BACKGROUND: Currently, reversed phase liquid chromatography (RP LC) relies heavily on acetonitrile (ACN) and methanol (MeOH), presenting significant environmental and safety risks. This study evaluates green organic modifiers, specifically propylene carbonate (PC), dimethyl carbonate (DMC), and ethyl lactate (EL), as alternatives in RP LC using monodisperse stationary phases (Diphenyl, C18/AR, and RP18-Amide). The research was designed as a comparative study to determine the elution strength, efficiency and separation performance of three different mixtures.
RESULTS: By integrating chromatogr

Cation exchange chromatography (CEX) of therapeutic monoclonal antibodies is generally performed with either salt gradient (MES buffer + NaCl) or using commercial pH gradient buffer. The goal of this study was to find out some alternative buffer systems for CEX separation of mAbs, which may offer alternative selectivity, while maintaining similar peak shapes. Among the new buffers that were tested, (N-morpholino)ethanesulfonic acid (MES) / 1,3-diamino-2-propanol (DAP), and citric acid / 2-(cyclohexylamino)ethanesulfonic acid (CHES) systems were particularly promising, especially when combining

Microalgal lipids were separated into two fractions, triacylglycerols (TAGs) and free fatty acids (FFAs), by solid-phase extraction employing sodium carbonate as the sorbent and dichloromethane (20% by volume) in n-hexane as the extracting solvent. The TAG fraction was then saponified, followed by acidification, extraction and tert-butyldimethylsilyl esterification. The FFA fraction was directly acidified, extracted and derivatized. From the lipid extracts of eight microalgal species examined, a total of 13 fatty acids were detected in the TAG fractions and nine were found in the FFA fractions

Companion of Cellulose Synthase (CC), has previously been shown to sustain cellulose synthesis during salt stress. However, the role of CCs in this process is not well-understood. In this study, we show that the intrinsically disordered N-terminal domain of Arabidopsis CC1 (CC1NTD) can undergo liquid-liquid phase separation (LLPS) under defined reducing conditions in the presence of trimethylamine N-oxide (TMAO), a naturally occurring plant stress osmolyte. The observed biomolecular condensates are enriched in CC1NTD and display liquid-like behavior, including spherical morphology, fusion, and

UNLABELLED: Nuclear paraspeckles play critical roles in orchestrating gene transcription to support tumor development and progression. Elucidating regulators of their dynamic compositions and formation could provide potential targets for treating cancer. In this study, we discovered that the short isoform of Wilms tumor gene on the X chromosome (WTX-S) specifically disrupted paraspeckle stability. WTX-S selectively interacted with and sequestered NONO (also as p54nrb) from paraspeckles. Mechanistically, interaction between WTX-S and NONO induced formation of nuclear bodies via liquid-liquid ph

Lipid droplets (LDs) were long thought to be passive organelles merely for neutral lipid storage. Mounting evidence redefines LDs as dynamic metabolic signaling hubs orchestrating cellular stress adaptation, with multifaceted roles in organelle crosstalk, metabolic reprogramming, redox balance and immune signaling. LD function is tightly intertwined with liquid-liquid phase separation (LLPS) and epigenetic remodeling, bridging cellular metabolism to gene expression and cell fate control. LD biogenesis relies on ER lipid structures, phase-separated protein assemblies and lipid regulatory protei

RIG-I-like receptors (RLRs) are essential for antiviral innate immunity through sensing viral RNA. Although liquid-liquid phase separation (LLPS) has emerged as a key mechanism in fine-tuning immune signaling, its role in the initial sensing of viral RNA remains poorly understood. Here, we identify the RNA-binding protein CSDE1 as a negative regulator of RLR signaling that facilitates viral immune evasion. Macrophages deficient in CSDE1 exhibit enhanced interferon production, and CSDE1-knockout mice show increased resistance to viral infection. Mechanistically, CSDE1 forms RNA-dependent biomol

Local flow patterns determine the uneven distribution of atherosclerotic lesions, yet the underlying mechanism remains poorly defined. We previously reported that protein tyrosine phosphatase nonreceptor type 14 (PTPN14) is implicated in atherosclerosis. Its role in flow-dependent endothelial dysfunction and the upstream regulatory mechanisms remain incompletely understood. Here, we show that PTPN14 functions as a flow-sensitive regulator in endothelial cells, with its protein stability reduced under disturbed flow. Endothelial-specific deletion of PTPN14 led to exacerbated endothelial inflamm

Luminal breast cancer is characterized by a persistent risk of recurrence and dysregulated lipid metabolism. However, the role of phase separation, a novel mechanism for the spatial compartmentalization of proteins, in lipid remodeling within the context of breast cancer remains largely unexplored. Utilizing the multiomics data from our large breast cancer cohort (n = 773), we revealed that aberrant lipid metabolism negatively impacts the prognosis of patients with luminal breast cancer. Furthermore, we deciphered that the copy number alteration-driven cis-regulation of DDHD domain containing

In this work we harness liquid-liquid phase separation to organize the internal content of protocell models based on water-in-oil droplets. Our approach is based on the spatial segregation of two polymers, polyethylene glycol (PEG) and dextran, within the aqueous phase to produce biphasic droplets. We demonstrate that magnetic field-responsive nanoparticles functionalized with dextran are preferentially sequestered within the dextran subphase. This encapsulation enables droplet heating by application of alternating magnetic fields (AMFs). Meanwhile, the PEG subphase of the droplets remains ava

Liquid-liquid phase separation (LLPS), a fundamental biological process that drives the formation of membraneless organelles (MLOs) through non-covalent interactions, has emerged as a central regulatory mechanism in cellular physiology and stress responses. Increasing evidence links dysregulated LLPS to the pathogenesis of cardiovascular diseases (CVDs). However, the conserved molecular mechanisms underlying LLPS dysfunction across diverse CVDs, as well as the translation of LLPS-targeted strategies into clinical applications, remain poorly understood. In this review, we propose a novel hypoth

Liquid-liquid phase separation (LLPS) is a metastable state in which a highly supersaturated solution phase separates into a drug-rich nanoaggregate phase dispersed in a drug-lean phase. LLPS-driven formation of drug-rich nanoaggregates has emerged as a key determinant of oral drug absorption. Despite this growing recognition, the influence of LLPS on ocular delivery remains unexplored. In this study, we investigated whether the nanoaggregate phase generated by LLPS modulates transcorneal permeation and compared the resulting drug flux with that of a dialysis membrane. Liquid-liquid phase separation (LLPS), a biophysical driver of membraneless organelle assembly, is central to pathological aggregation in neurodegenerative diseases. Initially linked to amyotrophic lateral sclerosis (ALS), LLPS dysregulation has now been implicated in Alzheimer's, Parkinson's, and frontotemporal dementia, where aberrant transitions convert dynamic condensates into insoluble fibrils. To systematically map this landscape, we employed CiteSpace-based bibliometrics to analyze 784 Web of Science articles from 2009 to 2024. Our analyses reveal dominant contributions from the United

Chimeric antigen receptor (CAR) T cell therapy faces challenges, such as tumor relapse due to antigen loss and poor CAR T cell expansion in vivo. Optimal tonic signaling (spontaneous CAR signaling) is crucial for CAR T cell fitness and antitumor function, requiring precise calibration to avoid dysfunction or exhaustion. Although CAR phase separation can enhance CAR clustering and tonic signaling to improve cytotoxicity and antigen sensitivity, constitutively high signaling could lead to potential side effects, including cytokine storm and neuronal toxicity. To overcome this limitation, we deve

Conventional hydrogen-bonded molecularly imprinted polymers (MIPs) suffered from significantly compromised adsorption performance in aqueous environments due to competitive interference from water molecules. To overcome this limitation, this study developed a novel imprinting strategy based on reversible Schiff base bonding. Schiff base covalent imprinting utilizes aldehyde groups and amino groups to form reversible covalent bonds (C = N), achieving specific recognition between template molecules and functional monomers. Schiff base has strong affinity and selectivity, which can effectively re

Splicing factor proline- and glutamine-rich (SFPQ) is an RNA binding protein that broadly regulates RNA metabolism. Although its nuclear roles are well studied, evidence of SFPQ's cytoplasmic functionality is emerging. Altered expression and nuclear-to-cytoplasmic redistribution of SFPQ have been recognized in amyotrophic lateral sclerosis (ALS) pathology, yet the mechanistic bases for these phenomena remain undetermined. We identified altered SFPQ splicing in ALS, increasing the expression of an alternative mRNA isoform lacking a nuclear localization sequence, which we termed "altSFPQ." We fi

The development of environmentally friendly oil-absorbing fibrous materials is crucial, as conventional separation materials contribute to secondary pollution due to their nondegradability. In this study, highly hydrophobic and superoleophilic porous polylactic acid (PLA) fibers were fabricated via centrifugal spinning combined with nonsolvent-induced phase separation. The fiber porosity was controlled by adjusting the ratio of good solvents to nonsolvent in the spinning solution. The morphology and physical properties of the PLA fibers were systematically analyzed. Among the prepared samples,

Skeletal muscle plays a crucial role in maintaining motor function and metabolic homeostasis, with its loss or atrophy leading to significant health consequences. Long non-coding RNAs (lncRNAs) have emerged as key regulators in muscle biology; however, their precise roles in muscle function and pathology remain to be fully elucidated. This study demonstrates that lncRNA maternally expressed gene 3 (MEG3) is preferentially expressed in slow-twitch muscle fibers and dynamically regulated during muscle development, aging, and in the context of Duchenne muscular dystrophy (DMD). The formation of phase-separated structures in hydrogels plays a crucial role in determining their optical and mechanical properties. Traditionally, phase-separated hydrogels are prepared through a two-step process involving initial hydrogel synthesis followed by post-treatment. In this study, we present an approach for temperature-governed phase separation microstructure modulation in hydrogels, harnessing the cononsolvency effect. This method allows the phase-separated structure to develop during hydrogel synthesis, significantly simplifying the preparation process. Importantly, we found tha

Solid stress, originating from rigid and elastic components of extracellular matrix and cells, is a typical physical hallmark of tumors. Mounting evidence indicates that elevated solid stress drives metastasis and affects prognosis. However, the molecular mechanism of how cancer cells sense solid stress, thereby exacerbating malignancy, remains elusive. In this study, our clinical data suggest that elevated stress in metastatic solid tumors is highly associated with the expression of cytoskeleton-associated protein 4 (CKAP4). Intriguingly, CKAP4, as a sensitive intracellular mechanosensor, res
