Joint associations of grip strength, lipid levels, and lipid-related genetic risk with atrial fibrillation.

INTRODUCTION: The associations between lipid levels and atrial fibrillation (AF) remain inconsistent, and whether functional status modifies these relationships is unclear. In this prospective cohort study, the joint associations of grip strength, lipid levels, and lipid-related genetic risk with incident AF were evaluated. METHODS: A prospective cohort analysis was conducted, and Cox models were used to evaluate the associations of lipid levels, lipid-related polygenic risk scores (PRSs), and grip strength with incident AF. Joint exposure categories were constructed according to lipid status

What Proportion of Antibody-Negative Type 1 Diabetes Cases Have a Nonautoimmune Etiology? A Genetics-Based Estimate.

UNLABELLED: Among patients with young-onset type 1 diabetes, 10-15% are autoantibody (aAb) negative. How many of these are autoimmune versus nonautoimmune is unknown. To estimate this proportion, we used the only high-confidence evidence of autoimmunity in these cases, the presence of an aAb(+) sibling. In the 2,500 affected sibling pairs of the Type 1 Diabetes Genetics Consortium cohort, we compared the Type 1 Diabetes Genetic Risk Score (GRS2) between 379 aAb(-) patients with an aAb(+) sibling (autoimmune cause) and 253 aAb(-) patients with no aAb(+) siblings (unknown cause), under the assum

DeepU-PRS: nonlinear and uncertainty-aware SNP heritability modeling for annotation-informed polygenic risk scoring.

Polygenic risk scores (PRSs) are widely used to quantify genetic predisposition to complex traits, and their predictive accuracy can be enhanced by incorporating functional annotations into single-nucleotide polymorphism (SNP) effect-size estimation. Existing approaches typically assume linear relationships between annotations and SNP effects, neglecting predictive aleatoric variance and potential nonlinear regulatory interactions. To evaluate the gains of nonlinear and uncertainty-aware modeling, we developed DeepU-PRS, a deep ensemble framework that extends the linear BLD-LDAK model with a n

PGS-GS: a framework integrating polygenic scores and genomic selection in animal breeding.

Genomic prediction has become a central paradigm in biology, enabling quantitative inference of genetic contributions to complex traits across humans, animals, and plants. Although genomic research in human genetics and animal breeding shares a highly homologous methodological foundation, significant barriers persist in their analytical paradigms and application scenarios. This study aims to promote cross-disciplinary integration by introducing human-derived polygenic scores (PGS) algorithms into animal genomic selection (GS) and proposing a PGS-GS framework with a preliminary weighting-based

Consensus meta-analysis of genome-wide association studies for Alzheimer's disease and related dementias.

To better characterize the genetic architecture underlying Alzheimer's disease (AD) and related dementias (ADRD), we performed a meta-analysis of European-ancestry genome-wide association studies in 128,681 cases or proxy cases of ADRD and 849,833 (proxy) controls. We identified 91 genetic loci associated with ADRD risk, of which 16 are new and 56 are specifically detected in clinically diagnosed AD cases. We also provide a list of 18 loci (15 new) requiring further external validation. A polygenic score combining the effects of ADRD loci other than APOE was primarily associated with AD rather

Additive value of polygenic risk and family history for coronary heart disease risk stratification in two diverse US cohorts.

Whether polygenic risk, monogenic familial hypercholesterolemia (FH), and family history (FamHx) are additively informative for coronary heart disease (CHD) risk prediction across self-identified race/ethnicity (SIRE) groups has not been established. In two diverse cohorts-Electronic Medical Records and Genomics (eMERGE) phase IV (eIV; n = 19,348) and All of Us (AoU; n = 239,645)-we quantified the associations of a polygenic risk score (PRSCHD), pathogenic/likely pathogenic variants in genes associated with FH, and FamHx with CHD and evaluated their incremental value when added to the pooled c

Methods for modeling gene-environment interplay using polygenic risk scores.

Polygenic risk scores (PRS) are increasingly recognized as pivotal tools for quantifying disease risk through the aggregation of multiple genetic variants. As sample sizes in genome-wide association studies (GWAS) continue to expand and PRS become more powerful, they are set to play a key role in translational research and personalized medicine. Understanding the interplay of PRS with environmental factors is critical for interpreting and applying PRS in diverse contexts. This interplay manifests in two forms: PRS-by-environment interaction (PRS × E) and gene-environment correlation (rGE). How

Genetic risk in Alzheimer's disease.

Alzheimer's disease (AD) has a strong genetic predisposition. Genome-wide association studies have identified multiple risk loci, yet many non-coding variants remain uncharacterized. Machine learning-based polygenic risk scores (PRS) enhance prediction by modeling genetic epistasis and sex-specific risks. This review summarizes AD genetic risk factors, PRS methodologies, and ML-based AD risk prediction. It also highlights challenges such as population bias, functional validation, and integrating multi-omics for precision medicine.
