Cryptosporidium infecting wild cricetid rodents from the subfamilies Arvicolinae and Neotominae.

We undertook a study on Cryptosporidium spp. in wild cricetid rodents. Fecal samples were collected from meadow voles (Microtus pennsylvanicus), southern red-backed voles (Myodes gapperi), woodland voles (Microtus pinetorum), muskrats (Ondatra zibethicus) and Peromyscus spp. mice in North America, and from bank voles (Myodes glareolus) and common voles (Microtus arvalis) in Europe. Isolates were characterized by sequence and phylogenetic analyses of the small subunit ribosomal RNA (SSU) and actin genes. Overall, 33·2% (362/1089) of cricetids tested positive for Cryptosporidium, with a greater

Procedural learning and cognitive flexibility in a mouse model of restricted, repetitive behaviour.

Restricted, repetitive behaviours (e.g., stereotypies, compulsions, rituals) in neurodevelopmental disorders have been linked to alterations in cortico-basal ganglia circuitry. Cognitive processes mediated by this circuitry (e.g., procedural learning, executive function) are likely to be impaired in individuals exhibiting high rates of repetitive behaviour. To test this hypothesis, we assessed both procedural learning and cognitive flexibility (reversal learning) using a T-maze task in deer mice (Peromyscus maniculatus) exhibiting various rates of repetitive behaviour (vertical jumping and bac

Novel potential reservoirs for Borrelia sp. and the agent of human granulocytic ehrlichiosis in Colorado.

Previous work demonstrated that Ixodes spinipalpis ticks maintained an enzootic cycle of Borrelia bissettii and the agent of human granulocytic ehrlichiosis (aoHGE) within woodrats (Neotoma mexicana) and deer mice (Peromyscus maniculatus) in northern Colorado (USA). Because I. spinipalpis is the only known vector of B. bissettii and aoHGE in Colorado, this study was designed to determine the reservoir status of other hosts of I. spinipalpis in five distinct ecological zones along the front range and foothills of Colorado. One hundred and twelve rodents of nine species were examined and 11 (10%

Genetic structure of the white-footed mouse in the context of the emergence of Lyme disease in southern Québec.

The white-footed mouse (Peromyscus leucopus) has expanded its northern limit into southern Québec over the last few decades. P. leucopus is a great disperser and colonizer and is of particular interest because it is considered a primary reservoir for the spirochete bacterium that causes Lyme disease. There is no current information on the gene flow between mouse populations on the mountains and forest fragments found scattered throughout the Montérégie region in southern Québec, and whether various landscape barriers have an effect on their dispersal. We conducted a population genetics analysi

Evaluation of a modified next-generation sequencing assay to detect Borrelia, Anaplasma, and Babesia in blood and ear tissue samples from field-collected rodents.

Efforts to monitor the expanding geographic distribution of pathogens associated with Ixodes scapularis Say (Acari: Ixodidae), and to evaluate the outcomes of environmental control interventions aiming to reduce the intensity of enzootic transmission of these pathogens, are dependent on sensitive and specific molecular assays for detection of pathogens in ticks and their rodent hosts. In this study, we modified a highly sensitive and specific multiplex PCR amplicon sequencing assay developed to identify pathogens in Ixodes ticks for use in screening blood or ear tissue from rodents (Peromyscus

Ixodes scapularis density and Borrelia burgdorferi prevalence along a residential-woodland gradient in a region of emerging Lyme disease risk.

The environmental risk of Lyme disease, defined by the density of Ixodes scapularis ticks and their prevalence of Borrelia burgdorferi infection, is increasing across the Ottawa, Ontario region, making this a unique location to explore the factors associated with environmental risk along a residential-woodland gradient. In this study, we collected I. scapularis ticks and trapped Peromyscus spp. mice, tested both for tick-borne pathogens, and monitored the intensity of foraging activity by deer in residential, woodland, and residential-woodland interface zones of four neighbourhoods. Everglades virus: an underrecognized disease-causing subtype of Venezuelan equine encephalitis virus endemic to Florida, USA.

Everglades virus (EVEV) is subtype II of the Venezuelan equine encephalitis virus (VEEV) complex (Togaviridae: Alphavirus), endemic to Florida, USA. EVEV belongs to a clade that includes both enzootic and epizootic/epidemic VEEV subtypes. Like other enzootic VEEV subtypes, muroid rodents are important vertebrate hosts for EVEV and certain mosquitoes are important vectors. The hispid cotton rat Sigmodon hispidus and cotton mouse Peromyscus gossypinus are important EVEV hosts, based on natural infection (virus isolation and high seropositivity), host competence (experimental infections), and fre

High Proportion of Unfed Larval Blacklegged Ticks, Ixodes scapularis (Acari: Ixodidae), Collected From Modified Nest Boxes for Mice.

The blacklegged tick, Ixodes scapularis Say, is the primary vector of several tick-borne pathogens, including those causing Lyme disease and babesiosis, in the eastern United States and active collection methods for this species include dragging or wild animal sampling. Nest boxes targeting mice may be an alternative strategy for the surveillance and collection of immature I. scapularis feeding on these hosts and would be much safer for animals compared to small mammal trapping. We constructed double-walled insulated nest boxes (DWINs) with collection tubes mounted below the nesting chamber an

Ixodes scapularis (Acari: Ixodidae) Reservoir Host Diversity and Abundance Impacts on Dilution of Borrelia burgdorferi (Spirochaetales: Spirochaetaceae) in Residential and Woodland Habitats in Connecticut, United States.

The dilution effect in the zoonotic disease transmission cycle theorizes that an increased diversity of host species will alter transmission dynamics, result in a decrease in pathogen prevalence, and potentially lower human disease incidence. The interrelationship of Borrelia burgdorferi (Johnson, Schmid, Hyde, Steigerwalt, and Brenner) (Spirochaetales: Spirochaetaceae), the etiological agent of Lyme disease (LD), and its primary vector, blacklegged ticks (Ixodes scapularis Say) (Acari: Ixodidae), is a commonly used example of the dilution effect, suggesting that an increased diversity of host

Impact of Sylvatic Plague Vaccine on Non-target Small Rodents in Grassland Ecosystems.

Oral vaccination is an emerging management strategy to reduce the prevalence of high impact infectious diseases within wild animal populations. Plague is a flea-borne zoonosis of rodents that often decimates prairie dog (Cynomys spp.) colonies in the western USA. Recently, an oral sylvatic plague vaccine (SPV) was developed to protect prairie dogs from plague and aid recovery of the endangered black-footed ferret (Mustela nigripes). Although oral vaccination programs are targeted toward specific species, field distribution of vaccine-laden baits can result in vaccine uptake by non-target anima

Minimal role of eastern fence lizards in Borrelia burgdorferi transmission in central New Jersey oak/pine woodlands.

The eastern fence lizard, Sceloporus undulatus , is widely distributed in eastern and central North America, ranging through areas with high levels of Lyme disease, as well as areas where Lyme disease is rare or absent. We studied the potential role of S. undulatus in transmission dynamics of Lyme spirochetes by sampling ticks from a variety of natural hosts at field sites in central New Jersey, and by testing the reservoir competence of S. undulatus for Borrelia burgdorferi in the laboratory. The infestation rate of ticks on fence lizards was extremely low (prevalence = 0.087, n = 23) compare

When is a parasite not a parasite? Effects of larval tick burdens on white-footed mouse survival.

Many animal species can carry considerable burdens of ectoparasites: parasites living on the outside of a host's body. Ectoparasite infestation can decrease host survival, but the magnitude and even direction of survival effects can vary depending on the type of ectoparasite and the nature and duration of the association. When ectoparasites also serve as vectors of pathogens, the effects of ectoparasite infestation on host survival have the potential to alter disease dynamics by regulating host populations and stabilizing transmission. We quantified the impact of larval Ixodes scapularis tick

Evidence for feedback control of pineal melatonin secretion.

Melatonin is the principle hormonal product of the pineal gland. It is secreted with a robust daily rhythm, peaking near the middle of the night. During the daytime, concentrations remain very low, as exposure to light robustly suppresses its secretion. The regulation of melatonin by light is well-characterized, but an interesting feature of the daily melatonin rhythm is that its peak occurs near the middle of the night and then levels begin to drop hours before morning light exposure. The mechanism underlying the light-independent drop in melatonin during late night remains unspecified. Feedb

Selective blockade of spontaneous motor stereotypy via intrastriatal pharmacological manipulation.

Abnormal repetitive behavior, including stereotypy, is often observed in conjunction with developmental, neuropsychiatric, and genetic disorders. The present work employed the deer mouse model of spontaneous and persistent stereotypy to identify basal ganglia involvement in the mediation of these abnormal behaviors. To evaluate the hypothesis that stereotypy is expressed in these mice due to alterations in the activity of cortico-basal ganglia motor circuits, intrastriatal pharmacological manipulations aimed at attenuating the spontaneously emitted stereotypy were performed. Bilateral striatal

Interference between the agents of Lyme disease and human granulocytic ehrlichiosis in a natural reservoir host.

Agents of Lyme disease (Borrelia burgdorferi) and human granulocytic ehrlichiosis (Ehrlichia phagocytophila) are perpetuated in a natural cycle involving the black-legged tick (Ixodes scapularis) and its vertebrate hosts. Using I. scapularis nymphs as the mode of infectious challenge, we studied how infection with one pathogen in white-footed mice (Peromyscus leucopus) affects their ability to acquire the other agent and subsequently to infect larvae, which these agents would do in nature. Two groups of mice were infected with either B. burgdorferi or E. phagocytophila. One week later, B. burg

Differential distribution of immature Ixodes dammini (Acari: Ixodidae) on rodent hosts.

Ectoparasites such as ixodid ticks that remain attached to hosts for several days while feeding on blood are able to overcome the inflammatory and immune responses of some hosts and not others. The immature stages of the deer tick Ixodes dammini are found more frequently on the white-footed mouse, Peromyscus leucopus, than on other rodents. We propose that P. leucopus is more tolerant to I. dammini than is a less common host, the meadow vole, Microtus pennsylvanicus. To test this hypothesis, the distribution patterns and engorgement indices were determined for larval and nymphal I. dammini col

Amblyomma auricularium (Ixodida: Ixodidae) in Florida: New Hosts and Distribution Records.

Previous published evidence for the occurrence of an exotic armadillo tick, Amblyomma auricularium (Conil), in Florida is scant, but we found it is fully established and integrated into the state's tick fauna. We collected 11,192 specimens of this tick from naturalized nine-banded armadillos, Dasypus novemcinctus L., and 14 other species of wild native mammals and birds in Florida, while sampling statewide during 2004 through 2007. In all, we document its specific presence only in 14 contiguous South Florida counties. Moreover, we report the first collections of A. auricularium from the Virgin

Prevalence of Yersinia pestis in rodents and fleas associated with black-tailed prairie dogs (Cynomys ludovicianus) at Thunder Basin National Grassland, Wyoming.

Rodents (and their fleas) that are associated with prairie dogs are considered important for the maintenance and transmission of the bacterium (Yersinia pestis) that causes plague. Our goal was to identify rodent and flea species that were potentially involved in a plague epizootic in black-tailed prairie dogs at Thunder Basin National Grassland. We collected blood samples and ectoparasites from rodents trapped at off- and on-colony grids at Thunder Basin National Grassland between 2002 and 2004. Blood samples were tested for antibodies to Y. pestis F-1 antigen by a passive hemagglutination as

A comparative study of mammalian and reptilian alternative pathway of complement-mediated killing of the Lyme disease spirochete (Borrelia burgdorferi).

The potential bactericidal activity of the alternative complement pathway of mammalian and reptilian sera to Borrelia burgdorferi sensu stricto (s.s.) was evaluated in vitro. Complement-mediated killing was observed when cultured spirochetes were inoculated into sera from the western fence lizard (Sceloporus occidentalis) and from the southern alligator lizard (Elgaria multicarinata), but not when they were inoculated into serum from either the deer mouse (Peromyscus maniculatus) or from humans. Spirochetes were still alive after 4 hr in lizard serum that had been preheated at 56 C for 30 min

Impacts of Tick Parasitism on the Rodent Gut Microbiome.

Host microbiota may impact disease vector behavior and pathogen transmission, but little is known about associations between ectoparasites and microbial communities in wildlife reservoir species. We used Illumina metagenomic sequencing to explore the impacts of tick parasitism on the rodent fecal microbiome in both a field and laboratory setting. We found that tick parasitism on wild hosts was associated with variation in the fecal microbiota of both the white-footed deermouse, Peromyscus leucopus, and the southern cotton rat, Sigmodon hispidus. In a lab experiment, we detected significant cha
