review, we focus on describing the important role of ECM stiffness and its composition in multiple ocular structures (including cornea, retina, optic nerve, tra

The mineralization of the extracellular matrix (ECM) is an essential and crucial process for physiological bone formation and pathological calcification. The abnormal function of ECM mineralization contributes to the worldwide risk of developing mineralization-related diseases; for instance, vascular calcification is attributed to the hyperfunction of ECM mineralization, while osteoporosis is due to hypofunction. AnnexinA6 (AnxA6), a Ca2+-dependent phospholipid-binding protein, has been extensively reported as an essential target in mineralization-related diseases such as osteoporosis, osteoar

Angiogenesis is essential for cardiac repair after myocardial infarction. Promoting angiogenesis has been demonstrated as an effective approach for myocardial infarction treatment. Several different strategies for inducing myocardial angiogenesis have been explored, including exogenous delivery of angiogenic genes, proteins, microRNAs, cells, and extracellular vesicles. Various types of injectable hydrogels have been investigated for cardiac tissue repair. One of the most promising injectable hydrogels in cardiac regeneration is a cardiac extracellular matrix hydrogel that is derived from dece

How cells adjust their growth to the spatial and mechanical constraints of their surrounding environment is central to many aspects of biology. Here, we examined how extracellular matrix (ECM) rigidity affects cell division. We found that cells divide more rapidly when cultured on rigid substrates. While we observed no effect of ECM rigidity on rounding or postmitotic spreading duration, we found that changes in matrix stiffness impact mitosis progression. We noticed that ECM elasticity up-regulates the expression of the linker of nucleoskeleton and cytoskeleton (LINC) complex component SUN2,

The optic nerve head (ONH) is a place of vulnerability during glaucoma progression due to increased intraocular pressure damaging the retinal ganglion cell axons. The molecular signaling pathways involved in generating glaucomatous ONH damage has not been fully elucidated. There is a great deal of evidence that pro-fibrotic TGFβ2 signaling is involved in modulating the ECM environment within the lamina cribrosa (LC) region of the ONH. Here we investigated the role of signaling crosstalk between the TGFβ2 pathway and the toll-like receptor 4 (TLR4) pathway within the LC. ECM deposition was exam
