Idiopathic pulmonary fibrosis (IPF) is a chronic progressive fibrotic lung disease with an extremely poor prognosis, and the rate of disease progression varies significantly among patients. Therefore, accurate prognostic assessment is crucial for clinical management. Traditional pulmonary function parameters, such as the absolute value of forced vital capacity (FVC) and the rate of decline in FVC as a percentage of predicted value (FVC%), serve as important bases for predicting disease progression and mortality. In recent years, prognostic prediction models have evolved from single indicators

Although mutant-specific KRAS inhibitors are approved to treat cancer, a deeper understanding of intratumoral changes driven specifically by KRAS inhibition is needed to maximize therapeutic responses. In this study, we used single-cell RNA sequencing, flow cytometry, and spatial transcriptomics to distinguish mechanisms of tumor control after KRASG12C inhibition [KRAS(G12C)i] or MEK inhibition (MEKi). Despite both inhibiting the MAPK pathway, KRAS(G12C)i and MEKi drive the adaptation of distinct neoplastic cell fates affecting metabolism and cell-cycle regulation, and additive tumor suppressi

Exosomes have emerged as promising therapeutic carriers, with over 40 000 scientific publications reflecting their exponential growth in biomedical research. Cardiovascular diseases (CVDs) are the leading cause of mortality worldwide. Among these, myocardial infarction (MI) represents the most prevalent and devastating form, contributing significantly to global morbidity and mortality. Reducing the mortality and long-term complications associated with MI is therefore both necessary and urgent. Exosomes, naturally occurring extracellular vesicles, offer a promising cell-free alternative for dru

Few conditions in reproductive medicine rival polycystic ovary syndrome (PCOS) in terms of clinical breadth and global impact. Affecting roughly 6-21% of women of childbearing age depending on which diagnostic criteria are applied PCOS sits at the intersection of endocrinology, metabolism, and gynecology, making it difficult to capture within any single disciplinary lens. Its hallmarks are well rehearsed: excess androgens, disrupted ovulation, and the characteristic follicular architecture seen on pelvic ultrasound. Yet what makes PCOS genuinely challenging is the degree to which these reprodu

Frontotemporal Dementia and Parkinsonism linked to chromosome 17 (FTDP-17) is a rare, early-onset, autosomal-dominant neurodegenerative tauopathy caused by mutations in the Microtubule-Associated Protein Tau (MAPT) gene. A subset of these mutations selectively disrupts the normal alternative splicing of MAPT exon 10, altering the physiological ratio of 3-Repeat (3R) and 4-Repeat (4R) tau isoforms; this imbalance, driving pathological tau aggregation and progressive neurodegeneration, is a pathogenic hallmark of FTDP-17. This review explores the molecular mechanisms regulating exon 10 splicing,

The relentless emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants continues to challenge global health, as high mutation rates and complex pathogenicity obscure molecular mechanisms and impede clinical progress. Despite extensive research across viral evolution, structural biology, immunology, diagnostics, and therapeutics, the resulting vast and rapidly outdated literature has widened the gap between fundamental discovery and medical application. Here, we systematically mined 439,724 coronavirus disease 2019 (COVID-19) publications using fine-tuned large langua

Exercise provides health benefits to multiple metabolic tissues through complex biological pathways and interactions between organs. However, investigating these complex mechanisms in humans is still limited, making mouse models extremely useful for exploring exercise-induced changes in whole-body metabolism and health. In this review, we focus on gaining a broader understanding of the metabolic phenotypes and molecular mechanisms induced by exercise in mouse models. We first discuss the differences in adaptations induced by aerobic and resistance exercise, and compare voluntary wheel running

The dissection of the veins is the trickiest step of Uterine transplantation (UTx). Performing the anastomosis of a single uterine vein could bring a therapeutic benefit and simplification of surgery and serve for managing unilateral venous thromboses. The objectives of this project were to evaluate the expression of early markers of ischemia-reperfusion and to compare findings following one or two vein anastomoses. Orthotopic uterine auto-transplantations were performed on an ovine model with anastomosis of either two (group 1) or one utero-ovarian veins (group 2). Blood gases, histology and

Ellis-van Creveld syndrome (EVC; MIM ID #225500) is a rare congenital disease with an occurrence of 1 in 60,000. It is characterized by remarkable skeletal dysplasia, such as short limbs, ribs and polydactyly, and orofacial anomalies. With two of three patients first noted as being offspring of consanguineous marriage, this autosomal recessive disease results from mutations in one of two causative genes: EVC or EVC2/LIMBIN. The recent identification and manipulation of genetic homologs in animals has deepened our understanding beyond human case studies and provided critical insight into diseas

Feingold syndrome is a skeletal dysplasia caused by loss-of-function mutations of either MYCN (type 1) or MIR17HG that encodes miR-17-92 microRNAs (type 2). Since miR-17-92 expression is transcriptionally regulated by MYC transcription factors, it has been postulated that Feingold syndrome type 1 and 2 may be caused by a common molecular mechanism. Here we show that Mir17-92 deficiency upregulates TGF-β signaling, whereas Mycn-deficiency downregulates PI3K signaling in limb mesenchymal cells. Genetic or pharmacological inhibition of TGF-β signaling efficiently rescues the skeletal defects caus

We present an original hybrid QM/MM scheme merging the many-body Green's function GW formalism with classical discrete polarizable models and its application to the paradigmatic case of a pentacene crystal. Our calculated transport gap is found to be in excellent agreement with reference periodic bulk GW calculations, together with properly parametrized classical microelectrostatic calculations, and with photoionization measurements at crystal surfaces. More importantly, we prove that the gap is insensitive to the partitioning of pentacene molecules in QM and MM subsystems, as a result of the

Polyglutamine (PolyQ) diseases have common features that include progressive selective neurodegeneration and the formation of protein aggregates. There is growing evidence to suggest that critical nuclear events lead to transcriptional alterations in PolyQ diseases such as spinocerebellar ataxia type 7 (SCA7) and Huntington's disease (HD), conditions which share a cerebellar degenerative phenotype. Taking advantage of laser capture microdissection technique, we compared the Purkinje cell (PC) gene expression profiles of two transgenic polyQ mouse models (HD: R6/2; SCA7: P7E) by microarray anal

PURPOSE: Acute renal tubular necrosis (ATN), a common cause of acute renal failure, is a dynamic, rapidly evolving clinical condition associated with apoptotic and necrotic tubular cell death. Its early identification is critical, but current detection methods relying upon clinical assessment, such as kidney biopsy and functional assays, are insufficient. We have developed a family of small molecule compounds, ApoSense, that is capable, upon systemic administration, of selectively targeting and accumulating within apoptotic/necrotic cells and is suitable for attachment of different markers for

The analysis of heart rate variability (HRV) provides information on neural control of the heart. We investigated HRV in normal rats and in models of experimental cardiac hypertrophy using the Holter monitoring and peak/trough method. In normal rats, two heart rate oscillations with different wavelengths, high frequency (HF) and low frequency (LF) oscillations, were detected. The HF oscillations were insensitive to propranolol and suppressed by atropine. The LF oscillations were sensitive to both antagonists. Thyrotoxicosis resulted in cardiac hypertrophy (+20%) and tachycardia. The HF oscilla

The interfacial electronic structure of biopolymer-metal oxide hybrids determine their stability, yet this structure is still not well understood. Here, we report a density functional theory study of 10 model systems consisting of cellulose and chitosan functionalized with ZnO and V2O5 separately and in ternary combination at the B3LYP/SDD level. According to frontier molecular orbital analysis, hybridization reduces the energy gap in the ternary composites from 7.106 eV for cellulose and 6.908 eV for chitosan to 3.559 eV and 3.496 eV, while the dipole moment increases from 0.536 D to 26.697 D

Dihydroquercetin (DHQ) is a promising object for the development of a treatment for patients with obesity and prediabetes requiring a moderate therapeutic effect. This paper reports a clinical case of DHQ application in a 30-year-old Caucasian male and proposes a molecular mechanism of its anti-obesity effect. DHQ was administrated as a dietary supplement at a dose of 100-200 mg/day during 3 months with treatment interruption for 1 month. The data collected one month before the treatment were used as a control. The molecular aspects were studied via molecular docking with β3-adrenoceptor (ADRB

BACKGROUND: The omentum is a vascularized, immune-active tissue with regenerative potential, particularly when activated by intraperitoneal stimuli. Its secreted factors may promote lymphangiogenesis, offering a novel approach to lymphedema treatment.
METHODS: The omentum was activated in mice using an intraperitoneal polydextran slurry. Gene expression was evaluated over time to assess inflammatory and lymphatic markers. Culture supernatants from the activated omentum were collected, and vascular endothelial growth factor-C ( VEGF-C ) levels were measured. Therapeutic potential was tested in

Phosphorodiamidate morpholino oligonucleotides (PMOs) are a class of antisense oligonucleotides. While PMOs have enabled several nucleic acid therapeutics, their structural and energetic solution properties remain poorly understood. Using solution viscosity measurements combined with computational molecular modeling, we explored interactions of therapeutic 22-mer, 25-mer, and 30-mer PMOs in concentrated solutions. Self-association of PMO monomers involves non-specific interactions, is energetically favorable (with estimates of -32 to -67 kcal/mol interaction energies), and exhibits biphasic ki

Plant-herbivore interactions follow zig-zag-like cycles in which herbivore- and damage-derived cues activate conserved defense signaling hubs, while insect effectors suppress or redirect these pathways, creating recurrent molecular bottlenecks that shape plant resistance. Plant-herbivore interactions are orchestrated through a dynamic molecular dialogue at the feeding interface, wherein insect attack triggers the generation of damage-derived signals from plant tissues and herbivore-derived signals from oral or salivary secretions. These cues can activate plant defense by sensing the damage- an
