Derivation of the MUSIe002-A human embryonic stem cell line.

The MUSIe002-A cell line was established from in vitro fertilization of human sperm and oocytes donated for research with informed consent. This cell line exhibited normal human embryonic stem cell (hESC) characteristics, including typical cell morphology, expression of all pluripotent stem cell markers, and potential to differentiate into three germ layers. A karyotyping analysis revealed 46 XY chromosome and cells that did not have mycoplasma contamination. MUSIe002-A represents a valuable unlimited cell source and is of potential interest for human in vitro stem cell based-models, genetic m

Chronic myelomonocytic leukaemia stem cell transcriptomes anticipate disease morphology and outcome.

BACKGROUND: Chronic myelomonocytic leukaemia (CMML) is a clinically heterogeneous stem cell malignancy with overlapping features of myelodysplasia and myeloproliferation. Over 90% of patients carry mutations in epigenetic and/or splicing genes, typically detectable in the Lin-CD34+CD38- immunophenotypic stem cell compartment in which the leukaemia-initiating cells reside. Transcriptional dysregulation at the stem cell level is likely fundamental to disease onset and progression. METHODS: We performed single-cell RNA sequencing on 6826 Lin-CD34+CD38-stem cells from CMML patients and healthy co

Optimization of adenoviral gene transfer in human pluripotent stem cells.

Human pluripotent stem cells, such as embryonic stem cells and induced pluripotent stem cells, have the potential to differentiate into a wide variety of cells in vitro and have applications in basic developmental biology research and regenerative medicine. To understand the process of differentiation from pluripotent stem cells to functional cells, it is necessary to efficiently and safely transfer and express exogenous genes. We attempted to optimize the efficient transfer of genes into pluripotent stem cells using adenoviral vectors. Comparative study of the activities of three representati

Neural stem cells and the quest for restorative neurology.

A great deal of interest has attracted the attention of researchers on the potential use of (neural) stem cells in cell replacement or restorative therapies for heretofore incurable CNS pathologies such as brain stroke, spinal cord injury, Parkinson's disease or multiple sclerosis. This short perspective illustrates our view of neural stem cell research with a focus on the stem cell concept, on the in situ identity of neural stem cells and on selected aspects of embryonic and adult neurogenesis. A brief survey of current stem cell-based experimental literature tries to provide a realistic pict

Gene delivery to embryonic stem cells.

Since the establishment of embryonic stem (ES) cells and the identification of tissue-specific stem cells, researchers have made great strides in the analysis of the natural biology of such stem cells for the development of therapeutic applications. Specifically, ES cells are capable of differentiating into all of the cell types that constitute the whole body. Thus, ES cell research promises new type of treatments and possible cures for a variety of debilitating diseases and injuries. The potential medical benefits obtained from stem cell technology are compelling and stem cell research sees a

Ex Vivo Imaging and Genetic Manipulation of Mouse Hair Follicle Bulge Stem Cells.

Stem cells that reside in the bulge of adult mouse hair follicles are a leading model of tissue stem cell research. Ex vivo culturing, molecular and cell biological characterizations, as well as genetic manipulation of fluorescence-activated cell sorting-isolated bulge stem cells offer a useful experimental pipeline to complement in vivo studies. Here we describe detailed methods for culturing, immunostaining, live cell imaging, and adenoviral infection of bulge stem cells for downstream applications such as in vitro clonal and in vivo patch assays.

Bridging the gap to clinical translation: the advanced role of PET molecular imaging in stem cell therapy.

In the past few decades, stem cell therapy has achieved remarkable progress in preclinical and clinical trials, especially for treating major diseases, such as stroke, Parkinson’s disease (PD), myocardial infarction, diabetes, etc (Temple, Cell Stem Cell 30:512-29, 2023). However, the current challenge in clinical translation lies in the difficulty in non-invasively, dynamically, and quantitatively assessing the survival, proliferation, homing, migration, and function of stem cells in vivo. This lack of assessment leads to inconsistent therapeutic effects, unclear mechanisms, and difficulties

Pluripotent stem cells models for Huntington's disease: prospects and challenges.

Pluripotent cellular models have shown great promise in the study of a number of neurological disorders. Several advantages of using a stem cell model include the potential for cells to derive disease relevant neuronal cell types, providing a system for researchers to monitor disease progression during neurogenesis, along with serving as a platform for drug discovery. A number of stem cell derived models have been employed to establish in vitro research models of Huntington's disease that can be used to investigate cellular pathology and screen for drug and cell-based therapies. Although some

The brain and eye: Treating cerebral and retinal ischemia through mitochondrial transfer.

UNLABELLED: Stroke remains a devastating disease with limited treatment options, despite our growing understanding of its pathology. While ischemic stroke is traditionally characterized by a blockage of blood flow to the brain, this may coincide with reduced blood circulation to the eye, resulting in retinal ischemia, which may in turn lead to visual impairment. Although effective treatment options for retinal ischemia are similarly scarce, new evidence suggests that deleterious changes to mitochondrial structure and function play a major role in both cerebral and retinal ischemia pathologies.

Development of stem cell therapy for atherosclerosis.

Cardiovascular disease (CVD) has a high incidence and low cure rate worldwide, and atherosclerosis (AS) is the main factor inducing cardiovascular disease, of which lipid deposition in the vessel wall is the main marker of AS. Currently, although statins can be used to lower lipids and low-density lipoprotein (LDL) in AS, the cure rate for AS remains low. Therefore, there is an urgent need to develop new therapeutic approaches, and stem cells are now widely studied, while stem cells are a class of cell types that always maintain the ability to differentiate and can differentiate to form other
