Muscimol prevents long-lasting potentiation of dorsal horn field potentials in rats with chronic constriction injury exhibiting decreased levels of the GABA transporter GAT-1.

The inhibitory activity of gamma-aminobutyric acid (GABA) is considered critical in setting the conditions for synaptic plasticity, and many studies support an important role of GABA in the suppression of nociceptive transmission in the dorsal horn. Consequently, any injury-induced modification of the GABA action has the potential to critically modify spinal synaptic plasticity. We have previously reported that chronic constriction injury of the sciatic nerve was accompanied by long-lasting potentiation of superficial spinal dorsal horn field potentials following high-frequency tetanus. Long-term potentiation of silent synapses in substantia gelatinosa neurons.

In this study, we observed quantal changes in single synapse excitatory postsynaptic currents to characterize N-methyl-D-aspartate receptor-mediated silent synapses between primary afferents and spinal substantia gelatinosa neurons. The failure rate of primary afferent quantal excitatory postsynaptic currents was lower at depolarized holding potentials than at hyperpolarized potentials. This lower failure rate at depolarized potentials was due to the activation of N-methyl-D-aspartate receptor-mediated excitatory postsynaptic currents. Pairing primary afferents with the postsynaptic depolariza

Computation of frequency-to-spatial transform by olfactory bulb glomeruli.

A physiological simulation of 2.5% of the input and inhibitory neurons and 25% of the primary mitral/tufted cells in a single mammalian olfactory bulb glomerulus was constructed. This physiological simulation used the integrate-and-fire paradigm with realistic activation curves and synaptic delays. The dendritic integration incorporated non-linear interactive effects of individual cell excitatory and inhibitory post-synaptic potentials (PSPs) from both axodendritic and dendro-dendritic synaptic contacts. Refractory periods for granule-cell inhibition of mitral/tufted cell activity lead to rela

Synaptic and membrane properties of parasympathetic ganglionic neurons innervating mouse trachea and bronchi.

The pathophysiology of airway diseases, such as asthma, is increasingly studied using transgenic mice and other mouse models of airway inflammation where allergen-induced changes in airway smooth muscle tone and mucous secretion is due, in part, to activation of preganglionic airway parasympathetic nerves. Ganglionic parasympathetic neurons located in the airways in several species, including humans, have anatomical and electrophysiological properties that limit transmission of preganglionic synaptic input. In this study, intracellular recordings were made from neurons in parasympathetic gangl

Effects of bupivacaine and ropivacaine on field potential in rat hippocampal slices.

BACKGROUND: In spite of more than 20 yr of research, the mechanism whereby local anaesthetics act on the brain to mediate anaesthesia still remains unclear. Furthermore, the effect of local anaesthetics on neuronal excitability and synaptic transmission in the hippocampus has not been reported. Thus, the purpose of the present study was to find out the differences between the local anaesthetics, bupivacaine and ropivacaine, in their actions on synaptic transmission of brain in the context of hippocampal field potential. METHODS: Brains were removed from 3- to 4-week-old rats and transverse sl

Intrinsic conductances actively shape excitatory and inhibitory postsynaptic responses in olfactory bulb external tufted cells.

The initial synapse in the olfactory system is from olfactory nerve (ON) terminals to postsynaptic targets in olfactory bulb glomeruli. Recent studies have disclosed multiple presynaptic factors that regulate this important linkage, but less is known about the contribution of postsynaptic intrinsic conductances to integration at these synapses. The present study demonstrates voltage-dependent amplification of EPSPs in external tufted (ET) cells in response to monosynaptic (ON) inputs. This amplification is mainly exerted by persistent Na(+) conductance. Larger EPSPs, which bring the membrane p

Recruitment of crab gastric mill neurons into the pyloric motor pattern by mechanosensory afferent stimulation.

1. The gastropyloric receptor (GPR) cells are stretch-sensitive muscle receptors in the crab stomatogastric nervous system that use both 5-hydroxytryptamine (serotonin) and acetylcholine as cotransmitters. Brief stimulation of these afferent neurons causes two gastric mill neurons to be recruited into the pyloric motor pattern. 2. The GPR cells evoke complex synaptic potentials in the lateral gastric (LG) and medial gastric (MG) motor neurons, two component neurons of the gastric mill central pattern generator. When the gastric mill is quiescent (as often happens in vivo), GPR stimulation tran

Changes in spinal inhibitory networks induced by furosemide in humans.

During neural development in animals, GABAergic and glycinergic neurons are first excitatory, and then become inhibitory in the mature state. This developmental shift is due mainly to strong expression of the cation-chloride K-Cl cotransporter 2 (KCC2) and down-regulation of Na-K-Cl cotransporter 1 (NKCC1) during maturation. The down-regulation of co-transporter KCC2 after spinal cord transection in animals leads to the depolarising (excitatory) action of GABA and glycine and thus results in a reduction of inhibitory synaptic efficiency. Furosemide, a loop diuretic, has been shown to selective

Effects of L-DOPA on nigral dopamine neurons and local field potential: comparison with apomorphine and muscimol.

L-DOPA is more effective than direct dopamine (DA) agonists in relieving the motor deficits in Parkinson's disease. Using in vivo recording, we compared the effect of l-DOPA and the direct DA agonist apomorphine on DA neurons in rat substantia nigra (SN). L-DOPA (50-100 mg/kg i.v.) decreased the firing rate as well as the variability and slow oscillation (SO) of firing. All effects were blocked by raclopride and mimicked by quinpirole, suggesting that they are mediated through D2-like receptors. Autoreceptor-selective doses of apomorphine (5-20 μg/kg i.v.) also inhibited all three parameters.

Effects of dopamine and fluphenazine on field potential amplitude in the salamander olfactory bulb.

The effects of dopamine (DA) and fluphenazine (FLU), a phenothiazine DA receptor antagonist, were examined in the salamander olfactory bulb. Field potentials were recorded in the granule cell layer of superfused hemibrain preparations, and the amplitude of the large positive peak was compared following stimulation of the olfactory nerve (ON) and lateral olfactory tract (OT). In preparations superfused with normal amphibian Ringer's solution, the large peak occurred 14-21 ms after either ON or OT stimulation. It therefore appeared to reflect the activation of granule cell synapses with mitral c

Dendritic shaft constrictions shape synaptic integration in neurons.

For nearly 150 years, textbooks have described dendritic morphology as optimized for efficient synaptic voltage transfer from spines to the soma, implemented as a tubular design respecting Rall's 3/2 rule for impedance matching at branch points. Here, we reveal that this view is an oversimplification. Using multiple high-resolution imaging techniques, we demonstrate that dendrites in cortical and hippocampal neurons exhibit nanoscale constrictions, termed dendritic shaft constrictions (DSCs), with diameters ranging from ∼100 to 500 nanometers in mice. We also identified DSCs in human hippocamp

Alpha-pompilidotoxin (alpha-PMTX), a novel neurotoxin from the venom of a solitary wasp, facilitates transmission in the crustacean neuromuscular synapse.

A new neurotoxin, named alpha-pompilidotoxin (alpha-PMTX) has been found in the venom of the solitary wasp Anoplius safnariensis. In the neuromuscular synapse of the lobster walking leg preparation, alpha-PMTX (10-100 micro/M) caused great enhancement of both the excitatory and inhibitory postsynaptic potentials. Recordings of the excitatory post synaptic currents (EPSCs) at the synaptic sites showed that alpha-PMTX reversibly and dose-dependently potentiates EPSCs. Alpha-PMTX may act primarily on the presynaptic membrane but the mode of action of the toxin is clearly different from other know

Recruitment of local excitatory circuits in the superior colliculus following deafferentation and the regeneration of retinocollicular inputs.

The local synaptic connectivity in the superficial gray layer of the superior colliculus (SC) was assessed following retinal ganglion cell axonal regeneration through a peripheral nerve graft into the SC of Lister Hooded rats, using in vitro brain slice techniques. Repair was effected between the ipsilateral eye and SC, following bilateral lesion of optic nerves and ablation of ipsilateral occipital cortex. Deafferentation surgery alone resulted in a complete loss of synaptic potentials of extrinsic origin, once both retinal and cortical inputs were removed. Stimulation of graft insertion site

Tomosyn-2 is required for normal motor performance in mice and sustains neurotransmission at motor endplates.

Tomosyn-1 (STXBP5) is a soluble NSF attachment protein receptor complex-binding protein that inhibits vesicle fusion, but the role of tomosyn-2 (STXBP5L) in the mammalian nervous system is still unclear. Here we generated tomosyn-2 null (Tom2(KO/KO)) mice, which showed impaired motor performance. This was accompanied by synaptic changes at the neuromuscular junction, including enhanced spontaneous acetylcholine release frequency and faster depression of muscle motor endplate potentials during repetitive stimulation. The postsynaptic geometric arrangement and function of acetylcholine receptors

Microglial CR3-mediated synaptic pruning in the dmPFC promotes the generation and maintenance of chronic muscle pain via glutamatergic dysfunction.

Chronic muscle pain (CMP) is highly prevalent, frequently comorbid with emotional disorders and characterized by a high risk of recurrence. Yet, the complex mechanisms underlying the generation and maintenance of CMP remain unclear, limiting the development of therapy. Here we identified suppressed glutamatergic neuronal excitability and reduced synaptic plasticity in the dorsomedial prefrontal cortex (dmPFC) of CMP rats using fiber photometry, patch-clamp, in vivo recording of field potentials and other techniques. The optochemical genetical activation of dmPFC glutamatergic neurons alleviate

Temporary Versus Permanent Synaptic Loss from Repeated Noise Exposure in Guinea Pigs and C57 Mice.

A single brief noise exposure can cause a significant loss of cochlear afferent synapses without causing permanent threshold shift. Previously we reported that the initial synaptic loss is partially reversible in Guinea pigs, indicating that synaptic loss can be categorized as either temporary or permanent. Since synaptic loss is biased to innervating auditory nerve fibers (ANFs) with low spontaneous spike rates (SSR), which are critical to the coding of in-background noise, coding-in-noise deficits (CIND) have been predicted to result from noise-induced synaptic damage. However, recent study

Effects of 5α-cholestan-3-one on the synaptic vesicle cycle at the mouse neuromuscular junction.

We have investigated the effects of 5α-cholesten-3-one (5Ch3, 200 nM) on synaptic transmission in mouse diaphragm. 5Ch3 had no impact on the amplitude or frequency of miniature endplate currents (MEPCs, spontaneous secretion), but decreased the amplitude of EPCs (evoked secretion) triggered by single action potentials. Treatment with 5Ch3 increased the depression of EPC amplitude and slowed the unloading of the dye FM1-43 from synaptic vesicles (exocytosis rate) during high-frequency stimulation. The estimated recycling time of vesicles did not change, suggesting that the decline of synaptic e

Functionally distinct evoked and spontaneous neurotransmission operate via a shared pool of synaptic vesicles in viscerosensory afferents.

Viscerosensory afferent neurons of the vagus nerve form excitatory synapses onto second-order neurons in the brainstem nucleus of the solitary tract (NTS). Vagal afferent neurons release glutamate via functionally distinct action potential-dependent (evoked) and -independent (spontaneous) release pathways. Current models remain conflicted regarding whether synapses maintain physically and functionally separate vesicle pools underlying each release pathway or if they are unified via a common pool of vesicles. Most evidence has been derived from neurons in the CNS, so it remains unclear whether

Synaptic integration in hippocampal CA1 pyramids.

Excitatory synapses on hippocampal pyramids are exclusively located to dendritic spines, usually in a 1:1 proportion. The number of spines indicates a convergence of as many as 25,000-30,000 excitatory boutons per CA1 pyramidal cell in rats. Activation of a single afferent fibre produces a unitary excitatory postsynaptic potential (EPSP) of about 150 microV, probably produced by a single quantum of transmitter. The release probability is normally low, but may be increased by facilitatory processes. On the average, each afferent fibre has few boutons (mostly 1, but up to 5) in contact with a gi
