BACKGROUND: ABO histocompatibility plays a critical role in red blood cell (RBC) transfusions and is of great importance in solid organ and hematopoietic stem cell transplantation. ABO is primarily typed from red blood cell samples using serological reagents. However, stem cell donors are often screened using buccal swabs which cannot be used for serologic testing. Furthermore, serologic testing can be complicated by recent transfusions, weak reactions, or unusual reaction patterns, which require resolution. ABO genotyping can help in these circumstances. Here we show the development and valid

Non-small cell lung cancer (NSCLC) remains a leading cause of cancer mortality, and many patients derive limited benefit from current systemic therapies. Until now, clinical and translational efforts have largely focused on recurrent genomic alterations in the Kelch-like ECH-associated protein 1-nuclear factor erythroid 2-related factor 2 (NRF2) axis, yet mutation status alone often fails to capture the biological heterogeneity and context dependence of NRF2 pathway dysregulation. In recent years, growing evidence has highlighted the NRF2 activation state as a clinically relevant feature that

Cryptosporidium genotyping has made significant contributions to the species structure and population genetics of Cryptosporidium spp. In addition, the standardized method has shown utility in epidemiologic investigations identifying case linkages and contamination sources that could not be determined with traditional epidemiologic tools. The standardized method for Cryptosporidium genotyping from stool specimens described here comprises multiple individual protocols to amplify and sequence regions of the small subunit ribosomal ribonucleic acid (SSU rRNA) and gp60 genes of Cryptosporidium.

Albinism is a phenotypically and genetically heterogeneous condition characterized by a variable degree of hypopigmentation and by ocular features leading to reduced visual acuity. Whereas numerous genotypic studies have been conducted throughout the world, very little is known about the genotypic spectrum of albinism in Africa and especially in sub-Saharan Western Africa. Here we report the analysis of all known albinism genes in a series a 23 patients originating from Mali. Four were diagnosed with OCA 1 (oculocutaneous albinism type 1), 17 with OCA 2, and two with OCA 4. OCA2 variant NM_000

BACKGROUND: Variants in the dynamin-1 (DNM1) gene typically cause synaptopathy, leading to developmental and epileptic encephalopathy (DEE). We aimed to determine the genotypic and phenotypic spectrum of DNM1 encephalopathy beyond DEE.
METHODS: Electroclinical phenotyping and genotyping of patients with a DNM1 variant were conducted for patients undergoing next-generation sequencing at our centre, followed by a systematic review.
RESULTS: Six patients with heterozygous DNM1 variants were identified in our cohort. Three had a typical DEE phenotype characterised by epileptic spasms, tonic seiz

Certain genetic polymorphisms of UDP glucuronosyltransferase 1 family, polypeptide A1 (UGT1A1) can reduce gene expression (*28, *60, *93) or activity (*6), thereby altering the pharmacokinetics, pharmacodynamics, and the risk of toxicities of UGT1A1 substrates, of which irinotecan is a widely-described example. This review presents an overview of the clinical effects of UGT1A1 polymorphisms on the pharmacology of UGT1A1 substrates, with a special focus on the novel histone deacetylase inhibitor belinostat. Belinostat, approved for the treatment of peripheral T-cell lymphoma, is primarily glucu

This study explored whether seasonal and/or climatic factors influenced detection of specific genotypes of Pneumocystis jirovecii. Between 1989 and 2001, 155 isolates of P. jirovecii were obtained from patients undergoing bronchoscopic alveolar lavage. For each isolate, the month and climatic conditions were noted. Genotypes of P. jirovecii were distinguished by polymorphisms in the mitochondrial large-subunit rRNA gene. There were monthly and seasonal variations in the frequency of detection of mixed genotypes (p 0.018 and p 0.031, respectively) and genotype 2 (p 0.029 and p 0.086, respective

Strain identification of Chlamydia trachomatis has historically been accomplished using serotyping as a phenotypic marker to differentiate chlamydial isolates (1). The target for serotyping is the major outer membrane protein (MOMP) which is the most antigenically diverse and abundant surface protein of the organism. Polyclonal antibodies (PAbs) were initially used for typing and were able to identify serovars D through K and LI, L2, and L3 as primarily genital pathogens, and serovars A, B, Ba, and C as trachoma pathogens. However, these groupings are somewhat imprecise. As immunotyping method

Neurodegeneration, childhood-onset, stress-induced, with variable ataxia and seizures, is a rare genetic cause of childhood neuroregression and ataxia. A girl in early childhood, born of third-degree consanguinity, presented with paroxysmal torticollis starting around 2 years of age, which worsened subsequently, accompanied by ataxia, titubation, dystonia, dyskinesias, seizures and encephalopathy. The brain MRI initially showed signal abnormalities in the periventricular white matter and bilateral putamina, with restricted diffusion seen in the latter. While the putaminal changes disappeared i

A flow cytometry assay for sex genotyping at single cell resolution is described. This assay is based on the detection of a Y chromosome-specific gene by in situ PCR. The protocol requires the isolation of lymphocytes from human blood, their fixation, permeabilization, and mild protein digestion, and the amplification of a Y chromosome-specific gene SRY, which is present only in male cells, by in situ PCR with a universal energy transfer-labelled primer (UniPrimer). Male cells can be detected by flow cytometry because the SRY gene is amplified and fluorescently labelled solely in male cells.

Babesia caballi is an intra-erythrocytic parasite causing equine piroplasmosis. Three B. caballi genotypes (A, B, and C) have been identified based on the 18 S rRNA and rhoptry-associated protein (rap-1) gene sequences. These variant parasite genotypes compromise the diagnostic utility of the WOAH-recommended serological assays in declaring horses free of equine piroplasmosis. Although a gene encoding a spherical body protein 4 (sbp4) has recently been identified as a potential antigen for the serological detection of B. caballi, the ability of this antigen to detect the different geographical

Over the past decade, there have been significant advances in our understanding of the immunopathogenesis and pharmacogenomics of severe immunologically-mediated adverse drug reactions. Such T-cell-mediated adverse drug reactions such as Stevens-Johnson syndrome/toxic epidermal necrolysis (SJS/TEN), drug-induced liver disease (DILI) and other drug hypersensitivity syndromes have more recently been shown to be mediated through interactions with various class I and II HLA alleles. Key examples have included the associations of HLA-B*15:02 and carbamazepine induced SJS/TEN in Southeast Asian popu

Listeriosis could occur as sporadic case as well as epidemic outbreak. For that reason it is important to subtype the Listeria monocytogenes strains using the fastest and most effective method. Genetic techniques are the most effective methods for differentiation of this species. Basing on latest available data eight selected subtyping genetic methods of Listeria monocytogenes strains were characterized: REA-PFGE, rybotyping, AFLP, PCR-RFLP, multiplex PCR, and methods from RAPD groups, such as AP-PCR, rep-PCR and ERIC -PCR. Analysis of available information allowed to assess that the most effe

Fabry's disease is an X-linked disorder due to mutations in the GLA gene encoding the lysosomal enzyme alpha-galactosidase A. Clinically, most patients present with the "classical" form, though "variant" forms with inaugural or preminent heart or kidney involvement have been described. Heterozygous women are most often symptomatic though generally less severely affected than men. We performed mutation analysis in 170 patients from 65 families and identified 55 different mutations. Our results confirm the wide molecular heterogeneity at this locus. Molecular study allows to confirm the diagnosi

Aicardi syndrome was originally described as a triad of partial or complete agenesis of the corpus callosum, infantile spasms, and pathognomic chorioretinal lacunae. Of approximately 200 cases reported since it was originally described in 1965, there have been no undisputed reports of Aicardi syndrome in a 46 XY male. Thus a dominant X-linked inheritance, presumed lethal in males, has been proposed. Herein we report a 5 year-old 46 XY male with the classic clinical triad of Aicardi syndrome.
