Human DNA-PK activates a STING-independent DNA sensing pathway.

Detection of intracellular DNA by the cGAS-STING pathway activates a type I interferon-mediated innate immune response that protects from virus infection. Whether there are additional DNA sensing pathways, and how such pathways might function, remains controversial. We show here that humans-but not laboratory mice-have a second, potent, STING-independent DNA sensing pathway (SIDSP). We identify human DNA-dependent protein kinase (DNA-PK) as the sensor of this pathway and demonstrate that DNA-PK activity drives a robust and broad antiviral response. We show that the E1A oncoprotein of human ade

A Rare Presentation of Pancreatic Panniculitis-like Reaction After a Wasp Sting.

Localized skin necrosis after bee and wasp stings is an exceptionally rare phenomenon, with few reported cases documented in the literature. Hymenoptera stings, including those from wasps, bees, and stinging ants, are common worldwide and typically result in localized reactions such as pain, erythema, and swelling. Although systemic complications, including anaphylaxis and multiorgan damage, are well recognized, localized skin necrosis remains poorly understood. In this study, we present a rare case of localized dermal necrosis in a 75-year-old woman with no systemic involvement after a yellow

SEL1L3 suppresses colorectal cancer cell growth and metastasis by preventing endoplasmic reticulum-associated degradation of STING.

Endoplasmic reticulum associated degradation (ERAD) plays pivotal role in protein homeostasis and quality control in normal and cancer cells, yet the regulatory mechanism of ERAD remains elusive, especially regarding its ubiquitination function mediated by hydroxymethylglutaryl reductase degradation protein 1 (HRD1). Here, we report that Sel-1 Suppressor of Lin-12-Like 3 (SEL1L3) protein resided on the ER membrane can effectively prevent HRD1-mediated ERAD process via dual mechanisms: SEL1L3 disrupts SEL1L-HRD1 complex by mutually exclusively interacting with SEL1L and HRD1 respectively, resul

Gut Microbiota-Derived TMAO Drives MC3T3-E1 Senescence and Osteogenic Dysfunction via cGAS-STING-NF-κB Signaling: Implications for Age-Related Bone Loss.

Age-related osteoporosis is closely associated with osteoblast dysfunction, in which cellular senescence plays a key role. Trimethylamine N-oxide (TMAO), a gut microbiota-derived metabolite, is implicated in aging and metabolic diseases and has been linked to bone metabolism. However, whether TMAO impairs bone formation by regulating osteoblast senescence remains unclear. This study investigated the effects of TMAO on osteoblast senescence and osteogenic function, focusing on the cGAS-STING-NF-κB signaling axis. MC3T3-E1 cells were treated with TMAO to evaluate proliferation, cell cycle progre

Dual-functional catalytic photothermal immunological agent for ferroptosis induction and cGAS-STING pathway activation: Enhancing tumor immunotherapy and generating robust immune memory.

The resistance to apoptosis and immune evasion of tumor cells significantly increase the risk of cancer treatment failure. Ferroptosis has emerged as a new strategy to address apoptosis resistance in tumor therapy. However, traditional ferroptosis inducers often lead to drug resistance due to the compensatory mechanism of GPX4, while the synergistic effect of photothermal-immunotherapy can circumvent this limitation. Additionally, relying solely on innate immunotherapy may fail to effectively suppress aggressive tumors with "cold" and immunosuppressive microenvironments, thereby inducing immun

Tumor-associated macrophage-derived exosomes promote radio-resistance in nasopharyngeal carcinoma cells accompanied by suppression of the cGAS/STING pathway.

PURPOSE: Radiotherapy resistance remains a major obstacle in nasopharyngeal carcinoma (NPC), leading to poor prognosis. While tumor-associated macrophages (TAMs) are implicated in radio-resistance, their mechanistic role is unclear. This study investigates the pivotal function of TAM-derived exosomes (TAM-Exos) and their underlying molecular mechanisms in conferring radio-resistance to NPC cells. MATERIALS AND METHODS: HK-1 cells were co-cultured with M0 macrophages derived from phorbol 12-myristate 13-acetate-induced THP-1 cells under conditions with or without ionizing radiation (IR). Gelsevirine ameliorates sepsis-associated encephalopathy by inhibiting the STING signalling-mediated pyroptosis pathway in microglia.

BACKGROUND: Sepsis-associated encephalopathy (SAE) is among the most prevalent and deadly complications associated with sepsis, but satisfactory treatments and therapeutic agents are lacking. Gelsevirine, an active ingredient derived from Gelsemium elegans Benth., has shown promising effects in animal models of anxiety, ischaemic stroke and osteoarthritis. However, its protective effect against SAE and its mechanism of action are still unknown. PURPOSE: To elucidate the efficacy of gelsevirine against SAE and the mechanism of its protective effect through the STING signalling-mediated pyropto

A novel STING agonist for cancer immunotherapy and a SARS-CoV-2 vaccine adjuvant.

A novel STING agonist, CDGSF, ipsilaterally modified with phosphorothioate and fluorine, was synthesized. The phosphorothioate in CDGSF might be a site for covalent conjugation. Injection of CDGSF generated an immunogenic ("hot") tumor microenvironment to suppress melanoma, more efficiently than dithio CDG. In particular, immunization with SARS-CoV-2 spike protein using CDGSF as an adjuvant elicited an exceptionally high antibody titer and a robust T cell response, overcoming the drawbacks of aluminum hydroxide. These results highlighted the therapeutic potential of CDGSF for cancer immunother

Hepatitis B virus polymerase disrupts K63-linked ubiquitination of STING to block innate cytosolic DNA-sensing pathways.

UNLABELLED: The cellular innate immune system recognizing pathogen infection is essential for host defense against viruses. In parallel, viruses have developed a variety of strategies to evade the innate immunity. The hepatitis B virus (HBV), a DNA virus that causes chronic hepatitis, has been shown to inhibit RNA helicase RIG-I-mediated interferon (IFN) induction. However, it is still unknown whether HBV could affect the host DNA-sensing pathways. Here we report that in transiently HBV-transfected Huh7 cells, the stably HBV-producing cell line HepAD38, and HBV-infected HepaRG cells and primar

Mangiferin inhibits cGAS-STING pathway-related inflammation via Nrf2 activation to protect against sepsis-induced heart injury.

BACKGROUND: Septic cardiomyopathy is characterized by oxidative stress and inflammation, and accounts for its associated high mortality. Mangiferin is a naturally occurring xanthonoid found abundantly in Anemarrhena asphodeloides Bunge, a traditional Chinese herb widely used for treatment of cardiovascular diseases. This study was designed to investigate the cardioprotective role of mangiferin against sepsis-induced heart injury with a focus on mitochondrial DNA (mtDNA) release and cGAS-STING pathway-related inflammation. METHODS: The septic cardiomyopathy model in mice was established by int

Decursin protects against DSS-induced experimental colitis in mice by inhibiting the cGAS-STING signaling pathway.

While studies have shown that Angelica gigas Nakai (A. gigas) can alleviate ulcerative colitis in mice, the therapeutic role of its main active ingredient, decursin, is uncertain. Therefore, we aimed to investigate the protective effect and mechanism of decursin against inflammatory bowel disease (IBD) in vivo using mice. IBD was simulated via induction with 3% dextran sodium sulfate (DSS), with or without daily treatment with decursin (10 mg/kg or 20 mg/kg) or 5-amino salicylic acid (5-ASA; 100 mg/kg) for 14 days. Mice were weighed and monitored daily for disease activity index (DAI) scoring.

Hierarchical engineering of mesoporous polydopamine for "homologous targeting-cascade blasting" biomimetic phototheranostic nanotrident.

The activation of the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway represents a promising strategy for eliciting host immune responses to eradicate tumors. However, the clinical application of STING agonists is severely hindered by the tumor immunosuppressive microenvironment (TIME) and non-specific delivery. Herein, a biomimetic phototheranostic nanotrident (mPDXZ@M) is meticulously designed through hierarchical engineering of mesoporous polydopamine (mPDA). Upon near-infrared laser irradiation, mPDA-mediated photothermal therapy (PTT) evokes robust cell apopt

Interferon stimulatory DNA activates the DNA damage signaling through ATM and DNA-PK sensing.

In eukaryotic cells, DNA is normally confined in the nucleus and mitochondria and the presence of DNA in the cytoplasm is a danger signal that activates innate immune responses. Upon detection of cytoplasmic dsDNA in mammalian cells, the cGAS-STING pathway induces type I-Interferon and inflammatory responses, a key step in innate immune activation. Since its discovery, Interferon Stimulatory DNA (ISD), a linear double-stranded DNA, has been largely used to study the cGAS-STING pathway and its regulation. Here, we show that ISD also stimulates DNA damage signaling. We show that ISD activates bo

Ginsenoside Rb3 attenuates skin flap ischemia-reperfusion damage by inhibiting STING-IRF3 signaling.

We investigate the protective effect of ginsenoside Rb3 on skin flap microvasculature following ischemia-reperfusion (I/R) injury and its regulatory mechanism. We used a rat model of I/R injury with the right iliolumbar artery and oxidative stress model of human dermal microvascular endothelial cells. The effects of Rb3 on skin flap tissue and endothelial cell survival, STING-IRF3 pathway activation, and endothelial cell adhesion were measured. Following reperfusion, the survival rate of rat perforator flaps in the Rb3-treated group gradually increased with increasing Rb3 concentration. The tr

Glucagon-like peptide-1 receptor agonists rescued diabetic vascular endothelial damage through suppression of aberrant STING signaling.

Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) protect against diabetic cardiovascular diseases and nephropathy. However, their activity in diabetic retinopathy (DR) remains unclear. Our retrospective cohort study involving 1626 T2DM patients revealed superior efficacy of GLP-1 RAs in controlling DR compared to other glucose-lowering medications, suggesting their advantage in DR treatment. By single-cell RNA-sequencing analysis and immunostaining, we observed a high expression of GLP-1R in retinal endothelial cells, which was down-regulated under diabetic conditions. Treatment of GLP-1

GPR162 activates STING dependent DNA damage pathway as a novel tumor suppressor and radiation sensitizer.

In the treatment of most malignancies, radiotherapy plays a significant role. However, the resistance of cancer cells to ionizing radiation (IR) is the main reason for the failure of radiotherapy, which causes tumor recurrence and metastasis. In this study, we confirmed that GPR162, an orphan receptor in the G-protein-coupled receptor family, acted as a novel radiotherapy sensitizer by interacting with the stimulator of interferon genes (STING), which targeted DNA damage responses, activated IRF3, accelerated the activation of type I interferon system, promoted the expression of chemokines inc

Aucubin administration suppresses STING signaling and mitigated high-fat diet-induced atherosclerosis and steatohepatosis in LDL receptor deficient mice.

The rising obesity epidemic in developed countries is associated with many chronic inflammatory diseases including atherosclerosis and nonalcoholic steatohepatitis (NASH). Consuming aucubin may benefit health by suppressing inflammation. Herein, we studied the effects of aucubin consumption on atherosclerosis and NASH progression induced by high-fat diet (HFD) in LDL receptor deficient (LDLr-/-) mice. Adult LDLr-/- mice were fed with HFD for 12 weeks and received oral administration of aucubin for the last 6 weeks. Aucubin did not alter body weight or dyslipidemia, but lowered hyperglycemia an

The conserved poxvirus membrane entry-fusion apparatus component OPG147 targets MITA/STING for immune evasion.

Monkeypox virus (MPXV) causes severe diseases in immunocompromised individuals. How MPXV evades the host defense remain enigmatic. We performed expression screens and identified MPXV OPG147, a membrane fusion machinery protein, as an inhibitor of cGAS-MITA/STING-mediated innate immunity. OPG147 from other poxviruses including the prototypic vaccinia virus (VACV) shows similar functions. OPG147 is associated with MITA/STING and STIM1, a calcium sensor that retains MITA/STING in the ER. OPG147 does not block cGAMP binding to MITA, but inhibits its ISGylation, dimerization/oligomerization and tra
