Fumigant Activity of 6 Selected Essential Oil Compounds and Combined Effect of Methyl Salicylate And Trans-Cinnamaldehyde Against Culex pipiens pallens.

We studied the knockdown activity and lethal toxicity of 6 essential oil compounds-methyl salicylate, linalool, 2-phenethyl alcohol, eugenol, β-citronellol, and trans-cinnamaldehyde-as fumigants against adult female Culex pipiens pallens in the laboratory. Of the 6 products tested, trans-cinnamaldehyde was the most toxic (LC50  =  0.26 µl/l air, 24 h) with a slow knockdown time (KT95  =  176.5 min at 0.5 µl/l air). Methyl salicylate displayed a lower toxicity (LC50  =  1.17 µl/l air, 24 h) but the fastest knockdown activity (KT95  =  16.8 min) at the sublethal concentration 0.5 µl/l air. Furth

Clearance of intracellular bacterial infections by hyaluronic acid-based ROS responsive drug delivery micelles.

Pathogenic bacteria residing inside cells could cause disruption of cellular metabolic balance. Therefore, basing on high oxidative stress response of the intracellular bacteria infected micro-environment, a novel amphipathic micelle (HATAD-TCS) was developed consisting of hyaluronic acid-derivative and reactive oxygen species (ROS) - responsive group and antibacterial agent triclosan (TCS). ROS-generating cinnamaldehyde (CA) was incorporated into ROS-cleavable linkages which are future linked to the 1-decylamine to form hydrophobicity. The cinnamaldehyde released did not just killed bacteria

Assessing the hazard of E-Cigarette flavor mixtures using zebrafish.

Since 2007, electronic cigarette (e-cigarette) sales in the U.S. have surpassed those of tobacco cigarettes. This is due, in part, to manufacturer's claims that they are a safer alternative to tobacco cigarettes. However, formaldehyde, acrolein, and diacetyl have been detected in e-cigarettes and public knowledge of e-cigarette composition and ingredient bioactivity is conspicuously lacking. We evaluated the toxicity of nine e-cigarette flavor mixtures and their constituents in the developmental zebrafish, an excellent whole animal biosensor of chemical hazard. Seven of the nine flavors (78%)

Enzymatic toxicogenation of "activated" cyclophosphamide by 3'-5' exonucleases.

3'-5' Exonucleases from various sources were found to toxicogenate 4-hydroxycyclophosphamide ("activated" cyclophosphamide) by splitting the oxazaphosphorinane ring and releasing an alkylating moiety and acrolein. Neither cyclophosphamide (CP) nor the deactivated metabolites of CP, 4-keto-CP and carboxyphosphamide nor 4-(S-ethanol)-sulfido-CP were attacked by 3'-5' exonucleases. DNA polymerases with proofreading activity, such as DNA polymerase I from E. coli or DNA polymerase delta from rabbit bone marrow, exhibited a tenfold higher specific activity with "activated" CP than "plain" 3'-5' pho

An improved synthesis of lysosomal activated mustard prodrug for tumor-specific activation and its cytotoxic evaluation.

Cyclophosphamide, an alkylating agent widely used as anticancer agent, biotransformed in vivo to unstable phosphoramidic mustard and acrolein, where the latter metabolite has been found responsible for hemorrhagic cystitis and renal toxicity. Being one of the most popular strategies to avoid these deleterious effects, prodrug design has been attempted, which can, in addition, enable selective drug targeting. Our efforts to design, synthesize and evaluate the enzymatically activated prodrug phosphorodiamidic mustard as potential candidate for selective chemotherapy in antibody-directed enzyme p

Glu504Lys Single Nucleotide Polymorphism of Aldehyde Dehydrogenase 2 Gene and the Risk of Human Diseases.

Aldehyde dehydrogenase (ALDH) 2 is a mitochondrial enzyme that is known for its important role in oxidation and detoxification of ethanol metabolite acetaldehyde. ALDH2 also metabolizes other reactive aldehydes such as 4-hydroxy-2-nonenal and acrolein. The Glu504Lys single nucleotide polymorphism (SNP) of ALDH2 gene, which is found in approximately 40% of the East Asian populations, causes defect in the enzyme activity of ALDH2, leading to alterations in acetaldehyde metabolism and alcohol-induced "flushing" syndrome. Evidence suggests that ALDH2 Glu504Lys SNP is a potential candidate genetic

Yields of tar and other smoke components from UK cigarettes.

Since 1972, regular six-monthly surveys of cigarettes on sale in the UK have determined the yields of tar, nicotine and carbon monoxide in mainstream smoke. During 1983-1990, a series of special studies investigated the yields of a range of additional analytes and their inter-relationships with the routinely monitored components. The results for total cyanide (as hydrogen cyanide), total aldehydes (including acetaldehyde), acrolein, formaldehyde and nitrogen monoxide are summarized, and the full data for six low-molecular weight phenols and three polycyclic aromatic hydrocarbons (PAHs) are rep

Relevance of gas and particulate phases of tobacco smoke for lung cancer formation: an experimental study in Syrian golden hamsters.

The lower boiling constituents of the gas phase of cigarette smoke such as acrolein, hydrogen cyanide, and formaldehyde are cilia toxic. If such compounds can inhibit the self-purifying capacity of the bronchi and thus act as pacemakers for the action of carcinogens, such as aromatic hydrocarbons, then filtration of these cilia toxic compounds by means of carbon filters should yield protection against the carcinogenic activity of hydrocarbons. To elucidate this hypothesis, benzo(a)pyrene [B(a)P] was instilled intratracheally to Syrian golden hamsters in addition to chronic inhalation of cigare

Influence of caffeine on allyl alcohol-induced hepatotoxicity in rats. I. In vivo study.

Cotreatment of rats with a low hepatotoxic dose (30.7 mg/kg, i.p.) of allyl alcohol (AA) and a higher, but nontoxic, dose (150 mg/kg, oral) of caffeine (CF) potentiated the hepatotoxicity of AA. This was verified by significantly higher levels of plasma alanine aminotransferase (ALT) activity and histopathologically greater severity of lesions in the periportal hepatocytes than those due to AA alone. Treatment of rats with 4-methylpyrazole (4-MP) (0.5 mmol/kg, i.p.) (an inhibitor liver alcohol dehydrogenase) for 30 minutes, followed by similar cotreatment with AA and CF, completely prevented t

Catalytic elevation effect of methylglyoxal on invertase and characterization of MGO modification products.

Reactive carbonyl species can modify digestive enzymes upon intake due to their electrophilic nature. This study evaluated the effects of methylglyoxal (MGO), glyoxal, acrolein, and formaldehyde on invertase, an enzyme presents in digestive tract. Unexpectedly, MGO enhanced, rather than inhibited, invertase activity. Moreover, MGO counteracted the inhibitory effects of the other three carbonyls on invertase activity. Kinetic analyses revealed that 150 mmolLexp.-1 MGO resulted in a 2-fold increase in the Km and a 3.3-fold increase in Vmax, indicating that MGO increased the turnover rate of sucr

Cytomegalovirus-related Cystitis in a Patient with Membranous Glomerulonephritis Receiving Cyclophosphamide: A Case Study.

Hemorrhagic cystitis is a familiar complication of cyclophosphamide therapy in patients receiving high doses of intravenous cyclophosphamide, which is commonly used as part of a chemotherapeutic regimen and as an immunosuppressant for various malignancies and connective tissue diseases. Acrolein, an active and urotoxic metabolite of cyclophosphamide, is the leading cause of this hazardous complication. However, there are very few case reports indicating the role of pathogens such as BK virus, adenovirus, cytomegalovirus (CMV), Escherichia coli, Proteus mirabilis, Klebsiella, and Candida as the

Outcome of Canine Multicentric Lymphoma after Single or Divided Treatment with Cyclophosphamide in Multidrug Chemotherapy.

Cyclophosphamide is commonly used in combination chemotherapy to treat dogs with lymphoma. The metabolite of cyclophosphamide, acrolein, can irritate urinary bladder and cause sterile hemorrhagic cystitis. Dividing the administration of cyclophosphamide across multiple days may reduce the concentration of this metabolite in urinary bladder and reduce the possibility of cystitis. However, the impact of the therapeutic effect of this modification is not evaluated and compared to traditional single maximum-tolerated dose regimen. Seventy-two dogs with multicentric lymphoma received either bolus d

Association of East Asian Variant Aldehyde Dehydrogenase 2 Genotype (ALDH2*2*) with Coronary Spasm and Acute Myocardial Infarction.

Coronary spasm plays an important role in the pathogenesis of ischemic heart disease, including angina pectoris, acute myocardial infarction (AMI), silent myocardial ischemia, and sudden death. The prevalence of coronary spasm is higher among East Asians probably due to genetic as well as environmental factors. ALDH2 eliminates toxic aldehydes including 4-hydroxy-2-nonenal (4-HNE) derived from lipid peroxidation and acrolein in tobacco smoking as well as ethanol-derived acetaldehyde and thereby protects tissues and cells from oxidative damage. Deficient variant ALDH2*2 genotype is prevalent am

In situ structural organization of encapsidated and unencapsidated Ectromelia virus DNA.

The structural organization of Ectromelia virus DNA in infected mouse liver cells has been studied by using thin sections stained with the Feulgen-like osmium-ammine reaction. We found that in the cytoplasmic factories, free viral DNA was structured into completely extended filaments 2-3 nm thick. Viral DNA in immature virions, however, appeared to have a structural organization that superimposed that of eukaryotic chromatin. This was constituted by roundish subunits, with a diameter of 11-13 nm, composed of a DNA ring encircling an unstained inner core. The mature virion was composed of the s

HERBICIDE TRIALS IN THE SNAIL HABITATS OF THE EGYPT 49 PROJECT.

The molluscan hosts of Schistosoma haematobium, Schistosoma mansoni and Fasciola spp. have been found to flourish in association with certain submersed, floating and emergent weeds in the irrigation system of the Egypt 49 project area. The efficacy of acrolein in killing Potamogeton crispus, an important submersed weed, has been previously established. The present paper deals with field trials of different herbicides, alone or combined, against the other predominant types of floating and emergent weeds-Eichhornia crassipes, Panicum repens, Typha ungustata and Scirpus inclinatus. The trials sho

Relationship of Urinary Mercapturic Acids of Selected Tobacco Smoke Constituents to Chronic Obstructive Pulmonary Disease in Cigarette Smokers of the Singapore Chinese Health Study.

α,β-Unsaturated carbonyl compounds in cigarette smoke, such as acrolein and acrylamide, are considered as possible causative factors for chronic obstructive pulmonary disease (COPD) in people who smoke cigarettes. A major route of metabolism of these compounds is reaction with glutathione to form adducts that are further metabolized and excreted in the urine as mercapturic acids, which serve as useful dosimeters of exposure. In this study, we analyzed urine samples from the Singapore Chinese Health Study, a prospective epidemiology study of 63,257 men and women 45-74 years old with Chinese ori

Modulation of key antioxidant enzymes and cell cycle arrest as a possible antifungal mode of action of cinnamaldehyde based azole derivative.

Although Candida auris was only identified in the year 2009, it has rapidly spread in more than a dozen countries and is proving more deadly and notorious. In our previous studies, we reported on the tremendous antifungal potential of a series of cinnamaldehyde based azole derivatives against fluconazole susceptible and resistant clinical isolates of Candida albicans and identified a promising lead molecule (6f). In this study, the effect of this compound on the viability and physiology of cell death in C. auris was assessed. The impact of compound 6f on cell cycle, oxidative stress enzymes an

Pyrido[1,2-c][1,2,4]triazol-3-ylidene: reactivity and its application in organocatalysis and organometallic catalysis.

The reactivity and catalytic performance of 2-ethylpyrido[1,2-c][1,2,4]-triazol-3-ylidene 6 have been comprehensively investigated. The carbene 6 has shown unusual properties owing to the effect of the pyrido-annulation. While formohydrazide 8 and hydrazine 9 are obtained via the destruction of the triazole skeleton of the carbene, 3-((1Z,3E)-4-(1H-pyrrol-1-yl)buta-1,3-dienyl)-1-ethyl-1H-1,2,4-triazole 10 is achieved through the cleavage of the C-N bond of the pyridine ring. The carbene has turned out to be a powerful catalyst in a variety of organocatalyzed and Pd(II)-catalyzed transformation
