Arsenic-sensing domain controls ACR3 transporter trafficking in Marchantia polymorpha.

Arsenic is a toxic, carcinogenic metalloid that threatens human health through contaminated water and food. Arsenite [As(III)] efflux mediated by ACR3 transporters is an ancient detoxification mechanism conserved across prokaryotes, fungi, algae, and non-angiosperm land plants, yet plant ACR3 proteins remain poorly characterized. Here, we show that MpACR3, the ACR3 ortholog from the model liverwort Marchantia polymorpha, contains an arsenic-responsive N-terminal domain that regulates intracellular trafficking. Under non-stress conditions, MpACR3 is detected in Golgi bodies as well as at the pl

Cerium dioxide nanoparticles protect Catharanthus roseus against arsenic (As) toxicity by enhancing oxidative tolerance, modulating gene expression and minimizing As uptake.

Arsenic (As) is a toxic metalloid, and its increasing contamination in soil has become an important public health concern. Nevertheless, cerium dioxide nanoparticles (CeO2 NPs) have shown potential for phytostabilization of metal polluted soil. Currently during pot trial, As toxicity (As30, As70 mg kg-1) decreased growth, photosynthetic pigments, gas exchange attributes, membrane stability indices (MSIs) and relative water contents (RWCs) of Catharanthus roseus. Moreover, As stress obviously increased, malondialdehyde (MDA), As, and hydrogen peroxide (H2O2) levels in C. roseus plants. Converse

Impacts of arsenic contamination on plants and the role of microalgae in arsenic stress mitigation for sustainable agriculture.

Arsenic (As) is a toxic metalloid that is accumulating in agricultural soils and irrigation water. As is occurring from both natural geochemical processes and various anthropogenic activities. Consequently, arsenic contamination is a serious global concern, negatively affecting plant productivity, food safety, and human health. Plants absorb As from the soil through their roots via various transport mechanisms. The absorbed As is further translocated to the shoots and other tissues, where it involved in several metabolic activities and hinders in various physiological and biochemical processes

Aqueous Extract of Saposhnikoviae Radix Antagonizes Arsenic Neurotoxicity via the NLRP3/Caspase-1/GSDMD Pyroptotic Axis: Translating Environmental Exposure into Neuropathology and Herbal Recalibration.

Chronic arsenic exposure causes neurological damage, but the underlying mechanisms remain incompletely understood. This study investigated whether Saposhnikoviae Radix (SR) aqueous extract protects against arsenic-induced neurotoxicity by modulating pyroptosis. Kunming mice were exposed to NaAsO2 (10 mg/kg) with or without SR (3, 6, 12 g/kg). NaAsO2 exposure induced anxiety-like behaviors and spatial memory deficits, accompanied by widespread neuronal damage (hippocampal pyramidal cell disarray, cerebellar Purkinje cell loss, and cortical vacuolation) and upregulation of pyroptosis-related pro

Novel Role of Gut-Derived Roseburia Intestinalis in Safeguarding Intestinal Barrier Integrity and Microenvironment Homeostasis During Arsenic Exposure.

As a well-known metalloid, arsenic usually causes human intestinal disorders via contaminated drinking water. However, the mechanisms underlying how arsenic induces intestinal injury remain unresolved, and the effective means of intervention are very limited. By establishing an acute arsenic exposure animal model, this work shows that arsenic disrupts the mechanical, chemical, immunological, and biological barriers of the intestine, and thereby changes the microenvironment in the gut. We further verify that the administration of fecal microbiota transplantation with a healthy gut microbiome al

Arsenic Impairs Wound Healing Processes in Dermal Fibroblasts and Mice.

Inorganic arsenic (NaAsO2) is a naturally occurring metalloid found in water resources globally and in the United States at concentrations exceeding the U.S. Environmental Protection Agency Maximum Contamination Level of 10 ppb. While exposure to arsenic has been linked to cancer, cardiovascular disease, and skin lesions, the impact of arsenic exposure on wound healing is not fully understood. Cultured dermal fibroblasts exposed to NaAsO2 displayed reduced migration (scratch closure), proliferation, and viability with a lowest observable effect level (LOEL) of 10 µM NaAsO2 following 24 h expos

Nod-like receptor protein 3 inflammasome-mediated pyroptosis contributes to chronic NaAsO2 exposure-induced fibrotic changes and dysfunction in the liver of SD rats.

The metalloid arsenic, known for its toxic properties, is widespread presence in the environment. Our previous research has confirmed that prolonged exposure to arsenic can lead to liver fibrosis injury in rats, while the precise pathogenic mechanism still requires further investigation. In the past few years, the Nod-like receptor protein 3 (NLRP3) inflammasome has been found to play a pivotal role in the occurrence and development of liver injury. In this study, we administered varying doses of sodium arsenite (NaAsO2) and 10 mg/kg.bw MCC950 (a particular tiny molecular inhibitor targeting N

Arsenic exposure induces intestinal barrier damage and consequent activation of gut-liver axis leading to inflammation and pyroptosis of liver in ducks.

Arsenic is an important hazardous metalloid commonly found in polluted soil, rivers and groundwater. However, few studies exist regarding the effect of arsenic trioxide (ATO) on the gut-liver axis and consequent hepatotoxicity in waterfowl. Here, we investigated the influence of ATO on duck intestines and livers, and explored the role of the gut-liver axis in ATO-induced hepatotoxicity and intestinal toxicity. Our results demonstrated that ATO-exposure induced intestinal damage, liver inflammatory cell infiltration and vesicle steatosis. Additionally, the intestinal microbiota community in ATO

Reproductive disorders in female rats after prenatal exposure to sodium arsenite.

Arsenic is a metalloid widely found in the environment in organic and inorganic forms. Exposure to inorganic arsenic forms via drinking water has been associated with an increased incidence of negative health effects, including reproductive disorders and dysfunction of the endocrine system. However, the impact of arsenic exposure on female reproductive development is still unclear. Therefore, in the present study, we evaluated the effects of prenatal exposure to arsenic on the initial sexual development and puberty onset, and in the morphology of the female reproductive organs, estrous cycle r

Adsorptive removal of As(V) by crawfish shell biochar: batch and column tests.

As a toxic and metalloid substance, excess arsenic (As) can cause serious harm to the environment and public health. In this work, crayfish shell biochar (CFS450) and modified biochar (MCFS450) were prepared to remove As(V) from aqueous solutions under various conditions. Compared to CFS450, MCFS450 had a higher specific surface area, better pore structure, and higher As(V) adsorption capacity. Based on the Langmuir model, its maximum As(V) adsorption capacity was 17.2 mg/g. The biochar had a large number of surface functional groups such as C-O, O-H, and -OH. After modification, a certain mas

Effects of arsenic and cadmium on bioaccessibility of lead in spiked soils assessed by Unified BARGE Method.

The bioaccessibility of lead (Pb) in contaminated soils has been extensively studied, including the influence of soil properties on Pb bioaccessibility. However, little is known about the effects of other metals/metalloid, such as arsenic (As), cadmium (Cd) on the bioaccessibility of Pb, i.e. whether As or Cd could increase or decrease the solubility of Pb in human gastrointestinal tract when Pb-contaminated soil and As-contaminated (or Cd-contaminated) soil are ingested simultaneously. Furthermore, it is far from clear that if soil property could make a difference to these effects. In this st

Arabidopsis and the genetic potential for the phytoremediation of toxic elemental and organic pollutants.

In a process called phytoremediation, plants can be used to extract, detoxify, and/or sequester toxic pollutants from soil, water, and air. Phytoremediation may become an essential tool in cleaning the environment and reducing human and animal exposure to potential carcinogens and other toxins. Arabidopsis has provided useful information about the genetic, physiological, and biochemical mechanisms behind phytoremediation, and it is an excellent model genetic organism to test foreign gene expression. This review focuses on Arabidopsis studies concerning: 1) the remediation of elemental pollutan

The cancer metallome continuum: a framework for understanding metal dysregulation from carcinogenesis to therapeutic resistance.

Metals and metalloids play essential roles in cancer biology, influencing redox balance, epigenetic regulation, immune responses, and cell death throughout tumour development, progression, and resistance to therapy. Despite this broad biological relevance, the systematic clinical integration of metallomics into oncology remains in its early stages. This review introduces the Cancer Metallome Continuum, a framework illustrating how metallomic reprogramming evolves from cancer initiation through progression to therapy-resistant states, and evaluates the analytical, systems-level, and translation

An integrative analysis of elementomics and oxidation-reduction potentials identifies redox-related metals associated with coronary artery disease and post-PCI outcomes in type 2 diabetic patients.

Exposure to metals has been associated with elevated oxidative stress, which may contribute to diabetic macrovascular complications. However, current methods for evaluating oxidative stress, which mainly rely on quantifying individual byproducts of oxidative damage, may not fully characterize the effects of metal exposure on global redox imbalance, especially in the setting of type 2 diabetes mellitus (T2DM). Here, by assessing global redox status using the novel marker oxidation-reduction potential (ORP) and performing elemental profiling of 35 plasma metals/metalloids in a large cohort of 31

The association among metals and changes in their concentrations in the treatment of lead poisoning in lead-exposed workers.

BACKGROUND: High-throughput metallomics studies on lead-exposed workers are rare, and the effects of lead exposure on other metals, as well as the loss of trace metals during such treatments, remain unclear. METHODS: We recruited 1091 lead-exposed and 75 lead-poisoned workers undergoing calcium disodium ethylenediaminetetraacetate (CaNa2EDTA) treatment. Urine samples were analyzed for 33 metals/metalloids using inductively coupled plasma mass spectrometry (ICP-MS). In lead-poisoned workers, first morning urine was collected daily over a 15-day two-course of lead elimination treatment, plasma

Advances in immunology of male reproductive toxicity induced by common environmental pollutants.

Humans are exposed to an ever-increasing number of environmental toxicants, some of which have gradually been identified as major risk factors for male reproductive health, even associated with male infertility. Male infertility is usually due to the reproductive system damage, which may be influenced by the exposure to contaminants such as heavy metals, plasticizers, along with genetics and lifestyle. Testicular immune microenvironment (TIM) is important in maintaining normal physiological functions of the testis, whether disturbed TIM after exposure to environmental toxicants could induce re
